      • Archaebacterium Thermococcus profundus DT5432가 생성하는 고도호열성 Pullulanase의 특성

        A hyperthermophilic Thermococcus profundus DT5432, anaerobic archaebacterium which was isolated from a deep-sea hydrothermal vent system(depth, 1,395m), metabolized starch at 80℃, and produced an active and thermostable extracellular pullulanase. Optimum condition for thermophilic pullulanase production was found to be at 80℃, pH 7.0 within 12hr incubation. The three bands of pullulanase activity were visible by activity staining on native-polyacrylamide gel electrophoresis. The pullulanase exhibited maximal activity at pH 7.0∼7.5 and 90℃, and was stable in the range of pH 5.0∼10.0 and up to 80℃ for 5hr. Thermostability of the enzyme was enhanced in the presence of 5mM Ca^(2+) at temperature above 90℃. The pullulanase activity increased in the presence of Ca^(2+), which acts as an activator and stabilizer in case of aerobic enzymes. The enzyme was resistant to various chemical reagents, particularly. detergents such as SDS. tween 40, 60, triton X-100 and urea. The pullulanase displayed a relative substrate secificity for hydrolysis of pullulan(100%) versus 30% for amylopectin, 17% for glycogen and 12% for starch.

      • 영지버섯 단백다당체 GLB-A의 UA에 대한 보호효과

        In the previous study we described the antitumor and immunomodulatory activities of GLB-A. an acidic protein-polysaccharide fraction of the growing tips of Ganoderma lucidum. In this study we used flow cytometry to elucidate the protective effect of GLB-A on the splenocytes and thymocytes of BALB/c mouse exposed to ultraviolet irradiation. GLB-A increased the thymocytes of BALB/c mouse exposed to ultraviolet irradiation. GLB-A increased the viability of the UV-irradiated splenocytes in a dose-dependent pattern (17.1% increase at 200 ㎍/㎖) and its protective effect was remarkable only during UV irradiation. When the UV absorption spectrum was analyzed, GLB-A showed a maximal absorbance at 257 nm. Based on these observations, the protective effect of GLB-A against UV radiation seemed to originate at least partly from its UV-absorbing property.

      • 韓國産 擔子菌類의 抗菌成分에 관한 硏究 (II) : 10種의 擔子菌에 대한 抗菌成分 檢索 A Screening Test for the Antibiotic Activities of Ten Basidiomycetous Fungi

        The ether, methanol, and physiological saline extracts of the dried carpophores of ten basidiomycetous fungi, which were collected in the conifer woods of Chungnam National University at Taechon and Keyryoung Mountain Area in Chungcheungnamdo, were subjected to an antibiotic activity test by disc-plate method. Among them, the methanol extracts of Inocybe trechispora, Lactarius velumis, and Laccaria laccata and the ether extracts of I.trechispora showed antibiotic activities against Sarcina lutea; and the methanol extracts of I. trechispora and L. velumis and physiological saline extract of Coriolus versicolor showed antibiotic activities against Staphylococcus aureus; and the methanol extract of L. velumis showed antibiotic activity against Escherichia coli. None of the extracts, however, showed any activity against Serratia marcescens and Proteusvulgaris.

      • 건강식품 바이탈 및 비지놀의 抗疲勞 效果에 관한 연구

        As a result of extensive research efforts on the oriental medicinal herbs, some of their interesting biological activities have been elucidated. In the present study, antifatigue effects of two natural food supplements consisting of oriental herbs, Vital and Vigilor, were investigated using the ICR mice. The mice were fed with antibiotic-free normal mouse feed containing Vital or Vigilor for 14,28, or 31 days and then subjected to forced swim tests. In a test carried out in melting-ice water, Vital and Vigilor significantly increased the exhaustion times of the 28 day-fed mice by 26.9 % (p < 0.01) and 16.2% (p < 0.05), respectively. However, in the other test performed at 15±0.5℃ using 31 day-fed mice, Vigilor failed to increase the exhaustion time significantly (at p< 0.05), while Vital showed antifatigue effect by increasing the swimming time by 29.9% (p<0.01). Neither of Vital nor Vigilor exerted antifatigue effects in a swim test carried out on day 14 after the initiation of feeding. These results suggest that Vital and Vigilor exhert antifatigue effets when administered for a sufficient period of time by enhancing the health of the host animals.

        As an investigation to develop novel antitumor immunotherapeutic agents, four protein-poly-saccharide fractions from Korean wild mushrooms were subjected to a flow cytometric analysis for their in vitro lymphoblastogenic activities on the BALB/c mouse splenic lymphocytes. Of these, the protein-poly-saccharide fractions of Russula alboareolata, Russula compacta and Laccaria amethystea not only showed lymphoblastogenic activity, but also increased the viability of the lymphocytes, while that of Russula nitrata exerted marked cytocidal activity.

