        A clinical analysis was done for the 110 pediatric patients with positive urine culture during Jun. 1992 to Dec. 1995 retrospectively. The patients who were hospitalized were classified group Ⅰ, under 29 days of age, and group Ⅱ, number of 29 days of age to 15 years old, respectively. The diagnostic criteria of urinary tract infection were first, the numbers of colonies of single organism isolated from clean urine collected by catheter was over 100,000/㎖. second, the numbers of colonies of single organism isolated from by supurapubic aspiration was over 1,000/㎖. third, two or more cultures yielding more than 100,000 colonies of a single identical organism from obtained by urine bag after aseptic preparation of genitalia. The results are summarized as follows : 1. The mean age of group Ⅰ was 9 days and 2.7 years in group Ⅱ. 2. Male to female ratio of group Ⅰ was 2.4 : 1 and 1.7 :1 in group Ⅱ. 3. The most common symptom was fever in both group Ⅰ(37%) and group Ⅱ(62%). 4. The pyuria(>5WBC/hpf) was detected in 21 cases(51%) of group Ⅰ and 32 cases(46%) of group Ⅱ. 5. The bacteriuria was detected in 22 cases(55%) of group Ⅰ and 25 cases(36%) in group Ⅱ. 6. The most commonly cultured bacterial species were E. coli in both group. 7. The most sensitive antibiotics for cultured E.coli was amikin.

        Effects of Triterpenoid Glycosides from Fresh Ginseng Berry on SW480 Human Colorectal Cancer Cell Line

        Purpose The pharmacological activities, notably the anticancer properties, of bioactive constituents from fresh American ginseng berry have not yet been well studied. In this study, we investigated the antiproliferative effects of fresh American ginseng berry extract (AGBE) and its representative triterpenoid glycosides using the human colorectal cancer cell line SW480. Materials and Methods Using high performance liquid chromatography (HPLC), the contents of 8 ginsenosides in AGBE were determined. The cell growth inhibitory effects of AGBE and three triterpenoid glycosides (ginsenosides Rb3, Re, and Rg3) were evaluated by proliferation assay and 3H-thymidine incorporation assay. Cell cycle and apoptotic effects were analyzed by using flow cytometry after staining with propidium iodide and annexin V. Results HPLC analysis data showed that AGBE has a distinct ginsenoside profile. AGBE inhibited SW480 cell growth significantly in a time-dependent (24-96 hours) and concentration-dependent (0.1-1.0 mg/mL) manner. Ginsenosides Rb3, Re, and Rg3 also possess significant antiproliferative activities on SW480 cells. 3H-thymidine incorporation assay indicated that AGBE and ginsenosides Rb3, Re, and Rg3 might inhibit the transferring and duplication of DNA in SW480 cells. Flow cytometric assay data suggested that AGBE arrested SW480 cells in S and G2/M phases, and significantly induced cell apoptosis. Conclusion AGBE and ginsenosides Rb3, Re, and Rg3 possessed significant antiproliferative effects and induced changes of morphological appearance on SW480 cells. The mechanisms of the antiproliferation of AGBE and tested ginsenosides involved could be cell cycle arrest and induction of apoptosis.

        Heterologous expression of two Physcomitrella patens group 3 late embryogenesis abundant protein (LEA3) genes confers salinity tolerance in arabidopsis

        Salinity stress is a major limiting factor in agriculture and adversely affecting the whole plant. As a halophyte, the moss Physcomitrella patens, has been suggested to be an ideal model plant to study salinity tolerance and adaption. Two abiotic stress-responsive Group 3 Late Embryogenesis Abundant protein genes had been identified in P. patens and named as PpLEA3-1 and PpLEA3-2, respectively. Functions of these two genes were analyzed by heterologous expressions in Arabidopsis, driven either by their native P. patens promoters or by the 35S CaMV constitutive promoter. Phenotype analysis revealed that pLEA3::LEA3, pLEA3::LEA3::GFP and 35S::LEA3::GFP transgenic lines had stronger salinity resistance than that in the wild type and empty-vector control. Further analysis showed that the contents of proline and soluble sugar were increased and the malondialdehyde (MDA) were repressed in these transgenic plants after exposure to salinity stress. Our observations indicate that these two Group 3 PpLEA genes played a role in the adaption to salinity stress.

        MRTF-A-NF-κB/p65 axis-mediated PDL1 transcription and expression contributes to immune evasion of non-small-cell lung cancer via TGF-β

        PD-L1 is abnormally regulated in many cancers and is critical for immune escape. Fully understanding the regulation of PD-L1 expression is vital for improving the clinical efficacy of relevant anticancer agents. TGF-β plays an important role in the low reactivity of PD-1/PD-L1 antibody immunotherapy. However, it is not very clear whether and how TGF-β affects PD-L1 expression. In the present study, we show that TGF-β upregulates the expression of the transcriptional coactivator MRTF-A in non-small-cell lung cancer cells, which subsequently interacts with NF-κB/p65 rather than SRF to facilitate the binding of NF-κB/p65 to the PDL1 promoter, thereby activating the transcription and expression of PD-L1. This leads to the immune escape of NSCLC cells. This process is dependent on the activation of the TGF-β signaling pathway. In vivo, inhibition of MRTF-A effectively suppresses the growth of lung tumor s y ngrafts with enrichment of NK and T cells in tumor tissue. Our study defines a new signaling pathway that regulates the transcription and expression of PD-L1 upon TGF-β treatment, which may have a significant impact on research into the application of immunotherapy in treating lung cancer.

