      • 소 수정란의 공배양 및 동결이 체외발생에 미치는 영향에 관한 연구

        The studies on the carried out to investigate the effects of co-culture with cumulus cells and oviduct epithelial cells on the in vitro fertilization and cleavage rate of bovine follicular oocytes, and to determine the optimum thawing temperature and equilibration time of frozen bovine embryos and on survival rate and in vitro developmental rate of bovine embryos. The ovaries were obtained from slaughtered Korean native cows. The follicular oocytes were cultured in TCM-199 medium containing 10 IU/㎖의 PMSG(Sigma, USA), 10 IU/㎖의 hCG, 1 ㎍/㎖의 β-estradiol(Sigma, USA) and 10% FCS(Sigma, USA) for 24-48 hrs in incubator with 5% CO_2 in air at 38.5℃ and then matured oocytes were again cultured for 12-18 hrs with motile capacitated sperm by preincubation of heparin. The bovine embryos following dehydration by cryoprotective agents and a various concentrations of sucrose were directly plunged into liquid nitrogen and thawed in 30℃ water. The results are summarized as followes : 1. The in vitro fertilization and in vitro developmental rates of bovine oocytes co-cultured with cumulus cells in TCM-199 media were 75.0%-76.8% and 17.3%-27.6%, respectively. And in-vitro fertilization rates of cumulus-enclosed oocytes(55.4%) were significantly(p<0.05) higher than cumulus-denuded oocytes(23.1%). 2. The in vitro fertilization and in vitro developmental rates of bovine oocytes co-cultured with 1 ×10^4 cells/㎖, 1 ×10^6 cells/㎖, 1 ×10^8 cells/㎖ and 1 ×10^15 cells/㎖ oviduct epithelial cells in TCM-199 media were 74.5%-77.8% and 15.7%-21.2%, respectively. 3. The in-vitro fertilization and in vitro developmental rates of bovine oocytes co-cultured in TCM-199 media containing PMSG, hCG, PMSG + hCG, PMSG + β-estradiol, hCG + β-estradiol 0 to 40 hrs after insemination were 74.0%-77.4% and 18.9% - 23.1, respectively. 4. The survival rates of bovine embryos thawed after rapid freezing in the media containing various kinds of cryoprotective agents added 0.25M and 0.50M sucrose were 14.7% - 35.1% and 17.6% - 31.6%, respectively. The survival rates of bovine embryos thawed after rapid freezing in the freezing media containing a various concentration of sucrose added 1.5M and 2.0M glycerol, 1.5M and 2.0M DMSO, and 1.5M and 2.0M propanediol were 23.5% -31.4% and 20.6% - 34.1%, respectively. 5. The temperature thawed at 30℃ after rapid freezing of bovine embryos resulted in a significantly higher embryos survival rate than did at 20℃ and 35℃. 6. The equilibration time on the survival rates of bovine embryos was attained after short period of time(2.5-5 min.) in the freezing medium higher than ling period of time(10-20min.).

        1년간의 자발적인 운동참여가 중년 남성 제2형 당뇨병 환자의 혈중 지질 성분에 미치는 영향

        The purpose of this study is to investigate the effects of voluntary exercise participation on blood lipids in type II diabetes mellitus middle-aged men. Eighteen male volunteers were recruited and exercised, jogging, cycle ergometer, swimming, in the sports center for 12 months without any supervision. They had educated heart rate calculation method to keep their target heart rate during exercise. Before and after the training, their body composition, maximal oxygen uptake, and blood lipids profiles were analyzed. Body weight was not changed during experimental periods; however body fat ratio was statistically decreased at the same periods. Blood glucose, TC, LDL-C, and TG were decreased significantly and HDL-C was significantly increased (p<.05). RQ was significantly lower compared to the before exercise and their maximal oxygen consumption and exercise time to fatigue were increased significantly (p<.05). These results suggested that voluntary exercise with simple exercise prescription was also effectual to enhance blood lipids in type II diabetes mellitus middle-aged men.