      • 인삼사포닌의 마우스 비장 임파구에 대한 면역활성

        To ellucidate immunologic activities of ginseng saponin, the splenic leukocytes of BALB/c mice were incubated in RPMI medium for 48hr with or without ginseng total saponin(GTS), and then analyzed flow cytometrically. Upto the concentration of 100㎍/㎖, GTS exherted stimulatory effect on the immune cells most prominently on B cells: the percentage of the surface Immunoglobulin (sIg) -positive cells (B cells) of the control was 47.l%, while that of GTS treated was 58.9%. The percentages of monoclonal antibody(mAb) J1j-reactive cells (T cells) were 28.8% and 3l.3%, those of mAb Gkl.5-reactive cells (CD4' T cells) were l8.5% and l9.l%, and those of mAb M1/70-reactive cells (mostly macrophages) were 4.4 and 5.0% at the concentration of 0 and 100㎍/㎖, respectively. However, at the concentration of 300㎍/㎖ or higher, GTS was found to be toxic to the immune cells especially to B cells in that the flow rate of the cells (gated viable cells) significantly decreased and the percentage of B cells droped from 58.9% to 31.2% and 25.4% at l00, 300, and 1000㎍/㎖, respectivelly. These data strongly suggests that GTS would exhert B-cell mitogen-like activity at physiological concentrations of 100㎍/㎖ or lower.

        The ether, methanol, and physiological saline extracts of the dried carpophores of ten basidiomycetous fungi, which were collected in the conifer woods of Chungnam National University at Taechon, were subjected to an antibiotic activity test by disc-plate method. Among them, the methanol extracts of Marasmius maximus, Hirschioporus abietinus, and Russula sanguinea and the the ether extracts of M. maximus and H. abietinus showed antibiotic activities against Sarcina lutea; and the methanol and ethanol extract of M. maximus showed antibiotic activity against Staphylococcus aureus. None of the extracts, however, showed any activity against gram negative bacteria, Serratia marcescens and Proteus vulgaris.

      • Phellinus linteus 균사 배양물로부터 분리한 단백다당체 Kp의 항암활성

        A protein-polysaccaride fraction Kp(53.9% polysaccharide, 14.2% protein) was separated from the shake-cultured mycelia of a basidiomycetous fungus, Phellinus linteus, and its antitumor activity against sarcoma 180 in ICR mice was investigated. When administered after the tumor implantation. Kp exerted antitumor activity by inhibiting the growth of the sarcoma 180 solid tumor by 71.5% and increasing the life span of the sarcoma 180 ascitic mice by 51.5% at 100 ㎎/㎏. In pretreatment tests, in which Kp was administered once daily for 9 days before the tumor implantation. Kp inhibited the growth of the solid and ascites form of sarcoma 180, respectively, by 35.4% and by 80.3% at 100 ㎎/㎏. However, Kp showed no in vitro cytotoxic activity against a murine leukemia L1210 and a human gastric tumor SNU.1 upto the concentration of 200 ㎍/㎖. From these results, it is clear that the antitumor activity of Kp is exerted through its immunomodulating activity on the host's immune system.

      • Phellinus baumi 상황버섯의 면역증강활성

        Certain mushrooms belonging to the genus Phellinus, such as P. linteus and P. igniarius, have been known for their antitumor and immunopotentiating activities. These mushrooms have collectively been called "Sang-Whang" in oriental medicine. In Korea, Phellinus baume has been cultivated from mid 1990's and are known as "Sang-Whang" mushrooms. However, its antitumor or immunomodulatory activity has never been carried out. In this study, two protein-polysaccharide fractions, one from hot water extract and the other from alkaline extract, were prepared and their stimulatory activities on the in vitro cultured BALB/c mouse splenic leukocytes were investigated using flow cytometric analysis. As a result, the water soluble protein-polysaccharide fraction, PBD, was found to exert stronger activities than the alkali-soluble protein-polysaccharide fraction, PMA, in in vitro lymphoblastogenic assay. PBD also showed in vitro stimulatory activity on T lymphocytes in that it enhanced the level of CD25 expression of the in vitro cultured splenic leukocytes.

        The ether, methanol, and physiological saline extracts of the dried carpophores of ten basidiomycetes, which were collected from several locations of Korea, were subjected to a screening test for antibiotic activity by disc-plate method using Sarcina lutea, Bacillus subtilis, Escherichia coli, and Serratia marcescens as target organisms. As the result, the methanol extract of five basidiomycetes-Amanita rebescens, Amanita solitaria, Lactarius vellereus, Latarius subzonarius and Scleroderma verrucosum-were found te have antibiotic activity. Among them, the methanol extracts of A. rubescens and L. vellereus exerted strong activity against both of the gram positive and negative bacteria. The ether extracts of A. rubescens and S. verrucosum also showed activity against S. lutea. The physiological saline extract, however, showed no activity at all.
