        방풍의 항염 효과 기전

        목적 : 방풍은 임상적으로 관절염을 포함한 다양한 염증성 질환 치료제로 사용되어 왔다. 본 연구에서는 인간 비만세포를 이용하여 세포 독성에 영향을 주지 않는 농도에서 방풍의 항염 효과 및 그 기전을 검토했다. 방법 : 인간의 HMC-1세포를 IMDM에서 페니실린, 스트렙토마이신, 모노티오글리세린를 첨가하여 배양하고 방풍추출액을 투여하였다. 그 다음 MTT, ELISA, RT-PCR, 세포내 칼슘측정, 핵단백분석을 이용하여 TNF-α, IL-6, IL-8 각각의 형성과 mRNA발현, 세포내 칼슘 수준, NF-κB 발현에 대한 방풍추출액의 반응을 측정하고 통계처리 하였다. 결과 : 방풍은 PMA와 calcium ionophore A23187로 활성화된 비만세포에서 세포내 칼슘 수준과 NF-κB, TNF-α와 IL-6의 발현을 억제 시켰고 RT-PCR을 이용한 mRNA 발현에서 TNF-α와 IL-6의 발현을 억제하였다. 결론 : 방풍은 비만세포내 칼슘 수준 및 NF-κB의 활성을 억제하고 염증성 세포 활성 물질인 TNF-α와 IL-6의 분비도 억제하여 항염 효과를 나타냄을 암시하고 있다.

        Activation of NKT Cells in an Anti-PD-1–Resistant Tumor Model Enhances Antitumor Immunity by Reinvigorating Exhausted CD8 T Cells

        <P>These findings provide mechanistic insights into the application of NKT cell stimulation as a potent adjuvant for immunotherapy against advanced cancer.</P><P>PD-1–based cancer immunotherapy is a successful example of immune checkpoint blockade that provides long-term durable therapeutic effects in patients with cancer across a wide spectrum of cancer types. Accumulating evidence suggests that anti-PD-1 therapy enhances antitumor immunity by reversing the function of exhausted T cells in the tumor environment. However, the responsiveness rate of patients with cancer to anti-PD-1 therapy remains low, providing an urgent need for optimization and improvement. In this study, we designed an anti-PD-1–resistant mouse tumor model and showed that unresponsiveness to anti-PD-1 is associated with a gradual increase in CD8 T-cell exhaustion. We also found that invariant natural killer T cell stimulation by the synthetic ligand α-galactosylceramide (αGC) can enhance the antitumor effect in anti-PD-1–resistant tumors by restoring the effector function of tumor antigen–specific exhausted CD8 T cells. IL2 and IL12 were among the cytokines produced by αGC stimulation critical for reinvigorating exhausted CD8 T cells in tumor-bearing mice and patients with cancer. Furthermore, we observed a synergistic increase in the antitumor effect between αGC-loaded antigen-presenting cells and PD-1 blockade in a therapeutic murine tumor model. Our study suggests NKT cell stimulation as a promising therapeutic strategy for the treatment of patients with anti-PD-1–resistant cancer.</P><P><B>Significance:</B> These findings provide mechanistic insights into the application of NKT cell stimulation as a potent adjuvant for immunotherapy against advanced cancer. <I>Cancer Res; 78(18); 5315–26. ©2018 AACR</I>.</P>

        침전극저주파 자극이 해마의 세포사 차단에 미치는 영향

        Objectives: This study was performed to investigate the effects of Needle Electrode Electrical Stimulation (NEES) on ischemia-induced cerebrovascular accidents. After obstruction and reperfusion of ** arteries in white mice, the amounts of necrosis and inflammation related substances IL-6, Caspase-3, and PARP, C-fos were measured in neurons of the hippocampus. The following results were obtained. Methods: This study used 21 male specific pathogen free (SPF) SD (Sprague Dawley) rats, 8 weeks of age and approximately 300 g in weight, that were given at least 1 week to adapt to the lab environment Each exposed artery was completely occluded with non-absorbent suture thread and kept in that state for 5 minutes. The sutures were then removed to allow reperfusion of blood. Test group is control group for comparison with the common carotid artery occlusion models, a GI group that underwent common carotid artery occlusion, and a needle electrode electrical stimulation (NEES) group that underwent NEES after artery occlusion. The GI and NEES groups were given 12, 24, or 48 hours of reperfusion before NEES. NEES device (PG6, ITO, Japan, 9V, current, 2Hz) was used to stimulate the right and left acupoint ST36 of the SD rats for 30 minutes while they were sedated with 3% isoflurane. An immunohistochemistry test was done on the forebrains of the GI induced rats. All the data collected from this study was symbolized and analyzed using a statistics processing program (SPSS 12.0K/PC). The level of significance was set at α=0.05 and a T-TEST analysis was used to find out the effects of treatment on each of the groups: the normal group, the CVA induced group, and the treatment after CVA induction group. Results: Both PARP and C-fos immuno-reactive cells, related to apoptosis, were greater in the GI groups than the NEES group. Caspase and IL-6 immuno-reactive cells, related to inflammation, were greater in the GI and NEES groups than the control group. Conclusions: This research was conducted to study the effects of NEES on CVA due to ischemia. Occlusion and reperfusion was performed on the common carotid arteries of white rats, after which amounts of substances related to neuron necrosis and inflammation-PARP, IL-6, Caspase-3, and C-fos - were measured in the Hippocampus.

