        Myricitrin exerts protective effect on retina in diabetic retinopathy via modulating oxidative stress expression of VEGF and apoptosis in experimental rats: a docking confirmation study

        Background The present study investigates the protective effect of myricitrin (MCT) on the retinal tissues in diabetic retinopathy of diabetic rats. Objective The diabetic rat model was created by treating the rats with streptozotocin along with high-energy diet. The blood glucose was evaluated by glucose estimation kit, and rats measuring glucose ≥ 12.00 mmol/L were considered diabetic and were selected for the study. The diabetes-induced rats were divided randomly as diabetic control rats (vehicle treated) (DB), diabetic rats MCT (100 mg/kg) (DB + MCT 100 mg/kg) and diabetic rats MCT 200 mg/kg (DB + MCT 200 mg/kg). Rats received MCT for 12 weeks via oral route daily. The rats after 12 weeks were sacrificed, and the eyes were removed. The retina tissue was evaluated for thickness, oxidative stress markers, levels of VEGF and Bax and Bcl-2. In silico docking analysis was done by Autodock Vina tools for target confirmation of MCT. Results MCT decreased the serum glucose levels and also improved the body weight in diabetic rats. MCT improved thickness and apoptosis of cells. It was also found that MCT decreased oxidative stress, and improved levels of VEGF in retinal tissues of diabetic rats. MCT also exerted anti-apoptotic activity as shown by over-expression of Bcl-2 and suppression of Bax in retinal tissues of rats. Docking study confirmed potential binding affinity of MCT with VEGF. Conclusions MCT could be a potential therapeutic molecule in treating diabetic retinopathy which could modulate glucose levels, oxidative stress and levels of VEGF in diabetic rats.

        Structures, biomimetic synthesis, and anti-SARS-CoV-2 activity of two pairs of enantiomeric phenylpropanoid-conjugated protoberberine alkaloids from the rhizomes of Corydalis decumbens

        (±)-Decumicorine A ( 1 ) and (±)- epi -decumicorine A ( 2 ), two pairs of enantiomeric isoquinoline alkaloids featuring a novel phenylpropanoid-conjugated protoberberine skeleton, were isolated and purifi ed from the rhizomes of Corydalis decumbens . The separation of (±)- 1 and (±)- 2 was achieved by chiral HPLC to produce four optically pure enantiomers. The structures and absolute confi gurations of compounds (−)- 1 , (+)- 1 , (−)- 2 , and (+)- 2 were elucidated by spectroscopic analysis, ECD calculations, and X-ray crystallographic analyses. The two racemates were generated from a Diels-Alder [4 + 2] cycloaddition between jatrorrhizine and ferulic acid in the proposed biosynthetic pathways, which were fully verifi ed by a biomimetic synthesis. Moreover, compound (+)- 1 exhibited an antiviral entry eff ect on SARS-CoV-2 pseudovirus by blocking spike binding to the ACE2 receptor on HEK-293T-ACE2 h host cells.

        Anti-toxoplasmosis Effects of Oleuropein Isolated from <i>Fraxinus rhychophylla</i>

        <P>Toxoplasmosis is a parasitic disease caused by <I>Toxoplasma gondii</I>, with very few therapeutic treatment options. Typically, the choices for treatment are pyrimethamine and sulfadiazine, however their utility is limited because of drug toxicity and serious side effects. For these reasons, new drugs with lower toxicity are urgently needed. In this study, the compound oleuropein isolated from <I>Fraxinus rhynchophylla</I> showed anti-<I>T. gondii</I> effects <I>in vitro</I> and <I>in vivo</I>. In Madin–Darby bovine kidney cells, the selectivity of oleuropein was 8.9, which was higher than sulfadiazine and pyrimethamine (3.8 and 2.5, respectively). In infected mice, the inhibition ratio of <I>T. gondii</I> in the peritoneal cavity was 55.4% compared to the negative control group after treatment with 300 mg/kg oleuropein. In addition, inhibitory effects on granuloma, apoptosis, necrosis and cyst-formation were shown in sections of spleen and liver. Oleuropein is therefore a potentially useful anti-<I>T. gondii</I> candidate for clinical application.</P>

