        Efficient Multicast Routing on BCube-Based Data Centers

        Multicast group communication has many advantages in data centers and thus is widely used by many applications. It can efficiently reduce the network traffic and improve the application throughput. For the multicast application in data centers, an essential problem is how to find a minimal multicast tree, which has been proved to be NP-hard. In this paper, we propose an approximation tree-building method for the minimal multicast problem, named HD(Hamming Distance)-based multicast tree. Consider that many new network structures have been proposed for data centers. We choose three representative ones, including BCube, FBFLY, and HyperX, whose topological structures can be regarded as the generalized hypercube. Given a multicast group in BCube, the HD-based method can jointly schedule the path from each of receiver to the only sender among multiple disjoint paths; hence, it can quickly construct an efficient multicast tree with the low cost. The experimental results demonstrate that our method consumes less time to construct an efficient multicast tree, while considerably reduces the cost of the multicast tree compared to the representative methods. Our approach for BCube can also be adapted to other generalized hypercube network structures for data centers after minimal modifications.

        Connectivity map identifies luteolin as a treatment option of ischemic stroke by inhibiting MMP9 and activation of the PI3K/ Akt signaling pathway

        This study aimed to explore potential new drugs in the treatment of ischemic stroke by Connectivity Map (CMap) and to determine the role of luteolin on ischemic stroke according to its effects on matrix metalloproteinase-9 (MMP9) and PI3K/Akt signaling pathway. Based on published gene expression data, differentially expressed genes were obtained by microarray analysis. Potential compounds for ischemic stroke therapy were obtained by CMap analysis. Cytoscape and gene set enrichment analysis (GSEA) were used to discover signaling pathways connected to ischemic stroke. Cell apoptosis and viability were, respectively, evaluated by flow cytometry and an MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5- diphenyl-2-H-tetrazolium bromide) assay. Quantitative real-time polymerase chain reaction (qRT-PCR) and western blot analysis were used to test the expression of MMP9 and the PI3K/Akt signaling pathway-related proteins in human brain microvascular endothelial cells (HBMECs) and tissues. Additionally, the infarct volume after middle cerebral artery occlusion (MCAO) was determined by a TTC (2,3,5-triphenyltetrazolium chloride) assay. The microarray and CMap analyses identified luteolin as a promising compound for future therapies for ischemic stroke. Cytoscape and GSEA showed that the PI3K/Akt signaling pathway was crucial in ischemic stroke. Cell experiments revealed that luteolin enhanced cell viability and downregulated apoptosis via inhibiting MMP9 and activating the PI3K/Akt signaling pathway. Experiments performed in vivo also demonstrated that luteolin reduced the infarct volume. These results suggest that luteolin has potential in the treatment of ischemic stroke through inhibiting MMP9 and activating PI3K/ Akt signaling pathway.

        Vacuum infiltration transformation of non-heading Chinese cabbage (Brassica rapa L. ssp. chinensis) with the pinII gene and bioassay for diamondback moth resistance

        Non-heading Chinese cabbage (Brassica rapa L. ssp. chinensis) is a popular vegetable in Asian countries. The diamondback moth (DBM), Plutella xylostella (L.), an insect with worldwide distribution, is a main pest of Brassicaceae crops and causes enormous crop losses. Transfer of the anti-insect gene into the plant genome by transgenic technology and subsequent breeding of insect-resistant varieties will be an effective approach to reducing the damage caused by this pest. We have produced transgenic non-heading Chinese cabbage plants expressing the potato proteinase inhibitor II gene (pinII) and tested the pest resistance of these transgenic plants. Non-heading Chinese cabbages grown for 45 days on which buds had formed were used as experimental materials for Agrobacterium-mediated vacuum infiltration transformation. Forty-one resistant plants were selected from 1166 g of seed harvested from the infiltrated plants based on the resistance of the young seedlings to the herbicide Basta. The transgenic traits were further confirmed by the Chlorophenol red test, PCR, and genomic Southern blotting. The results showed that the bar and pinII genes were co-integrated into the resistant plant genome. A bioassay of insect resistance in the second generation of individual lines of the transgenic plants showed that DBM larvae fed on transgenic leaves were severely stunted and had a higher mortality than those fed on the wild-type leaves.

