        Two farmed young ostrich chicks were applied to pathology laboratory at Chungnam National University. Grossly, one ostrich showed rotated tibiotarsus, the other showed rolled toes. Case 1 ostrich was hypertrophy of fibualis longus and tibial cranialis tendon and gastrocnemius tendon in crus, inflammation of flexor digitorum longus. In histopathological views, myocytes of fiburalis longus were showed hypertrophy and hyperplasia. It is not found any specific feature in X-ray of the case 2 ostrich. Limb deformities were detected frequently in farmed ostrich chicks in 2- to 3 weeks old. Limb deformities were an important cause of mortality in farmed ostrich chicks during the first 10weeks after hatch. Cause of limb deformities were considered a nutrition problem.

        Pilomatricoma was histopathologically diagnosed in a 2.5-year-old male Alaskan malamute. The patient was referred to Chungnam National University, Veterinary Medical Teaching Hospital with solitary, firm and well-circumscribed mass at shoulder. Grossly, the mass was dome shaped to tumor like and 10 x 8 x 5㎝ in size. Some lesions had gritty or bony consistency and was chalky when sectioned Histopathologic feature included mumerous clusters of tightly arranged, basaloid cells with evenly distributed chromatin and ghost cells were characterized by a central unstained zone. The ghost cells often become calcified and osseous metaplasia was also present.

      • 생물방제균 Bacillus subtilis YB-70이 생산하는 항진균성 항생물질의 분리 및 구조결정

        인삼근부균 Fusarium solani의 생육을 강력히 저해하는 생물방제균을 저병해 인삼경작지 근권토양에서 분리, 선발하여 Bacillus subtilis로 동정한 바 있고, 이 생물방제균 B. subtilis YB-70의 식물근부균 F. solani의 생육에 대한 길항기작이 항진균성 항생물질에 기인된다는 사실을 배양생산물의 분자량별 활성도 검증, 열처리에 의한 잔존 활성도 측정 등에 의해 열에 강한 저분자성 항생물질로 재확인하였고 in vivo test에서도 F.solani에 의한 근부병을 강력히 억제할 수 있음을 하였다. 생물방제균 Bacillus subtilis YB-70이 생산하는 항진균성 항생물질을 pH 침전, 용매추출, silica gel chromatography, LH-20 column chromatography 등을 통해 항생물질 YBS-1A, B로 분리, 정제 할 수 있었으며, 이 물질의 구조를 MS, NMR, amino acid analysis로 추정해 본 결과 YBS-1A는 5종 7개의 amino 산으로 구성된 itulin A 계열 항생물질로 규명되었으며, YBS-1B는 7종 11개의 amino 산으로 구성된 분자량 1,500정도의 itulin A 보다 좀 더 큰 항생물질로 추정되었다. A potential biocontrol bacterium, YB-70 was isolated from a rhizosphere in suppressive soil and identified as strain of Bacillus subtilis. In several biochemical and in vitro antibiosis tests on Fusarium solani with the culture filterates from B. subtilis YB-70, we found that antifungal mechanism of B. subtilis YB-70 was mediated by antibiotic substances produced from the bacterium. These antifungal substances were appeared to be beat-resistanl, micromolecular, and ethyl alcohol soluble. Antifungal agents produced by B. subtilis YB-70 showed strong inhibition against root-rotting fungi F. solani in in vivo pot test. An antifungal substance, YBS-1s, was purified from the culture broth of B. subtilis YB-70 by isoelectronic precipitation, silica gel column chromatography and Sephadex LH-20 column chromatography. Three antifungal agents were isolated from the culture broth of B. subtilis YB-70. Various instrumental analysis by Fab-MASS, ¹H-NMR, ¹³C-NMR, DEPT, and amino acid analyzet revealed that the YBS-1A was a peptide antibiotics of iturin class containing seven amino acids from five different groups, and the other(YBS-1B) was an analogue of iturin class containing seven amino acids from five different groups, and the other(YBS-1B) was an analogue of iturin group composed of 11 amino acids with larger molecular weight of about 1,500 dalton, which was lager than that of iturin A.

