        일부 한약재의 동종항원에 대한 세포증식 및 살세포반응 억제효과

        본 실험에서는 동양에서 예로부터 민간요법이나 한방에서 주로 많이 쓰여지고 있는 8가지 종류의 한약재에 대해서 면역억제제로써 사용 가능성을 실험하였다. 그 결과 당귀, 산사, 어성초, 오가피, 황기의 추출물은 동종항원에 반응하는 순수분리 T세포의 증식을 농도 의존적으로 억제시켰다. 또한 이들 T세포의 증식에 있어서 필수적인 IL-2를 포함한 cytokine 즉, IL-2, IL-4, IL-10, IFN-γ의 생산량은 대조군에 비해 실험군에서 유의한 차이가 없었고 특히 T세포 증식에 필수적인 IL-2의 생산량의 변화가 거의 없었다. 이는 한약재에 의한 T세포의 증식억제 효과가 T세포증식에 필수적인 IL-2의 생산량을 억제하기 때문에 일어나는 결과가 아님을 알 수 있었다. 그리고 T세포의 살세포작용 억제를 직접적으로 측정하기 위하여 세포내 LDH의 양을 조사한 결과 모든 대조군에서 50%이상의 살세포작용 억제가 일어났고, 그중 특히 오가피와 황기에서는 100% 살세포작용 억제가 일어났다. 따라서 본 실험에 사용된 당귀, 산사, 어성초, 오가피, 황기 등의 5가지 약재가 부작용 없는 면역억제로써 사용 가능성이 높은 것으로 생각된다. In this experiment, we showed the immunosuppressive effects of herbal plant extracts on the alloantigen reactive proliferation and cytotoxicity. The extracts of Angelica gigas, Crataegus pinnatifida, Houttuynia cordata, Acanthopanax sessiliflorum and Astragalus membranaceus markedly suppressed on the pro- liferation of primary T cells stimulated with allogeneic spleen cells in a dose-dependent manner. The production of IL-2, IFN-γ, IL-4 and IL-10 in the alloreactive primary T cells showed no significant difference in the presence or absence of herbal plants extracts. Also the result of mixed lymphocyte reaction (MLR)-induced cytotoxic T lymphocyte (CTL) showed what is above a certain point 50% inhibition. Specially, the extracts of Acanthopanax sessiliflorum and Astragalus membranaceus com- pletely suppressed the killing activity of CTL. Theses results suggest that the extracts of 5 herbal plants can be used as immunosuppressive agents.

        8주간의 유산소성 체조활동을 통한 뇌병변 장애인의 체력 및 염증반응지표 변화

        본 연구의 목적은 뇌병변 장애인을 대상으로 8주간의 유산소성 체조활동을 실시하여 신체조성, 체력 및 염증반응지표의 변화에 대하여 알아보는 것이다. 본 연구의 피험자는남·녀 뇌병변장애 중 대표적인 뇌졸중 장애인 17명을 대상으로 시작되었지만 9명이 중도포기하여 최종 8명이 참가하였다. 운동 프로그램 참여 전 사전 검사를 실시하였으며, 기본적 변인으로 신장, 체중, Body Mass Index(BMI), 안정시 심박수, 안정시 혈압을 측정하고, 체력 변인으로는 체간 굴곡과 신전, 견관절 굴곡과 신전의 유연성과 악력을 통한근력, 폐활량을 통한 폐기능을 측정하였다. 또한 혈액성분으로는 혈중지질, 그리고 심혈 관질환 위험인자인 염증반응지표로서 C-reactive protein(CRP), Interleukin-6(IL-6), Tumor Necrosis Factor-α(TNF-α)를 분석하였다. 8주 유산소성 체조활동 프로그램은 매회50분씩 주 2회 실시하였으며, 운동강도는 운동자각도(RPE) 수준 13으로 유지하였다. 뇌병변 장애인의 유산소성 체조활동 프로그램 참여 후 운동의 효과를 검증한 결과 전체 집단에서 안정시 수축기혈압과 이완기혈압이 유의하게 감소하였다(p<.01). 또한 체력 요인에서는 체간 굴곡과 신전(p<.05), 건측의 견관절 굴곡과 신전(p<.01), 환측의 견관절 굴곡(p<.05)이 유의하게 증가하였다. 혈액 성분의 분석 결과 혈중지질에는 유의한 변화가 나타나지 않았으며, 염증반응지표의 경우 IL-6가 유의하게 감소하였고(p<.05), 유의하지는 않으나 CRP도 감소하는 경향을 나타냈다(p<.088). 결론적으로, 지속적이고 규칙적인 유산소성 체조활동은 뇌병변 장애인의 안정시 심박수와 혈압 및 체력을 개선시키는 효과가 있을 뿐만 아니라 심혈관질환의 위험요소로 알려져 있는 염증반응지표를 개선하는 데에도 긍정적인 영향을 주는 것으로 나타났다. The purpose of this study was to analysis the effect of aerobic gymnastic exercise for eight weeks on physical fitness and inflammatory markers which are known as risk factors for cardiovascular diseases in hemiplegic disabled after cerebral stroke. For this purpose, stroke patients(n=8) participated in aerobic exercise training 2 days/week, 50 mins/day, for 8 weeks and maintained at 13 level of RPE. Body composition, resting heart rate and blood pressure were measured before physical fitness test. Trunk flexion/extension, shoulder flexion/extension, grip strength, pulmonary function were measured as physical fitness test. Blood sample of 15ml was drawn from antecubital veins for measuring inflammatory markers(c-reactive protein, interleukin-6, tumor necrosis factor-α). The result of this study is as follows: Resting blood pressure decreased significantly after 8-week aerobic exercise training in total stroke patients (p<.01). After completion of the aerobic exercise program, trunk extension/flexion(p<.05), shoulder extension/fexion(p<.01), were significantly improved in total stroke patients. There was no significant improvement on blood lipid profiles in total stroke patients. However, IL-6 was significantly improved in stroke patients after 8-week aerobic exercise(p<.05). In conclusion, These findings suggests that regular aerobic gymnastic exercise decreases resting heart rate and blood pressure, and improves physical fitness, inflammatory markers which are known as a risk factor of cardiovascular diseases in person with disability occurred due to hemiparetic stroke

