        Korean Red ginseng prevents endothelial senescence by downregulating the HO-1/NF-κB/miRNA-155-5p/eNOS pathway

        Background: Korean Red ginseng extract (KRGE) has beneficial effects on the cardiovascular system by improving endothelial cell function. However, its pharmacological effect on endothelial cell senescence has not been clearly elucidated. Therefore, we examined the effect and molecular mechanism of KRGE on the senescence of human umbilical vein endothelial cells (HUVECs). Methods: HUVECs were grown in normal or KRGE-supplemented medium. Furthermore, they were transfected with heme oxygenase-1 (HO-1) gene or treated with its inhibitor, a NF-κB inhibitor, and a miR-155-5p mimic or inhibitor. Senescence-associated characteristics of endothelial cells were determined by biochemical and immunohistochemical analyses. Results: Treatment of HUVECs with KRGE resulted in delayed onset and progression of senescence-associated characteristics, such as increased lysosomal acidic β-galactosidase and decreased telomerase activity, angiogenic dysfunction, and abnormal cell morphology. KRGE preserved the levels of anti-senescent factors, such as eNOS-derived NO, MnSOD, and cyclins D and A: however, it decreased the levels of senescence-promoting factors, such as ROS, activated NF-κB, endothelial cell inflammation, and p21 expression. The beneficial effects of KRGE were due to the induction of HO-1 and the inhibition of NF-κB-dependent biogenesis of miR-155-5p that led to the downregulation of eNOS. Moreover, treatment with inhibitors of HO-1, NF-κB, and miR-155-5p abolished the anti-senescence effects of KRGE. Conclusion: KRGE delayed or prevented HUVEC senescence through a signaling cascade involving the induction of HO-1, the inhibition of NF-κB-dependent miR-155-5p biogenesis, and the maintenance of the eNOS/NO axis activity, suggesting that it may protect against vascular diseases associated with endothelial senescence.

        ■ Objectives The purpose of this case study is to report that a hypertension with intracerebral hemorrhage of tae-eumin was treated with 'Cheongsim Yeunja-tang' and then the symptoms were improved and blood pressure was decreased. ■ Methods Although the patient who had hypertension with intracerebral hemorrhage took antihypertension drugs, intermittent increase of blood pressure was shown. We diagnosed him as Tae-eumin and treated with Cheongsim Yeunja-tang. We daily checked blood pressure and evaluated the symptoms. ■ Results After the treatment with Cheongsim Yeunja-tang, blood pressure was decreased and symptoms were improved. ■ Conclusion The result shows Cheongsim Yeunja-tang has antihypertensive effect and improve hypertension symptoms of Tae-eumin patient with intracerebral hemorrhage.

