Genome-wide analysis of histone modifications in latently HIV-1 infected T cells

<P><B>Objectives:</B></P><P>The transcriptional silencing of HIV type 1 (HIV-1) provirus in latently infected cells is a major hurdle on the pathway to HIV-1 elimination. The epigenetic mechanisms established by histone modifications may affect the transcriptional silencing of HIV-1 and viral latency. A systematic epigenome profiling could be applicable to develop new epigenetic diagnostic markers for detecting HIV-1 latency.</P><P><B>Design:</B></P><P>The HIV-1 latency cell lines (NCHA1, NCHA2 and ACH2] were compared with CD4<SUP>+</SUP> T-cell line (A3.01).</P><P><B>Methods:</B></P><P>The histone modification profiles obtained from chromatin immunoprecipiation followed by sequencing (ChIP-Seq) for histone H3K4me3 and H3K9ac were systematically examined and differential gene expression patterns along with levels of histone modifications were used for network analysis.</P><P><B>Results:</B></P><P>The HIV-1 latency gave rise to downregulation of histone H3K4me3 and H3K9ac levels in 387 and 493 regions and upregulation in 451 and 962 sites, respectively. By network analysis, five gene clusters were associated with downregulated histone modifications and six gene clusters came up with upregulated histone modifications. Integration of gene expression with epigenetic information revealed that the cell cycle regulatory genes such as <I>CDKN1A</I> (p21) and cyclin D2 (<I>CCND2</I>) identified by differentially modified histones might play an important role in maintaining the HIV-1 latency.</P><P><B>Conclusion:</B></P><P>The transcriptional regulation by epigenetic memory should play a key role in the evolution and maintenance of HIV-1 latency accompanied by modulation of signalling molecules in the host cells.</P>
A Survey on Characteristics and Trends of the WEEE Discharge in Korea

Since 2003, Extended Producer Responsibility (EPR) system has been continuously applied in the context of collection and recycling of Waste Electrical and Electronic Equipment (WEEE). This system has contributed to design the WEEE management directives and infra-structures for WEEE collecting and recycling in Korea. As an example, Ministry of Environment (MOE) achieved WEEE collection performance that approximately 297,610t of WEEE was collected in formal sector from the all nationwide in 2018. MOE takes this achievement a step further, encourage to expand the formal sector for the more WEEE collection and recycling performance. In this study, we investigate the current status of WEEE discharging trends and user behavior as well as systematical strategies that how to expand the formal sector in WEEE discharging step. A survey interviews were implemented using the telephone survey with computer-assisted telephone interviewing (CATI) method targeting the number of 2,000 interviewees. Survey results showed that 31.50% (n=630) of total respondents have the WEEE discharging experience within recent one year, and the most discharged item was refrigerator (n=221, 35.08%) from the interviewees. From the whole interviewee’s perspective, the top three discharge channels preferred were Local Authority (39.00%), Take-back (37.30%), and Personal Recyclers (10.20%). On the other hand, respondents who have responded had discharge experience within recent one year preferred the Local Authority (27.50%), Take-back (26.80%), and Door-to-Door service (18.7%) respectively. Result for survey on whether interviewees holding or not the end-of-life appliances which was potentially needed showed that the 38.05% (n=761) of the whole respondents have and hold the end-of-life appliances in their house. The most type of end-of-life appliances which held in their inhabitancy were refrigerator (48.10%), washing machine (16.03), television (9.46%), and computer (4.20%). This study can be utilized as a basic data for establishing more effective WEEE collecting strategies by the MOE based on WEEE discharge experience, routes, and potential item to be discharged.
A new flexible Weibull distribution

Many of studies have suggested the modifications on Weibull distribution to model the non-monotone hazards. In this paper, we combine two cumulative hazard functions and propose a new modified Weibull distribution function. The newly suggested distribution will be named as a new flexible Weibull distribution. Corresponding hazard function of the proposed distribution shows flexible (monotone or non-monotone) shapes. We study the characteristics of the proposed distribution that includes ageing behavior, moment, and order statistic. We also discuss an estimation method for its parameters. The performance of the proposed distribution is compared with existing modified Weibull distributions using various types of hazard functions. We also use real data example to illustrate the efficiency of the proposed distribution.

The waste of electrical and electronic equipment (WEEE) involves various types of products that have reached the end of their lifespan and can cause environmental contamination and health issues for humans when WEEE is treated in informal sectors. Since 2008, Korea has operated the Extended Producer Responsibility (EPR) system and the Eco-Assurance system (Eco-AS), which collect and recycle WEEE under related laws. Although the EPR and Eco-AS systems successfully dealt with about 376,000 tons of WEEE in 2020, approximately 58% remained in the informal sector, posing potential risks to the environment and humans. Consequently, this paper was designed to deliver information currently related to the status of WEEE recycling in Korea, mainly regarding legislation, management systems, recycling achievement, and potential problems. This study discussed the seriousness of the informal sector, in which WEEE is illegally treated without proper guidelines, and sought ways to reduce the proportion of the informal sector.
Surgical Outcomes of Sphenoid Wing Meningioma with Periorbital Invasion

Objective : The aim of this study was to evaluate the clinical outcome of sphenoid wing meningioma with periorbital invasion (PI) after operation. Methods : Sixty one patients with sphenoid wing meningioma were enrolled in this study. Their clinical conditions were monitored after the operation and followed up more than 5 years at the outpatient clinic of a single institution. Clinical and radiologic information of the patients were all recorded including the following parameters : presence of PI, presence of peri-tumor structure invasion, pathologic grade, extents of resection, presence of hyperostosis, exophthalmos index (EI), and surgical complications. We compared the above clinical parameters of the patients with sphenoid wing meningioma in the presence or absence of PI (non-PI), then linked the analyzed data with the clinical outcome of the patients. Results : Of 61 cases, there were 14 PI and 47 non-PI patients. PI group showed a significantly higher score of EI (1.37±0.24 vs. 1.00±0.01, p<0.001), more frequent presence of hyperostosis (85.7% vs. 14.3%, p<0.001), and lower rate of gross total resection (GTR) (35.7% vs. 68.1%, p=0.032). The lower score of pre-operative EI, the absence of both PI and hyperostosis, smaller tumor size, and the performance of GTR were associated with lower recurrence rates in the univariate analysis. However, in the multivariate analysis, the performance of GTR was the only significant factor to determine the recurrence rate (p=0.043). The incidences of surgical complications were not statistically different between the subtotal resection (STR) and GTR groups, but it was strongly associated tumor size (p=0.017). Conclusion : The GTR group showed lower recurrence rate than the STR group without differences in the surgical complications. Therefore, the GTR is strongly recommended to treat sphenoid wing meningioma with PI for the better clinical outcome.
