      • Effects of the pro-inflammatory milieu on the dedifferentiation of cultured fibroblast-like synoviocytes

        <P>The aim of this study was to determine whether the inflammatory milieu and/or hypoxia induces the dedifferentiation of synovial cells into mesenchymal stem-like cells, which may contribute to the tumor-like growth of synovial cells. Expression of mesenchymal stem cell markers (CD24, CD44, CD90, CD106, CD146 and Stro-1) was compared among cultured fibroblast-like synoviocytes (FLSs) from patients with rheumatoid arthritis (RA) or osteoarthritis (OA), bone marrow mesenchymal stem cells (BM MSCs) and normal dermal fibroblasts. After the cells were stimulated with pro-inflammatory cytokines for 3 days under hypoxia or normoxia, the stem cell markers were analyzed by FACS. CD44 and CD90 were expressed constitutively in all four cell types. Only the BM MSCs strongly expressed CD146. The expression of stem cell markers was similar between FLSs from RA and those from OA patients. In addition, the expression levels in FLSs were similar to those in normal dermal fibroblasts. The stimulation of FLSs and dermal fibroblasts with IL-1β or a mixture of cytokines under hypoxia did not induce a marked change in the expression of stem cell markers. These results indirectly suggest that the pro-inflammatory milieu may be not sufficient to induce the dedifferentiation of FLSs in arthritic joints.</P>

      • The whole-exome sequencing in three families with atopic dermatitis

        Background: The prevalence of atopic dermatitis (AD) has increased over a recent 10-year. AD tends to run in families that passed down from generation to generation and commonly starts in childhood. It is as high as 20% in children less than 5 years old. Thus, early discovery and therapy of AD are important. It has emerged the necessity of biomarker for early detection of AD. Objectives: The goal of this study is to find novel gene markers for diagnosis of early-onset atopic dermatitis in Koreans. Methods: Peripheral blood was obtained from three families (6 early-onset AD patients and 6 controls) with autosomal dominant condition. Whole Exome Sequencing (WES) was performed using SureSelect Human All Exon V4+UTR 71 Mb. Variants of atopic dermatitis were filtered step by step to gain the important candidate genes. Results: We have confirmed overlapping genes in common variants of filter 5 in three family. 15 genes were overlapped in filter 5 and two of them reached to filter 7 can be called “rare variant”. Uniquely, COL6A6 gene appeared in all of three family. We also found variants of filaggrin, filaggrin2 and IL4R that is well-known makers of atopic dermatitis. Conclusion: We were able to detect COL6A6 gene by finding overlapping genes in rare and common variants of three family. It may become a novel gene as early-onset AD marker.

        북강활 에틸아세테이트분획의 비만세포에서의 염증반응 억제효과

        Objectives: In this study, the pharmacological effects of the ethylacetate extract of Ostericum koreanum(North Kangwhal; NK) on allergic inflammation were investigated in activated human mast cells. Methods: North Kangwhal was extracted with 80% methanol for 24 h, and then fractionated with ethylacetate(NK-EtOAc extract). HMC-1 cells, an human mast line, were pre-incubated with different concentrations of NK-EtOAc extract for 30 min, and then stimulated with PMA(50 nM/ml) and A23187(1 μM/ml) at indicated times. The cell toxicity was determined by MTT assay. The concentrations of prostaglandin E2(PGE2) and cytokines(TNF-α, IL-8) were measured by enzyme-linked immunosorbant assay. Results: NK-EtOAc extract(10~50 ㎍/ml) significantly inhibited the productions of PGE2, TNF-α and IL-8 in PMA/A23187-stimulated HMC-1 cells without cell toxicity(0~50 ㎍/ml). NK-EtOAc extract also inhibited PMA/A23187-induced phosphorylation of ERK1/2 MAPK and the NF-κB p65 subunit translocation into the nuclear of HMC-1 cells. Conclusions: This study suggests that NK-EtOAc extract may have an anti-inflammatory property through suppressing the production of inflammatory mediators in activated mast cells and its molecular mechanism underlies the blocking of NF-κB pathway.

        활성 RAW 264.7 세포에서 항염증 및 자외선 유도 마우스 피부손상의 개선에 대한 새송이 추출물의 효과

        본 연구는 먼저 새송이버섯 추출물인 PEE가 활성화된 면역세포에서 항염증 효과가 있는지 알아보았다. 그 결과 LPS로 자극된 RAW 264.7 세포에 PEE를 처리했을 때 5과 15 mg/mL 등 높은 농도에서 NO를 효과적으로 억제하였다. 그리고 LPS가 유도하는 IL-1β와 IL-6는 5 mg/mL 농도에서는 억제 없었으나, 15 mg/mL 농도에서 현저히 억제하는 효과가 있었다. 다음은 UVB가 유도하는 C57BL/6 마우스 피부손상에 대한 PEE의 개선 효과에 대해서 알아보았다. PEE를 7일간 경구투여한 후 7일, 10, 13일 등 3일 간격으로 하루에 1회씩 UVB (250 mJ/cm²)로 마우스 등 피부에 조사하여 피부손상을 유발하였다. UVB 조사 후부터는 PEE를 경구투여와 함께 피부에 도포하여 PEE가 피부 손상에 대한 개선 효과를 조사하였다. 그 결과 PEE 투여군은 UVB로 유도되는 표피두께, 홍반지수 및 멜라닌 지수가 참고약물로 사용한 비타민 C(AA) 투여군과 유사한 개선되는 효과가 있었다. 또한 PEE투여군은 UVB가 유도하는 비만세포를 비롯한 염중세포 침윤이 현저히 억제되는 효과가 있었다. 이상의 결과를 종합해볼 때 PEE는 항염증 효과와 더불어 UVB가 유도하는 피부손상을 개선하는 우수한 효과가 있어 기능성 식품소재 및 화장품 소재로 활용할 수 있을 것으로 판단된다. The aim of this study was to investigate the anti-inflammatory and ultraviolet (UV)-protective effect of Pleurotus eryngii extract (PEE) in activated RAW 264.7 cells and UV-induced mouse skin damage. The results showed that PEE strongly inhibited the production of inflammatory mediators such as nitric oxide, interleukin (IL)-1β, and IL-6 at high concentrations in LPS-stimulated RAW 264.7 macrophages. In addition, PEE treatment suppressed erythema, melanin index, and epidermal thickness to a greater degree than ascorbic acid (AA) treatment in UV-irradiated mice. Finally, PEE treatment inhibited the infiltration of mast cell to a similar degree of AA treatment. Therefore, these results indicate that PEE could improve inflammation and skin damage in immune cells and UV-irradiated mice. This study may provide positive insights into PEE as a functional food and cosmetic ingredient for treatment of inflammation and skin damage.