        정신분열병에 대한 리스페리돈의 효과 및 안정성

        연구목적 : 본 시험의 목적은 임상시험 시작전에 연구자들을 대상으로 PANSS Workshop을 통하여 PANSS, ESRS에 대한 국내에서의 표준화 작업을 구축하고 새로운 정신병 치료제인 리스페리돈의 효과와 안정성을 재확인하여 리스페리돈 사용에 대한 적정화를 이루는데 있다. 연구방법 : 1996년 4월부터 1996년 9월까지 국내 39개 대학병원 정신과에 입원중인 혹은 증상이 악화되어 입원하는 정신분열병 환자 377명을 대상으로 다시설 개방 연구를 시행하였다. 1주일간의 약물 배설기간을 가진후, 리스페리돈을 8주간 투여하였고, 기준점, 1주, 2주, 4주, 그리고 8주후에 평가되었다. 용량은 제1일에는 리스페리돈 1mg씩 1일 2회, 제2일에는 2mg씩 1일 2회, 제3∼7일에는 3mg씩 1일 2회 투여하였다. 이후 환자의 임상상태에 따라 임의로 증량할 수 있으며, 최대 일일 16mg을 초과하지 않도록 하였다. 추체외로 증상을 조절하기 위한 투약을 허용하였다. 임상증상 및 부작용의 평가는 PANSS(Positive and Negative Syndrome Scale), CGI(Clinical Global Impression) 그리고 ESRS(Extrapyramidal Symptom Rating Scale)을 사용하였다. 연구결과 : 377명중 343명(91%)이 8주간의 연구를 완결하였다. 치료 종결시점인 8주후 PANSS 총점수가 20% 이상 호전된 경우를 약물 반응군으로 정의할때, 약물반응군은 81.3%였다. 리스페리돈에 반응하는 예측인자로는 발병연령, 이전의 입원 횟수, 유병기간이 관련 있었다. 리스페리돈은 1주후부터 PANSS양성, 음성, 및 일반정신병리 점수상에 유의한 호전을 보여 효과가 빨랐다. CGI의 경우도 기준점에 비해 1주후부터 유의한 감소를 나타내었다. ESRS의 경우, 파킨슨 평가점수는 기준점과 비교해 투여 1주, 2주, 4주후 유의하게 증가되었다가 8주후 기준점과 차이가 없었다. Dystonia 평가점수는 1주후만 유의한 증가를 보였으며, dyskinesia 평가점수는 유의한 차이가 없었다. 혈압, 맥박수의 생명징후 및 일반 혈액학 검사, 생화학적 검사, 심전도 검사에서 유의한 변화는 없었다. 결 론 : 이상의 다시설 개방 임상 연구를 통해 리스페리돈은 정신분열병 환자에서 양성증상뿐만 아니라 음성증상 및 전반적인 증상에도 효과적인 것으로 사료된다. 보다 명확한 평가를 위해서는 다른 항정신병약물과의 이중맹검 연구가 필요할 것으로 생각되며, 또한 장기적 치료에 대한 평가도 함께 이루어져야 하겠다. Objective : The purpose of this study was to investigate the efficacy and safety of risperidone in the treatment of Korean schizophrenic patients. Method : This multicenter open study included 377 schizophrenic patients drawn from 39 university hospitals. After a wash-out period of 1 week, the schizophrenic patients were treated with risperidone for 8 weeks and evaluated at 5 points ; at baseline, and 1, 2, 4 and 8 weeks of treatment. The dose was increased from 2mg/day(1mg twice daily) to 6mg/day(3mg twice daily) during the first week and adjusted to a maximum of 16mg/day over the next 7 weeks according to the patient's clinical response. Medication to control extrapyramidal symptoms was permitted. The psychiatric and neurological status of the patients was assessed by PANSS, CGI, and ESRS scales. Results : 343(91%) of 377 patients completed the 8-week trial period. Clinical improvement, as defined by a 20% or more reduction in total PANSS score at end point, was shown by 81.3% of patients. The predictors of response to risperidone were associated older age, shorter duration of illness, fewer previous hospitalization. Risperidone had rapid onset of action ; a significant decrease of the total PANSS and three PANSS factor(positive, negative, general), and CGI was already noticed at the end of first week. For the ESRS, parkinsonism rating scores were significantly increased until week 4 comparing with baseline. Dystonia rating scores were significantly increased until week 1, and dyskinesia rating scores were not significantly changed during the study. Laboratory parameters including vital sign, EKG, hematological, and biochemical values showed no significant changes during the trial. Conclusions : This study suggests that risperidone is generally safe and effective against both the positive and negative symptoms in our group of patients.

        나프타분해플랜트의 부탄추출공정에서 부탄증기의 연속누출에 의한 화재사고의 영향평가

        나프타분해플랜트의 부탄추출공정에서 부탄증기의 연속 누출에 의한 제트화재와 플래쉬화재의 사고결과에 대한 영향을 평가하고 사고결과에 미치는 공정변수들의 영향을 해석하였다. 877 kPa과 346.75K의 공정조건에서 부탄증기가 87.8 kg/s의 속도로 8m의 높이에서 연속누출되는 경우에 표준조건으로 설정한 대기 및 환경조건에서 사고결과를 해석한 결과, 제트화재에서는 화염의 형태와 크기를 예측할 수 있었으며 누출지점으로부터 200m에서 API모델로 산출한 목사열은 약 5kW/m^2이었고 플래쉬화재가 일어날 수 있는 연소범위는 11.2~120.2m이었다. 그리고 사고결과에 미치는 조업압력의 영향은 조업온도보다 크며 압력이 증가할수록 사고결과 값이 증가하였다. 이때 플래쉬화재에서 X_UFL에 미치는 공정압력의 영향은 X_LFL에서보다 약 1/10이하를 나타내었다. The consequence analysis for jet and flash fire accidents by the continuous release of butane vapor was performed and effects of process variables on consequences were analyzed in standard conditions. For the continuous release(87.8 kg/s) of butane vapor at 8 m elevated height in the debutanizing process of the naphtha cracking plant operating at 877 kPa, 346.75 K, we found that for the jet fire accident, shape and size of the flame could be predicted and thermal radiation estimated by API model at 200 m distance from release point was 1.5 kW/m^2, and that for the flash fire accident, effect range was 11.2~120.2m. Also, simulation results showed that effects of operating pressures on consequences were large than those of operating temperatures and results of accidents were increased with increasing operating pressures. At this time, effects of operating pressures on X_UFL. were smaller