        Screening of serum exosome markers for colorectal cancer based on Boruta and multi-cluster feature selection algorithms

        Background Early and timely diagnosis benefits the prognosis of patients with colorectal cancer (CRC). The purpose of this study was to explore biomarkers with diagnostic ability in colorectal cancer. Objective Boruta and multi-cluster feature selection (MCFS) algorithms were employed to analyze the expression data of serum exosome-sourced microRNAs (miRNAs) in CRC patients in the Gene Expression Omnibus (GEO) database to find candidate feature miRNAs that could distinguish between CRC and normal samples. To identify the feature miRNAs with the highest diagnostic ability, different support vector machine (SVM) classifiers were constructed, and the SVM classifier with the highest F1 score was selected based on IFS curve. To validate the clinical application value of the classifier, serum samples from 32 CRC patients and 19 healthy individuals were collected as external validation sets, and serum exosomes were extracted for quantitative real-time polymerase chain reaction (qRT-PCR) analysis. The data were imported into the model to verify the performance of the model. Result After feature selection by Boruta and MCFS algorithms, the first five candidate miRNAs (miR-21, miR-193b, miR-23a, miR-575, and miR-610) with sufficient ability to distinguish sample types were identified. In a series of classifiers constructed, the SVM classifier composed of the first four feature miRNAs (miR-21, miR-193b, miR-23a, and miR-575) was determined to have the best classification effect. qRT-PCR results of serum exosome miRNAs in clinical samples demonstrated that the expression of miR-21, miR-193b, and miR-23a in serum exosomes from CRC patients was significantly higher than that in normal samples, while that of miR-575 was significantly lower than that in normal samples. Subsequently, the receiver operating characteristics (ROC) curve of the diagnostic model based on four feature miRNAs was plotted. According to the results, the area under concentrations curves (AUC) value of the diagnostic model was 0.854, which suggested that the predictive performance of the model built on miR-21, miR-193b, miR-23a, and miR-575 was effective enough to distinguish healthy subjects from CRC patients. In addition, the expression of miR-21, miR-193b, miR-23a, and miR-575 was closely related to the tumor size, stage, and the presence of distant metastasis in CRC patients. Conclusion MiR-21, miR-193b, miR-23a, and miR-575 may be a new potential biomarker combination for the diagnosis of CRC. Clinical biopsy combined with these miRNA biomarkers is expected to promote the early diagnosis of CRC, thus optimizing the prognosis of CRC patients.

        Sensorless initial position estimation strategy for PMa‑SynRM drives based on dual rotating high‑frequency signal injection

        The rotating high-frequency (RHF) signal injection method is widely adopted for the initial position estimation in sensorless permanent magnet-assisted synchronous reluctance motor (PMa-SynRM) drives due to its simple implementation. However, the position estimation accuracy of the conventional method may be seriously deteriorated by the dead-zone effects of the inverter and the cross-saturation effects of the machine. Thus, to improve initial position estimation accuracy, a strategy to apply dual RHF signals is proposed in this paper. Different from the traditional solution, the proposed method sequentially injects dual RHF carrier voltages with the same frequencies but different amplitudes into the machine. Objective equations that consider the dead-zone and cross-coupling effects can be extracted from high-frequency response currents in turn. After the simple calculation, the initial position can be obtained directly and accurately, which effectively minimizes the influences of machine cross-saturation and inverter nonlinearities. The influence of the system delay on the rotor position estimation is also analyzed and deduced in detail. Finally, experimental results on a 5.5-kW PMa-SynRM indicate that the proposed method can achieve precise initial position estimation under any rotation position.

