Insulin-positive ductal cells do not migrate into preexisting islets during pregnancy

The adult pancreatic ductal system was suggested to harbor facultative beta-cell progenitors similar to the embryonic pancreas, and the appearance of insulin-positive duct cells has been used as evidence for natural duct-to-beta-cell reprogramming. Nevertheless, the phenotype and fate of these insulin-positive cells in ducts have not been determined. Here, we used a cell-tagging dye, CFDA-SE, to permanently label pancreatic duct cells through an intraductal infusion technique. Representing a time when significant increases in beta-cell mass occur, pregnancy was later induced in these CFDA-SE-treated mice to assess the phenotype and fate of the insulin-positive cells in ducts. We found that a small portion of CFDA-SE-labeled duct cells became insulin-positive, but they were not fully functional beta-cells based on the in vitro glucose response and the expression levels of key beta-cell genes. Moreover, these insulin-positive cells in ducts expressed significantly lower levels of genes associated with extracellular matrix degradation and cell migration, which may thus prevent their budding and migration into preexisting islets. A similar conclusion was reached through analysis of the Gene Expression Omnibus database for both mice and humans. Together, our data suggest that the contribution of duct cells to normal beta-cells in adult islets is minimal at best.

<P><B>Abstract</B></P> <P>Folate receptor targeted photo-luminescent quantum carbon dots (Fr-CDs) were successfully prepared from folic acid and phenylenediamine isomers through hydrothermal approaches. Fr-CDs were spherical particles smaller than 10 nm, and emit stable green, blue and red luminescence under ultraviolet region excitation (λex = 365 nm) with maximum emissive lengths at 530, 429, and 612 nm. And the corresponding photoluminescence quantum yield as 15.4%, 12.6% and 16.2% respectively. Up-converted photoluminescent properties in near infrared 800 nm spectral region located in green, blue and yellow region. In-vitro studies showed Fr-CDs had almost none cytotoxicity (cell viability over 80%) and high affinitive to the Hela celline highly-expressed-folate-receptor membranes, and lighted on cytoplasm as the fluorescent marker. It displayed long luminescent-stability with PL intensity above 90% in ultraviolet illuminant exposure over 24 h. In in-vivo studies, Fr-CDs were internalized and accumulated in targeted cancer tissues of cervical carcinoma and the emitting fluorescence maintains over 30 min.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Multicolor carbon dots was prepared from folic acid and phenylenediamin isomers. </LI> <LI> These carbon dots exhibit targeting abilities with high affinity to ovarian cancer cells and tissues. </LI> <LI> Ultra-violet and near-infrared light excited photo-luminescent spectrums were investigated, and related quantum yield was calculated. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>FA-doped CDs can effectively target ovarian cancer cells and aggregate in ovarian tumor tissue in a short 15 min.</P> <P>[DISPLAY OMISSION]</P>
Synthesis of Novel Poly(aryl ether sulfone)s Bearing Bulky Pendants for Gas Separation Membranes

A hydroxylated poly(aryl ether sulfone) (PES-OH) was synthesized by a demethylation reaction of a methoxy-functionalized PES (PES-OCH3) using the HCl/pyridine system. Derived from this hydroxylated PES precursor,bulky (2-trifluoromethyl-4-nitro) phenyl (TFN) and benzophenone (BZP) were introduced to the PES backbones as side groups via aromatic nucleophilic substitution reactions. A direct property comparison of PES-OH,PES-OCH3, PES-TFN and PES-BZP has been made, and these analogues exhibited different solubility, thermal properties and gas permeability. PES grafted with the trifluoromethyl side group (PES-TFN) demonstrated more attractive permeabilities and permselectivities for O2/N2 separation compared to the others, and it exhibited the best P(O2)/P(N2) value of 8.44.
