Application of Customized Industry-Specific Metrics to Assessment of Capital Projects Performances

The pharmaceutical industry is unique in its procedures and methods of manufacture, and thus requires a distinct project management approach. With the consideration of specific facility delivery processes of the industry, assessment of pharmaceutical construction project performance is required as a first step of improving their performance. To do so, industry-specific metrics tuned to the characteristics of the industry are developed, followed by establishment of a hierarchical structure enabling pharmaceutical projects to be properly grouped for performance assessment. Using collected data for producing metric values, descriptive and statistical analyses are performed to assess pharmaceutical project cost, schedule, and dimensional performances by three major types: bulk manufacturing, secondary manufacturing, and laboratory. By utilizing these industry-specific metrics and analyses results, the industry can find solutions to more effectively manage cost, schedule, and other resources for the delivery of pharmaceutical construction projects.

SUMMARY: The International Pancreas and Islet Transplant Association (IPITA), in conjunction with the Transplantation Society (TTS), convened a workshop to consider the future of pancreas and islet transplantation in the context of potential competing technologies that are under development, including the artificial pancreas, transplantation tolerance, xenotransplantation, encapsulation, stem cell derived beta cells, beta cell proliferation, and endogenous regeneration. Separate workgroups for each topic and then the collective group reviewed the state of the art, hurdles to application, and proposed research agenda for each therapy that would allow widespread application. Herein we present the executive summary of this workshop that focuses on obstacles to application and the research agenda to overcome them; the full length article with detailed background for each topic is published as an online supplement to Transplantation.

<P>The influenza endonuclease is an essential subdomain of the viral RNA polymerase. It processes host pre-mRNAs to serve as primers for viral mRNA and is an attractive target for antiinfluenza drug discovery. Compound L-742,001 is a prototypical endonuclease inhibitor, and we found that repeated passaging of influenza virus in the presence of this drug did not lead to the development of resistant mutant strains. Reduced sensitivity to L-742,001 could only be induced by creating point mutations via a random mutagenesis strategy. These mutations mapped to the endonuclease active site where they can directly impact inhibitor binding. Engineered viruses containing the mutations showed resistance to L-742,001 both in vitro and in vivo, with only a modest reduction in fitness. Introduction of the mutations into a second virus also increased its resistance to the inhibitor. Using the isolated wild-type and mutant endonuclease domains, we used kinetics, inhibitor binding and crystallography to characterize how the two most significant mutations elicit resistance to L-742,001. These studies lay the foundation for the development of a new class of influenza therapeutics with reduced potential for the development of clinical endonuclease inhibitorresistant influenza strains.</P>

Objective: Triplet-born lambs are less likely to survive to weaning than twin-born or single-born lambs. Appropriate ewe-lamb bonding behaviours and lamb vigour behaviours are necessary for survival of lambs. The aim of this experiment was to determine whether maternal nutrition during late pregnancy influenced behaviour of the ewe and her lambs soon after birth, and to determine whether mid-pregnancy body condition score (BCS) influenced any behavioural response. Methods: The experiments included ewes that were in BCS 2.0, 2.5, or 3.0 in mid-pregnancy and were fed either ad libitum or to pregnancy-maintenance requirements in late-pregnancy (day 115 until 136 in experiment one, and day 128 until 141 in experiment two). The time taken for lambs to stand, contact dam, suck from dam and follow dam was recorded three to 18 h after birth. The number of high- and low-pitched bleats emitted by the ewe and lambs was recorded, along with maternal behaviour score (MBS) of the ewe. Lambs in experiment two underwent a maternal-recognition test at 12 or 24 h. Results: There were significant effects of feeding treatment on bleating behaviour of ewes and lambs, but these were inconsistent among BCS groups and between experiments. Lamb vigour behaviours were not affected by feeding treatment. In experiment one, there was no effect of feeding treatment or BCS on MBS, but in experiment two, ewes in BCS3 in mid-pregnancy had greater MBS than ewes in BCS2 in mid-pregnancy (MBS 3.1/5 vs MBS 2.1/5; p<0.05). Conclusion: Given there were no repeatable effects on behaviour of ewes and lambs, ad libitum feeding rather than feeding for pregnancy-maintenance requirements cannot be used to improve behaviours soon after birth of triplet-bearing ewes in BCS 2-3 and their lambs in extensive pastoral conditions.