        낭성법랑모세포종, 함치성낭, 치성각화낭의 방사선소견과 Ki-67, PCNA, Cytokeratin 발현과의 연관성에 관한 연구

        Purpose : To compare the proliferation potential of the epithelial cells between unicystic ameloblastoma (UA), dentigerous cyst (DC), and odontogenic keratocyst (OKC) and to correlate this proliferation potential with the radiographic features of these three pathoses. Materials and Methods : Immunohistochemical expression of PCNA, Ki-67, and cytokeratin as a proliferation marker were assessed for 15 cases of UA, 15 cases of DC, and 15 cases of OKC. The degree of immunochemical expression of three proliferation markers were correlated with the radiographic features, especially cortical expansion (negative and positive) and shape of border (scalloped and round). Results : Using PCNA and Ki-67, OKC showed the highest proliferation potential and UA the lowest. Statistically significant differences were found between the OKC and the UA (p.0.05). However, no statistically significant difference was present according to the radiographic features in all pathoses. Using cytokeratin, there was no significant differences of proliferation potential among three pathoses. Conclusions : OKC epithelium has the most intense proliferation potential, followed by the dentigeous cyst and then unicystic ameloblastoma. There is no significant relation between the radiographic features and the proliferation potential of epithelium of these three pathoses.

        건진센터 수검자 자료를 이용한 비만지표에 대한 연구

        We have examined l5145 persons(7468 men 7677 women) to determine the role of Obesity Index(body mass index, WHR, body fat percent) on the risk of various chronic disease. All the subjects were pooled from Health Examination Center who took the annual health examination from June, 1994 through Jan, 1997, BMI(Body mass index, kg/㎡), WHR(Waist hip ratio) were measured and correlated with the frequency of chronic disease. In order to evaluated the BMI, WHR, Body Fat as an independent factors on rnorbidity, analysis of covariance was applied adjusted for age. The risk of variows chronic diseases were also analyzed with obesity, defined by BMI above 26kg/㎡ (men), 25kg/㎡ (women), by logistic regression adjusted age and sex. Risk of hypertension was 2.6 with 95% eonfidence interval of 1.9-2.8 in obese group. Risk of diabetes mel1itus, hypercholesterolemia, hypertriglyceridemia, gall stone, fatty liver were 3.2(2.3-4.4), 1.2(0.9-1.4), 2.1(1.7-2.8), 1.4(1.1-2.6) and 1.3(0.8-2.3) respectiveiy. The risk of various chronic diseases were also analyzed with obesity, WHR above 0.9(men), 0.81(women), by logistic regression adjusted age and sex. Risk of hypertension was 1.7 with 95% confidence interval of 1.4 -2.1 in obese group. Risk of diabetes mellitus, hypercholesterolemia, hypertriglyceridemia, gall stone, fatty liver were 3.1(2.3-4.0), 1.2(0.9-l.4), 1.4(1.1-2.6), 2.4(l.8-4.8) and 2.1(1.2-4.1) respectively. The BMI, WHR were independent risk factors for various chronic diseases.

        We report a case multiple piloleiomyoma in a 50-year-old man. About 30 years ago, multiple red colored firm papules with pain developed on the left lower leg. The skin lesions gradually increased in number and in and size arranging in a group. Some, lesion became more painful after cold exposure. About a few years ago, new lesions developed on the left back and flank abdomen, The histologic findings showed poorly demarcated and were composed of interlacing bundles of smooth muscle fibers with varying amounts of collagen bundles being intermingled. Masson`s trichrome and smooth muscle actin stain showed typical leiomyoma, and s-100 stains revealed an increased amount of nerve fibers interlacing within muscle fibers and the surrounding tissue. We diagnosed our case as unilateral multiple piloleiomyoma arising from arrector pilorum muscle. Pain was ameliorated with oral nifedipine alone.