        An empirical study on the determinants of air trade – The case of South Korea and Chinese provinces

        The air transportation industry is becoming an increasingly important growth factor for economic development as trade and logistics exchanges between South Korea and China continue to grow. The main objective of this article is to examine the factors that influence air trade between South Korea and 31 Chinese provinces from 2010 to 2019, with the aim of promoting the expansion of air trade between the two countries. The findings suggest that, with the exception of South Korea’s exports to the 31 Chinese provinces, the gross regional domestic product has a significantly positive effect. The effects of distance and personal income on total air trade, air imports and exports are notably negative and positive respectively. Furthermore, foreign direct investment has a positive impact only when South Korea exports to China. The study concludes that, enhancing the volume of air traffic between South Korea and China can be accomplished by directing investments towards inland areas that possess trade-potential, expanding airport infrastructure, and developing regional economic growth. Furthermore, it is suggested that emerging domestic markets should be under close observation due to the changes in the trade structure after the COVID 19.

        Investigation on the Absorption Kinetics of Chlorogenic Acid in Rats by HPLC

        Chlorogenic acid (ChA), a major phenolic compound in the Flos Lonicerae, is widely used in the traditional Chinese medicine practice. The purpose of this study is to report the pharmacokinetic parameters of ChA in rats after oral administration and explore its absorption profile briefly. A two-compartment model was proposed and validated through the program to explain the apparent triphasic phenomenon of ChA in rats after intragastric administration. A rapid absorption and a relatively slow distribution followed by a slower elimination phase were observed. At the administered doses of 200, 400 and 600 mg/kg, the values of absorption halflife (t1/2 Ka) were 10.23, 18.66 and 28.13 min. The values of distribution half-life (t1/2α) were 12.35, 31.04 and 39.19 min. And the values of elimination half-life (t1/2β) were 231.64, 337.23 and 420.81 min. The volume of distribution at the three doses were 55.26, 35.56, 32.22 L/kg, respectively. The AUC0→∞ (area under the concentration–time curve) was not proportional to the administered dose. In the range of the doses examined, the absorption pharmacokinetics of ChA in rats was based on nonlinear kinetics.

        Cloning, subcellular localization, and expression analysis of VvRING1a from grapevine

        Polycomb group (PcG) proteins within polycomb repressive complex 1 (PRC1) and PRC2 are signifi cant epigenetic regulationfactors involved in controlling plant developmental transitions. PRC2 catalyzes H3K27 trimethylation (H3K27me3),and PRC1 recognizes the site of H3K27me3 and catalyzes H2AK119 monoubiquitination to maintain transcription repression. PRC2 components are conserved from plants to animals and have been investigated for a long time. However, thecompositions and functions of PRC1 vary between animals and plants. Plant PRC1 has only been confi rmed in recent years. Very little research has been conducted on PcG in grapevine ( Vitis vinifera ‘Cabernet Sauvignon’). Here, we cloned the fulllengthcoding sequence of the VvRING1a gene, which is homologous with the Arabidopsis thaliana PRC1 core componentAtRING1a . We then analyzed the structure, subcellular localization, and expression pattern. The tissue-specifi c expressionanalysis showed that VvRING1a is expressed in various tissues, including in roots, stems, leaves, fl owers, buds, and fruits. This study will be a useful reference for further study of VvRING1a .

