        고지방식이 섭취 마우스를 이용한 복분자종자유의 혈행 개선 효과

        복분자종자유의 혈행 개선 기능성을 평가하기 위하여 8주 동안 일반식이와 고지방식이를 제공하는 그룹에 오메가-3 지방산과 복분자종자유를 일정량 제공하는 실험을 실시하였다. 실험군은 일반식이 그룹과 고지방식이 그룹으로 나누어 각 그룹 내에 음성대조군, 양성대조군(오메가-3 지방산), 복분자종자유(2 g, 4 g/2,000 kcal) 급이군으로 구성하였다. 양성대조군과 복분자종자유는 매일 경구 투여되었고 실험기간 종료 후 동물을 희생시켜 혈액과 장기를 얻어 혈중중성지방 및 콜레스테롤, 혈행 개선에 관련된 바이오마커(coagulation factor7, 12, serotonin과 TXB2)를 측정하였는데, 그 결과 복분자종자유는 혈중지질의 농도 개선을 통해 혈액의 점도를 감소시키고 혈액의 응고를 억제하여 혈행을 개선하는 기능성이 있는 것을 확인하게 되었다. 따라서 복분자종자유를 이용한 건강기능식품 개발은 혈관질환 환자들에게 혈행 개선을 통한 건강증진 효과를 보여줄 것으로 판단된다. Bokbunja (Rubus coreanus) is a Korean fruit and wild black raspberry that has antioxidant, anticancer, and beauty effects due to its abundant polyphenols and anthocyanins. The purpose of this study was to investigate the blood flow improvement effect of Bokbunja seed oil (BSO) in a high-fat diet-fed mouse model. We examined improvement of blood flow and its related biomarkers in vivo. Mice were divided into four groups; Control, high fat diet control (negative control, HFD), salmon oil control (positive control, HFD+commercial n-3 fatty acid), and BSO experiment groups (HFD+2 g/2,000 kcal, HFD+4 g/2,000 kcal). After the mice were sacrificed, plasma triglyceride, cholesterol, and blood flow-related biomarkers (coagulation factor 7, 12, serotonin, TXB2, PT, and aPTT) were measured in mouse blood and organs. BSO reduced blood viscosity through improvement of blood lipids (cholesterol and plasma triglycerides) as well as levels of blood coagulation factors and blood platelet activity. BSO also delayed blood coagulation time. Thus, we confirmed that BSO inhibits excessive blood clotting of blood vessels and improves blood flow. Taken together, these results suggest that BSO decreases plasma triglycerides and cholesterol and improves blood flow by regulating biomarkers.

        오메가-3 지방산 함유 복분자종자유에 의한 고지방식이 유도 고지혈증 마우스의 혈중 중성지방 및 콜레스테롤 감소 효과

        본 실험의 복분자종자유는 리놀렌산 238.3 mg/g, 리놀레산과 감마리놀렌산 427.1 mg/g을 포함하며 이는 고시된 범위내의 섭취량이므로 독성시험 없이 4주 동안 매일 경구 투여로 암컷과 수컷 쥐 모두에게 섭취하도록 하였다. 또한 성인정상체중을 약 60 kg으로 설정하고 60 kg의 성인이 하루 1 g, 2 g을 각각 섭취할 때의 혈중 지방의 감소 효과를 알아보기 위해 급이군의 설정을 1 g/60 kg BW/d(BSO 1 g), 2 g/60 kg BW/d(BSO 2 g)로 정하였다. 양성대조군의 경우에 판매되는 연어유 1,000 mg 캡슐을 2정 섭취하는 것을 권장하였고, 이에 맞추어 2 g/60 kg BW/d로 설정하게 되었다. 실험기간 종료 후 마우스를 희생시켜 혈액을 얻었으며, 복분자종자유(BSO 2 g)를 섭취한 마우스에서 총콜레스테롤 및 HDL, LDL/VLDL-콜레스테롤과 혈중 중성지방이 유의적으로 감소하는 것을 확인할 수 있었다(P<0.05). 이를 종합해 보았을 때 복분자종자유는 고지혈증 상태를 개선하고 미약하지만 항응고 활성을 통해 혈액의 항상성을 유지하도록 돕는 작용을 한다고 할 수 있다. 따라서 앞으로 동물유래가 아닌 식물유래 복분자종자유를 이용한다면 고지혈증 개선 효과와 혈액 항응고 활성을 조절할 수 있을 것이며, 더욱 다양한 표적 인자 분석을 통하여 복분자종자유의 고지혈, 항응고 및 혈행개선 기전연구가 가능할 것이라 판단된다. To investigate the effect of n-3 fatty acid from Bokbunja (Rubus coreanus Miq.) seed oil (BSO), we examined improvement of plasma triglycerides and cholesterol in vivo. Five-week-old ICR mice were divided into five groups of six mice each; Control, high fat diet (HFD) control (negative control), salmon oil control (positive control, HFD+commercial n-3 fatty acid), and BSO experimental groups (HFD+1 g/60 kg BW/d, HFD+2 g/60 kg BW/d). After 4 weeks of BSO treatment, we measured serum triglyceride and cholesterol levels. The levels of low-density lipoprotein/very-low-density lipoprotein-cholesterol, high-density lipoprotein-cholesterol, and total cholesterol were significantly (P<0.05) reduced in the group fed BSO at 2 g/60 kg BW/d compared to the negative control. Levels of triglycerides, which are similar to cholesterol, were also significantly (P<0.05) reduced in the same group. To investigate further, we tested blood coagulation parameters. Prothrombin time (PT) and activated partial thromboplastin time (aPTT) were not significantly different among the five groups according to BSO. However, the 2 g/60 kg BW/d BSO group treated with PT and aPTT showed a tendency to live longer than the negative control. Taken together, BSO might improve blood homeostasis mediated via hypo-lipidemic and anti-coagulation activities.