        Glucocorticoid-induced expansion of classical monocytes contributes to bone loss

        Classical monocytes are commonly involved in the innate inflammatory response and are the progenitors of osteoclasts. Excess endogenous glucocorticoids (GCs) can increase the levels of classical monocytes in blood and bone marrow. The role of this cell population in high-dose exogenous GC-induced osteoporosis (GIOP) remains to be elucidated. In this study, GIOP was established in rats and mice by daily methylprednisolone injection, and monocyte subsets were analyzed by flow cytometry. We demonstrated that classical monocytes accumulate in bone marrow during GIOP. Similarly, the monocyte proportion among bone marrow nucleated cells was also increased in patients with steroid treatment history. We sorted classical monocytes and analyzed their transcriptional profile in response to GCs by RNA sequencing. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis showed that classical monocytes isolated from GC-treated rats exhibited osteoclast differentiation potential. Deletion of classical monocytes by clodronate liposome treatment prevented GIOP via inhibition of osteoclastogenesis and restoration of CD31HiendomucinHi vessels. Regarding the molecular mechanism, classical monocytes express high levels of glucocorticoid receptors. In vitro treatment with GCs increased both the percentage and absolute number of monocytes and promoted their proliferation. In summary, classical monocytes mediated GC-induced bone loss and are a potential target for therapeutic intervention in GIOP treatment.

      • Molecular Cloning and Expression of Grass Carp MyoD in Yeast Pichia pastoris

        MyoD, expressed in skeletal muscle lineages of vertebrate embryo, is one of muscle-specific basic helix-loop-helix (bHLH) transcription factors, which plays a key role in the determination and differentiation of all skeletal muscle lineages. In this study, a cDNA of grass carp MyoD was cloned and characterized from total RNA of grass carp embryos by RT-PCR. The full-length cDNA of grass carp MyoD is 1597 bp. The cDNA sequence analysis reveals an open reading frame of 825 bp coding for a protein of 275 amino acids, which includes a bHLH domain composed of basic domain (1-84th amino acids) and HLH domain (98-142th amino acids), without signal peptide. Then the MyoD cDNA of grass carp was cloned to yeast expression vector pPICZ$\alpha$A and transformed into P. pastoris GS115 strain, the recombinant MyoD protein with a molecular weight of about 31KD was obtained after inducing for 2d with 0.5% methanol in pH 8.0 BMGY medium, and the maximum yield was about 250 mg/L in shaking-flask fermentation. The results were expected to benefit for further studies on the crystal structure and physiological function of fish MyoD.

      • Color Theorems, Chiral Domain Topology, and Magnetic Properties of Fe<sub><i>x</i></sub>TaS<sub>2</sub>

        <P>Common mathematical theories can have profound applications in understanding real materials. The intrinsic connection between aperiodic orders observed in the Fibonacci sequence, Penrose tiling, and quasicrystals is a well-known example. Another example is the self-similarity in fractals and dendrites. From transmission electron microscopy experiments, we found that FexTaS2 crystals with x = 1/4 and 1/3 exhibit complicated antiphase and chiral domain structures related to ordering of intercalated Fe ions with 2a x 2a and root 3a x root 3a superstructures, respectively. These complex domain patterns are found to be deeply related with the four color theorem, stating that four colors are sufficient to identify the countries on a planar map with proper coloring and its variations for two-step proper coloring. Furthermore, the domain topology is closely relevant to their magnetic properties. Our discovery unveils the importance of understanding the global topology of domain configurations in functional materials.</P>

      • Study on the Influence of UHVDC Dynamic Response on Recovery Characteristic of Receiving End Power Grid

        Aiming at the three UHVDC controller model in the electromechanical simulation software BPA which widely used in China, the structural features were analyzed. By the time-domain simulation, the influence of different simulation models and the key controller parameters on the recovery characteristics of the receiving end power system were studied. Simulation results show that the commutation failure simulated method impacts the recovery characteristic significantly for weak power grids, increasing the voltage measurement time constant of constant dc power control module and enhancing the start voltage threshold of voltage dependent current order limit (VDCOL) can decrease the reactive power consumption of inverter during the disturbance, and can help to improve the recovery characteristic of receiving end power grid.
