Characterization of a novel variant HMW‐glutenin gene from Elymus canadensis

High molecular weight (HMW) glutenin subunits (GS) play a key role in the determination of end‐use quality of wheat and other cereal crops. In this study, we report the isolation and characterization of both promoter region and ORF of novel HMW‐GS allele 1St1.3 from a perennial Triticeae species,Elymus canadensis. The amino acid (AA) sequences of E. canadensis 1St1.3 were deduced as 434 aa. Its protein primary structure comprises a signal peptide with a conserved N‐terminal domain, a central repetitive domain and a C‐terminal domain. E. canadensis 1St 1.3 possesses several distinct characteristics which are different from those of wheat HMW‐GSs. The N‐terminal domains of E. canadensis 1St 1.3 resemble that of y‐type subunits, while their C‐terminal domains are more similar to x‐type subunits. The deletion of 85 bp fragment has been observed in promoter region of 1St 1.3, however which has not interrupted the expression of this gene. Our results indicate that 1St 1.3 is novel HMW‐GS variants which will be valuable for enhancing our understanding of structural differentiation and the evolutionary relationship among HMW‐GSs in Triticeae species.
How Does Power Distance Belief Impact Variety-Seeking Behavior? A Moderated Mediation Approach

Purpose - This research aims to enhance knowledge of variety-seeking behavior by establishing a novel antecedent based on consumer internal characteristics instead of external stimuli. The report also argues that the insights gleaned can illuminate a method to analyze consumer decision-making based on intrinsic motivation. This study evaluated a potential connection between power distance belief and variety-seeking behavior. This study evaluates variety-seeking behavior by brand, color, taste, and fashion preferences. Varietyseeking behavior is essential in understanding consumer choices. This paper aims to find the mechanism of the relation between power distance belief with variety-seeking behavior, along with the boundary condition of variety-seeking behavior. Design/Methodology/Approach - This study performed three experiments for measurement and hypothesis testing. This study employed analysis of variance, multiple regression, and process macro for moderated mediation testing with SPSS 28.0. Findings - This research confirms the prediction that higher power distance beliefs tend to elect variety through Study 1, and that risk-seeking mediates the moderation of boredom between power distance belief and variety-seeking behavior through Study 2. The authors conclude by discussing the implications for the application of the theory for managers. Research Implications - These contributions provide professionals with practical consequences for a variety of marketing tasks, such as market segmentation, product extension and innovation, and advertising guides.

Background: Although the predictive value of the excision repair cross-complementing group 1 (ERCC1) C118T polymorphism in clinical outcomes of patients with colorectal cancer (CRC) receiving oxaliplatin-based chemotherapy has been evaluated in numerous published studies, the conclusions are conflicting. Therefore, we performed the present meta-analysis to determine the precise role of the ERCC1 C118T polymorphism in this clinical situation and help optimize individual chemotherapy. Materials and Methods: A multiple search strategy was used to identify eligible studies. Pooled odds ratios (ORs) and their 95% confidence intervals (CIs) were used to estimate objective response and oxaliplatin-induced toxicity, with hazard ratios (HRs) with 95%CIs for progression-free survival (PFS) and overall survival (OS). Results: A total of 22 studies including 2,846 CRC patients were eligible in the analysis. Overall, no significant correlation was found between the ERCC1 C118T polymorphism and objective response to oxaliplatin-based chemotherapy, in all patients or in the Asian and Caucasian subgroups. However, the pooled analysis showed that the PFS and OS were significantly shorter in patients who carried T/T or T/C genotypes of ERCC1 C118T as compared to the C/C genotype. On stratified analysis by ethnicity, the ERCC1 118T allele was associated with a favorable prognosis in Caucasians (PFS, HR=0.58, 95%CI: 0.24-1.44; OS, HR=0.38, 95%CI: 0.22-0.64) but an unfavorable prognosis in Asians (PFS, HR=2.49, 95%CI: 1.87-3.33; OS, HR=2.63, 95%CI: 1.87-3.69) based on a dominant model. In addition, we failed to find a statistically significant impact of ERCC1 C118T polymorphism on oxaliplatin-induced toxicity. Conclusions: The ERCC1 C118T polymorphism may have prognostic value in patients with CRC undergoing oxaliplatin-based chemotherapy.