        <i>In vivo</i> genotoxicity evaluation of lung cells from Fischer 344 rats following 28 days of inhalation exposure to MWCNTs, plus 28 days and 90 days post-exposure

        <P>Despite their useful physico-chemical properties, carbon nanotubes (CNTs) continue to cause concern over occupational and human health due to their structural similarity to asbestos. Thus, to evaluate the toxic and genotoxic effect of multi-wall carbon nanotubes (MWCNTs) on lung cells <I>in vivo</I>, eight-week-old rats were divided into four groups (each group = 25 animals), a fresh air control (0 mg/m<SUP>3</SUP>), low (0.17 mg/m<SUP>3</SUP>), middle (0.49 mg/m<SUP>3</SUP>), and high (0.96 mg/m<SUP>3</SUP>) dose group, and exposed to MWCNTs <I>via</I> nose-only inhalation 6 h per day, 5 days per week for 28 days. The count median length and geometric standard deviation for the MWCNTs determined by TEM were 330.18 and 1.72 nm, respectively, and the MWCNT diameters ranged from 10 to 15 nm. Lung cells were isolated from five male and five female rats in each group on day 0, day 28 (only from males) and day 90 following the 28-day exposure. The total number of animals used was 15 male and 10 female rats for each concentration group. To determine the genotoxicity of the MWCNTs, a single cell gel electrophoresis assay (Comet assay) was conducted on the rat lung cells. As a result of the exposure, the olive tail moments were found to be significantly higher (<I>p</I> < 0.05) in the male and female rats from all the exposed groups when compared with the fresh air control. In addition, the high-dose exposed male and middle and high-dose exposed female rats retained DNA damage, even 90 days post-exposure (<I>p</I> < 0.05). To investigate the mode of genotoxicity, the intracellular reactive oxygen species (ROS) levels and inflammatory cytokine levels (TNF-α, TGF- β, IL-1, IL-2, IL-4, IL-5, IL-10, IL-12 and IFN-γ) were also measured. For the male rats, the H<SUB>2</SUB>O<SUB>2</SUB> levels were significantly higher in the middle (0 days post-exposure) and high- (0 days and 28 days post-exposure) dose groups (<I>p</I> < 0.05). Conversely, the female rats showed no changes in the H<SUB>2</SUB>O<SUB>2</SUB> levels. The inflammatory cytokine levels in the bronchoalveolar lavage (BAL) fluid did not show any statistically significant difference. Interestingly, the short-length MWCNTs deposited in the lung cells were persistent at 90 days post-exposure. Thus, exposing lung cells to MWCNTs with a short tube length may induce genotoxicity.</P>

        KIOM-MA128 is a novel oriental herbal medicine which is for atopic dermatitis and asthma. The purpose of this study was to develop on analytical method of arctiin in rat plasma after oral administration of KIOM-MA128. Analyte was separated on a Atlantis dC18 reverse phase column, using gradient mobile phase (A:B = acetonitrile: 0.1% formic acid in water) at a flow rate of 0.3 mL/min. Detection was performed by electrospray positive ionization mass spectrometry using multiple reaction monitoring of the transitions of arctiin at m/z 552.4 → 372.8 and internal standard (carbamazepine) at m/z 237.0 → 194.5. The limit of quantification was 1 ng/mL for arctiin. The precisions were lower than 15% and the accuracy was between – 12.21 and 3.2%. The maximum concentration found in plasma samples was 4.1 ng/mL. The present method was successfully developed for detecting arctiin in plasma and this results would be utilized to the further study.

        Ga_(0.6)Sb_(l.4)Se₃single crystals were grown by the Bridgman technique method, These single crystals had orthorhombic structure, and lattice constant and electric conduction type of Ga_(0.6)Sb_(l.4)Se₃single crystals was a = 11.860Å , b = 12.057Å, c = 4.103Å and p-type, respectively. Direct band gap of Ga_(0.6)Sb_(l.4)Se₃single crystals obtained from optical absorption spectrum were 1.17 eV. Photoconductivity spectrum measured at 10K showed that the photoconductivity peak in Ga_(0.6)Sb_(l.4)Se₃single crystals was located in 960 nm.

