        Znf45l affects primitive hematopoiesis by regulating transforming growth factor-β signaling

        Znf45l, containing classical C2H2 domains, is a novel member of Zinc finger proteins in zebrafish. In vertebrates, TGF-βsignaling plays a critical role in hematopoiesis. Here, we showed that Znf45l is expressed both maternally and zygotically throughout early development. Znf45l-depleted Zebrafish embryos display shorter tails and necrosis with reduced expression of hematopoietic maker genes. Furthermore, we revealed that znf45l locates downstream of TGF-β ligands and maintains normal level of TGF-β receptor type II phosphorylation. In brief, our results indicate that znf45l affects initial hematopoietic development through regulation of TGF-β signaling. [BMB Reports 2014; 47(1): 21-26]

        The Wnt signaling pathway is well known to play major roles in skeletal development and homeostasis. In certain aspects, fracture repair mimics the process of bone embryonic development. Thus, the importance of Wnt signaling in fracture healing has become more apparent in recent years. Here, we summarize recent research progress in the area, which may be conducive to the development of Wnt-based therapeutic strategies for bone repair.

      • Facile Synthesis of Microsphere Copper Cobalt Carbonate Hydroxides Electrode for Asymmetric Supercapacitor

        Porous microspheres copper cobalt carbonate hydroxides (Cu<SUB>x</SUB>Co<SUB>2@?x</SUB>CH) pseudocapacitive electrode material comprised of nanoplates via a facile hydrothermal method were presented. Significantly, the crystalline structure, morphology and electrochemical performance of the Cu<SUB>x</SUB>Co<SUB>2@?x</SUB>CH can be readily manipulated by varying the Cu/Co molar ratios. Among various stoichiometries of Cu<SUB>x</SUB>Co<SUB>2@?x</SUB>CH porous microspheres studied, Cu<SUB>1.79</SUB>Co<SUB>0.21</SUB>CH consisted of nanoplates with a mean thickness of @?35nm showed a high specific capacitance of 789Fg<SUP>@?1</SUP> at a current destiny of 1Ag<SUP>@?1</SUP> and good rate ability. Furthermore, the optimized Cu<SUB>1.79</SUB>Co<SUB>0.21</SUB>CH electrode also exhibited remarkable high cycling stability, ca 77.5% after 3000 charge/discharge cycles at current density of 5Ag<SUP>@?1</SUP>. An asymmetric device was constructed from optimized Cu<SUB>1.79</SUB>Co<SUB>0.21</SUB>CH materials on nickel foam (NF) as cathode electrode and graphene on NF as anode electrode in a 6M KOH electrolyte. The asymmetric Cu<SUB>1.79</SUB>Co<SUB>0.21</SUB>CH/NF//graphene/NF supercapacitor device delivered a specific capacitance of 60.5Fg<SUP>@?1</SUP> in a potential range of 0@?1.6V. A high energy density of 21.5Whkg<SUP>@?1</SUP> was achieved at the power density of 200Wkg<SUP>@?1</SUP>. More significantly, the designed device exhibited excellent cycling stability with 73.3% capacity retention after 5000 cycles.

        Effects of Hardener Type on Cure Behavior and Physical Properties of TMP Modified UF Resin and Its Composites with Bamboo Powder

        In this study, effects of mixed hardeners (ammonium chloride mixed with ammonium persulfate, oxalate and phosphoric acid) on the properties of 2,4,6-trimethylolphenate modified UF resin (TMPUF) were studied, and the potlife, gel time and curing temperature of TMPUF/hardeners were characterized. The results indicated that the hardeners used have critical influence on the pH value of TMPUF and some proton donor can be used to lower the pH value of TMPUF with low formaldehyde content. Besides, bamboo particleboards were made with TMPUF/hardeners, and tensile properties, water absorption ratio and the swelling ratio of bamboo particleboards were characterized. The study demonstrated that the tensile strength of bamboo particleboards increased when partial ammonium chloride was replaced by ammonium persulfate to cure the TMPUF adhesives in our experiment.

        Chinese herbal injections in combination with radiotherapy for advanced pancreatic cancer: A systematic review and network meta-analysis

        Background: Advanced pancreatic cancer (APC) is a fatal disease with limited treatment options. This study aims to evaluate the effectiveness and safety of different Chinese herbal injections (CHIs) as adjuvants for radiotherapy (RT) in APC and compare their treatment potentials using network meta-analysis. Methods: We systematically searched three English and four Chinese databases for randomized controlled trials (RCTs) from inception to July 25, 2023. The primary outcome was the objective response rate (ORR). Secondary outcomes included Karnofsky performance status (KPS) score, overall survival (OS), and adverse events (AEs). The treatment potentials of different CHIs were ranked using the surface under the cumulative ranking curve (SUCRA). The Cochrane RoB 2 tool and CINeMA were used for quality assessment and evidence grading. Results: Eighteen RCTs involving 1199 patients were included. Five CHIs were evaluated. Compound Kushen injection (CKI) combined with RT significantly improved ORR compared to RT alone (RR 1.49, 95 % CrI 1.21–1.86). Kanglaite (KLT) plus RT (RR 1.58, 95 % CrI 1.20–2.16) and CKI plus RT (RR 1.49, 95 % CrI 1.16–1.95) were associated with improved KPS score compared to radiation monotherapy, with KLT+RT being the highest rank (SUCRA 72.28 %). Regarding AEs, CKI plus RT was the most favorable in reducing the incidence of leukopenia (SUCRA 90.37 %) and nausea/vomiting (SUCRA 85.79 %). Conclusions: CKI may be the optimal choice of CHIs to combine with RT for APC as it may improve clinical response, quality of life, and reduce AEs. High-quality trials are necessary to establish a robust body of evidence. Protocol registration: PROSPERO, CRD42023396828.

