Autophagy as a decisive process for cell death

Autophagy is an intracellular catabolic pathway in which cellular constituents are engulfed by autophagosomes and degraded upon autophagosome fusion with lysosomes. Autophagy serves as a major cytoprotective process by maintaining cellular homeostasis and recycling cytoplasmic contents. However, emerging evidence suggests that autophagy is a primary mechanism of cell death (autophagic cell death, ACD) and implicates ACD in several aspects of mammalian physiology, including tumor suppression and psychological disorders. However, little is known about the physiological roles and molecular mechanisms of ACD. In this review, we document examples of ACD and discuss recent progress in our understanding of its molecular mechanisms.

Nimesulide (NSAIDs) is often prescribed with antiulcer agents such as antacids or H2 receptor blockers to reduce the stomach irritation. We found that when nimesulide was compounded to powder with an antiulcer agent, composed of ranitidine, magnesium oxide, aluminum magnesium silicate and aluminum magnesium hydrate, the compounded powder's color was changed to hard yellow. So this study was planned to verify the pharmaceutical interaction between these two agents and to evaluate the clinical importance of this interaction. Nimesulide and antacid complex were triturated together by the ratio of 1:1 and 2:1 as the object. We measured the change of the powders color, contents of the main composition, pH and moisture content according to the compounding time and the storage period. For the compounding time, the experiment conditions were triturating each other separately then mixing, triturating together for 1, 3 and 5 minutes. The storage periods were 0, 1, 3, 7, 14, 21 and 28 days respectively. Compounded powder's color was considerably changed to yellow according to increase of the compounding time and the storage period. Though the color was changed, there was no relationship among the compounding time and contents of the main composition, pH level or moisture content (F0>F (1, 2; 0.05)). Also storage period didn't affect those factors(F0>F (1, 5; 0.05)). According to our results this pharmaceutical interaction did not alter the effect of each agent, and it was considered to be the tolerable incompatibility.

Hydrodynamic focusing technique to generate focused flow has been used for flow cytometry in microfluidic devices. However, devices with circular capillary tubes made of glass are not suitable for flow visualization or optical signal detection because the rays of light are distorted at the curved interface. We devised a new acrylic chamber assembled with a pulled micropipette and a rectangular microchannel made of glass. This new channel geometry enabled us to visualize the three-dimensional (3D) flow characteristics with confocal imaging technique. We analyzed the 3D hydrodynamic focusing in a circular capillary tube and a rectangular microchannel over a practical range of flow rates, viscosities and pressure drops.

High-throughput microscopy (HTM) was developed recently for the automatic detection of airborne asbestos fibers that can cause lung cancer, asbestosis and mesothelioma. The HTM method has been applied to couting the airborne asbestos fibers as an alternative to the conventional phase contrast microscopy (PCM). In this paper, we demonstrated that the HTM enabled us to obtain quantitative results for low-concentration airborne asbestos samples under detection limit, and we made a comparison between the results from HTM and PCM. In addition, a verification study was conducted using proficiency analytical testing (PAT) samples of chrysotile and amosite. The HTM method can be applied to the existing PCM method by reducing analysis time and labors. Potential applications can be extended to detection of asbestos fibers in soil and water.

삶의 최종단계는 죽음이다. 생물학적으로는 유기체가 생존능력을 상실한 것을 의미하며 신체기능의 정지로 간주되는 죽음은 노인의 주관심사인 동시에 노년기에 극복해야 할 주요 과업의 하나임에도 불구하고 제대로 주목받지 못한 영역으로 남아 있다. 일찍부터 에릭슨은 죽음을 잘 준비하고 대비하는 것이 노년기의 자아통합을 성취하고 한 인간으로서 완성된 삶을 가능케하는 핵심으로 보았다. 그러나 죽음을 터부시하는 우리나라의 문화에 영향을 받아 아직까지는 중요하게 다루어지지 못하고 있지만 죽음에 대한 논의가 조심스럽게 확대되어 가는 추세로 나타나고 있다. 이는 2010년 이후 죽음준비프로그램을 실시하는 노인복지관의 수가 많아지고 있는 것에서 유추할 수 있다. 이에 본 연구는 노인복지관 이용노인을 대상으로 죽음준비교육이 자아통합감을 매개로 하여 죽음불안에 미치는 영향을 분석하여 죽음준비교육프로그램에 자아통합감을 증가시킬 수 있는 요소를 보다 강화시킬 필요가 있음을 확인하였다. This study is to analyze how the elderly’s death educations considering ego-integrity as a parameter effect the elderly’s death anxiety. In the current aging society, there exist problems such as elder abuse or suicide and aggravation of their economic activities. At this point, the elderly’s ego-integrity possess a huge significance. And death education is expected to reduce death anxiety in older adults. Thus, this study is to look into the effects of the death education with mediation effects of ego-integrity on the elderly’s death anxiety. This research project was conducted in 10 senior welfare centre in Korea, from October 1st 2014 to 3rd targeting 840 elder people who were in their 60s or older, and with the data from final 656 targets, we did analysis using PASW 21.0. As a result, we can notice that elder death education affect the elderly’s death anxiety with Mediation effects of ego-integrity. That is to say, we are able to find out that if elder people take death eduction, there will be possibly a negative effect on death anxiety with positive effect on ego-integrity. Finally, this indicates that it is necessary to establish policies to develop death eduction on older adults and lead the elder people to participate death education in order to provide a positive influence on their ego-integrity and to reduce death anxiety.
