        MUC16 facilitates cervical cancer progression via JAK2/STAT3 phosphorylation‑mediated cyclooxygenase‑2 expression

        Objectives MUC16 (mucin 16, also known as CA-125, cancer antigen 125, carcinoma antigen 125, or carbohydrate antigen 125) has been predicted as tumor biomarker for therapy. We determined to investigate effects and regulatory mechanism of MUC16 on cervical tumorigenesis. Methods Expression levels of MUC16 in cervical cancer cell lines was analyzed via qRT-PCR (quantitative real-time polymerase chain reaction). Knockdown of MUC16 was conducted via shRNA (Short hairpin RNA) transfection. MTT and colony formation assays were used to investigate effect of MUC16 on cell proliferation. Wound healing assay was utilized to detect migration and transwell assay to detect invasion. The underlying mechanism was demonstrated via western blot analysis. Results MUC16 was elevated in cervical cancer cell lines. MUC16 knockdown inhibited cell proliferation, invasion and migration. Gain- and loss-of functional assays revealed that over-expression of MUC16 activated Janus Kinase 2 (JAK2)/ signal transducer and activator of transcription 3 (STAT3) via phosphorylation, thus facilitating cyclooxygenase-2 (COX-2) expression, while knockdown of MUC16 demonstrated the reverse effect on JAK2/STAT3 activation and COX-2 expression. Moreover, inhibition of JAK2/STAT3 attenuated the regulation of MUC16 on COX-2. Conclusions MUC16 enhanced proliferation and invasion of cervical cancer cells via JAK2/STAT3 phosphorylation-mediated cyclooxygenase-2 expression, suggesting the potential therapeutic target ability of MUC16 to treat cervical cancer.

        Oxidative stress-induced circKIF18A downregulation impairs MCM7-mediated anti-senescence in intervertebral disc degeneration

        Low back pain, triggered by intervertebral disc degeneration (IVDD), is one of the most common causes of disability and financial expenditure worldwide. However, except for surgical interventions, effective medical treatment to prevent the progression of IVDD is lacking. This study aimed to investigate the effects of circKIF18A, a novel circRNA, on IVDD progression and to explore its underlying mechanism in IVDD. In this study, we found that oxidative stress was positively correlated with nucleus pulposus cell (NPC) senescence in IVDD and that circKIF18A was downregulated in IVDD and attenuated senescent phenotypes such as cell cycle arrest and extracellular matrix degradation in NPCs. Mechanistically, circKIF18A competitively suppressed ubiquitin-mediated proteasomal degradation of MCM7, and the protective effects of circKIF18A on NPCs were partially mediated by MCM7 under oxidative stress. Intradiscal injection of adenoviral circKIF18A ameliorated IVDD in a rat model. This study revealed that circKIF18A regulates NPC degeneration by stabilizing MCM7 and identified a novel signaling pathway, the circKIF18A-MCM7 axis, for anti-senescence molecular therapy in IVDD.

        Effects of regeneration conditions on sulfated CuSSZ-13 catalyst for NH3-SCR

        To understand the role of regeneration conditions on sulfur-poisoned Cu/SSZ-13 catalysts for NH3-SCR, the physicochemical characterizations and NOx conversions were investigated. The sulfur-poisoned Cu/SSZ-13 catalysts were treated at different conditions as a function of temperature and duration. TGA results revealed that regeneration at 500 oC only removed parts of sulfur spices and at 700 oC can completely remove all sulfur spices. The physical structural characterizations results illustrate that regeneration below 700 oC has no significant impact on CHA structure for Cu/SSZ-13 catalysts, while dealumination occurs on poisoned Cu/SSZ-13 when regeneration temperature is higher than 700 oC. EPR and H2-TPR results show that the sulfate decomposition and Cu migration reactions involved during regeneration and, as a result, the content of Cu2+ gradually increased as the extent of regeneration increased. The kinetics tests support that NOx conversion recovery is related to the content of Cu2+ increase during regeneration. Our study reveals that the optimum regeneration temperature is 700 oC, because severe dealumination at 750 oC inhibited Cu2+ amount increase.

        An Enhanced Grouping Proof for Multiple RFID Readers and Tag Groups

        RFID authentication on large numbers of tags or tag groups has attracted many researchers’ attention as an extension of initial one-or-two-tags scenarios. Due to demands of RFID application in supply chain, various proofs are proposed in the purpose of solving existing problems of grouping tags identification. However, it is proved that verifiers in these proofs are not aware of abnormal relating devices instantly, which may cause both security and technical risks and lie heavy upon identification of the problematic tags and readers. In this paper, we propose an enhanced grouping proof for multiple RFID readers and tag groups, according to which the verifier is able to cope with multiple readers and tag groups simultaneously and know the detailed status of the problematic tags. In addition, privacy protection is provided by offering mutual authentication between the readers and tags. Moreover, it is hard for the malicious tags to pass the verification in the proposed proof.