        착한가격업소 인증제도 활성화의 영향요인 분석 : 서울특별시 자치구를 중심으로

        본 연구의 목적은 지역적 차원에서 착한가격업소 인증이 활성화 되는 요인을 서울시 기초자치단체를 대상으로 실증 분석하고자 한다. 구체적으로 2015년부터 2018년까지 25개 서울특별시 기초자치단체에 위치한 착한가격업소 인증 대상 업소들을 분석대상으로 하여 정보공개청구와 행정자료를 통해 패널자료를 형성하였다. 기초통계 분석을 통해 지역의 점포 생존율과 착한가격업소 인증의 수는 부정적 관계를 나타내어 초기 창업실패의 비중이 높은 곳에서 착한가격업소의 인증을 확보하려는 노력을 확인하였다. 패널회귀분석 결과에서는 모방적, 강압적 동형화 요인이 통계적으로 모두 유의미하게 지역 내 착한가격업소 인증에 유의미한 영항력을 나타냈다. 결론적으로 착한가격업소 인증제도는 지역의 일상물가안정화에 영향을 미칠 수 있는 대표적인 정책으로 해당 인증제도의 활성화를 위해서는 기초자치단체의 적극적인 관심과 함께 법적 근거를 제정하고 인증업소에 대한 다각적인 지원이 이루어져야 할 것이다. The purpose of this study is to empirically analyze the factors that activate the certification of the “Good Price Store Certification System” at the local level, targeting the basic municipalities of Seoul. Specifically, from 2015 to 2018, panel data were formed through requests for information disclosure and administrative data by targeting businesses subject to good price business certification in 25 basic municipalities in Seoul as a unit of analysis. Through descriptive analysis, the survival rate of local stores and the number of certifications of good-priced establishments have a negative relationship, confirming efforts to secure certification of good-priced establishments in places with a high proportion of initial start-up failures. In the results of panel regression analysis, both mimetic and coercive factors were statistically significant, indicating a significant influence on the certification of good-priced businesses in the region. In conclusion, the “Good Price Store Certification System” is a representative policy that can stabilize daily prices in the region. To revitalize the certification system, the local government should actively interest in the legal basis and provide various support to the certification business.

        Evaluation of the acute, sub-chronic and chronic oral toxicity, genetic toxicity, and safety of a Lomens-P0

        Extracts of Hordeum vulgare and Chrysanthemum zawadskii, two traditional herbal medicines, have long been used to treat women’s diseases. Our previous studies have confirmed that these extracts could help relieve the symptoms of premenstrual syndrome by inhibiting prolactin release. A mixture of these natural products was named Lomens-P0. In this study, we conducted three genotoxicity tests (bacterial reverse mutation, mammalian chromosome aberration, and mammalian erythrocyte micronucleus studies) and four oral toxicity tests (single-dose, 2-week repeated-dose, and 13-week repeated-dose studies in rodents, and a single-dose dose-escalation toxicity study in a non-rodent model) to confirm the potential toxicity and safety of Lomens-P0. The results of this series of tests indicated Lomens-P0 did not induce genotoxicity, and the NOAEL for the rodent was 2000 mg/kg BW/day. Similarly, no toxic effects were evident in the single-dose-escalation study in the nonrodent model. In conclusion, we confirmed that Lomens-P0 might have potential utility as a raw material for nutraceuticals and natural medicines.