        Simulated Moving Bed를 이용한 Bupivacaine 키랄분리 : (1) Preparative Chromatography를 이용한 기본조건 결정 (1) Optical Separation Conditions for Preparative Chromatography

        Racemate Bupivacaine의 키랄분리를 위한 크로마토그래피의 최적분리조건을 조사하였다. Kromasii^(�) 키랄고정상에서 최적이동상조성 성분은 n-hexane/2-propanol/acetic acid/triethylamine, 99/1/0.3/0.05(vol.%)이였다. 이동상 조성성분인 2-propanol의 부피비가 증가할수록 체류시간과 분리도가 동시에 감소되었다. 이동상 성분 중에서 미량으로 존재하는 triethylamine이 증가할수록 피크의 sharpness가 증가하고 체류시간 역시 감소하였다. 또한, acetic acid가 없으면, 크로마토그래피 광학분할이 불가능하였으며 부피 0.3%에서 분리도가 가장 높음을 확인할 수 있었다. 한편, 키랄의약품의 대량분리방법중의 하나인 SMB(simulated moving bed)크로마토그래피 시스템(직경 1.0cm×10cm×8개 칼럼으로 구성)의 최적분리조건을 전산모사에 의해 구하였으며, 이를 실험으로 확인하였다. 그 결과 switching time 15분, feed의 유량 0.1 ml/min, desorbent 0.86 ml/min, raffinate 0.3 ml/min, extract 0.66 ml/min, recycle 1.0 ml/min일 때 100% 순도의 R-form과 S-form을 얻을 수 있었으며, 실험치와 전산모사치간의 오차는 ??2%이내였다. The chiral separation of racemate Bupivacaine was carried out to find the optimum condition for chromatographic system. The optimum species and composition for mobile phase were n-hexane/2-propanol/acetic acid/triethylamine, 99/1/0.3/0.05(vol.%) in case Kromasil� chiral stationary phase was employed. The retention time and resolution decreased with the increase in the ratio of 2-propanol in the mobile phase. The sharpness of peak and retention time decreased, as the content of triethylamine increased. Without the existence of acetic acid, the chromatographic separation didn't occur, and the resolution was the highest at the acetic acid volume ratio of 0.3%. The optimum condition of SMB (simulated moving bed) was determined by simulation and its results was compared with exprimental data from lab-scale SMB (10 mm 1D×8 ea). Rased on the SMB simulation and experimental results, 100% purity of R and S Bupivacaine were obtained and the error between the calculated and experimental value was within ±2%.

      • P-Δ효과를 고려한 골조구조물의 극한해석

        본 연구는 P-Δ 효과를 고려한 철골조 골조구조물의 극한해석 알고리즘을 개발하는 것을 목적으로 한다. 극한해석에 사용되는 골조요소의 특성을 정의하기 위하여 직접해석법의 요소정식화를 사용하였다. 사용된 요소는 휨부재인 보요소와 휨-압축부재인 보-기둥요소이고, 이들 두 요소에 대하여 직접 해석법에 의한 정식화를 수행하여 내부파라메타와 변환파라메타를 정의하였다. 이들 두 파라메타를 사용하여 P-Δ효과를 고려한 철골조 골조구조물의 극한해석 알고리즘을 개발하였고, 개발된 알고리즘을 예제를 통하여 검증하였다. The purpose of this study was to develop a Limit Analysis Algorithm considering P-Δ effect. Direct Method approadch is used to define element characteristics. Two types of elements are used; beam and beam-column element. In Direct Method, two parameters are used for formulation of element; internal parameter and transformed parameter. Using these parameters, a algorithm for limit analysis including P- Δ effect is developed and the accuracy and effectiveness are verified by some examples analyzed using traditional method.