        Activating SIRT3 in peritoneal mesothelial cells alleviates postsurgical peritoneal adhesion formation by decreasing oxidative stress and inhibiting the NLRP3 inflammasome

        Peritoneal adhesions (PAs) are a serious complication of abdominal surgery and negatively affect the quality of life of millions of people worldwide. However, a clear molecular mechanism and a standard therapeutic strategy for PAs have not been established. Here, we developed a standardized method to mimic the pathological changes in PAs and found that sirtuin 3 (SIRT3) expression was severely decreased in adhesion tissues, which was consistent with our bioinformatics analysis and patient adhesion tissue analysis. Thus, we hypothesized that activating SIRT3 could alleviate postsurgical PAs. Sirt3-deficient (Sirt3−/−) mice exhibited many more PAs after standardized abdominal surgery. Furthermore, compared with wild-type (Sirt3+/+) mice, Sirt3-deficient (Sirt3−/−) mice showed more prominent reactive oxygen species (ROS) accumulation, increased levels of inflammatory factors, and exacerbated mitochondrial damage and fragmentation. In addition, we observed NLRP3 inflammasome activation in the adhesion tissues of Sirt3−/− but, not Sirt3+/+ mice. Furthermore, mesothelial cells sorted from Sirt3−/− mice exhibited impaired mitochondrial bioenergetics and redox homeostasis. Honokiol (HKL), a natural compound found in several species of the genus Magnolia, could activate SIRT3 in vitro. Then, we demonstrated that treatment with HKL could reduce oxidative stress and the levels of inflammatory factors and suppress NLRP3 activation in vivo, reducing the occurrence of postsurgical PAs. In vitro treatment with HKL also restored mitochondrial bioenergetics and promoted mesothelial cell viability under oxidative stress conditions. Taken together, our findings show that the rescue of SIRT3 by HKL may be a new therapeutic strategy to alleviate and block postsurgical PA formation.

        Mathematical models for manufacturing a novel gear shaper cutter

        The design principle and models for novel error-free shaper cutters are discussed to improve the accuracy and service life of gear cutting tools. The modified methods for designing the tooth profile of the shaper cutter are developed by including the inverse envelope method, midpoint-midline method and midpoint-displacement method based on the theory of geometrical reverse problem. The universal mathematical models for manufacturing optimal tooth profile of shaper cutters are presented. The feasibility and reliability of the proposed principle, methods and models are proved by combining the numerical examples with practical application. The design method suggested in this paper is superior to the traditional design scheme, and will be helpful in facilitating the design and manufacture of shaper cutters.

        Beyond design basis seismic evaluation of underground liquid storage tanks in existing nuclear power plants using simple method

        Nuclear safety-related underground liquid storage tanks, such as those used to store fuel for emergencydiesel generators, are critical components for safety of hundreds of existing nuclear power plants (NPP)worldwide. Since most of those NPP will continue to operate for decades, a beyond design base (BDB)seismic screening of safety-related underground tanks in those NPP is beneficial and essential to publicsafety. The analytical methodology for buried tank subjected to seismic effect, including a BDB seismicevaluation, needs to consider both soil-structure and fluid-structure interaction effects. Comprehensiveanalysis of such a soil-structure-fluid system is costly and time consuming, often subjected to availabilityof state-of-art finite element tools. Simple, but practically and reasonably accurate techniques for seismicevaluation of underground liquid storage tanks have not been established. In this study, a mechanicsbased solution is proposed for the evaluation of a cylindrical underground liquid storage tank using handcalculation methods. For validation, a practical example of two underground diesel fuel tanks in anexisting nuclear power plant is presented and application of the proposed method is confirmed by usingpublished results of the computer-aided System for Analysis of Soil Structural Interaction (SASSI). Theproposed approach provides an easy to use tool for BDB seismic assessment prior to making decision ofapplying more costly technique by owner of the nuclear facility

      • Mitochondria-mediated Apoptosis in Human Lung Cancer A549 Cells by 4-Methylsulfinyl-3-butenyl Isothiocyanate from Radish Seeds

        4-Methylsulfinyl-3-butenyl isothiocyanate (MTBITC) found in the radish (Raphanus sativus L.), is a wellknown anticancer agent. In this study, the mechanisms of the MTBITC induction of cell apoptosis in human A549 lung cancer cells were investigated. Our PI staining results showed that MTBITC treatment significantly increased the apoptotic sub-G1 fraction in a dose-dependent manner. The mechanism of apoptosis induced by MTBITC was investigated by testing the change of mitochondrial membrane potential (${\Delta}{\Psi}m$), the expression of mRNAs of apoptosis-related genes by RT-PCR, and the activities of caspase-3 and -9 by caspase colorimetric assay. MTBITC treatment decreased mitochondrial membrane potential by down-regulating the rate of Bcl-2/Bax and Bcl-xL/Bax, and activation of caspase-3 and -9. Therefore, mitochondrial pathway and Bcl-2 gene family could be involved in the mechanisms of A549 cell apoptosis induced by MTBITC.

        Application of a traction metal clip with a fishhook-like device in wound sutures after endoscopic resection

        Background/Aims: Endoscopic wound suturing is an important factor that affects the ability to remove large and full-thickness lesionsduring endoscopic resection. We aimed to evaluate the effect of a traction metal clip with a fishhook-like device on wound sutures afterendoscopic resection. Methods: From July 2020 to April 2021, patients who met the enrollment criteria were treated with a fishhook-like device during theoperation to suture the postoperative wound (group A). Patients with similar conditions and similar size wounds who were treatedwith a “purse-string suture” to suture the wounds were retrospectively analyzed as the control group (group B). Difference in the suturerate, adverse events, time required for suturing, and number of metal clips were compared between the two groups. Results: The time required for suturing was 7.72±0.51 minutes in group A and 11.50±0.91 minutes in group B. This difference was statisticallysignificant (F=13.071, p=0.001). The number of metal clamps used in group A averaged 8.1 pieces/case, and the number ofmetal clamps used in group B averaged 7.3 pieces/case. This difference was not statistically significant (F=0.971, p=0.331). Conclusions: The traction metal clip with the fishhook-like device is ingeniously designed and easy to operate. It has a good suture effecton the wound after endoscopic submucosal dissection and effectively prevents postoperative adverse events.