      • HFC-134a와 대체냉매 HFC-152a에 CF_3I를 혼합한 자동차냉매 특성

        These days, an attention of environment has been increasing all over the world. to prevent the green house effect and destruction of an ozone layer, an ozone destruction potential(ODP) be zero and a refrigerant for low global warming potential(GWP) is needed. HFC-134a, in which hydrogen is mixed instead of chlorine is a refrigerant used for automobile conditioners and its destruction potential is ecologically zero. However, it is not considered as a perfect substitutive refrigerant as its GWP is high. It is studied refrigerant mixtures in which HFC-134a, HFC-152a and CF_3I with low GWP and zero ODP are mixed by experimentally and concluded as follows; 1. with the variation of speed of compressor outside temperature and flow rate, the heat of evaporator and compressor and coefficient of performance was varied, and influenced the air conditioner. 2. The pressure of evaporator was decreased with increasing the speed of compressor and that of the refrigerant HFC-134a was higher 21% than that of azotrope refrigerant mixed with CF_3I. The overall capacity of condensor and evaporator was increased and HGC-134a was higher than azotrope refrigerant mixed with CF_3I 3. The capacity of evaporator with the refrigerant HFC-134a was higher 6% than azotrope refrigerant mixed with CF31 and the required power with zaotrope refrigerant mixed with CF_3I was higher 11% than that of HFC-134a. 4. The COP of the refrigerant FHC-134a was higher 31% in 1000RPM. when outside temperature was increased, COP was decreased. 5. Performance of HFC-134a is better than azotrope refrigerant mixed with CF_3I but prefered zaotrope refrigerant mixed with CF_3I considering the environment.

      • 폐경 후 여성에서 체성분과 골밀도와의 관계

        Background: Body weight is an important factor that influence the bone density in postmenopausal women except estrogen dificiency. However, different results are reported about the relationship between body composition and bone density in the postmenopausal women. We have studied the relationship between age, body weight, body mass index (BMI), waist hip ratio (WHR), muscle mass, fat mass, fat free mass and bone density. Materials and Methods: We have studied 127 persons of postmenopausal women who visited university medical center and examined the inbody 3.0 and Dual Energy X-ray Absorptiometry (DEXA) from Jan, 2001 to Jun, 2002. they didn't have any disease and didn't received homone therapy, osteoprosis therapy or other medication that influence the bone density. Results: The numbers of study subjects is total 127 persons. Mean age is 56.9±5.14, mean weight is 59.3±8.7 kg, mean BMI is 25.37±3.16 (kg/㎡), mean fat mass is 20.02±5.05 kg, mean muscle mass is 37.49±4.50 kg, mean fat free mass is 39.80±4.70, mean BMD is 0.828±0.148 (g/㎠). In the result of linear regression analysis, age, height, weight, muscle mass, fat free mass, fat mass, BMI are significant determinants of BMD. In stepwise multiple regression analysis, age is the most significant determinant of BMD and besides age, fat mass is the most significant determinant of BMD among body composition. Conclusion: In postmenopausal women, age, height, weight, BMI, muscle mass, fat free mass, fat mass are significant determinants of BMD and besides age, fat free mass is the most significant determinant of BMD among the body composition. So, diet and exercise that increase fat free mass will contribute to bone density increment.

