        Reaction of Mast Cells and Goblet Cells in the Small Intestine of C57BL/6 and C3H/HeN Mice Infected with Echinostoma hortense

        Mast cells and goblet cells have been known to protect the host against parasites. In this study, we examined the response of the mast cells and goblet cells over a period of 6 weeks in the duodenum, jejunum and ileum of C3H/HeN and C57BL/6 mice infected with Echinostoma hortense (E. hortense). In addition, we investigated whether the worm recovery rate of uninfected mice (the control group) or E. hortense-infected mice (the experimental group) was associated with the number of mast cells and goblet cells. The worm recovery rate was higher in the C3H/HeN mice than in the C57BL/6 mice. The number of goblet cells significantly increased in the experimental group of the C3H/HeN and C57BL/6 mice compared with the control group of both strains (P<0.005). Worm recovery peaked 3 weeks after the infection of the C57BL/6 mice and at 2 weeks after the infection of the C3H/HeN mice, and it was higher in the duodenum than in the jejunum and ileum. However, the infected site in the intestine had no relation with worm expulsion. In the C3H/HeN and C57BL/6 mice, the number of goblet cells in the experimental group was significantly higher than that in the control group (P<0.005). The number reached a peak 2 weeks after the infection and it even increased in duodenum, jejunum and ileum. The increased number of goblet cells was retained 6 weeks after infection. The number of goblet cells was higher in the C3H/HeN mice than in the C57BL/6 mice (P<0.01). These results indicate that goblet cells are related with the worm expulsion. Furthermore, immunohistostaining of the antral intestinal walls for lectin showed the significant increase of the number of goblet cells in the experimental group (P<0.001). The high infection rate in the duodenum was found during the early infection. An increased infection rate in the jejunum and ileum was found 3 weeks after infection and the infection rate was higher in the C3H/HeN mice than in the C57BL/6 mice. Taken together, the present study indicates that goblet cells, rather than mast cells, may play critical roles in parasite expulsion.

        An extremely rare case of prenatally diagnosed absent both aortic and pulmonary valves

        We describe a case of absent aortic and pulmonary valves, diagnosed at 16.4 weeks of gestation. Fetal echocardiography showed cardiomegaly with dilated both ventricles. No valve leaflets were observed in the aorta and pulmonary artery, and a typical to-and-fro flow pattern was noted in both great arteries on color Doppler imaging. Fetal hydrops was also detected. Follow-up ultrasonographic evaluation at 19 weeks demonstrated intrauterine fetal death. Postmortem autopsy revealed the absence of both aortic and pulmonary valve leaflets. To the best of our knowledge, this is the earliest diagnosed case of absent both aortic and pulmonary valves and only the second case to be diagnosed prenatally.

        A numerical study has been carried out on the optimization of an air cleaning system which was installed in a heating, ventilation and air conditioning system (HVAC) system of subway station for particle removal. Required particle removal efficiencies of three different air cleaning systems were calculated from ventilation rate, and indoor/outdoor concentration of PM10. Mass balance equations of PM10 were used to solve the required particle removal efficiencies. Fibrous filter was considered as an air cleaning system. Calculations were carried out about two different places which were waiting area and platform of subway station, respectively. This study proposed optimized design and operation condition of each air cleaning system.

        Mirror Syndrome Resulting from Metastatic Congenital Neuroblastoma to Placenta

        Congenital neuroblastoma is a rare disease. Placental metastasis is extremely rare and poor prognosis has been reported in neonates. Mirror syndrome could occur in mother with placental metastasis with possibilities of hypertension and edema. We report a case of detection of left suprarenal mass in fetus at 31<sup>+5</sup> weeks’ gestation. Mother presented with palpitation, edema, headache, and hypertension. Maternal 24 hours urine vanillylmandelic acid (VMA) and normetanephrine (NME) level at 34 weeks’ gestation were elevated. Consequently, emergent cesarean section was done. Based on abdominal ultrasonography and whole body magnetic resonance imaging, left adrenal tumor with liver metastasis was suspected. Neuroblastoma was confirmed by liver and placenta biopsy. Chemotherapy was started with Pediatric Oncology Group 9243 at day 7 and changed into Children's Oncology Group 3961 due to cholestasis and poor response during 2nd cycle. Plasma exchange was done for aggravated direct hyperbilirubinemia. The baby expired at 73 days due to multi-organ failure. Maternal symptoms were completely resolved in 2 weeks after delivery along with normalization of the elevated level of VMA and NME. We report a first case of mirror syndrome in Korean mother and fetus resulting from metastatic congenital neuroblastoma to placenta.