      • 수종 식물추출물의 항히스타민작용

        The antihistaminic action of eighteen herbal medicines was investigated by the radioligand binding and functional assays. The hexane fractions of Trichosanthis radix, Mori cortex radicis and Evodiac fructus dose-dependently inhibited [³H]mepyramine binding to H, receptor in guinea-pig brain homogenates and histamine-induced contraction of isolated guinea-pig ileum. Antihistaminic action of the hexane and ethyl acetate fractions of Mori cortex radicis and the hexane fraction of Evodiae fructus was more potent than their antimuscarinic action evaluated from the inhibition of [³H]QNB binding and carbachol response. The ethyl acetate and chloroform fractions from Scutcllariae radix also inhibited histamine-induced contraction, but antihistaminic potencies of these fractions were almost identical with their antimuscarinic potencies. The hexane fractions of Mori cortex radicis and Evodiae fructus inhibited selectively the increase of histamine-induced cutaneous vascular permeability in the rat dorsal skins. However, the ethyl acetate fraction from Scutellariae radix inhibited eqipotently the effects of histamine and serotonin on the vascular permeability. These results demonstrate that the hexane and ethyl acetate fractions of Mori cortex radicis and the hexane fraction of Evodiae fructus have the selective histamine H₁ receptor blocking activity.

        THE EFFECT OF PRIMING ETCHED DENTIN WITH SOLVENT ON THE MICROTENSILE BOND STRENGTH OF HYDROPHOBIC DENTIN ADHESIVE

        장기적인 상아질 접착의 내구성 악화는 접착층과 혼성층의 친수성 부위에서의 가수분해에 의해 일어나는 것으로 보고 되고 있다. 본 연구의 가설은 콜라겐 망상체를 유기용매로 프라이밍하면 콜라겐 조직을 붕괴시키지 않고 수분을 밀어내고 소수성 단량체와 유기용매로 이루어진 접착제가 침투하여 접착강도를 얻을 수 있다는 것이다. 두 소수성 단량체인 Bisphenol-A-glycidylmethacrylate (Bis-GMA)와 triethyleneglycol dimethacrylate (TEGDMA)를 아세톤, 에탄올 또는 메탄올에 용해시켜 세 가지의 실험용 접착제를 준비하였다. 산 부식과 수세과정 후에, 접착제를 습윤 상아질 표면(습윤 접착)이나 동일한 용매로 프라이밍 된 상아질 표면(용매 프라이밍 접착)에 적용하였다. 48시간 후와 1개월 후, 및 10.000회의 열순환 후에 미세인장결합강도를 측정하였다 접착계면은 주사전자현미경을 이용하여 관찰하였다. 접착방법에 무관하게 대부분의 시편의 접착계면예서 잘 발달된 혼성층을 관찰할 수 있었고 가장 높은 평균 미세인장결합강도는 에탄올을 포함하는 접착제의 48시간 후 시편에서 관찰되었다. 용매를 이용하여 프라이밍하는 접착 방법에서는 에탄올이나 메탄올을 포함하는 접착제에서 열순환 후에 미세인장접착강도가 증가하는 경향을 보였다. 그러나 습윤 접착의 경우에는 시효처리 후 미세인장결합강도의 증가가 관찰되지 않았다. 본 연구에서 에탄올을 이용한 상아질 프라이밍으로 우수한 접착력을 얻을 수 있었고. 열순환 후 접착력이 더욱 증가하였다 Deterioration of long-term dentin adhesion durability is thought to occur by hydrolytic degradation within hydrophilic domains of the adhesive and hybrid layers. This study investigated the hypothesis that priming the collagen network with an organic solvent displacc water without collapse and thereby obtain good bond strength with an adhesive made of hydrophobic monomers and organic solvents. Three experimentaladhesives were prepared by dissolving two hydrophobic monomers bisphenol-A-glycidylmethacrylate (Bis-GMA) and triethyleneglycol dimethacrylate (TEGDMA), into acetone, ethanol or methanol. After an entching and rinsing procedure, the adhesives were applied onto either wet dentin surfaces (wet bonding) or dentin .surfaces primed with the same solvent (solvent-primed bonding). Microtensile bond strength (MTBS) was measured at 48 hrs, 1 month and after 10,000 times of thermocycles. The bonded interfaces were evaluated using a scanning electron microscope (SEM) Regardless of bonding protocols, weIl-developed hybrid layers were observed at the bonded interface in most specimens. The highest mean MTBS was observed in the adhesive containing ethanol at 48 hrs. With solvent-primed bonding increased MTBS tendencies were seen with thermocycling in the adhesives containing ethanol or methanol. However in the case of wet bonding, no increase in MTBS was observed with aging.