Background: The predictive value of the xeroderma pigmentosum group D (XPD) Lys751Gln polymorphism regarding clinical outcomes of patients with colorectal cancer (CRC) receiving oxaliplatin-based chemotherapy has been evaluated in numerous published studies, but the results remain inconclusive. Therefore, we performed a meta-analysis to determine the precise role of the XPD Lys751Gln polymorphism in this clinical situation and optimize individual chemotherapy. Materials and Methods: A multiple search strategy was used to identify eligible studies. Pooled odds ratios (ORs), generalized odds ratio (ORG) and their 95% confidence intervals (CIs) were used to estimate the objective response, while hazard ratios (HRs) with 95%CIs were used for progression-free survival (PFS) and overall survival (OS). Results: A total of 17 studies including 2,286 patients met the inclusion criteria. Overall, the XPD 751Gln allele was associated with a non-significant reduced objective response to oxaliplatin-based chemotherapy in all patients or in the Asian and Caucasian subgroups. However, poor PFS and OS of CRC patients treated with oxaliplatin-based regimens were significantly related to the XPD 751Gln allele in the dominant model (PFS: HR=2.10, 95%CI: 1.65-2.67; OS: HR=3.18, 95%CI: 1.57-6.47). On stratified analysis by ethnicity, these relationships were more pronounced in Asians (PFS: HR=2.49, 95%CI: 1.79-3.47; OS: HR=5.25, 95%CI: 3.46-7.94) than in Caucasians (PFS: HR=1.73, 95%CI: 1.22-2.46; OS: HR=1.78, 95%CI: 1.06-2.99). Conclusions: The XPD Lys751Gln polymorphism may have prognostic value in patients with CRC undergoing oxaliplatin-based chemotherapy.
A study on the modified Hough algorithm for image processing in weld seam tracking

Nowadays, automatic Gas metal arc (GMA) welding is receiving a growing attention for its fast production speed and wide-rangingversatility. Furthermore, automatic and robotic welding systems are urgently needed for general application, which depends on the realizationof auto-weld seam tracking. Seam tracking system allows for not only robot or machine trajectory shifts, but also adaptive controlto change weld bead formation, making further improvement on the weld quality. Based on the laser vision system, this study aims fordevelopment of an image processing by the modified Hough algorithm which applied for seam tracking system in GMA welding. Toachieve this objective, image features in the procedure of welding processing were discussed and specially treated. Algorithms in theimage processing were investigated to maximize the processing effect based on the modified Hough algorithm, which was quite costefficientin line detection. Quantitative evaluations of the used algorithm were set up to acquire optimal image processing algorithm. Finally, a common image processing method was employed to verify efficiency of proposed image processing and extract the weldinglocation in a shorten time for industrial application. It can be concluded the detected feature points and centerlines proved that this algorithmcan be used for automatic GMA welding process as well as industrial applications.
Determining Inspection Intervals for One-shot Systems with Support Equipments

In this paper, we consider one-shot systems comprise one-shot devices and support equipments. The one-shot devices are stored for long periods of time, and its failure is detected only upon inspection. Support equipments are necessary for the operation of one-shot devices, and are maintained immediately upon failure. This paper deals with an inspection schedule problem for one-shot systems with support equipment and limited maintenance resources. Interval availability and life cycle costs are used as optimization criteria, and are estimated by simulation. Therefore, we aim to determine near-optimal inspection intervals of one-shot systems that minimize the expected life cycle cost and satisfy the target interval availability during inspection periods. An estimation of distribution algorithm (EDA) is proposed to find near-optimal solutions, and numerical examples are studied to investigate the effects of various model parameters.