        Centella asiatica extract prevents visual impairment by promoting the production of rhodopsin in the retina

        BACKGROUND/OBJECTIVE: Centella asiatica, also known as Gotu kola, is a tropical medicinal plant native to Madagascar, Southeast Asia, and South Africa. It is well known to have biological activities, including wound healing, anti-inflammatory, antidiabetic, cytotoxic, and antioxidant effects. The purpose of this study was to determine the efficacy of extracts of C. asiatica against age-related eye degeneration and to examine their physiological activities. MATERIALS/METHODS: To determine the effects of CA-HE50 (C. asiatica 50% EtOH extract) on retinal pigment cells, we assessed the cytotoxicity of CoCl₂ and oxidized-A2E in ARPE-19 cells and observed the protective effects of CA-HE50 against N-methyl-N-nitrosourea (MNU)-induced retinal damage in C57BL/6 mice. In particular, we measured factors related to apoptosis and anti-oxidation and the protein levels of rhodopsin/opsin. We also measured glucose uptake to characterize glucose metabolism, a major factor in cell protection. RESULTS: Induction of cytotoxicity with CoCl₂ and oxidized-A2E inhibited decreases in the viability of ARPE-19 cells when CA-HE50 was administered, and promoted glucose uptake under normal conditions (P < 0.05). In addition, CA-HE50 inhibited degeneration/apoptosis of the retina in the context of MNU-induced toxicity (P < 0.05). In particular, CA-HE50 at 200 mg/kg inhibited the cleavage of pro-caspase-3 and pro-poly (ADP-ribose)-polymerase and maintained the expressions of nuclear factor erythroid 2-related factor 2 and heme oxygenase-1 similar to normal control levels. Rhodopsin/opsin expression was maintained at a higher level than in normal controls. CONCLUSION: A series of experiments confirmed that CA-HE50 was effective for inhibiting or preventing age-related eye damage/degeneration. Based on these results, we believe it is worthwhile to develop drugs or functional foods related to age-related eye degeneration using CA-HE50.

        Lomens-P0 (mixed extracts of Hordeum vulgare and Chrysanthemum zawadskii) regulate the expression of factors affecting premenstrual syndrome symptoms

        BACKGROUND/OBJECTIVES: Premenstrual syndrome (PMS) is a disorder characterized by repeated emotional, behavioral, and physical symptoms before menstruation, and the exact cause and mechanism are uncertain. Hyperprolactinemia interferes with the normal production of estrogen and progesterone, leading to PMS symptoms. Thus, we judged that the inhibition of prolactin hypersecretion could mitigate PMS symptoms. MATERIALS/METHODS: Hordeum vulgare L. extract (HVE), Chrysanthemum zawadskii var. latilobum extract (CZE), and Lomens-P0 the mixture of these extracts were tested in subsequent experiments. The effect of extracts on prolactin secretion at the in vitro level was measured in GH3 cells. Nitric oxide and pro-inflammatory mediator expression were measured in RAW 264.7 cells to confirm the anti-inflammatory effect. Also, the hyperprolactinemic Institute for Cancer Research (ICR) mice model was used to measure extract effects on prolactin and hormone secretion and uterine inflammation. RESULTS: Anti-inflammatory effects of and prolactin secretion suppress by HVE and CZE were confirmed through in vitro experiments (P < 0.05). Treatment with Lomens-P0 inhibited prolactin secretion (P < 0.05) and restored normal sex hormone secretion in the hyperprolactinemia mice model. In addition, extracts significantly inhibited the expression of pro-inflammatory biomarkers, including interleukin-1β, and -6, tumor necrosis factor-α, inducible nitric oxide synthase, and cyclooxygenase-2 (P < 0.01). We used high-performance liquid chromatography analyses to identify tricin and chlorogenic acid as the respective components of HVE and CZE that inhibit prolactin secretion. The Lomens-P0, which includes tricin and chlorogenic acid, is expected to be effective in improving PMS symptoms in the human body. CONCLUSIONS: The Lomens-P0 suppressed the prolactin secretion in hyperprolactinemia mice, normalized the sex hormone imbalance, and significantly suppressed the expression of inflammatory markers in uterine tissue. This study suggests that Lomens-P0 may have the potential to prevent or remedy materials to PMS symptoms.

