한국인 정신분열병과 도파민 D5 수용체 유전자 연합연구

본 연구는 도파민D5 수용체 유전자와 정신분열병 병인과의 상관성을 밝히기 위하여 한국인 정신분열병 환자군 100명과 정상대조군 100명에서 다형성의 분포를 중합효소연쇄반응을 이용하여 연합연구를 시행하였다. 도파민 D5 수용체 유전자의 대립유전자는(CT/GT/GA)n의 반복으로 구성된 154, 152, 150, 148, 146, 144, 142, 140, 138, 134염기 쌍을 가진 11가지 종류였고 환자군과 정상대조군사이에 대립유전자의 분포는 통계적으로 유의한 차이를 보였다. 각각의 대립유전자 빈도는 환자군에서 대립유전자 A10이 유의하게 감소되어 있었다. 환자군과 정상대조군의 유전자형 분포는 두 군간에 유의한 차이는 없었다. 본 연구의 결과는 도파민 D5 수용체 유전자가 한국인 정신분열병 발생의 취약성에 기여할 가능성이 있다는 것을 제시하지만 본 연구의 결과만으로는 이 유전자부위를 정신분열병의 유전적 결정인자로 결론 내리기는 어려우며 앞으로 더 많은 연구대상을 이용하여 이 도파민 D5 수용체 유전자부위에 대해 본 연구결과를 확인하는 연구가 계속적으로 이루어져야 할 것으로 생각된다. Objective : This study was performed to define the genetic relationship between the microsatellite (CT/GT/GA)n polymorphism for the dopamine D5 receptor gene and schizophrenia. An association study in 100 schizophrenic patients and 100 normal controls of Korean was made by means of using polymerase chain reaction. Results : The microsatellite(D5(CT/GT/GA)n) had 11 alleles. These was a significant difference in the allele distribution between schizophrenic and normal controls(p〈0.05). In schizophrenic patients, the frequency of allele A10 was decreased. As to the genotype distribution, there was no difference in both groups. Conclusions : This finding suggests that dopamine D5 gene is likely to be related to the development of schizophrenia in Korea but with only this result, we cannot come to the conclusion that this genetic locus is the genetic determinant of schizophrenia. Further studies of dopamine D5 receptor genetic locus that can confirm this result should be made.

Endothelial progenitor cells (EPCs) and outgrowth endothelial cells (OECs) play a pivotal role in vascular regeneration in ischemic tissues; however, their therapeutic application in clinical settings is limited due to the low quality and quantity of patient-derived circulating EPCs. To solve this problem, we evaluated whether three priming small molecules (tauroursodeoxycholic acid, fucoidan, and oleuropein) could enhance the angiogenic potential of EPCs. Such enhancement would promote the cellular bioactivities and help to develop functionally improved EPC therapeutics for ischemic diseases by accelerating the priming effect of the defined physiological molecules. We found that preconditioning of each of the three small molecules significantly induced the differentiation potential of $CD34^+$ stem cells into EPC lineage cells. Notably, long-term priming of OECs with the three chemical cocktail (OEC-3C) increased the proliferation potential of EPCs via ERK activation. The migration, invasion, and tube-forming capacities were also significantly enhanced in OEC-3Cs compared with unprimed OECs. Further, the cell survival ratio was dramatically increased in OEC-3Cs against $H_2O_2$-induced oxidative stress via the augmented expression of Bcl-2, a pro-survival protein. In conclusion, we identified three small molecules for enhancing the bioactivities of ex vivo-expanded OECs for vascular repair. Long-term 3C priming might be a promising methodology for EPC-based therapy against ischemic diseases.

We, in this study, propose the effect of the sulfonated binder in electrochemical system. First, poly(3-methyl thiophene, P3MT) was polymerized by using FeC13 as both a dopant and an initiator. SEBS as a binder was sulfonated to enhance the electrochemical properties. The cathode was prepared to combine 3PhT: super-P: binder (10: 1: 1 wt.%) on Al mesh. The organic electrolyte and anode were used 1 M LiPF6 + PC/DMC and Li on Ni mesh, respectively. The specific discharge capacity of P3MT-SSEBS electrode is high as 80 mAh/g, while that of P3MT-SEBS or P3MT-PVDF one has lower values as 50 or 20 mAh/g, respectively. The reason for this is that the sulfonated SEBS acts as a binder as well as a dopant. In an advance, the effect of sulfonation promotes the miscibility, contact area and affinity.

