      • Species and gamete-specific fertilization success of two sea urchins under near future levels of pCO<sub>2</sub>

        Since the Industrial Revolution, rising atmospheric CO<SUB>2</SUB> concentration has driven an increase in the partial pressure of CO<SUB>2</SUB> in seawater (pCO<SUB>2</SUB>), thus lowering ocean pH. We examined the separate effects of exposure of gametes to elevated pCO<SUB>2</SUB> and low pH on fertilization success of the sea urchin Strongylocentrotus nudus. Sperm and eggs were independently exposed to seawater with pCO<SUB>2</SUB> levels ranging from 380 (pH7.96-8.3) to 6000ppmv (pH7.15-7.20). When sperm were exposed, fertilization rate decreased drastically with increased pCO<SUB>2</SUB>, even at a concentration of 450ppmv (pH range: 7.94 to 7.96). Conversely, fertilization of Hemicentrotus pulcherrimus was not significantly changed even when sperm was exposed to pCO2 concentrations as high as 750ppmv. Exposure of S. nudus eggs to seawater with high pCO<SUB>2</SUB> did not affect fertilization success, suggesting that the effect of increased pCO<SUB>2</SUB> on sperm is responsible for reduced fertilization success. Surprisingly, this result was not related to sperm motility, which was insensitive to pCO<SUB>2</SUB>. When seawater was acidified using HCl, leaving pCO<SUB>2</SUB> constant, fertilization success in S. nudus remained high (>80%) until pH decreased to 7.3. While further studies are required to elucidate the physiological mechanism by which elevated pCO<SUB>2</SUB> impairs sperm and reduces S. nudus fertilization, this study suggests that in the foreseeable future, sea urchin survival may be threatened due to lower fertilization success driven by elevated pCO<SUB>2</SUB> rather than by decreased pH in seawater.

        원발성 간암에서 PIVKA-II 및 Lens Culinaris Agglutinin-A 반응성 Alpha-fetoprotein(AFP-L3)의 임상적 유용성

        Background/Aims: Des-γ-carboxy prothrombin(DCP), a protein induced by vitamin K absence or antagonist-II(PIVKA-II), and Lens culinaris agglutinin-A reactive AFP-L3 have been reported to be useful markers for the diagnosis of hepatocellular carcinoma (HCC). In the present study, both the PIVKA-II and AFP-L3 were analyzed and compared with a conventional AFP to determine its usefulness, specificity and sensitivity in the diagnosis of HCC. Methods: Sera were collected from 108 patients consisting of 17 patients with chronic hepatitis, 22 patients with liver cirrhosis and 69 patients with HCC. The AFP-L3 was determined by an lectin affinity electrophoresis coupled with an antibody-affinity blotting. Level of DCP was measured by an enzyme immunoassay with an anti-DCP monoclonal antibody. Results: The sensitivity and specificity of PIVKA-II and AFP L3 were 49.3% and 89.5%, and 32.5% and 85.7%, respectively. No significant correlation was found between the PIVKA-II or AFP L3 and serum AFP. No correlation was found etween the PIVKA-II or AFP L3 and the characteristics of HCC.       • Potential Effect of S-Nitrosylated Protein Disulfide Isomerase on Mutant SOD1 Aggregation and Neuronal Cell Death in Amyotrophic Lateral Sclerosis

        Aggregation of misfolded protein and resultant intracellular inclusion body formation are common hallmarks of mutant superoxide dismutase (mSOD1)-linked familial amyotrophic lateral sclerosis (FALS) and have been associated with the selective neuronal death. Protein disulfide isomerase (PDI) represents a family of enzymatic chaperones that can fold nascent and aberrant proteins in the endoplasmic reticulum (ER) lumen. Recently, our group found that S-nitrosylated PDI could contribute to protein misfolding and subsequent neuronal cell death. However, the exact role of PDI in the pathogenesis of ALS remains unclear. In this study, we propose that PDI attenuates aggregation of mutant/misfolded SOD1 and resultant neurotoxicity associated with ER stress. ER stress resulting in PDI dysfunction therefore provides a mechanistic link between deficits in molecular chaperones, accumulation of misfolded proteins, and neuronal death in neurodegenerative diseases. In contrast, S-nitrosylation of PDI inhibits its activity, increases mSOD1 aggregation, and increases neuronal cell death. Specifically, our data show that S-nitrosylation abrogates PDI-mediated attenuation of neuronal cell death triggered by thapsigargin. Biotin switch assays demonstrate S-nitrosylated PDI both in the spinal cords of SOD1 (G93A) mice and human patients with sporadic ALS. Therefore, denitrosylation of PDI may have therapeutic implications. Taken together, our results suggest a novel strategy involving PDI as a therapy to prevent mSOD1 aggregation and neuronal degeneration. Moreover, the data demonstrate that inactivation of PDI by S-nitrosylation occurs in both mSOD1-linked and sporadic forms of ALS in humans as well as mice.

