This study was performed to assess anthropometric status, functional health and physical fitness among the elderly in Seoul and Kyunggi-do. Four hundred forty free-living elderly people(127 male, 331 female), aged 65 or more were interviewed by trained interviewers using structured questionnaires including demographic information, activities of daily living(ADL), instrumental activities of daily living(IADL), physical activity. And they measured anthropometic status and physical fitness. All the data were analyzed by student t-test, chi-square analysis using SPSS 9.0 version at p<0.05. The results can be summarized as follow : Of all the subjects, 46.3% were obesity, especially 92.4% of female were abdominal obesity. In functional health status, ADL score was 6.62±1.54, and IADL score was 25.94±6.92. Most of the subjects were difficult to walk and run. Female was a significantly higher flexibility and body fat than male(p<0.001), whereas female was a significantly lower muscular endurance than male(p<0.001).

As the use of preservatives in foods has been increased, the misuse and overuse of preservatives are occasionally occurred. As the variety of food has risen, it is necessary to establish the screening method to determine preservatives in various foods. In this study, benzoic acid, sorbic acid, methyl p-hydroxybenzoate, ethyl p-hydroxybenzoate, i-propyl p-hydroxybenzoate, n-butyl p-hydroxybenzoate were analyzed by TLC and HPLC. In order to determine the recoveries from food, these preservatives were spiked into foods. As a result, solvent systems of 8:2:0.45:0.05 and 12:2:0.45:0.05 of pentane:acetic acid:chloroform:ethanol were appropriate for TLC analysis of preservatives. All preservatives were well resolved with solvent system of 2% ammonium acetate in 2.5% acetic acid:methanol (90/10~10/90) by HPLC. Direct extraction of preservatives from food proved to be more proper than distillation and extraction method. The recoveries by direct extraction method showed 93.7~104.6% of preservatives studied.

<P>Calcineurin is a calcium-activated phosphatase to mediate lymphocyte activation and neuron signaling, but its role in inflammatory arthritis remains largely unknown. In this study, we demonstrate that calcineurin was highly expressed in the lining layer, infiltrating leukocytes, and endothelial cells of rheumatoid synovium. The basal expression levels of calcineurin were higher in the cultured synoviocytes of rheumatoid arthritis patients than those of osteoarthritis patients. The calcineurin activity in the synoviocytes was increased by the stimulation with proinflammatory cytokines such as IL-1beta and TNF-alpha. Moreover, rheumatoid arthritis synoviocytes had an enlarged intracellular Ca(2+) store and showed a higher degree of [Ca(2+)](i) release for calcineurin activity than osteoarthritis synoviocytes when stimulated with either TNF-alpha or phorbol myristate acetate. IL-10, an anti-inflammatory cytokine, failed to increase the Ca(2+) and calcineurin activity. The targeted inhibition of calcineurin by the overexpression of calcineurin-binding protein 1, a natural calcineurin antagonist, inhibited the production of IL-6 and matrix metalloproteinase-2 by rheumatoid synoviocytes in a similar manner to the calcineurin inhibitor, cyclosporin A. Moreover, the abundant calcineurin expression was found in the invading pannus in the joints of mice with collagen-induced arthritis. In these mice, calcineurin activity in the cultured synovial and lymph node cells correlated well with the severity of arthritis, but which was suppressed by cyclosporin A treatment. Taken together, our data suggest that the abnormal activation of Ca(2+) and calcineurin in the synoviocytes may contribute to the pathogenesis of chronic arthritis and thus provide a potential target for controlling inflammatory arthritis.</P>
