Development of a Virus Elution and Concentration Procedure for Detecting Norovirus in Oysters

Low levels of virus contamination and naturally occurring reverse transcription-polymerase chain reaction (RTPCR) inhibitors restrain virus detection in oysters. A rapid and efficient oyster-processing procedure that can be used for sensitive virus detection in oysters was developed. Poliovirus type 1 Sabin strain was used to evaluate the efficacy of virus recovery. The procedure included (a) acid-adsorption and elution with buffers (0.25M glycine-0.14 M NaCl, pH 7.5; 0.25M threonine-0.14M NaCl, pH 7.5); (b) polyethylene glycol (PEG) precipitation; (c) resuspension in Tween 80/Tris solution and chloroform extraction; (d) the second PEG precipitation; (e) viral RNA extraction with TRIzol and isopropanol precipitation; and (f) RT-PCR combined with semi-nested PCR. The overall recovery of elution/concentration was 19.5% with poliovirus. The whole procedure usually takes 19 hr. The overall detection sensitivity was 4 RT-PCR units of genogroup I norovirus (NoV) and 6.4 RT-PCR units of genogroup II Nov/25 g of oysters initially seeded. The virus-detecting method developed in this study should facilitate the detection of low levels of NoV in oysters.
Expression of translationally controlled tumor protein mRNA in human colon cancer

The translationally controlled tumor protein (TCTP) was first identified as a tumor-associated protein. Here we described the expression profile of TCTP mRNA using a panel of normal human tissues. We also investigated the cell-cycle-dependence expression of the TCTP mRNA using synchronized cultures of Saccahromyces cerevisiae. The TCTP gene was constantly expressed through the yeast cell cycle. The expression of TCTP mRNA from three human colon carcinoma cell lines and the tumors xenografted in a nude mouse was studied. Our data imply that the TCTP is a housekeeping gene and that its deregulation may be involved in the tumor development of colon carcinoma.
정규 모집단의 평균 비교를 위한 신뢰구간 겹치기 시각화

두 개의 정규 모집단의 평균이 같은가를 검증할 때 두 개의 신뢰구간이 겹치는 지를 시각적으로 판단하여 결정하는 방법은 매우 직관적이면서도 사용하기 쉽다. 그러나 신뢰구간이 겹칠 때도 두 집단의 평균은 통계적으로 유의하게 다를 수 있으므로 가설검증 결과와 다를 수 있다. 평균 차에 대한 신뢰구간을 각 평균의 신뢰구간으로 변환한 후에 두 신뢰구간이 겹치는지 여부를 시각적으로 판단하여 가설검증을 할 수 있는 방법을 제안한다. 또한 이 방법은 분산이 같은 k개의 정규 모집단의 평균을 비교할 경우에도 사용할 수 있음을 보인다. Data analysts sometimes test the equality of two normal population means by the inspection of the overlapping of two confidence intervals. This method seems simple to use; however, it is a common statistical misconception to suppose that two normal means are not significantly different because of no overlapping. This article will present transforming the confidence interval of the mean difference to individual confidence intervals that are visualized to inspect overlapping. It will also be shown that this technique can be extended when comparing the k normal population means with equal variances.

Internet gaming disorder (IGD) is a complicated behavioral and mental disorder. IGD is suggested as a one of the addictive dis-orders, also, it has unique characteristic features. Because of the bio-psycho-social model of addiction, IGD is part of a wide spectrum of comorbid psychiatric conditions and sociocultural contexts. Based on previous qualitative studies, we can define the intervention strategy based on the severity of the client’s addiction and characteristics of the problem. Therefore, we developed a multidimensional assessment protocol entitled K-PPC (Korean Patient Placement Criteria) for IGD and a tailored individual intervention approach entitled STEP (Stepped Tailored Empowerment Program) based on the concept proposed by the ASAM PPC (American Society of Addiction Medicine) model designed to help those who abuse alcohol and the Stepped model for ad-diction disorders. The K-PPC Assessment protocol is composed of 4 domains; internet game use behaviors, concurrent psychiatric problems, motivation, and the environmental factors affect-ing recovery and relapse. The STEP intervention matrix was composed of the severity of the addiction (basic and intensive) and concurrent psychiatric problems (introversion and extroversion). The treatment program was developed using information taken from evidence-based programs like cognitive behavioral treatment and motivational interviewing. The periods of the basic and intensive intervention are 8 weeks and 16 weeks, respectively. Because of the tailored approach of STEP, it is hypothesized that outcomes will be better compared to those in people treated with the current standard of care.