      • 센서응용을 이한 CMOS열전대의 공정기술 개발

        마이크로 열전센서는 마이크로히터와 열전소자로 구성된다. 열전도율이 낮고 내부응력이 최소화된 NON절연막 다이아프램을 LPCVD법, APCVD법 및 실리콘 이방성식각법을 각각 활용하여 형성하였다. Si_3N_4 및 SiO_2의 두께가 각각 1500Å 및 3000Å의 조합인 NON막이 가장 양호한 응력특성을 나타내었다. 고주파 스퍼터링법으로 형성된 백금박막을 이용한 히터와 온도감지기로 형성된 마이크로 가스유량센서를 제조하고 그 동작특성을 측정하였다. 유량센서의 동작특성은 100∼500sccm의 유량범위에서 양호한 선형성을 보였다. CMOS공정기술과 호환성을 갖는 마이크로 유량센서의 마스크패턴과 공정을 각각 설계하였다. A micro thermosensor is composed of a microheater and a thermo element. NON insulator diaphragm with low thermal conductivity and minimum internal stress was fabricated^by using LPCVD, APCVD and silicon anisotropic etch techniques. The deposited NON film represented the best stress characteristics when the thickness of Si_3N_4 and SiO_2 are 1500Å and 3000Å, respectively. A micro gas-flow sensor was fabricated and characterized that is consist of platinum thin film heater and temperature detector. The performance characteristics of the micro gas-flow sensor shows a good linearity within 100-500sccm flow rate range. Mask pattern and CMOS process of micro gas-flow sensor have been designed according to CMOS process technology.

      • β-Carotene 이 혈중 효소활성도에 미치는 영향

        This study was carried out in order to observe the effects of ethanol and β-carotene on the cholesterol, Triglyceride, LDL-cholesterol, HDL-cholesterol, glutathione peroxidase and catalase activities. The normal white S.D rats were classified into 4 group: 0.9% NaCl administration and 20% ethanol administration were referred to as group A, 20% ethanol and 50% β-carotene mixture administration was group B and 50% β-carotene administration was group C for the 30 day-feeding experiment. The results were obtained as following: 1. The blood lipid concentration was most highly increased in group A, was almost normal in group B, but significantly decreased in goup C. 2. The blood antioxidant enzyme activities were significantly decreased in group A, but significantly increased in goup B and C. 3. The blood β-carotene concentration were significantly decreased in group A, and slightly decreased in group B. Surprisingly it was increased in group C.

      • 담배 박각시나방 Manduca sexta의 계획된 세포죽음동안의 Proteasome의 변화

        모든 진핵세포의 주요 단백질 분해요소인 proteasome은 변성되거나 상해된 불필요한 단백질을 제거할 뿐만 아니라, 세포주기를 비롯한 다양한 대사과정들의 주요 효소들을 필요 시 적절히 분해함으로써 항상성을 유지시키는 필수적인 효소로 잘 알려져 있다. 또한 proteasome은 특정 포유동물 조직의 계획된 세포죽음(programmed cell death 또는 apoptosis)와 밀접하게 관련되어 있다고 보고된 바, 본 연구에서는 담배 박각시나방(Manduca sexta)의 변태과정에서 일어나는 마디간 근육(intersegmental muscle)의 급격한 파괴과정동안의 proteasome의 변화양상을 발현 및 단백질 분해활성을 중점으로 조사하였다. 정상근육과 파괴직전의 근육으로부터 proteasome을 분석한 결과 단백질 분해활성이 최소한 6배 이상 증가하였으며, proteasome의 주요 구성성분들의 발현 변화를 각각의 항체(antibody)를 이용하여 조사한 결과 세포죽음 직전에 급격히 증가함을 보였다. 이러한 구성성분들의 발현증가가 proteasome 복합체 형성을 통해 활성의 증가를 일으키는지 조사한 결과, 세포내에서 실질적으로 단백질 분해활성을 담당한다고 여겨지는 26S proteasome(2,000 kDa)의 위치에서 커다란 활성의 변화와 구성성분의 증가를 볼 수 있었다. 따라서, 담배 박각시나방의 계획된 세포죽음 현상은 특정 호르몬의 신호에 의해 proteasome의 발현이 급격히 증가하여 26S proteasome의 복합체 형태로 세포 내 대부분의 단백질들을 분해함으로써 30시간이내 근육조직을 완전히 소멸시키는 것을 알 수 있었다. 한편, proteasome을 구성하는 특정 구성성분의 과다 발현이 세포의 단백질 분해현상에 어떠한 영향을 미치는 지 조사하기 위하여 쥐의 fibroblast cell에 26S proteasome의 ATPase 구성성분인 m56 유전자를 삽입한 후 세포의 추출물로부터 proteasome의 양과 활성의 변화를 관찰하였다. Proteasome을 초원심분리로 추출하여 관찰한 결과, 담배 박각시나방의 경우와는 달리 유전자 삽입 세포는 모든 실험에서 정상 세포와 전혀 차이를 보이지 않았다. 이 결과로부터 세포의 생명 유지에 필수적이고 매우 다양한 기능을 가진 proteasome은 40여개의 구성성분들의 발현이 매우 정교하게 조절되고 있으며, 특정 세포죽음 자극(apoptotic signal)이 왔을 때 일련의 구성 성분들의 발현과 26S proteasome 복합체 형성이 증가함으로써 그 기능을 수행하는 것을 알 수 있으며, 보다 자세한 연구는 비정상적 단백질 분해나 세포죽음을 동반하는 여러 질환들의 원인과 현상을 규명할 수 있으며, 또한 그 활성을 조절함으로써 궁극적인 치료의 방향도 제시 가능할 것으로 기대된다.

