Multifunctional imaging of the deposition of amyloid-beta (Aβ) aggregates in the brain is of greatimportance in diagnosing Alzheimer’s disease. Herein, we report a multifunctional Aβ-targeting smallmolecular-weight gadolinium (Gd)-based contrast agent (CA), Gd-DO3A-Chal (a new Gd-chelateconjugated with chalcone), that showed 8 times higher binding affinity to Aβ aggregates than apreviously reported Gd-chelate conjugated with Pittsburgh compound B. Gd-DO3A-Chal showedmultimodal imaging capability. As a newfluorescence imaging probe, Gd-DO3A-Chal showed a goodmatch with immunostained images using 6E10 monoclonal antibodies for the detection of Aβ aggregatesin 5XFAD transgenic mouse brain sections. For in vivo magnetic resonance (MR) imaging without bloodbrainbarrier disruption, longitudinal relaxation time (T1)-weighted MR images after intravenousadministration of Gd-DO3A-Chal showed signal enhancement of the Aβ distribution in living 5XFADtransgenic mouse brain. Therefore, in vivo MR images for Aβ detection in addition tofluorescent opticalimages of Aβ aggregates with high specificity and sensitivity using this new multifunctional Aβ-targetingCA were successfully demonstrated.

The purpose of this study was to test whether elevated glycated hemoglobin A1c (HbA1c) levels are associated with cancer incidence in the Korean population. In cohorts of the Korea Genome and Epidemiology Study (KoGES) consortium, we tested whether plasma levels of HbA1c were associated with all-site cancer incidence in 7,822 participants without any known history of cancer or diabetes. Cancer developed in 117 participants during the follow-up period. Subjects were subdivided into 3 categories according observed levels of HbA1c(< 5.7%, low; ≥ 5.7% and < 6.5%, mid; and ≥ 6.5%, high). The adjusted hazard ratio for all-site cancer was 3.03 (95% confidence intervals, 1.54-5.96) for the high HbA1c group relative to the low HbA1c group after adjusting for covariates. Higher circulating HbA1c levels were associated with an increased risk of all-site cancer in Korean population.

Ultra-high-field (UHF) magnetic resonance imaging (MRI) can open up new avenues in clinical diagnosticimaging. Its sensitivity can be further improved using contrast agents (CAs). Therefore, developing low-molecular-weight MR CAs that are efficient under UHF is crucial to ensure taking full advantage of MRimaging at UHF. Moreover, designing highly stable UHF liver-specific CA has been challenging. Here, usingdysprosium (Dy3+), which relaxes water protons via a Curie relaxation that increases T2 relaxationsubstantially with the magneticfield strength, we report a new macrocyclic liver specific Dy3+ complex(Dy[DO3A-EOB]). We observed a 7.06 times higher r2/r1 ratio of Dy[DO3A-EOB] at 9.4 T than that of 1.5 T,which suggests a superior negative-enhancing MR CA at UHF. In vivo MR imaging results furtherdemonstrated that Dy[DO3A-EOB] showed a much strong negative enhancement at UHF (9.4 T) thanunder a lowfield (1.5 T). It was also found that, in addition to the ligand structure, the metal itself plays animportant role in determining the liver uptake pattern of the CA. Therefore, we successfullydemonstrated thefirst example of a small-molecular-weight liver-specific Dy3+ complex acting as aT2 CA with high sensitivity at UHF for diagnosis of liver diseases.
Inhibition of Wntless/GPR177 suppresses gastric tumorigenesis

Wntless/GPR177 functions as WNT ligand carrier protein and activator of WNT/β-catenin signaling, however, its molecular role in gastric cancer (GC) has remained elusive. We investigated the role of GPR177 in gastric tumorigenesis and provided the therapeutic potential of a clinical development of anti-GPR177 monoclonal antibodies. GPR177 mRNA expression was assessed in GC transcriptome data sets (GSE15459, n = 184; GSE66229, n = 300); protein expression was assessed in independent patient tumor tissues (Yonsei TMA, n = 909). GPR177 expression were associated with unfavorable prognosis [log-rank test, GSE15459 (P = 0.00736), GSE66229 (P = 0.0142), and Yonsei TMA (P = 0.0334)] and identified as an independent risk predictor of clinical outcomes: GSE15459 [hazard ratio (HR) 1.731 (95% confidence interval; CI; 1.103- 2.715), P = 0.017], GSE66229 [HR 1.54 (95% CI, 1.10-2.151), P = 0.011], and Yonsei TMA [HR 1.254 (95% CI, 1.049- 1.500), P = 0.013]. Either antibody treatment or GPR177 knockdown suppressed proliferation of GC cells and sensitized cells to apoptosis. And also inhibition of GPR177 suppresses in vitro and in vivo tumorogenesis in GC cells and inhibits WNT/β-catenin signaling. Finally, targeting and inhibition of GPR177 with antibody suppressed tumorigenesis in PDX model. Together, these results suggest GPR177 as a novel candidate for prognostic marker as well as a promising target for treatment of GC patients. [BMB Reports 2018; 51(5): 255-260]
Apolipoprotein A5 3`-UTR variants and cardiometabolic traits in Koreans

BACKGROUND/OBJECTIVES: This study aimed to test the association between APOA5 3"-UTR variants (rs662799) and cardiometabolic traits in Koreans. SUBJECTS/METHODS: For this study, epidemiological data, Apolipoprotein A5 (APOA5) genotype information, and lymphoblastoid cell line (LCL) biospecimens from a subset of the Ansung-Ansan cohort within the Korean Genome and Epidemiology study (KoGES-ASAS; n = 7,704) as well as epidemiological data along with genomic DNA biospecimens of participants from a subset of the Korea National Health and Nutrition Examination Survey (KNHANES 2011-12; n = 2,235) were obtained. APOA5 mRNA expression was also measured. RESULTS: APOA5 rs662799 genotype distributions in both the KoGES-ASAS and KNHANES groups were 50.6% for TT, 41.3% for TC, and 8.1% for CC, which are similar to those in previous reports. In both groups, minor C allele carriers, particularly subjects with CC homozygosity, had lower high-density lipoprotein (HDL) cholesterol and higher triglyceride levels than TT homozygotes. Linear regression analysis showed that the minor C allele significantly contributed to reduction of circulating HDL cholesterol levels [β = -2.048, P < 0.001; β = -2.199, P < 0.001] as well as elevation of circulating triglyceride levels [β = 0.053, P < 0.001; β = 0.066, P < 0.001] in both the KoGES-ASAS and KNHANES groups. In addition, higher expression levels of APOA5 in LCLs of 64 healthy individuals were negatively associated with body mass index (r = -0.277, P = 0.027) and circulating triglyceride level (r = -0.340, P = 0.006) but not significantly correlated with circulating HDL cholesterol level. On the other hand, we observed no significant difference in the mRNA level of APOA5 according to APOA5 rs662799 polymorphisms. CONCLUSIONS: The C allele of APOA5 rs662799 was found to be significantly associated with cardiometabolic traits in a large Korean population from the KoGES-ASAS and KNHANES. The effect of this genotype may be associated with post-transcriptional regulation, which deserves further experimental confirmation.