        광조사 방법에 따른 치과용 심미수복재의 중합수축

        Dental resin composites were introduced commercially in the mid-1960s for the restoration of anterior teeth. Since their advent, resin composites have undergone significant development, which continues to improve the longevity and application of resin composite restorations. Despite the development of new dentin-bonding agents, no system is currently available that completely withstands the formation of contraction gaps at the cementum/dentin-restoration junction, due to the unavoidable polymerization shrinkage of the resin composite during setting, therefore, a major aim of this study was to minimize or control the polymerization shrinkage of composites allowing flow of resin during setting reaction. A thermal dilatometer (DIL 402C, TASC 414/3A Controller, Netzche, Germany) was used to measure and compare conventional continuous cure and pulse cure polymerization shrinkage of seven light curing dental composites and three compomers. For conventional cure, restorative material were polymerized for 2 minutes using VIP (Bisco, U.S.A.) with 400 ㎽/㎠ intensity. After then, further polymerization was allowed for next 12 minutes that total 14 minutes of shrinkage was measured. For pulse cure, material was pre-polymerized irradiating light for 5 seconds with 200 ㎽/㎠ and post-ploymerized 3 minutes later with 400 ㎽/㎠ for 2 minutes. 9 minutes were allowed for further polymerization that total 14 minutes of shrinkage was measured. Polymerization shrinkage ranged from 3.758 ㎛/㎜ (SYC) to 6.897 ㎛/㎜ (P60) for conventional cure and 4.026 ㎛/㎜ (FLT) to 6.996 ㎛/㎜ (SRF) for pulse cure. The highest difference between the two methods was found at EAN (2.713 ㎛/㎜), followed by F20 and CRA which showed significant differences of 2.133, 1.274 ㎛/㎜ respectively (p<0.05). The least difference was found at SYC (0.489 ㎛/㎜). Universal resin composite group showed the highest polymerization shrinkage (6.514±0873 ㎛/㎜), and flowable composite showed the least shrinkage (4.139 ㎛/㎜). Packable composite showed 5.090±1.906 ㎛/㎜, and compomers showed 6.129±2.652 ㎛/㎜.

      • 열가소성 Wood-Plastic Composites의 기계적 물성에 미치는 커플링제의 영향

        목분 충진제의 함량을 달리 하고 PE수지를 매티릭스로 하여 wood plastic composites(WPC)를 제조하였다. 또한 매트릭스와 충진제간의 계면결합력을 증가시키기 위하여 커플링제로 phthalic anhydride(PA)를 사용하였으며, 충진제의 충진률과 커플링제 처리가 복합재의 기계적 물성에 미치는 영향과 계면현상을 관찰하였다. 목분의 충진률 30wt%, PA커플링제농도 3wt%일 때 인장강도는 26.37MPa로 최대값을 나타내었으며, 이때 충격강도는 46.24J/m의 최대값을 나타냈다. 또한 WPC는 주사전자현미경 관찰을 통해서 커플링제의 처리농도의 증가에 따라 목분과 PE 수지와의 분산과 결합이 잘 이루어졌으을 확인할 수 있었다. Wood powder as reinforcing fillers and polyethylene as a matrix have been used for wood plastic composites(WPC). In preparing WPC, the coupling agent, phthalic anhy dride(PA) was used in order to increase the interfacial bonding force between matrix and fillers. In this study, the effect of wood powder, PA concentration on the mechanical properties and interfcial phenomena on the composites was eveluated. The tensile strength of 3wt% PA-treated composites reached its maximum value of 25.91 MPa when the wood powder content was 30wt%, and the mximum impact strength of PA-treated composites was 46.24H/m. SEM observation showed that wood powder was well dispersed and bonded well with PE matrix by increasing the coupling agent concentration.

      • Lumped Model을 이용한 건물의 비탄성 동적해석

        In this study the Lumped model approach, which is widely used in nonlinear dynamic analysis of tall buildings under lateral loads, is investigated. The parameters needed to form the Lumped model were obtained from the static pushover tests of the three dimensional frame model. Tri-linear force-displacement relationships were used to represent the behavior of the Lumped model. The results from the three dimensional nonlinear dynamic analysis of the frame model are compared with those of Lumped model analysis. In the elastic region, the displacement, velocity and acceleration records are close to those of the Lumped model, but in the inelastic region the responses of Lumped model are overestimated.

      • 저먼 세퍼드에서 발생한 내측 갈고리돌기 유합부전을 컴퓨터 단층촬영술을 이용하여 진단한 증례

        A 4-year-old male German shepherd dog with reluctance to exercise, swelling of bilateral elbow joint and general depression was referred to the Veterinary Medical Teaching Hospital of Chungnam National University. Swelling, alopecia and lichenification of bilateral elbow joint were observed on physical examination. Osteophytes, sclerosis and joint mouse of bilateral elbow joint were found on radiography. Deformation of medical coronoid process and soft tissue density swelling at caudal part of bilateral elbow joint were also observed on radiography. Computed tomography revealed fragmented medial coronoid process and osteophytes of bilateral elbow joint. Therefore this case was diagnosed as fragmented medical coronois process and degenerative joint disease.
