        DIS80의 遺傳子型 決定을 위한 PCR法의 條件에 관한 硏究

        The polymerase chain reaction(PCR) is known to be a very sensitive method of nucleic acid synthesis by which a particular segment of deoxyribonucleic acid(DNA) can be specifically amplified. However, as no single protocol will be appropriate to all situations, it requires to optimize the PCR conditions for a given application, especially analytical procedure to amplified fragment length polymorphism of VNTR region. Recently, typing of VNTR regions in the field of forensic science is almost done by PCR, but this reaction ofter results in a number of potential problems including generation of recombinant alleles during the extension phase. The objective of this paper is to evaluate the cause of PCR products of a abnormal length, and to set up the guideline for the reliable production of D1S80VNTR region in the human genome by studying several parameters that influence polymerase chain reaction. In the study of D1S80 VNTR region to be amplified, the following PCR conditions are optimal in the 50㎕ of reaction volume and under the temperature conditions of 95℃ for 1 minute, 65℃ for 2 minutes and 72℃ for 2 minutes. 1. The adequate concentration of MgCl₂ is within the range of 1.0 to 2.0 mM. 2. The high activities of Taq polymerase show about 0.5 to 1.5 units without generation of the undesired products. 3. The adequate amount of template DNA is about 30 to 50ng, although the template DNA can be amplified with the minimum of 40pg. 4. With the 50ng of template DNA, the adequate numbers of PCR cycle range 25 to 27 cycles. 5. Artifact production by PCR is minimal within the range of 0.2 to 0.3μM of each primer concentration. With the above conditions, amplified fragment length polymorphism (AMP FLP) obtained by polymerase chain reaction from the genomic DNAs of 4 Korean families (one family with 8 offsprings and both parents, 3 families composed of each one child and both parents) were analyzed to test the applicability of this technique to paternity testing. The results obtained present reliable parent child relationships based on the correct genotypes of D1S80, without any undesired PCR products of abnormal length that are postulated as heteroduplex resulting from cross annealing or hybridization between the amplified target DNA.

        치환나프톨류(4-Cl-1-Naphthol, 6-Br-2-Naphthol, 8-NH_2-2-Naphthol)의 해리상수를 온도(25∼40℃)와 압력(1∼2000 bar)을 변화시키면서 분광학적 방법으로 측정했다. 이들의 해리상수는 치환기가 들어가므로서 모두 감소했다. 이것은 치환기의 I-효과로서 설명이 가능했으며 또 온도와 압력이 증가하면 해리상수도 증가하였다. 이들 해리상수로부터 반응의 열역학적 성질을 계산하여 반응의 성질을 알아보았다. The dissociation constants of 4-Cl-1-naphthol, 6-Br-2-naphthol, 8-NH_2-2-naphthol in aqueous solution were measured by spectroscopic method in the temperature range from 25 to 40℃ and pressure up to 2000bar. The dissociation constants were decreased as the substituents were inserted in naphthol from 4.4×10 exp(-10) to 5.82×10 exp(-11) as chloride compound and 2.5×10 exp(-10) to 3.44×10 exp(-11) or 4.21×10 exp(-11) as bromine or amino compounds, respectively. This decrease can be explained with the I- or R-effects of substituents. From the dissociation constants various thermodynamic properties were claculated and discussed the characteristics of the dissociation reaction.

        This paper presents a LTCC Balun-BPF, which is a BPF(band pass filter) with a Balun in a single LTCC chip for the direct interface with a MMIC chip having balanced inputs. The physical dimension of the designed Balun-BPF is 2.4x2.0x0.88 ㎣ and the used dielectric constant E_(r) is 36. A Balun of three-lines structure with striplines and a BPF of comb-line structure was combined into the Balun-BPF. The simulated result shows 4.8dB of insertion loss, 178~179 degree of the phase imbalance, 14dB of the return.