        Purification and Characterization of the Lipase from Acinetobacter sp. B2

        Industrial development has increase consumption of crude oil and environmental pollution. A large number of microbial lipolytic enzymes have been identified and characterized to date. To development for a new lipase with catalytic activity in degradation of crude oil as a microbial enzyme, Acinetobactor sp. B2 was isolated from soil samples that were contaminated with oil in Daejon area. Acinetobactor sp. B2 showed high resistance up to 10 mg/mL unit to heavy metals such as Ba, Li, Al, Cr, Pb and Mn. Optimal growth condition of Acinetobactor sp. B2 was confirmed $30^{\circ}C$. Lipase was purified from the supernatant by Acinetobactor sp. B2. Its molecular mass was determined to the 60 kDa and the optimal activity was shown at $40^{\circ}C$ and pH 10. The activation energies for the hydrolysis of p-nitrophenyl palmitate were determined to be 2.7 kcal/mol in the temperature range 4 to $37^{\circ}C$. The enzyme was unstable at temperatures higher than $60^{\circ}C$. The Michaelis constant $(K_{m})\;and\;V_{max}$ for p-nitrophenyl palmitate were $21.8{\mu}M\;and\;270.3{\mu}M\;min^{-1}mg\;of\;protein^{-1}$, respectively. The enzyme was strongly inhibited by $Cd{2+},\;Co^{2+},\;Fe^{2+},\;Hg^{2+},\;EDTA$, 2-Mercaptoethalol. From these results, we suggested that lipase purified from Acinetobactor sp. B2 should be able to be used as a new enzyme for degradation of crude oil, one of the environmental contaminants.

      • Expression of Soybean Nodule-specific Uricase (Nodulin-35) Gene in the Fission Yeast Schizosaccharomyces pombe

        대두와 유사한 ureide대사를 갖는 분열효모 S.pombe에 뿌리혹 특이성 uricase를 만드는 Nodulin-35 유전자의 1.1kb 의 작동역을 pART2 벡터에 크로닝하였다. 이 재조합 프라스미드의 S.pombe 형질전환체로부터 CAT 합성을 측정한 결과 뿌리혹에서만 발현되는 Nodulin-35 유전자가 유사한 대사를 갖고 있는 효모에서 항상성(constitutive) 발현을 하였다. Soybean nodulin-35(N-35) promoter region was cloned in Schizosac-charomyces vector, pART2. The chimaeric gene consisted of 1.1 kb of the N-35 promoter region, chloramphenicol acetyl transferase (CAT) and N-23 polyA. After transformation into S. pombe SP223 transformants were selected with auxortophic marker.CAT activity of the transformant was showed. These data indicate that N-35 gene can express in S.pombe.

        지모의 뿌리줄기로부터 분리된 nyasol의 미용효과

        지모 뿌리를 100% EtOH로 추출, 농축하여 medium pressure liquid chromatography 기반의 NO 분획물 활성 검정법을 이용하여 활성을 검정하였다. preparative high performance liquid chromatography를 반복적인 칼럼크로마토그래피를 수행하여 1개의 단일성분을 분리하였다. NMR, MS 등을 포함한 이화학 및 분광학적 자료에 근거하여 분리된 물질을 구조 분석한 결과 (-)-nyasol로 구조동정 하였다. 분리된 활성물질 nyasol은 NO 생성 억제, melanin과 tyrosinase를 억제하는 미백효과, thymus and activation-regulated chemokine 생성을 억제하는 활성에서 6.25-12.5 μg/mL의 IC50 값을 가지는 강한 저해 활성을 나타냈다. 특히 nyasol은 염증에 의한 항아토피 및 항미백활성에 있어 뛰어난 효능을 나타내었다. 이 결과로 보아 nyasol은 천연 기능성 화장품으로써 이용 가능성을 넓힐 수 있을 것으로 사료된다. The rhizomes of Anemarrhena asphodeloide was extracted with 100% ethanol and concentrated subfractions were separated with medium pressure liquid chromatography-based activity profiling. One compound was isolated from the subfraction 10 through the repeated preparative high performance liquid chromatography (prep-HPLC). According to physico-chemical and spectroscopic data including NMR and MS, the chemical structures of the compound was determined as nyasol (1). Nyasol was exhibited potent inhibitory activity for NO (IC50: 12.5 μg/mL), tyrosinase (IC50: 12.5 μg/mL), melanin contents (IC50: 12.5 μg/mL), thymus and activation-regulated chemokine (TARC) production (IC50: 6.25 μg/mL). As a result, nyasol has an excellent inflammation-dependent anti-whitening and TARC production activity. It could be used to a large range of functional cosmetics.

