        전안부 빛간섭단층촬영계를 이용한 연성콘택트렌즈의 산소투과율에 따른 중심각막두께 변화 비교

        목적: 전안부 빛간섭단층촬영계를 이용하여 산소투과율이 다른 연성콘택트렌즈를 착용하였을 때 각막의 저산소 상태에서 산소투과율에 따른 중심각막두께의 변화를 비교하여 알아보고자 하였다. 대상과 방법: 건강한 성인 30명을 대상으로 렌즈를 착용하지 않은 눈을 대조군으로 하고 산소투과율이 낮은 렌즈와 산소투과율이 높은 렌즈를 각각 착용하여 눈을 감은 상태를 유지하면서 120분 동안 30분 간격으로 전안부 빛간섭단층촬영계를 이용하여 중심각막 두께를 측정하였다. 결과: 상피층의 두께 변화는 세 그룹 간에 유의한 차이는 없었지만, 기질층 두께의 변화에서는 산소투과율이 낮은 그룹이 다른 두 그룹에 비해 증가하였으며 30분마다 약 1%씩 통계적으로 유의한 증가를 보였다(p<0.05, ANOVA). 전체 각막 두께의 변화에서도 산소투과율이 낮은 그룹이 다른 두 그룹에 비해 유의하게 증가하였고 산소투과율이 높은 그룹과 대조군은 두 그룹 간에 유의한 차이가 없었다. 중심각막두께의 최종변화는 산소투과율이 낮은 그룹은 27.69 ± 9.36 um, 산소투과율이 높은 그룹은 15.07 ± 11.49 um로 산소투과율이 낮은 그룹이 산소투과율이 높은 그룹에 비해 유의하게 증가하였다. 결론: 전안부 빛간섭단층촬영계를 이용하여 저산소 상태에서 산소투과율이 낮은 렌즈가 산소투과율이 높은 렌즈에 비해 기질층의 부종을 통하여 중심각막두께를 증가시키는 것을 알 수 있었다. Purpose: With anterior segment optical coherence tomography(OCT), changes of central corneal thickness should be compared by contact lens oxygen transmissibility when wearing different soft contact lens under the corneal hypoxic and closed-eye condition. Methods: Control group is eyes which don t wear lens for 30 healthy adults. They wore lens with low oxygen transmissibility (Lens A) and lens with high oxygen transmissibility (Lens B). Central corneal thickness was measured by anterior segment OCT every 30 minutes maintained closed-eye during 120 minutes. Results: There was no significant change among the three groups for the changes of epithelial thickness. But, in the changes of stromal thickness, Lens A group was increased in comparison with other two groups and showed statistically significant increase of about 1% every 30 minutes (p < 0.05). In changes of the whole corneal thickness, Lens A group showed significant increase in comparison with other two groups and there was no significant difference between Lens B group and control group. In the final changing rate of the central corneal thickness, Lens A group was 5.07 ± 3.06% and Lens B group was 2.68 ± 1.67%. Lens A group showed more significant increased than Lens B group. Conclusions: Under the hypoxic condition, when wearing lens with low oxygen transmissibility for 120 minutes, central corneal thickness showed significant increase. And, usages of anterior segment OCT should be gradually increased for several diseases of various purposes because it can grasp with the whole anterior video under the condition wearing lens.

      • 反應性染料의 着色拔染에 對한 顔料 및 拔染劑의 處理條件에 關한 硏究

        For this study, colcor discharge was made of the discharge agent Emulsion paste following the dip dyeing by applying the reaction dyes to the cotton fabrics, and given steam heat treatment by printing it on dip dyed fabric, and then the spectrophotometer was used to find out dyeing effect depending on its respective density, through which the color differences were obtained, the clearness were compared with each other, light fastness and washing fastness were measured and the optimal discharge agent and treatment conditions were viewed. With the results of above process, the following phenomenon have been noticed. 1) It was found that the color blots with the thicker density of the discharge agent. The optimal density could be best obtained from Decroline 5%, Rongarite 10%, and Stannous chloride 3%. 2) The clearness measured by the spectrophotometer proved that Stannous chloride 3% and 30 minute heat treatment steaming showed 1.1 color differences, the best effect of color differences. 3) In the light fastness, the Stannous chloride was effective more than 5th grade in the color fastness. In the light fastness, the color in pigment was 1 grade better than the dip dyed fabric. 4) In the washing fastness, the dip dye4 fabric could obtained 4 grade better effect. However, the color in pigment is 1 or 2 grade lower in the Decroline and Rongalite, and the fabrics ironed by using of Yeomwhaje 1 suk showed 4 grade superior. 5) In preparation of color discharge paste gives a significant affection on Viscosity depending on the mixing process. And it is recommendable to following process in its preparation: color discharge-glycerine-pigment-emulsion in that order. with the result above, it was found that among the 3 color discharge paste, the Stannous chloride was superior in all the clearness, light fastness and washing fastness. Also it can be viewed that the Rongalite or Decroline were not so effective in the adherence of Emulsion. I wish to continue further studies on the scientific optimal conditions of the color discharge paste to contribute to the growth of dyeing industry.

        Development of bacteria as diagnostics and therapeutics by genetic engineering

        Bacteria sense and respond to the environment, communicate, and continuously interact with their surroundings, including host bodies. For more than a century, engineers have been trying to harness the natural ability of bacteria as live biotherapeutics for the treatment of diseases. Recent advances in synthetic biology facilitate the enlargement of the repertoire of genetic parts, tools, and devices that serve as a framework for biotherapy. This review describes bacterial species developed for specific diseases shown in in vitro studies and clinical stages. Here, we focus on drug delivery by programing bacteria and discuss the challenges for safety and improvement.