      • 저에너지 사료 급여가 거세 비육돈의 성장과 도체 품질에 미치는 영향

        Barrows eat excessively more feed than gilts and as a result, their carcass grades are, in general, lower than those of the latter. The present study was conducted to investigate the effect of a low-energy diet on growth and carcass quality in finishing barrows and thereby to draw a guideline for the usage of it. In experiment I(Exp. I), 60 LYYD cross-bred barrows weighing ~70kg were randomly assigned into four pens and fed ad libitum either of two finishing diets containing 103%(control diet) and 87%(low-energy diet) of the NRC digestible energy(DE) requirements, respectively, up to slaughter at ~105kg. Exp. Ⅱ was same as Exp. Ⅰ except that average initial weight of the barrows(48 heads) was ~60kg and feed intake was measured. In Exp. I, by feeding the low-energy diet, average daily gain(ADG) was decreased(P<0.05); backfat thickness corrected for 110kg of live weight tended to be decreased(P=0.11); and the ratio of A,B grade carcass was not apparently affected[93%(low-energy diet) vs 90%(control)]. In Exp. II, as in Exp. I, ADG was decreased by feeding the low-energy diet vs control, which is interpreted to have resulted primarily from a 14% decrease in total DE intake. Compared with control, low-energy diet group had a similar average daily feed intake(3.10 vs 3.04kg) and a 14% higher feed/gain. In somewhat contrast to the result of Exp. I, backfat thickness corrected for the live weight was not different between the two groups in Exp. II, which is presumed to have resulted mainly from a genetic ununiformity of the animals with respect to this trait. The A,B carcass ratio, on the other hand, was not appreciably different between the two groups(73 vs 83%). In summary, under the current domestic carcass grading system allowing a relatively wide range of backfat thickness for A and B grades, a low-energy diet is considered to be useful only for a barrow population where the A, B carcass ratio is too low for excessive fackfat. More studies are needed, however, to determine whether a low-energy diet will be useful under the situation of international pork market demanding a higher lean meat ratio.

        As the exiting school facilities become old and unfit for new curriculum, the systematical remodeling process based on its educational and physical functions is required, due to the inadequate maintenance and repairing practices, unnecessary costs are expended. Therefore, this study aims to establish a remodelling decision-making model for improving the educational environment of the existing school facilities. According to the budget system, it proposes the concept of remodelling that includes the activities of extension, reconstruction, repair and improvement. This study classifies the performance evaluation for school facilities as that of safety, durability and educational function, and articulates the assessment standards, methods and elements. In the end, it suggests a rational model for remodelling decisions making that can provide efficient and comprehensive remodelling process, economic and sustainable school development.