      • Proteomic Characterization of the Outer Membrane Vesicle of <i>Pseudomonas putida</i> KT2440

        <P>Outer membrane vesicles (OMVs) are produced by various pathogenic Gram-negative bacteria such as <I>Escherichia coli</I>, <I>Pseudomonas aeruginosa</I>, and <I>Acinetobacter baumannii</I>. In this study, we isolated OMVs from a representative soil bacterium, <I>Pseudomonas putida</I> KT2440, which has a biodegradative activity toward various aromatic compounds. Proteomic analysis identified the outer membrane proteins (OMPs) OprC, OprD, OprE, OprF, OprH, OprG, and OprW as major components of the OMV of <I>P. putida</I> KT2440. The production of OMVs was dependent on the nutrient availability in the culture media, and the up- or down-regulation of specific OMPs was observed according to the culture conditions. In particular, porins (e.g., benzoate-specific porin, BenF-like porin) and enzymes (e.g., catechol 1,2-dioxygenase, benzoate dioxygenase) for benzoate degradation were uniquely found in OMVs prepared from <I>P. putida</I> KT2440 that were cultured in media containing benzoate as the energy source. OMVs of <I>P. putida</I> KT2440 showed low pathological activity toward cultured cells that originated from human lung cells, which suggests their potential as adjuvants or OMV vaccine carriers.         Pseudomonas sp. G314가 생산하는 생물 계면활성제의 특성

        대전일원의 유류오염 지역의 토양에서 분리된, 생물 계면활성제 생성능이 우수한 Pseudomonas sp. G314균주 [23]가 생산하는 생물 계면활성제의 특성을 조사하고 그 성분을 확인하였다. Pseudomonas sp. G314가 생산하는 생물 계면활성제는 상온에서 10일 보관 후에도 26.2 dyne/㎝ 정도의 표면장력을 유지해 냉장 보관한 계면활성제와 비슷하게 안정하였고, 5 l 발효조를 이용한 배양에서 회분배양의 25 dyne/㎝ 보다는 약간 높은 27 dyne/㎝ 정도의 계면활성제를 생산해 대량 배양 할 수 있음을 확인하였다. 또 이 계면활성제는 acetone과 methanol에 잘용해 되고 benzene과 toluene에 약하게 용해되어 glycolipid 계열의 생물계면활성제임이 추정되었고[23], 이를 silica gel column을 통해 용출하고, TLC로 전개하여 확인된 Rf 0.58인 spot이 bial’s reagent와 rhodamine 6G에서 양성반응을 나타내 Pseudomonas sp. G314가 생산하는 생물 계면활성제는 탄수화물과 지질이 함유된 glycolipid 계열의 생물 계면활성제임을 확인하였다. The purpose of this paper is to analyze the characteristics and chemical components of biosurfactant produced by Pseudomonas sp. G314. Pseudomonas sp. G314 was isolated from soil samples which were contaminated with oil in Daejon area. As such, it produced quality biosurfactant [23]. One type of biosurfactant was kept in a refrigerator, whereas another type of biosurfactant was kept in room temperature. The surface tension activities were then compared. As a result, the biosurfactant from Pseudomonas sp. G314 that was kept at room temperature was stable for 10 days, showing 26.2 dyne/㎝ of surface tension activity. This result was found to be similar to that of the refrigerator storage. The surface tension of batch culture was 25 dyne/㎝, but the culture in the 5 l fermentor was 27 dyne/㎝. Therefore, it can be suggested that the large-scale culture is feasible via the fermentor. Biosurfactant from Pseudomonas sp. G314 was estimated to be a kind of glycolipid because it dissolved in acetone and methanol much better than in benzene and toluene [23]. A spot was detected through the elution of silica gel column and the spread of TLC, and the Rf value was 0.58. This spot has a positive reaction with Bail’s reagent and rhodamine 6G. Hence, we can conclude that biosurfactant from Pseudomons sp. G314 was a glycolipid containing carbohydrate and lipid.