<P>Notch is a transmembrane protein that acts as a transcriptional factor in the Notch signaling pathway for cell survival, cell death and cell differentiation. Notch1 and Fbw7 mutations both lead the activation of the Notch1 pathway and are found in the majority of patients with the leukemia T-ALL. However, little is known about the mechanisms and regulators that are responsible for attenuating the Notch signaling pathway through Fbw7. Here, we report that the serum- and glucocorticoid-inducible protein kinase SGK1 remarkably reduced the protein stability of the active form of Notch1 through Fbw7. The protein level and transcriptional activity of the Notch1 intracellular domain (Notch1-IC) were higher in SGK1-deficient cells than in SGK1 wild-type cells. Notch1-IC was able to form a trimeric complex with Fbw7 and SGK1, thereby SGK1 enhanced the protein degradation of Notch1-IC via a Fbw7-dependent proteasomal pathway. Furthermore, activated SGK1 phosphorylated Fbw7 at serine 227, an effect inducing Notch1-IC protein degradation and ubiquitylation. Moreover, accumulated dexamethasone-induced SGK1 facilitated the degradation of Notch1-IC through phosphorylation of Fbw7. Together our results suggest that SGK1 inhibits the Notch1 signaling pathway via phosphorylation of Fbw7.</P>

<B>ABSTRACT</B><P>Integrin-linked kinase (ILK) is a scaffold and protein kinase that acts as a pivotal effector in integrin signaling for various cellular functions. In this study, we found that ILK remarkably reduced the protein stability of Notch1 through Fbw7. The kinase activity of ILK was essential for the inhibition of Notch1 signaling. Notably, the protein level and transcriptional activity of the endogenous Notch1 intracellular domain (Notch1-IC) were higher in ILK-null cells than in ILK wild-type cells, and the level of endogenous Notch1-IC was increased by the blocking of the proteasome, suggesting that ILK enhances the proteasomal degradation of Notch1-IC. ILK directly bound and phosphorylated Notch1-IC, thereby facilitating proteasomal protein degradation through Fbw7. Furthermore, we found down-regulation of Notch1-IC and up-regulation of ILK in basal cell carcinoma and melanoma patients but not in squamous cell carcinoma patients. These results suggest that ILK down-regulated the protein stability of Notch1-IC through the ubiquitin-proteasome pathway by means of Fbw7.</P>

Background: Cutaneous warts are a common complaint to visit dermatologic clinic and its course is variable, ranging from spontaneous resolution to a chronic condition re-fractory to treatment. Objective: To evaluate the efficacy and safety of punch biopsy for cutaneous warts. Methods: Thirty-nine patients who received punch biopsy for warts were reviewed through charts and photos. Among them, 15 were matched with cryotherapy-only controls in terms of size and location of the wart. We compared the number and cost of treatments between the two groups. Results: Eleven of the total 39 patients were treated with cryotherapy in addi-tion to punch biopsy and the average number of treatments was 4.1±3.3 (mean±standard deviation). In a case-control study, the ratio value of cost was 2.9±3.6 in the experi-mental group and was 5.9±4.1 in controls (p<0.05). Con-clusion: Punch biopsies can decrease the number and cost of treatment by reducing the size of warts and inducing local in-flammation to accelerate resolution. Therefore, punch re-duction should be considered as a viable measure to treat warts. (Ann Dermatol 29(2) 200∼205, 2017)

온라인베팅은 누구나 언제 어디서나 접속이 가능하며 ICT기술의 발전에 따라 베팅의 편리성과 다양한 기회로 국내 불법 도박규모는 합법 사행산업의 4배 이상에 달하며, 불법 온라인베팅 시장 또한 가파르게 커지고 있다. 이에, 국내에서 운영되고 있는 체육진흥투표권과 전자복권 외에 다른 유형의 온라인베팅도 확대허용하고 관리해야 할 필요성이 제기되고 있다. 본 연구는 온라인베팅 확대허용에 대한 인식, 온라인베팅 확대허용 시 운영방안과 불법 온라인베팅 축소방안에 대한 인식을 온라인베팅 이용자, 중독자 및 일반인 대상으로 파악하였다. 분석결과, 온라인베팅 확대허용에 대한 긍정적・부정적 인식에 집단별 유의한 차이가 있는 것으로 검증되었으며, 합법 온라인베팅 종목으로 경주류 베팅 확대허용 시 운영방안에 대한 인식 차이도 의미 있는 차이를 보였다. 이를 통해 온라인베팅 확대허용으로 양성화하게 되면 불법 온라인베팅 이용자가 합법 온라인베팅 이용자로 전환하게 될 가능성과 건전하게 이용할 수 있는 가능성이 있다는 것이 확인된 만큼 온라인베팅 확대허용의 긍정적 효과를 기대할 수 있을 것이다. Since online betting occurs on online where anybody can access through internet, thus, the online gambling market has been expanding. Domestic illegal gambling market size is 4 times bigger than that of legal gambling market, and illegal online betting market is rapidly growing as well. Currently online gambling does exist domestically in form of Sports promotion voting rights, also known as, Toto and Proto and internet lottery. However, the need for other types of online gambling to be permitted, expanded and managed has been suggested. This research has analyzed online gambling users, online gambling addicts and general population’s awareness on expansion and operation of online gambling, contraction measures of illegal online gambling and operation measures in case of permitted expansion of online gambling. The result of this research’s analysis showed that each group has significantly different awareness level on positive and negative effects of permitting online gambling’s expansion. Also, it showed that each group has significantly different awareness on permitting expansion of racing betting as a legal gambling item. Since it is the proven fact that illegal online gambling users could turn into legal users and that users could use the service more positively if online gambling is legalized through allowance and expansion process; one could expect the positive effects from permitting expansion of online gambling.
