      • Redox-responsive biodegradable nanogels for photodynamic therapy using Chlorin e6

        <P>Recent advances in drug carrier design in the field of photodynamic therapy (PDT) have stimulated the development of numerous sophisticated drug delivery carriers. We developed novel biodegradable and biocompatible nanogels as PDT carriers. The nanogels were synthesized by ATRP technique using inverse miniemulsion and their biodegradability was determined in the presence of glutathione. The model photosensitizer (PS) was encapsulated in the biodegradable nanogels by simple mixing and sonication. The cellular uptake of the PS-loaded nanogels and the cytotoxicity of the nanogels before and after laser irradiation were determined using HeLa cells. The results showed that the Ce6-nanogel complex was well internalized into the tumor cells, and the Ce6-loaded nanogels did not influence on cellular viability of the cells before light irradiation. Under light exposure, however, the Ce6-nanogel complex-treated HeLa cells revealed strong photoactivity. These nanogels may enhance therapeutic efficacy of PSs without any complex chemical modifications with PSs.</P>

      • Short communication: Bacteriocin KC24 produced by Lactococcus lactis KC24 from kimchi and its antilisterial effect in UHT milk

        The severity of Listeria monocytogenes infections emphasizes the need for prevention or elimination of the pathogen from dairy products. Lactococcus lactis KC24, isolated from kimchi, exhibited an antimicrobial effect against food pathogens, including L. monocytogenes ATCC 15313. Lactococcus lactis KC24 was cultured in a 5-L jar fermenter at 35<SUP>o</SUP>C, and bacteriocin activity was maximal at 4h of incubation and persisted for 20h. Bacteriocin KC24 was inactivated by protease XIV, indicating that it has a proteinaceous nature. Bacteriocin activity was maintained at pH 3.0 to 9.0 and at temperatures of 50 to 121<SUP>o</SUP>C. The mode of inhibition against L. monocytogenes ATCC 15313 was shown to involve a bactericidal effect by treatment with 100 and 200 arbitrary units (AU)/mL of bacteriocin KC24. To test the activity of bacteriocin KC24 in a food product, bacteriocin KC24 and nisin (100 and 200AU/mL) with 4 log cfu/mL of a mixed culture of L. monocytogenes (ATCC 15313, ScottA, H7962, and H7762) were applied to UHT milk. Compared with the control, treatment with bacteriocin KC24 completely inhibited the growth of L. monocytogenes and resulted in no detectable L. monocytogenes after 14d at 4<SUP>o</SUP>C, whereas nisin moderately inhibited L. monocytogenes, resulting in a final concentration after 14d at 4<SUP>o</SUP>C higher than the initial inoculum. Bacteriocin KC24 may prove useful in improving the safety of dairy products.

      • 韓牛의 肩胛骨 發生에 關한 形態學的 硏究

        This study was undertaken to obtain basic data on the fetal endochondral ossification in the native Korean cattle scapula, using histochemical observation to determine the earliest stages of both chondrification and ossification. Thirty scapulae, from a series of embryos and fetuses of the korean native cattle ranging from 11 to 70mm in crown-rump (C-R) length were used The ossification center was observed in the supra and infraspinus fossa in the 5th group (CRL 51-60mm), markedly ossified in the 6th group (CRL 61-70mm) by Alizatin red S stain. The chondrogenic center was observed in the 1st group (CRL 11-20mm), and the primary ossification center was presented in the 4th group (CRL 41-50mm) by H-E stain. Osteocytes, Osteoblasts, Myeloid cells, and irregular ladder shaped trabeculae were observed in actively developing areas of endochondral ossification in the 5th group (CRL 51-60mm)by H-E stain. Alcianophility was increased markedly in the intererritorial matrix in the 3rd group (CRL 31-40mm). Also this reaction was decreased markedly in the interterritorial matrix, the adjacent portion to the marrow cavity, and trabecula in the 5th group (CRL 51-60mm).

      • 한국에서 유행하는 �냑鵝뵉퓌竄叢�관한 혈청역학적 연구

        A seroepidemiological survey was performed to estimate the prevalence of leptospirosis in Korean population. The study population consisted of 967 randomly selected people from 17 counties out of 5 provinces. Macroscopic slide agglutination test was used for serology of leptospirosis. 117 sera out of 967 were positive (11.7%) for leptospirosis. Kang Won Province showed highest positive rate (17.1%), followed by Kyunggi (16.2%), Chung Book (10.2%), Kyung Book (6.2%), Chun Nam Province (2.2%), Hoeng Sung County showed highest prevalence rate (26.2%) and Chang Sung County Showed lowest prevalence (0%). There was no difference in prevalence rate between male and female. But older people (41∼60 years) showed higher prevalence rate than the younger people (21∼30 years). Some activities, including weeding in the field or grave and working barefoot in the rice field were associated with higher prevalence. History of recent flu-like symptoms suggestive of leptospirosis (chill, headache, cough, walking difficulty, dyspnea, diarrhea, conjunctival suffusion) was more common in antibody positive group. Leptospirosis is wide spread infection throughout Korea, though considerable variance in prevalence rate was noted.

