Development of orthotopic patient-derived tumor xenograft models in endometrial cancer

목적: To evaluate orthotopic xenografting of endometrial cancer(EM Ca) in mice for development of novel translational research platform in endometrial cancer. 방법: Fresh surgical specimens were taken from patients with EM Ca after surgery at Department of Obstetrics and Gynecology, Samsung Medical Center, Seoul, Korea between December, 2011 to February, 2016. We successfully established PDXs by orthotopic and subrenal capsule implantation of patient’s EM Ca tissues into female BALB/C-nude mice. The subrenal capsule implatation method were that pieces of EM Ca specimens from patients were meticulosely grafted in the subrenal capsule. Mean while, estrogen preconditioning was done for orthotopic implatation by given a daily s.c injection of 0.1μg estradial for three days. After then, pieces of EM Ca specimens from patients were meticulosly grafted in the enlarged mice’s uterus. The H&E staining, short tandem repeat, array comparative genomic hybridization, human and mouse albumin test and cancer panel were conducted to verifythe histopathologic and genetic similarity between the PDXs and primary patient’s tissues. 결과: Orthotopic PDX engraftment rate were sucessful in 100% (3/3 cases). While, Successful rate of subrenal capsule implantation method was 55.6% (10/18 cases). In orthotopic PDXs, median duration of first generation of PDX develoment from implantation of patient’s tissues was 5.43month (range: 0.83-6.63 month). And, in subrenal capsule implantation PDXs, median duration of first generation was 5.05month (range : 4.17-9.33 month). Both methods were proved to preserve histopathologic and genetic silimarity between PDXs and the primary patient’s tissues. 결론: The PDXs for endometiral cancer with histopathologic and genetic stability can be efficiently developed by an orthotopic implantation method rather than a subrenal capsule implantation method. This method can a promising platform for conducting translational research and future precision medicine in endomerial cancer.

온라인 신분증명에는 공인인증서, I-PIN, 휴대전화 등이 사용된다. 서비스 제공자는 사용자의 신분을 증명한 후 사용자의 정보를 회원가입과 같은 절차를 통해 수집하고 저장한다. 이와 같이 중앙에서 관리하는 것은 관리적 측면의 편리성은 존재하지만 관리시스템 자체가 단일 실패 지점으로서의 위험성과 해킹 등으로 인한 개인정보 유출의 위험성이 상존한다. 최근 중앙관리의 한계를 급복하기 위해 분산 원장관리 기술인 블록체인 기술이 등장하였다. 블록체인 기술은 각각의 정보의 블록을 체인으로 연결하여 블록체인에 참여하는 각 노드에 의해 관리하는 분산관리 기술이다. 본 논문에서는 블록체인 기술을 기반으로 하는 온라인신분증명 스킴을 제안한다. 블록체인을 기반으로 하기 때문에 개인의 정보를 중앙에서 관리할 필요가 없다. 또한 블록체인의 특성상 위변조로부터 안전하다는 장점이 있다. 마지막으로, 제안하는 방법에서 노드에 저장되는 개인정보는 암호화하는 방법을 사용하여 타인의 정보를 볼 수 없도록 하는 기밀성을 제공할 수 있어 노출에 대한 안전성을 제공한다.

본 연구는 버스 정보 서비스를 대상으로 탐색적 연구를 진행하여 모바일 정보 서비스 디자인 지침을 제안하는 것을 목적으로 하였다. 이를 위해 대중 교통 정보 시스템 사용자를 대상으로 인터뷰 및 정황 조사를 실시한 결과를 정보 검색 상호작용의 계층적 모델에 근거하여 분석, 목적 분류 별 정보 니즈를 도출하였다. 그 결과, 탐색적 목적의 정보 사용시 사용자 측의 인지적 수준이 가장 중요한 영향을 주었으며, 확증적 목적의 정보 사용 시 감성적, 상황적 수준이 중요한 영향을 주고 있기에 이에 기반한 디자인 구성이 필요함을 밝혔다. This study aims to provide design guidelines of mobile information service for public transportation system. Interview and contextual inquiry was conducted. The transcripts are analyzed to identify users' general goals for using mobile information service, and to induce more specific informational needs upon each goal based on stratified model of information retrieval interaction. Findings are that a design emphasizing on cognitive level of information is necessary for users with exploratory goals, and for users with confirmatory goals, design emphasis should be put on affective and situational level of information.

<P/><P>Protein therapeutics have gained attention recently for treatment of a myriad of human diseases due to their high potency and unique mechanisms of action. We present the development of a novel polymeric thermosponge nanoparticle for efficient delivery of labile proteins using a solvent-free polymer thermo-expansion mechanism with clinical potential, capable of effectively delivering a range of therapeutic proteins in a sustained manner with no loss of bioactivity, with improved biological half-lives and efficacy in vivo.</P>

The switching off characteristics of the power bipolar transistor, with the LDBC (low doped base contact), is presented. The simulation results for ihc resistive load circuit show that the transistor with LDBC has a shorter storage and fall time for the large reverse bias current. It is concluded that the LDBC offers a lower minimum of power loss and wider range of biasing flexibility to the designer because of the lack of tailing phenomena seriously occurred in the normal base power transistor.<br/> Experimental results proved the improvement of switching speed for the LDBC structure and make it possible to design switching transistor without life time killing technique.

<P>Methicillin-resistant <I>Staphylococcus aureus</I> (MRSA) is difficult to treat using available antibiotic agents. Honeybee venom has been widely used as an oriental treatment for several inflammatory diseases and bacterial infections. The venom contains predominantly biologically active compounds, however, the therapeutic effects of such materials when used to treat MRSA infections have not been investigated extensively. The present study evaluated bee venom and its principal active component, melittin, in terms of their antibacterial activities and <I>in vivo</I> protection against MRSA infections. <I>In vitro</I>, bee venom and melittin exhibited comparable levels of antibacterial activity, which was more marked against MRSA strains, compared with other Gram-positive bacteria. When MRSA-infected mice were treated with bee venom or melittin, only the latter animals were successfully rescued from MRSA- induced bacteraemia or exhibited recovery from MRSA-infected skin wounds. Together, the data of the present study demonstrated for the first time, to the best of our knowledge, that melittin may be used as a promising antimicrobial agent to enhance the healing of MRSA-induced wounds.</P>
