        Aerobic exercise training enhances cell proliferation and reduces IL-2 and IL-6 cytokine production in spleen of high fat diet-induced obese mice

        This study investigated the effect of aerobic exercise training on immune cell proliferation and inflammatory cytokine production in the spleen of high fat diet-induced obese mice. C57B1J6 male mice (4 weeks aged. n20) were fed a high fat diet (45% fat) for 5 weeks so that obesity was led intentionally. Then, these obese mice were divided into 2 groups: control group (CON. n10) or exercise group (EXE, n=l0). EXE performed treadmill running for 30-60 min/day at 10-22 m/min, 0% grade, five times per week for 8 weeks. After 13 weeks, all the splenocyte was collected and Con A (Concanvalin A 10 ㎍/ml) was used to stimulate the cell proliferation. MTS and BIOPLEX assay were used for cell proliferation and cytokine production. Independent t-test was used and a p-value under 0.05 was considered as statistically significant. In the results, body weight, IL-2 and IL-6 production were significantly reduced and Splenocyte proliferation was significantly increased after 8 weeks of exercise training. These findings suggest that aerobic exercise training has a positive effect for improving the obese-induced immune dysfunction.

        FSCB phosphorylation in mouse spermatozoa capacitation

        It is generally accepted that spermatozoa capacitation is associated with protein kinase A-mediated tyrosine phosphorylation. In our previous study, we identified the fibrous sheath CABYR binding protein (FSCB), which was phosphorylated by PKA. However, the phosphorylation status of FSCB protein during spermatozoa capacitation should be further investigated. To this aim, in this study, we found that phosphorylation of this 270-kDa protein occurred as early as 1 min after mouse spermatozoa capacitation, which increased over time and remained stable after 60 min. Immunoprecipitation assays demonstrated that the tyrosine and Ser/Thr phosphorylation of FSCB occurred during spermatozoa capacitation. The extent of phosphorylation and was closely associated with the PKA activity and spermatozoa motility characteristics. FSCB phosphorylation could be induced by PKA agonist DB-cAMP, but was blocked by PKA antagonist H-89.Therefore, FSCB contributes to spermatozoa capacitation in a tyrosine-phosphorylated format, which may help in further elucidating the molecular mechanism of spermatozoa capacitation.         In the present study, we investigated possible mechanism for the triggered global CaH wave from local Ca2+ wave in atrial myocytes under shear stress. Shear stress was applied onto single cells isolated from rat atria and two-dimensional confocal Ca2+ imaging was performed to measure Ca2+ signal. Shear stress of 16 dyn/cm2 induced local Ca2+ wave, which was followed by a rapid transverse global Ca2+ wave inhibition of Na + -ci+ exchange using KB-R7943 suppressed the occurrence of rapid transverse wave ob served under shear stress, but not the shear-induced local wave. in the presence of this chemical local wave was developed to global wave under shear stress which results in slower time to peak of peripheral and central CaH transients. As the preceding local Ca"+ wave signal was higher, the latency of the occurrence of transverse global wave was shorter, suggesting Ca2+ dependence of transverse Ca2+ wave. The results sug-gest that shear stress may activate Na+ -Ca2+ exchange via local CaZ+ wave, triggering the rapid global Ca2+ wave

      • 고립성 폐결절의 조직학적 진단에 있어서 경기관지폐생검의 진단적 가치

        Transbronchial lung biopsy(TBLB) is a relatively simple and convenient diagnostic tool in solitary pulmonary nodule(SPN) on chest radiograph, which is cannot be diagnosed through routine tests. To investigate the diagnostic value and diagnostic rate of TBLB according to the variable factor, we analyzed 69 patients with radiologically SPN lesion on chest radiograph which was confirmed histologically. A histologic diagnosis of TBLB was made in 47 of 69 patients(68.1%), in which 26 of 37(70.3%) with malignancy and 21 of 32(65.6%) with benign lesion. There were no significant differences in diagnostic yield according to age, sex and location of the lesion. The diagnostic rate tended to increase with the more than 5 pieces to acquired by TBLB. When the size of SPN was less than 3 ㎝, it cannot be diagnosed. In conclusion, TBLB is a diagnostic procedure with good yield and safety, but percutaneous needle aspiration, transbronchial needle aspiration, and bronchial washing fluid cytology may be a complementary procedure to overcome the limitation of TBLB and get higher diagnostic yield for SPN.

