        Paricalcitol attenuates lipopolysaccharide-induced inflammation and apoptosis in proximal tubular cells through the prostaglandin E<sub>2</sub> receptor EP4

        Background: Vitamin D is considered to exert a protective effect on various renal diseases but its underlying molecular mechanism remains poorly understood. This study aimed to determine whether paricalcitol attenuates inflammation and apoptosis during lipopolysaccharide (LPS)-induced renal proximal tubular cell injury through the prostaglandin E<sub>2</sub> (PGE<sub>2</sub>) receptor EP4. Methods: Human renal tubular epithelial (HK-2) cells were pretreated with paricalcitol (2 ng/mL) for 1 hour and exposed to LPS (1 μg/mL). The effects of paricalcitol pretreatment in relation to an EP4 blockade using AH-23848 or EP4 small interfering RNA (siRNA) were investigated. Results: The expression of cyclooxygenase-2, PGE<sub>2</sub>, and EP4 were significantly increased in LPS-exposed HK-2 cells treated with paricalcitol compared with cells exposed to LPS only. Paricalcitol prevented cell death induced by LPS exposure, and the cotreatment of AH-23848 or EP4 siRNA offset these cell-protective effects. The phosphorylation and nuclear translocation of p65 nuclear factor-kappaB (NF-κB) were decreased and the phosphorylation of Akt was increased in LPS-exposed cells with paricalcitol treatment. AH-23848 or EP4 siRNA inhibited the suppressive effects of paricalcitol on p65 NF-κB nuclear translocation and the activation of Akt. The production of proinflammatory cytokines and the number of terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling-positive cells were attenuated by paricalcitol in LPS exposed HK-2 cells. The cotreatment with an EP4 antagonist abolished these anti-inflammatory and antiapoptotic effects. Conclusion: EP4 plays a pivotal role in anti-inflammatory and antiapoptotic effects through Akt and NF-κB signaling after paricalcitol pretreatment in LPS-induced renal proximal tubule cell injury.

        안모의 수직적 양태에 따른 골격 및 치조골의 위치 관계에 관한 연구

        교정 증례의 분석 예후 추정은 두개골의 수평 및 수직적인 관계가 동시에 고려되는 입체적 관점에서 평가되어야 한다. 따라서 본 연구에서는 성장이 완료된 성인 남녀 각 100명을 추출하여 두부 방사선 사진을 채득한 후 하안면 고경과 상안면 고경의 비율로 안모의 수직적 수직적 형태를 구분하여 군을 설정하였으며 이들 군간의 골격 및 치조골의 크기와 위치 관계를 비교하고 상호 관련성을 조사하여 다음과 같은 결과를 얻었다. 1. 치조고경, 안면 고경, lower gonial angle, 그리고 FMA 항목에서 하안면 고경이 큰 군의 것이 작은 군에 비하여 유의하게 크게 나타났다. 2. 치조고경, 안면고경, 하악지 고경, 그리고 Jarabak ratio 항목에서 남서이 여성에 비하여 유의하게 크게 나타났다. 3. 치성 및 골격성의 수직계측항목과 UAFH/LAFH의 상관관계 조사에서 치조고경,안면고경, lower gonial angle, FMA 그리고 Bjok's Sum 항목이 UAFH/LAFH와 유의한 상관관계를 보였다. The purpose of this study was to investigate the variations of the skeletal and dentoalveolar dimensions in relation to vertical facial patterns. Lateral cephalogram of 200 cases (100 cases of male and 100 cases of female, average age of which was 23.2 years) were traced and some measurements of skeletal and dentoalveolar dimensions were measured. The ratio of UAFH/LAFH was employed to classify the samples into groups of excess and short lower anterior facial height. And the comparison between two groups were taken statistacally. The following results were obtained. 1. The dentoalveolar height, lower anterior facial height, lower gonial angle, and FMA in the excess-lower-anterior-facial-height group were significantly larger than those in short-lower-anterior-facial-height group. 2. The dentoalveolar height, ramus height, and Jarabak ratio in the male subjects were significantly larger than those in the female subjects. 3. The UAFH/LAFH ratio showed a significant correlation to upper, lower facial height, AUDH, PUDH, ALDH, PLDH, Lower gonial angle, FMA, and Bjork's Sum.