        Hepatoprotective effect of Centella asiatica 50% ethanol extract against acetaminophen-induced acute liver injury in BALB/c mice

        N-acetyl-p-aminophenol (acetaminophen, APAP) is a well-known component of analgesic and antipyretic monotherapy products. However, exceeding the recommended dose can lead to serious injury to the liver. We conducted this study to determine the potential of Centella asiatica as a natural substance to protect against APAP-induced liver injury. When acute hepatotoxicity was induced in mice by APAP overdose, their liver weight decreased significantly (p < 0.05). However, mice treated with C. asiatica 50% ethanol extract (CA-HE50, 200 mg/kg) for a week before induction of hepatotoxicity by APAP had similar liver weights to those of mice in which hepatotoxicity was not induced. In particular, levels of aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase, which are biomarkers of liver injury, were significantly increased by APAP and dose-dependently decreased by CA-HE50 (p < 0.05). Glutathione and malondialdehyde, indicators of oxidative stress, were significantly changed by APAP and CA-HE50 (p < 0.05). In addition, hepatic necrosis and expression of genes encoding pro-inflammatory cytokines (tumor necrosis factor-α, interleukin (IL)-1β, and IL-4) induced by APAP were inhibited by CA-HE50, and these results were dose-dependent. Through our in vivo studies, we found that CA-HE50 can help prevent APAP-induced hepatic tissue injury in BALB/c mice. Furthermore, CA-HE50 was effective at protecting RAW 264.7 cells from lipopolysaccharide-induced cytotoxicity and inhibiting the release of nitric oxide from these cells; in particular, asiaticoside was found to be a key component of CA-HE50 responsible for these effects. Therefore, we suggest that CA-HE50 has potential applications in functional health foods and drugs.

        유자 에탄올 추출물의 면역력 증진 효과

        본 연구에서는 유자가 선천성 면역력에 미치는 효과를 알아보기 위하여 유자 30% 주정추출물(CJE)을 사용하였으며 선천성 면역에 중요한 역할을 하는 대식세포를 이용해 실험하였다. CJE는 마우스 대식세포인 RAW264.7에서 1,000 μg/mL의 최고 농도까지 세포독성을 보이지 않았고, 전사인자 NF-κB와 염증성 매개물질인 COX-2, PGE2의 활성 및 발현 증강에 영향을 미치며, 특히 300 μg/mL의 농도에서부터 유의적 차이를 보이는 것으로 확인되었다. 산화질소 생성능과 대식세포에서 분비되는 사이토카인인 TNF-α, IL-1β의 발현을 대조군에 비해 농도 의존적으로 증가시킨다는 결과를 얻었으나, IL-6에서는 통계적으로 약간의 유의성이 있는 증가를 보였고 IL-10은 정상대조군에 비해 거의 유의적인 차이를 보이지 않았다. CJE는 또한 NK 세포의 활성을 농도의존적으로 증가시키고 비장세포의 증식능도 농도 의존적으로 증가시킨다는 것을 확인하였다. 이러한 결과로 미루어보아 CJE는 인체의 대식세포 활성의 증가를 통해 선천성 면역력을 증가시킬 것으로 판단된다. Citrus junos Sieb. ex Tanaka has been traditionally called Yuza in Korea and is used as a cuisine material or tea as well as medicinal herb. In this study, we evaluated the immune-enhancing effect of Citrus junos ethanol extract (CJE) on RAW264.7 mouse macrophage and primary immunocytes. CJE treatment showed increased macrophage activity in a dose-dependant manner. CJE also enhanced natural killer (NK) cell activity. We measured lactate dehydrogenase (LDH) level as a measurement of NK cell cytotoxicity against YAC-1 lymphoma cells. CJE treatment showed an increased LDH level in a dose-dependent manner. Finally, we evaluated the effect of CJE on mouse primary splenocyte proliferation. CJE treatment slightly increased splenocyte proliferation compared to the control. The results of this study suggest that CJE can help immune function via macrophage cytokine production, increased NK cell activity, and splenocyte proliferation.