        Development of Self-microemulsifying Drug Delivery System for Enhancing the Bioavailability of Atorvastatin

        The objective of the study was to prepare self-microeulsifying drug delivery system (SMEDDS) incorporating atorvastatin calcium and evaluate its properties and oral bioavailability. Solubility of atorvastatin in various vehicles was determined. Pseudo-ternary phase diagrams were constructed to identify the good self-emulsification region. The droplet size distributions of the resultant emulsions were determined by dynamic light scattering measurement. The mean droplet size of chosen formulation (20% ethyl oleate, 40% tween-80, 40% Carbitol(R)) was 23.4±1.3 nm. The SMEDDS incorporating atorvastatin calcium appeared to be associated with better performance in dissolution and pharmacokinetic studies, compared with raw atorvastatin calcium. In dissolution test, the release percentage of atorvastatin from SMEDDS mixture could rapidly reach more than 95% within 3 min. Oral AUC0→8(hr)values in SD rats was 1994±335 ng·hr/mL, which significantly increased (P<0.05) compared with raw atorvastatin calcium. The SMEDDS formulation was relatively stable when stored at 4˚C during 3 months. Our studies illustrated the potential use of SMEDDS for the delivery of hydrophobic compounds, such as atorvastatin, by the oral route.

      • Ex Vivo Live Full-Thickness Porcine Skin Model as a Versatile In Vitro Testing Method for Skin Barrier Research

        Since the European Union (EU) announced their animal testing ban in 2013, all animal experiments related to cosmetics have been prohibited. Here, we investigated whether an ex vivo live porcine skin model can be employed to study the safety and skin barrier-improving effects of hydroxyacids widely used in cosmetics for keratolytic peels. Glycolic acid (1-10%), salicylic acid (0.2-2%), and lactobionic acid (1.2-12%) were used as representative substances for α-hydroxyacid (AHA), β-hydroxyacid (BHA), and polyhydroxyacid (PHA), respectively. When hydroxyacids were applied at high concentrations on the porcine skin every other day for 6 days, tissue viability was reduced to 50-80%, suggesting that the toxicity of cosmetic ingredients can be evaluated with this model. Based on tissue viability, the treatment scheme was changed to a single exposure for 20 min. The protective effects of a single exposure of hydroxyacids on skin barrier function were evaluated by examining rhodamine permeability and epidermal structural components of barrier function using immunohistochemistry (IHC) and immunofluorescence (IF) staining. Lactobionic acid (PHAs) improved skin barrier function most compared to other AHAs and BHAs. Most importantly, trans-epidermal water loss (TEWL), an important functional marker of skin barrier function, could be measured with this model, which confirmed the significant skin barrier-protective effects of PHAs. Collectively, we demonstrated that the ex vivo live full-thickness porcine skin model can be an excellent alternative to animal experiments for skin studies on the safety and efficacy of cosmetic ingredients.         The High Performance of Polydopamine-coated Electrospun Poly(ether sulfone) Nanofibrous Separator for Lithium-ion Batteries

        In this work, electrospun poly(ether sulfone) (PES) nanofibrous membranes coated with hydrophilic polydopamine (PES-PDA) were successfully prepared. As a separator of lithium-ion batteries, the prepared PESPDA nanofibrous membrane shows ultrahigh porosity, due to the extra-large surface area of the nanofibers. Moreover, good thermostability of PES and polydopamine also results in the low shrinkage of the membrane under high temperature. As a result of the large number of amine groups and phenolic hydroxyl groups from the polydopamine, superior electrolyte wettability and high liquid electrolyte uptake capacity of the PES-PDA nanofibrous membrane have also been disclosed through comprehensive study, which would decrease the electrolyte absorption time and improve the performance of the battery. In the cell evaluation, an expected enhancement of cell performance with reduced polarization phenomenon and improved cycling stability is observed after coating with polydopamine, which certifies the validity of the polydopamine coating.

      • Biochar-supported nZVI (nZVI/BC) for contaminant removal from soil and water: A critical review

        <P><B>Abstract</B></P> <P>The promising characteristics of nanoscale zero-valent iron (nZVI) have not been fully exploited owing to intrinsic limitations. Carbon-enriched biochar (BC) has been widely used to overcome the limitations of nZVI and improve its reaction with environmental pollutants. This work reviews the preparation of nZVI/BC nanocomposites; the effects of BC as a supporting matrix on the nZVI crystallite size, dispersion, and oxidation and electron transfer capacity; and its interaction mechanisms with contaminants. The literature review suggests that the properties and preparation conditions of BC (e.g., pore structure, functional groups, feedstock composition, and pyrogenic temperature) play important roles in the manipulation of nZVI properties. This review discusses the interactions of nZVI/BC composites with heavy metals, nitrates, and organic compounds in soil and water. Overall, BC contributes to the removal of contaminants because it can attenuate contaminants on the surface of nZVI/BC; it also enhances electron transfer from nZVI to target contaminants owing to its good electrical conductivity and improves the crystallite size and dispersion of nZVI. This review is intended to provide insights into methods of optimizing nZVI/BC synthesis and maximizing the efficiency of nZVI in environmental cleanup.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Aggregation and passivation of nZVI can be alleviated by surfactants and doping methods. </LI> <LI> BC hinders corrosion and improves the dispersion and electron transfer of nZVI. </LI> <LI> Properties of nZVI depend on those of the BC, feedstock and pyrogenic temperature. </LI> <LI> BC enhances electron transfer from nZVI to the contaminants due to the presence of quinone and graphene moieties. </LI> <LI> nZVI/BC shows strong ability to remove HMs, nitrates, and organic contaminants in soil and water. </LI> </UL> </P>