      • 병원식당 직원급식을 위한 조리방법의 개선 : 직원급식에 종사하는 조리원을 대상으로

        This study was carried out to evaluate hospital cook`s cooking method and to suggest the more proper food-service guideline for hospital staff. Questionnaire method to hospital cooks were practiced and the data were analyzed by SAS package program for the frequency, percentage, Anova and the Duncan multiple test. The summarized results are as like these ; 1. Hospital cooks' recognition degree for kitchen management was very excellent. 2. Hospital cooks who are in charge of hospital staff were very positive to participate cooking method improvement program. 3. The more careered cook, the more sincerely have done for standard recipe in hospital kitchen. 4. The degree of cooks` desire for cooking method education programs was indicated by significance.

      • 한국 청소년에서 혈청 Adiponectin 농도와 비만도와의 상관관계

        연구배경: Adiponectin은 지방세포에서 특이적으로 발현되고 분비되는 adipocytokine이다. 아직 adiponectin 과 소아 비만과의 관련성을 연구한 보고는 많지 않다. 본 연구에서는 한국 청소년을 대상으로 혈청 adiponectin농도와 비만도 및 심혈관계 질환의 위험인자들과의 상관관계를 보고자 하였다. 방법: 2003년 4월 서울특별시 서대문구 소재의 한 중학교에서 12∼15세의 무작위 선발된 164명을 연구 대상으로 하여 신장, 체중, 허리둘레, 체지방을 측정하고 체질량지수를 계산하였다. 또한 공복채혈을 통하여 콜레스테롤, HDL-콜레스테롤, 중성지방, 혈당 및 인슐린 농도를 측정하고 혈청 adiponectin 농도는 ELISA 방법을 이용하여 측정하였다. 결과: 혈청 adiponectin 농도에 따라 삼등분하여 저 adiponectin군, 중간 adiponectin군, 고 adiponectin군으로 나누고 각 군간에 신체계측치 및 임상적 특성을 비교 분석하였다. 저 adiponectin군에서 다른 군에 비해 체질량지수와 허리둘레가 의미있게 크게 나타났다. 또한 저 adiponectin군에서 공복혈당 및 공복혈청 인슐린 농도가 높게 나타났으며, HOMA-IR 값도 높게 나타났다. 저 adiponectin군에서 총콜레스테롤과 중성지방치는 높게, 그리고 HDL 콜레스테롤치는 낮게 나타났다. 다변량회귀분석을 시행한 결과 혈청 adiponectin농도와 가장 상관관계가 높은 변수는 허리둘레와 성별로 나타났다. 결론: 소아에서도 성인과 마찬가지로 혈청 adiponectin농도와 복부비만 및 심혈관계 질환의 위험인자들과의 강한 음의 상관관계를 보임을 알 수 있었다. 향후 사춘기 성호르몬의 변화가 adiponectin에 미치는 영향 및 소아비만의 치료에 따른 adiponectin의 변화 등에 대한 연구가 필요하리라 사료된다. Background: Adiponectin is an adipocytokine that is highly specific to adipose tissue. In contrast to other adipocytokine, the adiponectin levels are decreased in obesity and/or type 2 diabetes. There are few studies regarding the correlation between the adiponectin concentration and obesity in children. Thus, whether the serum adiponectin concentrations are associated with adiposity in children was investigated. Methods: One hundred and sixty four subjects were selected from the participants in an ongoing study on the relationship between birth weight and insulin resistance in children. The current weights, heights, body fat percentages, waist circumferences, blood pressures, lipid profiles and insulin resistance, by the HOMA method, were measured in all the subjects. The serum adiponectin concentrations were determined by a validated sandwich ELISA, using a human adiponectin-specific antibody. Results: The serum adiponectin concentration was negatively correlated with the body mass index, waist circumference, systolic blood pressure, insulin resistance by HOMA and serum triglycerides, and positively correlated with the serum HDL cholesterol level. The serum adiponectin concentrations in the boys were significantly lower than in the girls. In a multiple regression analysis, the serum adiponectin concentration was strongly associated with waist circumference and gender. Conclusion: It is concluded that there was an inverse relationship between the serum adiponectin concentration and abdominal adiposity in children. However, further studies on independent gender differences on adiponectin are needed (J Kor Soc Endocrinol 18:473∼480, 2003).