      • Emergence of Panton-Valentine leucocidin-positive ST8-methicillin-resistant Staphylococcus aureus (USA300 clone) in Korea causing healthcare-associated and hospital-acquired bacteraemia

        <P>Panton-Valentine leucocidin (PVL)-positive sequence type (ST)8-MRSA-SCCmec IVa (USA300) is the epidemic strain of community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) in North America. USA300 is extremely rare in South Korea, and PVL-negative ST72 SCCmec type IVc is the predominant CA-MRSA clone. In a multicentre, prospective cohort study of S. aureus bacteraemia, we identified PVL-positive ST8-MRSA isolates by performing multilocus sequence typing and PCR for PVL. We analyzed the clinical characteristics of patients with PVL-positive ST8-MRSA bacteraemia, and performed SCCmec, spa, and agr typing, PCR for arginine catabolic mobile element (ACME), virulence gene profiling, and pulsed-field gel electrophoresis (PFGE). Among a total of 818 MRSA isolates, we identified ten isolates of PVL-positive ST8-MRSA (USA300) (3 from Hospital D, 4 from Hospital G, and 3 from Hospital A), all of which involved exclusively healthcare-associated (5 isolates) and hospital-acquired bacteraemia (5 isolates). This strain accounted for 8 similar to 10 % of the hospital-acquired MRSA bacteraemia in Hospitals D and G. Bacteraemia of unknown origin was the most common type of infection followed by pneumonia. All the isolates were SCCmec type IVa, spa type t008, and agr group I. Eight of the isolates harboured ACME. In a PFGE analysis, four isolates were identical to the USA300 control strain, five differed by a single band, and the remaining one differed by two bands. All the isolates were pulsed-field type USA300. This is the first report of healthcare-associated and hospital-acquired bacteraemia caused by USA300 in South Korea. USA300 seems to be an emerging hospital clone in this country.</P>

        Impact of White Matter Changes on Activities of Daily Living in Mild to Moderate Dementia

        <P>The association between white matter changes and activities of daily living (ADL) in a large, well-defined cohort of patients with mild-to-moderate dementia (either Alzheimer’s disease or subcortical vascular dementia) were investigated. A total of 289 patients were divided into three groups (140 mild, 99 moderate, and 50 severe) depending on the degree of white matter changes as indicated on brain magnetic resonance image scans. Further, we analyzed the three groups’ performances on basic and instrumental ADL. The degree of white matter changes was associated with greater age, hypertension, previous history of stroke, higher Hachinski Ischemic Score, worse global cognitive and functional status, and an increased impairment of basic ADL and instrumental ADL. The increased impairment with regard to the severe group’s performance on both the basic and instrumental ADL remained significant after adjustment for age and hypertension. Tasks involving physical activities were most significant. This was the first study investigating the association between white matter changes and ADL in a large, well-defined dementia cohort. The present study suggests that severe white matter changes may be associated with higher impairment on both basic and instrumental ADL.</P><P>Copyright © 2010 S. Karger AG, Basel</P>

        We have performed an EMTDC simulation for the current limiting effects of a superconducting fault current limiter (SFCL). A three-phase fault in the 154 kV transmission line between the gaepo and Sungnam substations produced a fault current of up to 60kA. The SFCL with 100 Ω impedance after quench limited the current to 17kA within a half cycle. This limited current is well below the upper limit of a circuit breaker, suggesting the impedance of the SFCL in the transmission line is sufficient.

      • 커피찌꺼기를 이용한 활성탄의 제조 및 흡착특성

        The exhausted coffee is a kind of biomass resources. In this study Activated carbon was prepared from ex.hausted coffee by activation with potassium Hydroxide. Processes of this study were as following : rosting step and carbonizaton step and chemical activation step. The rostiong step was canied out at more than 300 °C. The carbonization and activation step was simultaneously caπied out at 500-700°C. The purpose of this study is to evaluate the feature and usability of activated cofee char through SEM photographs, Iodine absorbability, TGA analysis, and technical analysis. For the activated carbon produced under optimal condition, iodine adsorptivity was 774mg/g, specific surface area was foπned well. In case of making activated carbon using exhausted coffee, it is considered that it has value enough as an activated carbon.
