The isolation and in vitro differentiation of neural progenitor cells (NPCs) provide an approach to study the regulation of development and differentiation in cell culture. This paper describes a culture system of embryonic-derived NPCs. NPCs from mouse cortex of embryonic day 14 (E14) can be maintained and expanded using a serum free defined medium containing basic fibroblast growth factor (bFGF). In the highly purified secondary cultures, most of these cells were positive for nestin, a specific marker for NPCs in developmental brain. Immunocytochemistry analysis showed the expression of metabotropic glutamate receptor, mGluR3 in the nestin-immunoreactive cultured embryonic-derived NPCs. Over 99.7% of the nestin-expressing cells co-expressed mGluR2/3. These results suggest that mGluR3 may play a role in the early neurogenetic events. Here, this method will facilitate studies using embryonic-derived NPCs as cell culture system for neural development and differentiation.

Deubiquitinating enzymes reversibly control the state of protein ubiquitination. Ubiquitin carboxy-terminal hydrolase L1 (UCH-L1) is a deubiquitinating enzyme that is exclusively expressed in neuronal cells and in the testis/ovary. UCH-L1 is a constituent of cellular aggregates that are commonly observed in neurodegenerative diseases such as Parkinson’s disease (PD) and Alzheimer’s disease (AD). The UCH-L1 gene reportedly causes familial PD and is the basis for the gracile axonal dystrophy (gad) mouse. These facts suggest that the function of UCH-L1 is closely related to the survival and maintenance of the neuron. This review summarized the role of UCH-L1 in the pathogenesis of neurodegenerative diseases, with particular emphasis on the gad mouse, which offers unique insights into the relationship of UPS to neurodegeneration in an in vivo setting.
Specific Expression of Ep-CAM during Mouse Spermatogenesis

Analysis of the biochemical events and the genes at various developmental stages in testis of mammal is an important prerequisite for understanding the characterization of spermatogenesis. The present study was demonstrated the specific expression of epithelial cellular adhesion molecule (Ep-CAM) in the spermatocytes of mouse testis. In addition, FACS analysis showed the nearly homogeneous population of isolated spermatocytes using immunomagnetic beads. These results may enable an easy and accurate analysis of the developmental stage during mammalian spermatogenesis, especially spermatocytes.

본 연구에서는 HPL-04가 골관절염의 예방 및 치료 약물로서의 가능성을 탐색하여 다음과 같은 결론을 얻었다. HPL-04는 연골세포 생존율과 연골형성과 관련된 collagen type Ⅱ, SOX 9 그리고 aggrecan의 유전자 발현을 유의성 있게 증가시킬 뿐만 아니라 염증성 인자와 관련 있는 MMP-2, 9도 유의성 있게 감소시켰다. HPL-04는 MIA에 의해 유도된 퇴행성 골관절염에서 관절연골의 파괴와 골 침식 등 연골의 변성을 억제했으며 proteoglycan의 소실을 유의성 있게 감소시켰다. 이에 본 연구는 HPL-04가 부작용이 적고 약리 효과가 뛰어나 골관절염 예방 및 치료제 개발에 활용될 수 있을 것으로 본다. HanPoong Leading (HPL)-04 were prepared with different oriental medicines (balk of Kalopanax pictus balk, Chaenomelis Fructus, Angelica gigas root, Zingiber officinale, Raphanus sativus Linne and Saururus chinensis Baill.) to investigate the protective effects of HPL-04 on cartilage degradation in knee osteoarthritis (OA). Rat articular chondrocytes incubated with rhIL-1α markedly increased matrix metalloproteinase (MMP)-2 and 9 activities, decreased cell viability and reduced chondrogenic gene expression. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, MMP-2 and 9 activities and real time RT-PCR indicated that HPL-04 counteracted these harmful effects in dose-dependent manner. In addition, for experimental OA in vivo, monosodium iodoacetate (MIA, 0.5 mg/50 μL) was injected into knee joints of rats and administered HPL-04 to rats for 4 consecutive weeks after MIA treatment. The experimental data showed that treatment with HPL-04 significantly prevented of MMP-2 and 9 activities in articular cartilage. Histopathological and micro-CT evaluations of the knee joints also revealed that HPL-04 effectively ameliorated MIA-induced degenerative OA. In conclusion, HPL-04 has potential applicability for the prevention and treatment of degenerative OA.
급성염증성 발열 모델에서의 항염증성 약재 혼합 추출물(YG-1)의 효과

YG-1의 항염증 및 해열 치료 약물로서의 가능성을 검증하기 위하여 대식세포와 동물모델에서 염증성 사이토카인 활성 및 해열 관련 인자들을 연구하여 다음과 같은 결론을 얻었다. YG-1은 염증성 사이토카인 인자인 IL-1β의 발현을 효과적으로 억제하였으며, iNOS 및 COX-2의 발현을 억제시킴으로써 염증인자인 NO 및 PGE2 생성을 유의적으로 감소시키는 것으로 나타났다. 또한 YG-1은 LPS 유도성 발열모델에서 염증을 완화시키고 해열의 효과를 보여 주었다. 이상의 실험 결과로부터 본 실험에 사용된 YG-1은 염증에의해 유발되는 인후염과 같은 질환을 예방 및 치료할 수 있을 것으로 기대된다. Traditional herbs, such as Lonicera japonica, Arctii Fructus, and Scutellariae Radix have been used as traditional drug due to their anti-inflammatory and anti-oxidant activities. The aim of this study was to investigate the anti-inflammatory effects of extract mixture (YG-1) in a model of lipopolysaccharide (LPS)-induced acute inflammation in both macrophage (RAW 264.7) cells and Sprague-Dawley rats. YG-1 did not show specific cellular toxicity in RAW 264.7 cells until a concentration of 100 μg/mL. YG-1 reduced various markers related to inflammation such as IL-1β, COX-2, and iNOS caused by LPS in RAW 264.7 cells. Consistent with these results, YG-1 exerted significant anti-inflammatory effects in an acute inflammation rat model. Acute fever and high concentration of IL-1β in serum induced by LPS were significantly reduced by YG-1. These results were similar to flubiprofen, a commercial anti-inflammatory and anti-febrile drug. Therefore, these results indicate that YG-1 has beneficial effects on LPS-induced acute inflammation and suggest that YG-1 can serve as an effective anti-inflammatory and anti-febrile drug.

<P>Ubiquitination is required throughout all developmental stages of mammalian spermatogenesis. The two ubiquitin C-terminal hydrolase (UCH) enzymes, UCH-L1 and UCH-L3, deubiquitinate ubiquitin-protein conjugates and control the cellular balance of ubiquitin. These two UCH isozymes have 52% amino acid identity and share significant structural similarity. A new function of these two closely related UCH enzymes during spermatogenesis which is associated with germ cell apoptosis has been analyzed. Apoptosis, in general, is thought to be partly regulated by the ubiquitin-proteasome system. During spermatogenesis, apoptosis controls germ cell numbers and eliminates defective germ cells to facilitate testicular homeostasis. In this paper, I review the distinct function of the two UCH isozymes in the testis of gad and Uchl3 knockout mice, which are strongly but reciprocally expressed during spermatogenesis. In addition, the importance of UCHL1-dependent apoptosis for normal spermatogenesis and sperm quality control is discussed.</P>
