        An alternative method for internal stability prediction of gravelly soil

        Experimental observations have shown that geometric characteristics of the soil have significant impact on the internal stability. Due to the diversity of grain composition, the degree of correlation between geometric control variables and internal stability fordifferent types of soil may not be the same. Based on the statistical information of 140 different soils collected from the literature, analternative method is proposed to assess the internal stability of gravelly soil with the maximum grain size reaching 60 mm. According to the values of d90, all the 140 soils are first classified into three clusters through hierarchical clustering method. Averagevalues of d90 for cluster I, II and III are 8.16 mm, 58.66 mm and 152.33 mm, respectively. Then, the experimental results are taken asdichotomous variable (0 refers to internally unstable and 1 refers to internally stable). Point-biserial correlation coefficient isintroduced to measure the correlation between geometric control variables and internal stability of the soils. The results show that Cu(coefficient of uniformity) and (d15c/d85f)max of cluster II present the highest degree of correlation with internal stability of soils amongthe three clusters. Therefore, an assessing method is suggested which unites two control variables (Cu and (d15c/d85f)max) to identify theinternal stability of gravelly soil, and the boundary of the method is proposed as well. At last, laboratory tests are carried out to assessthe internal stability of 8 gravelly soil samples which were gathered locally. The proposed method, together with five commonly usedcriteria, is used for the prediction of internal stability. By comparing the predictive results with the test results, the reliability of thesuggested method is verified.

        Neoadjuvant Chemotherapy-Guided Bladder-Sparing Treatment for Muscle-Invasive Bladder Cancer: Results of a Pilot Phase II Study

        Purpose Reduced quality of life after cystectomy has made bladder preservation a popular research topic for muscle-invasive bladder cancer (MIBC). Previous research has indicated significant tumor downstaging after neoadjuvant chemotherapy (NAC). However, maximal transurethral resection of bladder tumor (TURBT) was performed before NAC to define the pathology, impacting the real evaluation of NAC. This research aimed to assess real NAC efficacy without interference from TURBT and apply combined modality therapies guided by NAC efficacy. Materials and Methods Patients with cT2-4aN0M0 MIBC were confirmed by cystoscopic biopsy and imaging. NAC efficacy was assessed by imaging, urine cytology, and cystoscopy with multidisciplinary team discussion. Definite responders (≤ T1) underwent TURBT plus concurrent chemoradiotherapy. Incomplete responders underwent radical cystectomy or partial cystectomy if feasible. The primary endpoint was the bladder preservation rate. Results Fifty-nine patients were enrolled, and the median age was 63 years. Patients with cT3-4 accounted for 75%. The median number of NAC cycles was three. Definite responders were 52.5%. The complete response (CR) was 10.2%, and 59.3% of patients received bladder-sparing treatments. With a median follow-up of 44.6 months, the 3-year overall survival (OS) was 72.8%. Three-year OS and relapse-free survival were 88.4% and 60.0% in the bladder-sparing group but only 74.3% and 37.5% in the cystectomy group. The evaluations of preserved bladder function were satisfactory. Conclusion After stratifying MIBC patients by NAC efficacy, definite responders achieved a satisfactory bladder-sparing rate, prognosis, and bladder function. The CR rate reflected the real NAC efficacy for MIBC. This therapy is worth verifying through multicenter research.

        Static and Seismic Experimental Study of Novel Prefabricated Beam-Column Joints with Elongated-Hole Brackets

        To enhance the structural connectivity of prefabricated steel frame systems and augment their construction effi ciency, this study introduces an innovative prefabricated joint design tailored for square steel columns and H-beams characterized by varying beam heights. This study includes both static loading tests and seismic tests performed on full-scale joints featuring two diff erent beam heights. This investigation involved a comprehensive analysis of the static and seismic performance of the joints, employing various performance metrics such as ultimate load capacities, ultimate rotation angles, hysteresis curves, skeleton curves, stiff ness degradation curves, and ductility coeffi cients, in alignment with established structural codes and standards. The results indicate that the plasticity of the H-beam is fully developed, exhibiting a relative slip phenomenon. Additionally, the joints demonstrate commendable rotational capacity, with hysteresis curves consistently manifesting an inverse S-shape and exhibiting noteworthy stiff ness degradation. Furthermore, the comparison with the unimproved joint shows that the novel joint, in addition to being easy to construct, has better ductility and energy dissipation capacity. The results of the study will provide a technical reference for further optimization and application of prefabricated beam-column joints.

        Identification of MicroRNAs as potential biomarkers for detecting ischemic stroke

        Background: Increasing epidemic of ischemic stroke (IS) makes it urgent to understand the pathogenesis and regulatory mechanism, previous studies have described microRNAs (miRNAs) is part of the brain's response to ischemia. Objective: The aim of this study was to screen potential biomarkers for the prediction and novel treatment of IS. Methods: Differentially expressed miRNAs were screened from three newly diagnosed IS patients and three controls by RNA sequencing technology. Furthermore, target prediction databases were then used to analysis the target genes of different expressed miRNAs, and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway database were used to identify the functions and the main biochemical and signal pathways of differentially expressed target genes. Results: Our results revealed that 27 miRNAs were differentially expressed in IS, among which, hsa-miR-659-5p was the most highly increased and was first found to be associated with IS. In addition, KEGG pathway analyses showed that differentially expressed miRNAs were mainly significantly enriched in lysosome pathway, cytokine-cytokine receptor interaction pathway, spliceosome pathway, base excision repair pathway. Conclusions: miRNAs were involved in IS pathogenesis, and hsa-miR-659-5p, hsa-miR-151a-3p and hsa-miR-29c-5p as the three highest |log2FoldChange| regulation in this study, which may be the biomarkers of IS and need further study.