        Identification and sex expression profiles of olfactory-related genes in Mythimna loreyi based on antennal transcriptome analysis

        To better understand the olfactory mechanism of Mythimna loreyi, a worldwide migratory pest, we for the first time conducted a large scale identification of olfactory-related genes and investigation of their sex expression profiles by transcriptomic analysis. A total of 42,832 unigenes were obtained by transcriptome sequencing, as sembly and annotation, with an average length of 1,229 bp and N50 of 2,086 bp. In particular, 138 olfactoryrelated genes were identified by homologous blasting, including 33 odorant binding proteins (OBPs), 16 che mosensory proteins (CSPs), 63 odorant receptors (ORs), 24 ionotropic receptors (IRs) and two sensory neuron membrane proteins (SNMPs). Further, by using differential gene expression (DGE) and fragments per kilobase per million fragments (FPKM) values to compare the transcript levels between female and male antennae, we found that 22 olfactory-related genes (9 OBPs, one CSP and 12 ORs) were sex biased, with 10 genes being male biased and 12 genes female biased. In addition, sex and tissue expression profiles determined by qPCR of 15 selected genes confirmed the reliability of sex expression profiles obtained by the transcriptomic analysis, and demonstrated that most olfactory-related genes were specifically or primarily expressed in antennae, suggesting their roles in olfaction, while a few genes were highly expressed in other tissues, implying their non-olfaction functions. This study provides an important basis for further functional study of olfactory genes in M. loreyi.

        Novel Variants in the FIG4 Gene Associated With Chinese Sporadic Amyotrophic Lateral Sclerosis With Slow Progression

        Background and Purpose Mutations in the FIG4 gene have been linked to amyotrophic lateral sclerosis (ALS) type 11 in Caucasian populations. The purpose of this study was to identify FIG4 variants in a cohort of 15 familial ALS (FALS) indexes and 275 sporadic ALS (SALS) patients of Han Chinese origin. Methods All 23 exons of FIG4 were sequenced using targeted next-generation sequencing. An extensive literature review was performed to detect genotype-phenotype associations of FIG4 mutations. Results No FIG4 variants were identified in the FALS patients. One novel heterozygous missense variant (c.352G>T [p.D118Y]) and one novel heterozygous nonsense variant (c.2158G>T [p.E720X]) in FIG4 were identified in two SALS patients. The p.E720X variant is interpreted as likely pathogenic while the p.D118Y variant is a variant of uncertain significance. The patient carrying the p.E720X mutation developed lower-limb-onset slowly progressive ALS, and survived for 11.5 years. The patient harboring the FIG4 p.D118Y variant also presented with progressive ALS, with the score on the ALS Functional Rating Scale–Revised (ALSFRS-R) decreasing by 0.4 per month. The rate of decrease in the ALSFRS-R scores from symptom onset to diagnosis seemed to be lower in the patients carrying FIG4 variants than the no-FIG4-mutation ALS patients in this study. Conclusions Our findings suggest that ALS patients carrying FIG4 mutations are not common in the Chinese population and are more likely to exhibit slow progression.

        Serum Retinol-Binding Protein Levels Are Associated with Nonalcoholic Fatty Liver Disease in Chinese Patients with Type 2 Diabetes Mellitus: A Real-World Study

        Background: The association of serum retinol-binding protein (RBP) levels with nonalcoholic fatty liver disease (NAFLD) remains controversial. Furthermore, few studies have investigated their relationship in type 2 diabetes mellitus (T2DM) patients. Therefore, the aim of the present study was to explore the association between serum RBP levels and NAFLD in Chinese inpatients with T2DM.Methods: This cross-sectional, real-world study included 2,263 Chinese T2DM inpatients. NAFLD was diagnosed by abdominal ultrasonography. The subjects were divided into four groups based on RBP quartiles, and clinical characteristics were compared among the four groups. The associations of both RBP levels and quartiles with the presence of NAFLD were also analyzed.Results: After adjustment for sex, age, and diabetes duration, there was a significant increase in the prevalence of NAFLD from the lowest to the highest RBP quartiles (30.4%, 40.0%, 42.4%, and 44.7% for the first, second, third, and fourth quartiles, respectively, P<0.001 for trend). Fully adjusted multiple logistic regression analysis revealed that both increased RBP levels (odds ratio, 1.155; 95% confidence interval, 1.012 to 1.318; P=0.033) and quartiles (P=0.014 for trend) were independently associated with the presence of NAFLD in T2DM patients.Conclusion: Increased serum RBP levels were independently associated with the presence of NAFLD in Chinese T2DM inpatients. Serum RBP levels may be used as one of the indicators to assess the risk of NAFLD in T2DM patients.