        The Impact of Gate Leakage Current on PLL in 65 ㎚ Technology

        For CMOS technology of 65 nm and beyond, the gate leakage current can not be negligible anymore. In this paper, the impact of the gate leakage current in ring voltage-controlled oscillator (VCO) on phaselocked loop (PLL) is analyzed and modeled. A voltage-to-voltage (V-to-V) circuit is proposed to reduce the voltage ripple on Vctrl induced by the gate leakage current. The side effects induced by the V-to-V circuit are described and optimized either. The PLL design is based on a standard 65 ㎚ CMOS technology with a 1.8 V power supply. Simulation results show that 97 % ripple voltage is smoothed at 216 ㎒ output frequency. The RMS and peak-to-peak jitter are 3 ps and 14.8 ps, respectively

      • Tumor Markers for Diagnosis, Monitoring of Recurrence and Prognosis in Patients with Upper Gastrointestinal Tract Cancer

        To evaluate the value of combined detection of serum CEA, CA19-9, CA24-2, AFP, CA72-4, SCC, TPA and TPS for the clinical diagnosis of upper gastrointestinal tract (GIT) cancer and to analyze the efficacy of these tumor markers (TMs) in evaluating curative effects and prognosis. A total of 573 patients with upper GIT cancer between January 2004 and December 2007 were enrolled in this study. Serum levels of CEA, CA19-9, CA24-2, AFP, CA72-4, SCC, TPA and TPS were examined preoperatively and every 3 months postoperatively by ELISA. The sensitivity of CEA, CA19-9, CA24-2, AFP, CA72-4, SCC, TPA and TPS were 26.8%, 36.2%, 42.9%, 2.84%, 25.4%, 34.6%, 34.2% and 30.9%, respectively. The combined detection of CEA+CA199+CA242+CA724 had higher sensitivity and specificity in gastric cancer (GC) and cardiac cancer, while CEA+CA199+CA242+SCC was the best combination of diagnosis for esophageal cancer (EC). Elevation of preoperative CEA, CA19-9 and CA24-2, SCC and CA72-4 was significantly associated with pathological types (p<0.05) and TNM staging (p<0.05). Correlation analysis showed that CA24-2 was significantly correlated with CA19-9 (r=0.810, p<0.001). The levels of CEA, CA19-9, CA24-2, CA72-4 and SCC decreased obviously 3 months after operations. When metastasis and recurrence occurred, the levels of TMs significantly increased. On multivariate analysis, high preoperative CA72-4, CA24-2 and SCC served as prognostic factors for cardiac carcinoma, GC and EC, respectively. combined detection of CEA+CA199+CA242+SCC proved to be the most economic and practical strategy in diagnosis of EC; CEA+CA199+CA242+CA724 proved to be a better evaluation indicator for cardiac cancer and GC. CEA and CA19-9, CA24-2, CA72-4 and SCC, examined postoperatively during follow-up, were useful to find early tumor recurrence and metastasis, and evaluate prognosis. AFP, TPA and TPS have no significant value in diagnosis of patients with upper GIT cancer.

        In this paper, we study the eigenvalue problem of Schr$\ddot{o}$dinger-type operator on bounded domains in strictly pseudoconvex CR manifolds and obtain some universal inequalities for lower order eigenvalues. Moreover, we will give some generalized Reilly-type inequalities of the first nonzero eigenvalue of the sub-Laplacian on a compact strictly pseudoconvex CR manifold without boundary.

        The Investigation of Cell Culture Conditions to Maintain Chicken Embryonic Stem Cells as Totipotent Cells

        The ES cell can provide a useful system for studying differentiation and development in vitro and a powerful tool for producing transgenic animalds. To investigate the culture condition of chicken embryonic stem (CES) cells which can retain their multipotentiality or totipotency, three kinds of feeder layer cells, SNL cells, primary mice embryonic fibroblasts (PMEF) cells and primary chicken embryonic fibroblasts (PCEF) cells, were used as the feeder cells in media of DMEM supplemented with leukemia inhibitory factor (LIF), basic fibroblast growth factor (bFGF) and stem cell factor (SCF) for co-culture with blastoderm cells from stage X embryos of chicken. The alkaline phosphatase (AKP) test, differentiation experiment in vitro and chimeric chicken production were carried out. The results showed that culture on feeder layer of PMEF yielded high quality CES cell colonies. The typical CES cells clone shape revealed as follows: nested aggregation (clone) with clear edge and round surface as well as close arrangement within the clone. Strong alkaline phosphatase (AKP) reactive cells were observed in the fourth passage cells. On the other hand, the fourth passage CES cells could differentiate into various cells in the absence of feeder layer cells and LIF in vitro. The third and fourth passage cells were injected into the subgerminal cavity of recipient embryos at stage X. Of 269 Hailan embryos injected with CES cells of Shouguang Chickens, 8.2% (22/269) survived to hatching, 5 feather chimeras had been produced. This suggests that an effective culture system established in this study can promote the growth of CES cells and maintain them in the state of undifferentiated and development, which lays a solid foundation for the application of CES cells and may provide an alternative tool for genetic modification of chickens.