        Risk Factors for Intraprocedural Rerupture during Embolization of Ruptured Intracranial Aneurysms

        Background: Intraprocedural rupture (IPR) is a devastating complication in endovascular treatment of ruptured intracranial aneurysms, but its risk factors have not been fully assessed. This study was performed to explore the risk factors for IPR during embolization of ruptured cerebral aneurysms. Methods: A total of 1,494 patients with ruptured intracranial aneurysms who underwent endovascular interventional embolization were enrolled. Clinical characteristics were collected for each patient. Univariate and multivariate logistic regression analysis was employed to identify the factors independently associated with IPR. A receiver operating characteristic (ROC) curve analysis was performed to determine the cutoff values of continuous variables predicting IPR. Results: Forty-one patients suffered from IPR (2.7%). Multivariate logistic regression analysis indicated that aneurysm size (odds ratio [OR], 0.819; 95% confidence interval [CI], 0.732–0.916), aneurysms with irregular morphology (OR, 2.162; 95% CI, 1.143–4.091), time from symptom onset to intervention (OR, 1.615; 95% CI, 1.207–2.161), and vasospasm during embolization (OR, 2.021; 95% CI, 1.038–3.934) were the independent risk factors of IPR. ROC curve analysis showed that the area under the curve for aneurysm size and time from onset to intervention were 0.697 (cutoff value, 3.4 mm; sensitivity, 78.8%; and specificity, 53.7%) and 0.659 (cutoff value, 2 days; sensitivity, 78.0%; and specificity, 45.2%), respectively. Conclusion: Aneurysms with irregular morphology, aneurysms ≤ 3.4 mm in diameter, time from onset to intervention > 2 days and cerebral vasospasm during embolization are independent risk factors for IPR during coil embolization of ruptured aneurysms. More attention should be paid to the factors increasing the risk of IPR in patients with ruptured aneurysms so as to minimize this complication.

        Clinical Impact of Drug Adherence of Tyrosine Kinase Inhibitors in Children with Ph-Positive Acute Lymphoblastic Leukemia

        Purpose This study aimed to explore the impact of ABL1–tyrosine kinase inhibitors (TKIs) adherence on the survival of chromosome-positive (Ph+) acute lymphoblastic leukemia (ALL) children and clarify the potential predictors of patients’ prognosis from TKIs intake practices.Materials and Methods Ninety newly diagnosed Ph+ ALL patients who received TKIs were enrolled. We collected the baseline characteristics and adverse events in all children; moreover, TKIs adherence was measured by an eight-item Morisky medication adherence scale (MMAS-8). Progression-free survival (PFS) and overall survival (OS) analysis were performed, and risk factors for PFS and OS were evaluated.Results Among all patients, 69 cases were regarded as adherers, while 21 were non-adherers. The median duration of TKIs interruption was significantly prolonged in the non-adherence group than in the adherence group (13 [0-101] vs. 56 [11-128], p < 0.001). Additionally, dose reduction occurred in 55.2% of non-adherers versus 23.0% of adherers (p=0.002). The PFS and OS in adherers were significantly higher versus non-adherers (p=0.020 and p=0.039). MMAS-8 score was an independent risk factor for PFS (p=0.010) and OS (p=0.031). Among non-adherers, the median OS was only 23.1% (4.2%-42%) in patients aged ≤ 10 years versus 54.4% (38.8%-70%) in adolescents. Most of the patients who experienced TKIs non-adherence suffered pancytopenia.Conclusion TKIs adherence during treatment significantly influenced the survival of pediatric Ph+ ALL patients, and non-adherers with age ≤ 10 years were more vulnerable to TKIs disruption. The cumulative TKIs dose should be especially emphasized to patients with age ≤ 10 years, which may result in an inferior achievement of relevant treatment milestones.