        Long noncoding RNA UCA1 inhibits ischaemia/reperfusion injury induced cardiomyocytes apoptosis via suppression of endoplasmic reticulum stress

        Background Ischemia heart disease is one of the major causes of death worldwide which often associated with tissue infarction and limit the recovery of function. Multiple factors involved in the I/R-induced cardiomyocyte dysfunction which were consistent with a role of oxidative stress and altered endothelium-dependent responses. However, the pathogenic mechanisms in I/R injury remain unclear. Materials and methods The H9C2 cells were in the ischaemia/reperfusion (I/R) condition. After I/R, the cells were transfected with or without adenovirus-urothelial carcinoma associated 1(Ad-UCA1). Then qRT-PCR analysis was performed to quantify mRNA expression of different treatment groups. Cell apoptosis rate was assessed using flow cytometry and ER stress biomarker expression were measured by immunoblotting. Intracellular and mitochondrial ROS generation were assayed by fluorescence microscope after staining with the DCFDA or MitoSOX. Results I/R conditions trigger lncRNAs UCA1 expression, cellular and mitochondria ROS production, resulting in cell apoptosis through the induction of oxidative and ER stress. Overexpression of UCA1 protects H9C2 cells from I/R-induced ER stress and cell apoptosis. Moreover, UCA1 might be a potential regulator in the protective effect of I/R‑induced oxidative stress and mitochondria dysfunction. Subsequently, ER stress inhibitor attenuated the effect of siUCA1 induced injury in H9C2 cells. Conclusion The expression of UCA1 against I/R induced oxidative stress and mitochondria dysfunction via suppression of endoplasmic reticulum stress. UCA1 might be a biomarker to improved diagnosis of I/R injury.

        The hydrogen storage nanomaterial MgH2 improves irradiation-induced male fertility impairment by suppressing oxidative stress

        Objective: This study aimed to reveal the protective effect of hydrogen storage nanomaterial MgH2 on radiationinduced male fertility impairment. Methods: The characterization of MgH2 were analyzed by scanning electron microscopy (SEM) and particle size analyzer. The safety of MgH2 were evaluated in vivo and in vitro. The radioprotective effect of MgH2 on the reproductive system were analyzed in mice, including sperm quality, genetic effect, spermatogenesis, and hormone secretion. ESR, flow cytometry and western blotting assay were used to reveal the underlying mechanisms. Results: MgH2 had an irregular spherical morphology and a particle size of approximately 463.2 nm, and the content of Mg reached 71.46%. MgH2 was safe and nontoxic in mice and cells. After irradiation, MgH2 treatment significantly protected testicular structure, increased sperm density, improved sperm motility, reduced deformity rates, and reduced the genetic toxicity. Particularly, the sperm motility were consistent with those in MH mice and human semen samples. Furthermore, MgH2 treatment could maintain hormone secretion and testicular spermatogenesis, especially the generation of Sertoli cells, spermatogonia and round sperm cells. In vitro, MgH2 eliminated the [·OH], suppressed the irradiation-induced increase in ROS production, and effectively alleviated the increase in MDA contents. Moreover, MgH2 significantly ameliorated apoptosis in testes and cells and reversed the G2/M phase cell cycle arrest induced by irradiation. In addition, MgH2 inhibited the activation of radiation-induced inflammation and pyroptosis. Conclusion: MgH2 improved irradiation-induced male fertility impairment by eliminating hydroxyl free radicals.

        QTL Mapping of Kernel Oil Content of Chromosome 6 in a High Oil Maize Mutant (Zea mays L.)

        An infrequent high oil maize mutant CE03005 (white seed color and high oil content) from ethyl methane sulphonate pollen mutagenesis was investigated in this research. A F2 population derived from CE03005×B73 was used to analysis the genetic effects influencing kernel oil content and to map kernel oil content QTL. Ears from three generations were harvested and the individual kernel oil content of each generation was measured with nuclear magnetic resonance. Genetic analysis showed that the average oil content of white kernels was significantly higher than that of yellow kernels. Genotypic and phenotypic analyses were compared using composite interval mapping. A QTL (oilc6-m1)associated with high kernel oil content was detected in the three generations at bin 6.01 of chromosome 6 near the phenotypic marker SC (seed color). The mapping results were congruent with the genetic analysis. The results explained the reason why the oil content of the white colored kernels was higher than that of the yellow ones because the oil content QTL linked tightly with the seed color gene in chromosome 6 in the mutant. The way of QTL mapping was credible. Molecular and phenotypic markers identified could be used for breeding purpose and would lend convenience in cloning QTL.
