<P>An immunochromatographic assay for the detection of hepatitis B surface antigen (HBsAg) was developed using Fe<SUB>3</SUB>O<SUB>4</SUB> nanoparticles as signal materials. Anti-HBsAg antibodies were labeled with carboxylated Fe<SUB>3</SUB>O<SUB>4</SUB> particles with an average diameter of 153.6 nm, and an immunochromatography strip was constructed. The detection capability of the assay developed here is 1 ng/mL HBsAg.</P>
Regulation of MicroRNA-Mediated Developmental Changes by the SWR1 Chromatin Remodeling Complex

<P>The ATP-dependent SWR1 chromatin remodeling complex (SWR1-C) exchanges the histone H2A-H2B dimer with the H2A.Z-H2B dimer, producing variant nucleosomes. Arabidopsis thaliana SWR1-C contributes to the active transcription of many genes, but also to the repression of genes that respond to environmental and developmental stimuli. Unlike other higher eukaryotic H2A.Z deposition mutants (which are embryonically lethal), Arabidopsis SWR1-C component mutants, including arp6, survive and display a pleiotropic developmental phenotype. However, the molecular mechanisms of early flowering, leaf serration, and the production of extra petals in arp6 have not been completely elucidated. We report here that SWR1-C is required for miRNA-mediated developmental control via transcriptional regulation. In the mutants of the components of SWR1-C such as arp6, sef, and pie1, miR156 and miR164 levels are reduced at the transcriptional level, which results in the accumulation of target mRNAs and associated morphological changes. Sequencing of small RNA libraries confirmed that many miRNAs including miR156 decreased in arp6, though some miRNAs increased. The arp6 mutation suppresses the accumulation of not only unprocessed primary miRNAs, but also miRNA-regulated mRNAs in miRNA processing mutants, hyl1 and serrate, which suggests that arp6 has a transcriptional effect on both miRNAs and their targets. We consistently detected that the arp6 mutant exhibits increased nucleosome occupancy at the tested MIR gene promoters, indicating that SWR1-C contributes to transcriptional activation via nucleosome dynamics. Our findings suggest that SWR1-C contributes to the fine control of plant development by generating a balance between miRNAs and target mRNAs at the transcriptional level.</P>
Electrochemical Immunosensor Using a Gas Diffusion Layer as an Immobilization Matrix

The modification of a gas diffusion layer (GDL), a vital component in polymer electrolyte fuel cells, is described here for use in the electrochemical detection of antibody-antigen biosensors. Compared to other substrates (gold foil and graphite), mouse anti-rHBsAg monoclonal antibody immobilized on gold-coated GDL (G-GDL) detected analytes of goat anti-mouse IgG antibody-ALP using a relatively low potential (−0.0021 V vs. Ag/AgCl 3 M NaCl), indicating that undesired by-reactions during electrochemical sensing should be avoided with G-GDL. The dependency of the signal against the concentration of analytes was observed, demonstrating the possibility of quantitative electrochemical biosensors based on G-GDL substrates. When a sandwich method was employed, target antigens of rHBsAg with a concentration as low as 500 ng/mL were clearly measured. The detection limit of rHBsAg was significantly improved to 10 ng/mL when higher concentrations of the 4-aminophenylphosphate monosodium salt (APP) acting on substrates were used for generating a redox-active product. Additionally, it was shown that a BSA blocking layer was essential in improving the detection limit in the G-GDL biosensor.
데이팅 앱의 사용 동기의 변화와 그에 따른 사용 양상의 변화 - Tinder 국내 사용자 경험을 중심으로

최근 데이팅 앱은 연애 상대를 찾기 위한 목적에서 나아가 다양한 관계의 만남을 지원하는 개념으로 확장되었다. 하지만 대다수의 기존 연구에서는 데이팅 앱 사용의 다양한 동기 양상에 대해 다루기 보다는, 성적이고 단기적인 관계를 찾는 앱으로서만 다뤄져 왔다. 따라서 본 연구는 국내의 Tinder(틴더) 사용자를 대상으로 이들의 다층적인 틴더 사용 동기에 초점을 추고 그에 따른 행동 양상 및 전략의 변화를 반구조적 인터뷰를 통해 탐구하였다. 연구 결과 대다수의 사용자들이 틴더 사용 초기 특정한 관계에 대한 동기를 가지고 시작하기 보다는, 오락과 같은 비관계적 동기를 더 크게 가지는 것으로 나타났다. 하지만 대다수의 사용자들은 틴더를 지속적으로 사용함에 따라, 관계적 동기를 동시에 추구하는 경향을 보였으나, 자신이 추구하는 관계의 유형에 대해 명확한 경계를 짓지 않으려는 태도가 나타났다. 위와 같은 연구 결과에 기반하여, 데이팅 앱 디자인의 개선방안에 대해 논의한다.
