The intracytoplasmic sperm injection (ICSI) procedure has recently been utilized to produce transgenic animals and may serve as an alternative to the conventional pronuclear microinjection in species such as pigs whose ooplasm is opaque and pronuclei are often invisible. In this study, the effects of sperm membrane disruption and electrical activation of oocytes on in vitro development and expression of transgene green fluorescent protein (GFP) in ICSI embryos were tested to refine this recently developed procedure. Prior to ICSI, sperm heads were treated with Triton X-100+NaCl or Triton X-100+NaCl+NaOH, to disrupt membrane to be permeable to exogenous DNA, and incubated with linearized pEGFP-N1 vector. To induce activation of oocytes, a single DC pulse of 1.3 kV/cm was applied to oocytes for $30{\mu}sec$. After ICSI was performed with the aid of a micromanipulator, in vitro development of embryos and GFP expression were monitored. The chemical treatment to disrupt sperm membrane did not affect the developmental competence of embryos. 40 to 60% of oocytes were cleaved after injection of sperm heads with disrupted membrane, whereas 48.6% (34/70) were cleaved without chemical treatment. Regardless of electrical stimulation to induce activation, oocytes were cleaved after ICSI, reflecting that, despite sperm membrane disruption, the perinuclear soluble sperm factor known to mediate oocyte activation remained intact. After development to the 4-cell stage, 11.8 (2/17, Triton X-100+NaCl+NaOH) to 58.8% (10/17, Triton X-100+NaCl) of embryos expressed GFP. The expression of GFP beyond the stage of embryonic genome activation (4-cell stage in the pig) indicates that the exogenous DNA might have been integrated into the porcine genome. When sperm heads were co-incubated with exogenous DNA following the treatment of Triton X-100+NaCl, GFP expression was observed in high percentage (58.8%) of embryos, suggesting that transgenic pigs may efficiently be produced using ICSI.

Information system data improvement was conducted by the system reform for a discriminatory element elimination, and information system improvement plan along with a working environment change for the serial number unification and system standardization of company employees. The study preceded with an analysis of the DB table of the operational information system of all DTaQs. Data transfer, validation, and tuning were also conducted. Data unification is a simple DB transfer job that cooperates with the management department of the entire information system by carefully communicating preshared possible issues and influence analyses. These studies can help discriminate element elimination and better working conditions. This thesis summarizes the major activities and achievements of data transfer and standardization of employees serial numbers.
자료교환체계 개선 및 OTP 다중인증을 통한 보안강화 메일시스템 구현

IT 기술이 발달함에 따라 해킹 및 개인정보 유출과 같은 보안 사고가 지속적으로 일어나고 있다. 이는 국방기술품질원의 사내 전산메일의 경우에도 마찬가지로 예방적 차원의 대책 강구가 필요하였다. 이메일을 통한 기술자료 유통시 ‘내외부 메일’ 및 ‘자료교환시스템’ 보안 강화가 필요해짐에 따라 기존 사용중이던 사내메일 시스템을 개선하여, ‘부서장 결재선 강화’, ‘반출메일 암호화 설정’, ‘다단계 로그인 인증 절차 도입’ 등 보안 취약 및 위해 요소를 제거하였다. 물론 보안 측면만 강조해서는 시스템 사용자 입장에서 불편함을 감수해야만 하는 결과가 초래되는 경우가 발생하기도 하므로 이를 만회하기 위한 사용자 편의성 측면에서 보완적인 개선조치도 수행하였다. 성능이 업그레이드된 최신 이메일 솔루션, 키보드보안/모바일OTP/암호화 프로그램 등 보안 SW 적용, 표준기술(Non-Active X) 적용, 유관시스템 연동, 망연계 설치, 그리고 데이터 이관 및 검증 등을 거쳐 개선하였고, 보다 지능화되고 발전되어 가는 해킹기술 발전과 같은 보안 위협요소들로 인해 향후에도 지속적인 모니터링 및 이메일 보안기능 강화와 성능 업데이트가 이루어짐이 적절하다. As advanced technology has developed recently, security problems such as hacking or personal information leakage are continuously and increasingly occurring. Accordingly, information systems in DTaQ are also affected by these threats and need to be prepared to respond to possible security problems. This would enable new DTaQ e-mail and data exchange systems to adopt technology standardization such as a non-active-X security enhancement and user-friendly function improvement. This would be helpful to eliminate a security-threat element and make a safe and user-friendly e-working environment. For example, the newest e-mail solution, keyboard security solution, mobile one-time password, and encoding program make the DTaQ e-mail and data exchange system not only improve security, but also strengthen the efficiency of its performance. Also, system improvement is conducted by related systems linkage, network connection, data transfer, tuning, etc. This study summarizes the major activities and achievements of adopting the new e-mail and data exchange system with a security solution, standardization of technology, etc.
Expression of Recombinant Human Follicle-stimulating Hormone in the Chinese Hamster Ovary Cell

As an preliminary experiment for making transgenic animals producing human follicle stimulating hormone (hFSH), we tried to express recombinant hFSH gene in vitro. hFSH is a heterodimeric glycoprotein hormone produced in the anterior pituitary gland. The hormone is essential in the regulation of reproductive processes, such as follicular development and ovulation. Genes encoding the common gonadotrophin alpha subunit and FSH-specific beta subunit were inserted into retroviral vectors under the control of the rat beta actin promoter. Gene transfer to the Chinese hamster ovary (CHO) cells was done by infection of the retroviruses harvested from PT67 packaging cells transfected with recombinant retrovirus vector DNA. After selection with G4l8, PCR and RT-PCR analyses of the G4l8-resistant CHO cells showed successful transfer and expression of both and fragments of the FSH gene.

STUDY DESIGN.: In vivo study of a rat spinal cord injury model. OBJECTIVES.: To develop complete transection model of thoracic spinal cord using a polymer sheet and a microtube relevant for delayed transplantation of stem cells. SUMMARY OF BACKGROUND DATA.: Stem cell transplantation for the regeneration of spinal cord injuries has used animal models. However, current models suffer from inflammation and leakage, which lessens their usefulness in studying delayed stem cell transplantation. METHODS.: Thoracic spinal cord at T9 level of adult Sprague-Dawley rats was exposed and a 50:50 sheet of poly(D,L-lactic-coglycolic acid) was inserted, exposed spinal cord was completely transected, and collagen was filled between the gap between the proximal and distal stumps of transected spinal cord. A microtube was placed and fixed between the polymer surfaces facing each other. Behavior testing, magnetic resonance imaging, and myelography were performed to characterize the new complete transection with a gap formation and polymer insertion (GAP) model and to compare the GAP model with the control models. Human mesenchymal stem cells (hMSCs) were transplanted into 3 models and immunohistochemistry and western blot were performed. RESULTS.: The inserted poly(D,L-lactic-coglycolic acid) sheet was completely disappeared 10 weeks after operation, but the inserted microtube remained firmly fixed in its original position. Myelography of the GAP model showed no leakage of contrast medium around the injured spinal cord, whereas magnetic resonance imaging of the severe contusion and simple transection models showed some leakage of contrast medium. Immunohistochemistry and western blot after hMSCs transplantation indicated that transplanted hMSCs survived and migrated well in the GAP model, and the deposition of inflammatory cells in GAP model was less than a simple transection model or severe contusion model. CONCLUSION.: The developed GAP model is more relevant for delayed transplantation of stem cells for the study of regeneration of spinal cord injury of rats.