      • 냉매조건에 따른 열교환기의 성능특성

        자동차 공조용 핀형 열교환기의 성능특성에 관한 연구

        Fin-tube type(Fin-type) heat exchanger has been tested in order to replace the heat exchanger of parallel flow type(P.F-type) which is now widly used in automobile air conditioning system. The following conclusions are drawn by the comparison of the characteristics of the heat exchangers. Evaporator and condenser capacities and COP(Coefficience of performance) were varied as with the compressor speed, outdoor air temperature and air flow rate changed, which much influenced on the characteristics of the air conditioning system. Evaporator and condenser capacities were increased with increasing compressor speed and outdoor air temperature. Evaporator and condenser pressures of Fin- type were decreased by 7% and 5% respectively compared with those of P.F-type. The COP of Fin-type was decreased with increasing outdoor air temperature and compressor speed. The COP of P.F-type was decreased by 14% compared with that of Fin-type.

      • 필라델피아 염색체 양성 급성골수성백혈병에서 Imatinib Mesylate로 분자학적 관해를 유도한 후 성공적인 자가 말초혈액 조혈모세포이식이 시행되었던 1예

        저자들은 필라델피아 염색체 양성 급성골수성백혈병 환자에서 복합항암화학요법으로 혈액학적 완전관해를 유도하고, imatinib mesylate를 이용하여 분자학적 관해를 유도한 후, 성공적으로 자가 말초혈액조혈모세포이식을 시행하였던 1예를 경험하였기에 이를 보고하는 바이다. Philadelphia chromosome (Ph¹)-positive acute myeloid leukemia (AML) is very rare disease, comprising 3~4% of the AML, and is a clinically distinct entity with poor clinical outcome with conventional chemotherapy. We report a case of Ph¹-positive AML who underwent successful autologous peripheral blood stem cell transplantation after achieving a molecular remission with imatinib mesylate.

      • Olfactory modulation by neuropeptides in cockroaches

        Olfactory sense is an essential modality for insects to locate hosts, mates, oviposition sites, and food resources in nature. Based on many studies so far, insect olfactory systems in periphery as well as central nervous systems seem to be highly plastic with behavioral and physiological changes associated with learning and memory, sensory processes, and other developmental processes. Eventually, it is also evident that these plasticity and processes in olfactory systems are modulated by a various ensemble or sets of neuropeptides. The olfactory reception in the peripheral systems of the cockroach, Periplaneta americana, associated with learning behaviors, is fluctuated by the circadian rhythm, which is subsequently thought to control the titer of hormones and various neuropeptides concomitantly. However, it remains still elusive how neuropeptides and neuroendocrine systems modulate olfactory system in the peripheral systems in cockroaches as well as any other insects. Here, our aims to characterize the ultrastructure of these neuro-endocrine systems in the peripheral olfactory systems in American cockroach, Periplaneta americana. Using in situ hybridization methods, we found out that tachykinin and its receptors seem to be important neurotransmission machineries in the periphery to convey the arousal signals from the insect brain. Our findings also indicate complicated endocrine systems connected with central nervous systems may modulate the olfactory reception in the periphery.

      • DNA-free Genome Editing in Plants

        CRISPR/Cas9-based genome editing technology fast replaces the previous methods that require protein engineering such as Zinc Finger Nucleases (ZFNs) and TALE nucleases (TALENs). Conventional genome editing of plant cells using CRISPR/Cas9 technology largely depends on Agrobacterium-mediated transformation of the plant cells and subsequent regeneration of whole plants from the edited cells. During this process, unwanted foreign DNAs including the antibiotics gene and fragments of the T-DNA can be introduced into plant genome. Insertion of these unwanted DNA causes lots of regulatory restrictions when commercializing the LMO products. To step aside these issues, we designed DNA-free ribonucleoprotein-based method and regenerated whole plants from the successfully engineered cells. We will share our discovery on the successful implement of this technology in lettuce protoplasts.