      • 대규모 원단 도매 시장에서의 포름알데히드 노출에 관한 연구

        This study was conducted to assessment airborne formaldehyde concentration at dry-goods stores, 'G' market and additionally at the reconstructed lecture room in the university during November, 2001. 'NIOSH 2016' method, with 2,4-DNPH treated silica gel sampler and HPLC, was used for sampling and analysis of formaldehyde concentration. Because every merchant rejects to participate in study, volunteers, pretend to customer, were involved to monitor personal exposure level. The results are summarized as follows ; 1. The geometric mean(GM) and geometric standard deviation(GSD) of TWA concentrations from 'G' market were 0.037 ppm and 2.17 respectively. 2. The TWA concentrations of the personal samples from 'G' market ranged from 0.009 to 0.065 ppm and those of the area samples from '' market ranged from 0.06 to 0.071 ppm. 3. The TWA concentrations of the area samples from reconstructed lecture room ranged from 0.022 to 0.094 ppm. The TWA concentration from latest reconstructed lecture room was higher than that of older reconstructed lecture room. 4. All of samples except one were exceeding NIOSH TWA criteria, 0.016 ppm.

      • Protection of Mice against <i>Mycobacterium tuberculosis</i> Infection by Immunization with Aqueous Fraction of Triton X-100-soluble Cell Wall Proteins

        <P>Abstract</P><P>The aqueous fraction of Triton X-100-soluble proteins (TSP-Aq) of <I>Mycobacterium tuberculosis</I> cell wall was reported to stimulate T-cell responses in peripheral blood monocytes from tuberculosis (TB) patients and to induce Th1 cytokines, suggesting presence of protective antigens. In this study, therefore, we examined the protective efficacy of TSP-Aq against <I>M. tuberculosis</I> infection in a mouse model. C57BL/6 mice were immunized with TSP-Aq or culture filtrate proteins (CFP) mixed with incomplete Freund’s adjuvant or with BCG followed by i.v. challenge with <I>M. tuberculosis</I> H37Rv. TSP-Aq induced strong interferon-&ggr; production by spleen cells, and mice immunized with TSP-Aq antigens gave a significant reduction in <I>M. tuberculosis</I> CFU counts by 1.17–1.32 log<SUB>10 </SUB>CFU in the lungs and 1.31–2.08 log<SUB>10</SUB> CFU in the spleen from 6 to 28 weeks. The degree of protection offered by TSP-Aq was comparable to that of CFP and of the BCG vaccine. The results demonstrated that the TSP-Aq antigens confer a significant level of protection against the growth of the organism in the lungs and spleen in a mouse model of TB and indicate that TSP contains major protective antigens of <I>M. tuberculosis</I>. </P>

        흰쥐 지방세포 원형질막 단백질에 대한 다클른항체의 in vitro 세포 특성효과

        The objectives of the current study were to raise polyclonal antibodies in sheep against adipocyte plasma membrane(APM) proteins isolated from Sprague-Dawley rat, to investigate tissue specificity, and to determine cytotoxic effects of antiserum on cultured rat adipocytes. Plasma membrane proteins from adipocyte, heart, kidney, liver, muscle, and spleen were isolated using an iso-osmotic sucrose self-forming gradient. An adult male sheep was immunized three times at three week intervals with the purified Sprague-Dawley rat APM proteins. Antisera were taken from immunized sheep at 10, 12, and 14 days after the third immunization. Antiserum exhibited strong antigen-antibody reactivity against APM proteins determined by enzyme-linked immunosorbent assay(ELISA), and the reactivity could be detected at dilutions in excess of 1:81,000. Antisera showed very low binding affinity with proteins isolated from heart, kidney, liver, muscle or spleen whilst they showed very high reactivity with APM proteins in ELISA. Tissue specificity of the antisera was reconfirmed by Western immunoblotting using anti-sheep immunoglobulin G-alkaline phosphatase conjugate as a secondary antibody. Treatment of antisera on confluent Sprague-Dawley rat adipocytes in culture caused lysis of the cells and release of cytosolic lactate dehydrogenase whereas adipocytes treated with non-immunized serum maintained their integrity.