        Antiviral activity of gemcitabine against human rhinovirus in vitro and in vivo

        <P>Rhinovirus, a major causative agent of the common cold, is associated with exacerbation of asthma and chronic obstructive pulmonary disease. Currently, there is no antiviral treatment or vaccine for human rhinovirus (HRV). Gemcitabine (2',2'-difluorodeoxycytidine, dFdC) is a deoxycytidine analog with antiviral activity against rhinovirus, as well as enterovirus 71, in vitro. However, the antiviral effects of gemcitabine in vivo have not been investigated. In the current study, we assessed whether gemcitabine mediated antiviral effects in the murine HRV infection model. Intranasal administration of gemcitabine significantly lowered pulmonary viral load and inflammation by decreasing proinflammatory cytokines, including TNE-alpha and IL-1 beta, and reduction in the number of lung-infiltrating lymphocytes. Interestingly, we found that the addition of UTP and CTP significantly attenuated the antiviral activity of gemcitabine. Thus the limitation of UTP and CTP by the addition of gemcitabine may inhibit the viral RNA synthesis. These results suggest that gemcitabine, an antineoplastic drug, can be repositioned as an antiviral drug to inhibit HRV infection. (C) 2017 Elsevier B.V. All rights reserved.</P>

      • 少陰人 補中益氣湯과 後世方 補中益氣湯이 陽虛病證에 미치는 影響

        In order to investigate experimentally the effects of Soum-In Bojungikit'ang and Dong-Won Bojungikit'ang on Yang-Insufficient syndrome(陽虛證) induced by Hydrocortisone acetate(H.A.) in experimental animals(Mice and Rats), the author experimented with various activities. Body temperature(BT), Body weight(BW), Exercise time, Cyclic-AMP, the Rat's Hair condition was measured. The results obtained are as follows ; 1. In body temperature, the Dong-Won Bojungikit'ang treated group was increased significantly and the Soum-In Bojungikit'ang treated group showed no significance. 2. In body weight, the Soum-In Bojungikit'ang treated group had increased significantly and the Dong-Won Bojungikit'ang treated group showed no significance and there was a significant difference between the Soum-In Bojungikit'ang and the Dong-Won Bojungikit'ang. 3. In exercise time, the Soum-in Bojungikit'ang and the Dong-Won Bojungikit'ang treated groups were significantly increased. 4. In cyclic AMP, both experimental groups showed increasing tendancy, but showed no significance. 5. In the Rat's hair condition, the Soum-In Bojungikit'ang showed significance. 6. There was no significant difference between the Soum-In Bojungikit'ang and the Dong-Won Bojungikit'ang except the body weight. From the above findings, ① it has been demonstrated that the Soum-In Bojungikit'ang and Dong-Won Bojungikit'ang groups seem to produce the effectiveness on the recovery from depression of metabolism induced by Hydrocortisone acetate. ② In body temperature response the Dong-Won Bojungikit'ang had effectiveness during recovery. ③ In body weight response the Soum-In Bojungikit'ang had the effectiveness during recovery. ④ In exercise time, Both experimental group had the effectiveness during recovery. ⑤ In cyclic AMP response, Both experimental group had no effectiveness during recovery. ⑥ In the Rat's hair condition response the Soum-In Bojungikit'ang group had the effectiveness during recovery. Therefore it is suggested that the Soum-In Bojungikit'ang and Dong-Won Bojungikit'ang groups had effectiveness during recovery from Yang-Insufficient syndrome more or less.

      • TiN이 증착된 SiO/TiN 박막의 전압-전류 특성

        In this study, the evaporation of SiO layers with different thickness on Si wafer(n-100) is conducted by using the RF sputtering method. Thin films are made with the structure of Si/SiO and Si/SiO/TiN by evaporating TIN which is used as a diffusion barrier of non-resistant superintegrated semiconductor circuit. The research is performed to increase the I-V characteristics depending upon the SiO thickness and the reliability and reproducibility of the TIN thin film. It is shown that, since the TIN is diffused into SiO surface and fosters the potential barrier highly in the SiO/TiN thin film, the saturation voltage appears after the SiO thin film does. The thicker the SiO film becomes, the weaker the current variation does and the less leakage current the TiN deposited thin film has. The leakage current variation with a little slope is found in the negative voltage zone due to the effect of the trap state density near the electrode. The spiking which occurs when the film is too very thin can be reduced by adding TiN. The stability can be obtained due to no pervasion of A1 into SiO. The experimental result matches the Ohm s law and satisfies the equation of current.

        낭성법랑모세포종, 함치성낭, 치성각화낭의 방사선소견과 Ki-67, PCNA, Cytokeratin 발현과의 연관성에 관한 연구

        Purpose : To compare the proliferation potential of the epithelial cells between unicystic ameloblastoma (UA), dentigerous cyst (DC), and odontogenic keratocyst (OKC) and to correlate this proliferation potential with the radiographic features of these three pathoses. Materials and Methods : Immunohistochemical expression of PCNA, Ki-67, and cytokeratin as a proliferation marker were assessed for 15 cases of UA, 15 cases of DC, and 15 cases of OKC. The degree of immunochemical expression of three proliferation markers were correlated with the radiographic features, especially cortical expansion (negative and positive) and shape of border (scalloped and round). Results : Using PCNA and Ki-67, OKC showed the highest proliferation potential and UA the lowest. Statistically significant differences were found between the OKC and the UA (p.0.05). However, no statistically significant difference was present according to the radiographic features in all pathoses. Using cytokeratin, there was no significant differences of proliferation potential among three pathoses. Conclusions : OKC epithelium has the most intense proliferation potential, followed by the dentigeous cyst and then unicystic ameloblastoma. There is no significant relation between the radiographic features and the proliferation potential of epithelium of these three pathoses.