        복지권으로서 교육권 보장을 위한 『장애인 등에 대한 특수교육법』

        결핍에서 오는 필요의 개념은 장애인에게 복지권으로서 교육받을 권리를 가장 잘 말해주고 있다. 장애인은 신체적·인지적 손상으로 발생하는 기본적인 생존적 필요의 충족뿐만 아니라 동시에 교육기회 균등이나 개인차의 고려 등을 통해 무지로부터 벗어날 수 있는 보편적 필요가 충족되어야 함을 논의하였다. 장애인들에게 이러한 결핍에 따른 필요를 충족시켜 줄 이론적 근거가 롤즈(J. Rawls)의 정의론이라 할 수 있다. 정의론의 '차등의 원칙'에 따르면, 교육에 있어 비장애인과 장애인 중 먼저 최소 수혜자인 장애인의 교육복지를 우선하여 극대화할 필요성을 제시함으로써, 그들의 교육권 보장을 위한 이론적 근거에 대한 정당화를 논의하였다. 기존의 특수교육진흥법은 '국가가 교육할 권리'를 가지는 국가 주도적 교육이었다면, 「장애인 등에 대한 특수교육법」 제정은 수년간 장애인의 교육권 확보를 위해 애쓴 장애인 교육 주체들이 노력 끝에 '교육받을 권리'를 찾게 된 의미 있는 결실로 평가되어 진다. The concept of need which comes from lack represents well the right to education as welfare rights to individuals with disabilities. It is necessary to meet the universal need of individuals with disabilities such as an equal opportunity for education and the consideration for individual difference as well as their substantial need. The rationale which may satisfy the need associated with the lack can be J. Rawls's a Theory of Justice. The difference principle by Rawls presents the need of the educational welfare of individuals with disabilities(the least advantaged) to take precedence over that of the non-disabled and be maximized, it is considered that he created the rationale that makes secure their right to education. While established Special Education Promotion Law was national-driven education that state had to the education right, the enactment of 'the Special Education Law for the Individuals with Disabilities, etc.' can be a significant fruit which takes back 'the right to education by citizens' by the educational subjects of the individuals with disability who have taken pains to secure their right to education for years.

      • LTCC 기술을 이용한 광대역 3㏈ 결합기 설계 및 제작

        This paper presents a 3 dB wideband tandem quadrature coupler fabricated by LTCC technology. The 3 dB tandem coupler was designed with minimum numbers of layers and via holes to reduce the cost and to increase yield on manufacturing. Since tight coupling value is easily achievable with the tandem structure, any high impedance lines or tight coupling gaps can be avoided. The simulated and measured data shows wide bandwidth from 1.4 GHz to 2.4 GHz with 0.3 dB amplitude imbalance, phase difference within 1.5 degree, and the isolation characteristics about 20 dB. The size of fabricated 3dB coupler is 9.64×10.42×1.74㎣.

        흰쥐 대뇌피질에서 cholecystokinin octapeptide(CCK-8)의 분포를 관찰하기 위하여 간접면역형 광법을 행하여 관찰해 본 결과 다음의 많은 부위에서 CCK-8양성세포 및 양성섬유를 관찰할 수 있었다. 신피질: CCK-8양성세포는 주로 제2, 3층에서, 양성섬유는 제2, 3 그리고 6층에서 저밀도를 나타내었다. 후구: 전후핵에서 많은 CCK-8양성세포 및 양성섬유를 관찰하였다. 이상엽 및 후내야: 양성세포는 주로 제2층에서 관찰되었으며 양성섬유는 제2층에서 중밀도 제3층에서 저밀도로 관찰되었다. 해마체: 많은 양성세포 및 고밀도의 양성섬유가 원추세포층에서 관찰되었다. In order to investigate the distribution of immunoreactive cholecystokinin octapeptide (CCK-8) containing neurons in the cerebral cortex of the rat, brains of the rats were processed for indirect immunofluorescent technique. The results obtained were as follows. Neocortex; CCK-8 immunoreactive cell bodies were found in layer Ⅱ and Ⅲ and immunoreactive axon terminals were heaviest in layers Ⅱ, Ⅲ and Ⅵ. Olfactory bulb; The anterior olfactory nucleus contained numerous CCK-8 immunoreactive cell bodies and a moderate density of immunoreactive axon terminals. Piriform and entorrhinal cortex; CCK-8 immunoreactive neurons and axon terminals were observed in layer Ⅱ and Ⅲ. However, they were more frequent in layer Ⅱ. Hippocampus; CCK-8 immunoreactive cell bodies and axon terminals were found in pyramidal layer.

        The dissociation constants of 2, 3-, 2, 4-, 2, 5-and 3, 5-dimethylpyridine were measured by conductometric method at various temperatures (15∼40℃) and pressures (1∼2500 bar) in aqueous solution. The shift of dissociation constants of methylptridines from pyridine were explained by the I-effect and cooperative effect of the methyl groups. From the dissociation constants various thermodynamic properties were calculated and discussed the characteristics of the dissociation reactions.

        In spite of the economic regeneration of 1999 through the program of IMF in late 1997, a recent economic condition has been getting worse. Many private-and public-side companies faced the serious depression. The 3rd sector in particular is in difficulties for management, for the local government itself is framed to the economic problems and thus in a bad shape for supporting the financial benefits. The central and local government also are needed in downsizing and streamlining the system and the structure in the field and then try to examine those businesses invested previously. In the process, more importance is placed on the role-play and mission of the 3rd sector. The finding of this study is, firstly, to examine legal situations and, secondly, to review the comprehensive research on the Korean and foreign case studies and, finally, to suggest policy-making guidelines of the 3rd sectors.

        쌍발 복합재 비행기의 속도, 고도 보정 및 상승성능에 관한 연구

        Airspeed and altimeter calibration of "win Mee was conducted by the flight test. We have adopted system to system method. Flight test data is corrected for instrumented error and position error, and the resultin data was satisfied. Climb Performance flight test also was conducted. But we could not have all data because of limited flight time. The resulted data was satisfied compare with calculated dab.