        This paper reports some experimental results on the machining performance of non-ground cross hole type milling inserts. Six kinds of milling inserts were manufactured without using grinding process. Machining experiments were carried out to compare the machining performance of non-ground inserts with that of ground ones. The experimental results indicate that the cutting forces and surface roughnesses of machined surface of both ground and non-ground inserts are comparable.

        Orthosiphon stamineus Reduces Appetite and Visceral Fat in Rats

        To evaluate whether Orthosiphon stamineus (OS) reduces appetite and fat deposition, food intake and visceral fat mass following treatment with OS (450 mg/kg/day, per oral) for 2 weeks in Sprague-Dawley (SD) rats were measured. Hypothalamic neuropeptides associated with appetite regulation was also analyzed. Two weeks of OS treatment reduced food intake and visceral fat mass compared to saline-treated SD rats. OS treatment for 2 weeks elevated the expression of proopiomelanocortin (POMC) in the hypothalamus, whereas neuropeptide Y (NPY) expression was decreased, which may be associated with the elevation of plasma leptin concentration in OS-treated rats. Methanol and butanol extracts of OS increased leptin mRNA expression in 3T3-L1 cells in dose-dependent manner. Acute study showed intraperitoneal injection of various concentrations of OS increased POMC expression in the hypothalamus, whereas NPY expression decreased dose-dependently. These results suggest that OS reduces appetite, which may have resulted from the increased expression of POMC and decreased expression of NPY in the hypothalamus in association with the elevation of leptin. OS could be a medicinal food applicable for body weight control.

      • Ginkgetin, a Biflavone from <i>Ginko biloba</i> Leaves, Inhibits Cyclooxygenases-2 and 5-Lipoxygenase in Mouse Bone Marrow-Derived Mast Cells

        <P>Ginkgetin, a biflavone from <I>Ginkgo biloba</I> leaves, was previously reported to be a phospholipase A<SUB>2</SUB> inhibitor and this compound showed the potent antiarthritic activity in rat adjuvant-induced arthritis as well as analgesic activity. This investigation was carried out to find effects on cyclooxygenase-2 (COX-2) <I>in vitro</I> effect. Ginkgetin inhibits COX-2 dependent phases of prostaglandin D<SUB>2</SUB> (PGD<SUB>2</SUB>) generation in bone marrow-derived mast cells (BMMC) in a concentration-dependent manner with IC<SUB>50</SUB> values of 0.75 μ<SMALL>M</SMALL>. Western blotting probed with specific anti-COX-2 antibodies showed that the decrease in quantity of the PGD<SUB>2</SUB> product was accompanied by a decrease in the COX-2 protein level. In addition, this compound consistently inhibited the production of leukotriene C<SUB>4</SUB> (LTC<SUB>4</SUB>) in a dose dependent manner, with an IC<SUB>50</SUB> value of 0.33 μ<SMALL>M</SMALL>. These results demonstrate that ginkgetin has a dual cyclooxygenase-2/5-lipoxygenase inhibitory activity. Furthermore, this compound also inhibited degranulation reaction in a dose dependent manner, with an IC<SUB>50</SUB> value of 6.52 μ<SMALL>M</SMALL>. Therefore, this compound might provide a basis for novel anti-inflammatory agents.</P>