      • Egr1 is a critical transcription factor to mediate estrogen responses in mouse uterus

        The Egr family of zinc finger transcription factors consisting of 4 members (Egr1 to Egr4) regulates critical genetic programs involved in cellular growth, differentiation, and function. They are co-ex-pressed in many different tissues, suggesting that they may have some redundant functions. While it is clear that estrogen regulates Egr1 in estrogen sensitive breast cancer cells, function of Egr1 and mechanisms by which estrogen (E2) and/or progesterone (P4) regulates Egr1 in uterus still remain unexplored. Thus, we have examined regulatory mechanisms by which Egr1 is regulated in the uterus and abnormal uterine phenotypes of Egr1(-/-) mice. Eight-week-old female mice were ovariectomized (OVX) and rested for a week. Uteri of OVX mice treated with various concentrations of E2 and/or other hormones were collected at 2 h after hormone treatment unless otherwise indicated. ICI 182,780 [estrogen receptor (ER) antagonist] or RU486 [progesterone receptor (PR) antagonist] was injected to OVX mice 30 min prior to hormone treatments. OVX Egr1(+/+) and Egr1(-/-) mice were treated with E2 and/or P4 to examine expression patterns of genes important for estrogen responses, and steroid hormone-induced cell proliferation in the uterus. Collected uteri were utilized for RT-PCR, realtime RT-PCR, Western blotting and histological analyses. Egr1 mRNA was rapidly induced with the highest level at 2h after E2 treatment and gradually deceased to basal levels at 12 h. Pretreatment of ICI 182,780 significantly reduced E2-induced increase of Egr1. However, an agonist for GPR30, a membrane estrogen receptor failed to induce mRNA expression of Egr1, suggesting that E2-dependent Egr1 transcription is mainly regulated via nuclear estrogen receptor, ER. P4 effectively dampened E2-dependent Egr1 transcription and its antagonistic effects were partially interfered with RU486 pretreatment. Histological analyses with BrdU incorporation experiments showed that vascular permeability (an early estrogen response) but not cell proliferation (a late response) was significantly impaired in the uteri of E2 treated OVX Egr1(-/-) mice. Interestingly, some genes involved in early estrogen responses such as Bip and HIF-1a but not those in late responses are dysregulated in uteri of Egr1(-/-) mice. Collectively, our results show that E2 transiently induces Egr1 via activation of nuclear ER. P4 antagonizes E2-dependent Egr1 regulation via PR. Impaired early estrogenic responses in Egr1(-/-) uteri could be due to aberrant gene expression affected by loss of Egr1 which act as a master regulator of estrogen actions in the uterus.-ex

        Effects of Transcutaneous Electrical Nerve Stimulation(TENS), Self- Stretching and Functional Massage on the Muscle Fatigue by Maximum Muscular Strength

        The present study purposed to examine the effects of transcutaneous electrical nerve stimulation, self-stretching and functional massage on the recovery of muscle contraction force for muscle fatigue caused by sustained isotonic contraction. The subjects of this study were 45 healthy students. They were divided into transcutaneous electrical nerve stimulation group(n=15), self-stretching group(n=15) and functional massage group(n=15), and using Primus RS. We observed the pattern of changes in maximal voluntary isometric contraction force(MVIC) after causing muscle fatigue in quadriceps femoris muscle through sustained isotonic contraction. Maximal voluntary isometric contraction force(MVIC) were greatly increased after transcutaneous electrical nerve stimulation, self-stretching and functional massage. In the comparison of recovery rate of muscle contraction force for muscle fatigue caused by sustained isotonic contraction among the treatment groups, it did not show any significant differences. However, it showed that each treatment may be effective in recovery of muscle fatigue caused by sustained isotonic contraction.

        In Vitro Growth of Preantral Follicle and Maturation of Intrafollicular Oocyte from Aged Mice

        This study aimed to recover the ovarian function through in vitro culture of preantral follicles from aged mice. First, we isolated the preantral follicles from ovaries of sixty-seven-week old B6D2F1 mice with decreased fecundity to know how many follicles were present in them, which was 6 preantral follicles including 2 primary, 2 early secondary and late secondary follicles from 8 aged mice. It was confirmed that a few follicles (~2) were present in aged mice through histological analysis compared to adult mice as control. The 9 days of in vitro culture of preantal follicles showed in vitro growth and induced maturation after treatment with hCG (2.5 IU/mL) and EGF (5 ng/mL). Cumulus cells in the cumulus-oocyte complexes (COCs) were removed using hyaluronidase and oocytes at the germinal vesicle (GV) and GV breakdown (GVBD) were obtained from preantral follicle culture of aged mice in vitro. In conclusion, these observations demonstrated that there still were a few preantral follicles in the ovaries of 67 week-old mice, which we were able to culture in vitro and oocytes were obtained from them. This study proposed an in vitro culture system using preantral follicle as a therapeutic strategy for fertility preservation in humans for assisted reproductive medicine.