        DNA looping-mediated repression by histone-like protein H-NS: specific requirement of Esigma70 as a cofactor for looping.

        <P>Transcription initiation by RNA polymerase (RNP) carrying the house-keeping sigma subunit, sigma70 (Esigma70), is repressed by H-NS at a number of promoters including hdeABp in Escherichia coli, while initiation with RNP carrying the stationary phase sigma, sigma38 (Esigma38), is not. We investigated the molecular mechanism of selective repression by H-NS to identify the differences in transcription initiation by the two forms of RNPs, which show indistinguishable promoter selectivities in vitro. Using hdeABp as a model promoter, we observed with purified components that H-NS, acting at a sequence centered at -118, selectively repressed transcription by Esigma70. This selective repression is attributed to the differences in the interactions between hdeABp and the two forms of RNPs, since no other factor is required for the repression. We observed that the two forms of RNPs could form an open initiation complex (RP(O)) at hdeABp, but that Esigma70 failed to initiate transcription in the presence of H-NS. Interestingly, KMnO4 assays and high-resolution atomic force microscopy (AFM) revealed that hdeABp DNA wrapped around Esigma70 more tightly than around Esigma38, resulting in the potential crossing over of the DNA arms that project out of Esigma70 . RP(O) but not out of Esigma38 . RP(O). Based on these observations, we postulated that H-NS bound at -118 laterally extends by the cooperative recruitment of H-NS molecules to the promoter-downstream sequence joined by wrapping of the DNA around Esigma70 . RP(O), resulting in effective sealing of the DNA loop and trapping of Esigma70. Such a ternary complex of H-NS . Esigma70 hdeABp was demonstrated by AFM. In this case, therefore, Esigma70 acts as a cofactor for DNA looping. Expression of this class of genes by Esigma38 in the stationary phase is not due to its promoter specificity but to the architecture of the promoter . Esigma38 complex.</P>

        Eradication of drug-resistant Acinetobacter baumannii by cell-penetrating peptide fused endolysin

        Antimicrobial agents targeting peptidoglycan have shown successful results in eliminating bacteria with high selective toxicity. Bacteriophage encoded endolysin as an alternative antibiotics is a peptidoglycan degrading enzyme with a low rate of resistance. Here, the engineered endolysin was developed to defeat multiple drug-resistant (MDR) Acinetobacter baumannii. First, putative endolysin PA90 was predicted by genome analysis of isolated Pseudomonas phage PBPA. The His-tagged PA90 was purified from BL21(DE3) pLysS and tested for the enzymatic activity using Gram-negative pathogens known for having a high antibiotic resistance rate including A. baumannii. Since the measured activity of PA90 was low, probably due to the outer membrane, cell-penetrating peptide (CPP) DS4.3 was introduced at the N-terminus of PA90 to aid access to its substrate. This engineered endolysin, DS-PA90, completely killed A. baumannii at 0.25 μM, at which concentration PA90 could only eliminate less than one log in CFU/ml. Additionally, DS-PA90 has tolerance to NaCl, where the ~50% of activity could be maintained in the presence of 150 mM NaCl, and stable activity was also observed with changes in pH or temperature. Even MDR A. baumannii strains were highly susceptible to DS-PA90 treatment: five out of nine strains were entirely killed and four strains were reduced by 3–4 log in CFU/ml. Consequently, DS-PA90 could protect waxworm from A. baumannii-induced death by ~70% for ATCC 17978 or ~44% for MDR strain 1656-2 infection. Collectively, our data suggest that CPP-fused endolysin can be an effective antibacterial agent against Gramnegative pathogens regardless of antibiotics resistance mechanisms.

        Lipocalin2 as a potential antibacterial drug against Acinetobacter baumannii infection

        Available antibiotics to treat Acinetobacter baumannii infection is limited due to increasing resistance and the emergence of multiple drug-resistant strains. Hence, discovering effective agents against A. baumannii to reduce the number of infectionrelated deaths is imperative. In search of novel and alternative antibiotics, the antibacterial function of lipocalin2 (Lcn2) was investigated to treat systemic infections of A. baumannii using a mouse neutropenia model. We observed a significant increase in serum Lcn2 levels upon bacterial injection into the mouse, and the administration of recombinant Lcn2 (rmLcn2) extended their survival. Such protective effects were also observed in rmLcn2-pretreated macrophages, where rmLcn2 reduced the survival of the pathogen inside the macrophages. The underlying molecular mechanism of Lcn2 protection was also investigated. We observed that pretreatment of the Raw- 264.7 macrophages with rmLcn2 markedly altered the expression of tonB3, which encodes a component of the transporter for ferrisiderophores in A. baumannii. However, the expression of katG, the gene encoding catalase, remained unaffected. These indicate that Lcn2-mediated defense against the pathogen is related to nutritional immunity rather than reactive oxygen species (ROS) production. Furthermore, the addition of rmLcn2 in infected mice diminished bacterial burden in multiple organs and enhanced the expression of tonB3 in the liver, spleen, and lungs of the infected mice. Increased survival rate due to rmLcn2 treatment declined when the infection model was established using lcn2-defective (lcn2-/-) mice, which indicated the necessity of endogenous Lcn2. Therefore, the antibacterial function of Lcn2 can be exploited to develop an alternative therapeutic agent against A. baumannii.