      • Expression and regulation of inhibitor of DNA binding proteins <i>ID1</i>, <i>ID2</i>, <i>ID3</i>, and <i>ID4</i> at the maternal-conceptus interface in pigs

        <P><B>Abstract</B></P> <P>Inhibitor of DNA binding (ID) proteins, ID1, ID2, ID3, and ID4 are transcriptional regulators that have a helix-loop-helix (HLH) domain but not a basic DNA binding domain. ID proteins inhibit the functions of basic HLH transcription factors and regulate cell proliferation and differentiation. However, the expression and function of <I>ID1</I>, <I>ID2</I>, <I>ID3</I>, and <I>ID4</I> at the maternal-conceptus interface are not fully understood in pigs. Therefore, we determined the expressions of <I>ID1</I>, <I>ID2</I>, <I>ID3</I>, and <I>ID4</I> in porcine endometrium, conceptus, and chorioallantoic tissues. <I>ID1</I>, <I>ID2</I>, <I>ID3</I>, and <I>ID4</I> mRNAs were expressed in the endometrium, with lower levels of <I>ID1</I>, <I>ID2</I>, and <I>ID4</I> on Day 12 of pregnancy than during the estrous cycle. <I>ID1</I>, <I>ID2</I>, <I>ID3</I>, and <I>ID4</I> mRNAs were also detected in conceptus and chorioallantoic tissues during pregnancy. ID2 protein was mainly localized to luminal epithelia and weakly to vascular smooth muscle cells in the endometrium and conceptus trophectoderm. Increasing doses of interleukin-1β decreased levels of <I>ID2</I> mRNA, while estradiol-17β increased levels of <I>ID3</I> mRNA in endometrial explants. The expressions of <I>ID2</I> and <I>ID4</I> mRNAs were higher in endometria from gilts with somatic cell nuclear transfer-derived conceptuses compared with endometria from gilts carrying conceptuses derived from natural mating on Day 12. These results indicate that the expressions of ID family genes are dynamically regulated at the maternal-conceptus interface, suggesting that ID proteins may play critical roles in the regulation of endometrial epithelial cell function and conceptus development to support the establishment and maintenance of pregnancy in pigs.</P> <P><B>Highlights</B></P> <P> <UL> <LI> <I>ID1</I>, <I>ID2</I>, <I>ID3</I>, and <I>ID4</I> expression in porcine uterine endometrium is pregnancy status- and stage-specific. </LI> <LI> ID2 protein is detected primarily in endometrial LE cells. </LI> <LI> IL1B decreases the expression of <I>ID2</I> mRNA in endometrial tissues. </LI> <LI> The expressions of <I>ID2</I> and <I>ID4</I> mRNAs increase in endometria of gilts with SCNT-derived conceptuses. </LI> </UL> </P>

      • Aspergillus awamori와 Zymomonas mobilis로 구성된 고정화혼합 배양계의 에탄올 생산에 미치는 Triton, PVA 및 PEG의 영향

        The effects of triton-X 100, PVA and PEG on the ethanol production and raw starch hydrolysis by co-immobilized A. awamori and Z. mobilis (A-Z system) were investigated. When gel beads containing A. awamori(1.25 × 10 exp (9) spores/ℓ - gel) and Z. mobilis(0.5g cell/ℓ - gel) were cultured in 2% raw starch solution at 30℃ for 120 hours, ethanol concentration, ethanol yield (Y_p/s) and pH of culture broth were 2.8g/ℓ, 0.16 and 3.8~4.0, respectively. Ethanol production and starch hydrolysis by addition of PVA was efficient, compared to that of PEG and ethanol concentration in culture broth with 0.2% PVA was 3.56g/ℓ. At A-Z 36 culture system with changing silicon check valve for cotton plug at 36 hours in A-Z culture system, ethanol yield on glucose, Y_p/s = 0.36, showed 2 times higher than that of control culture system(cotton plug culture). Ethanol production and productivity by cultivating 85g gel beads at 30℃ in a 1ℓ jar fermentor(220rpm) of medium 500㎖ were 32g/ℓ and 1.8g/ℓ/day, respectively. After 12 hours of cultivation, gel beads were distinguished oxygen-rich surface for A. awamori from oxygen-deficient central part for Z. mobilis.

        韓茵蔯蒿湯이 알코올 투여로 유발된 흰쥐의 고지혈증과 간 손상의 예방에 미치는 영향

        Objectives : This study was performed to compare the effect of water extracts from Hanlnjinho-tang on hyperlipidemia and liver damage induced by alcohol. Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And each extract was administrated for the same period. We measured the serum component in rat's blood, body weight and weight of liver. Results : 1. At first, we observed effects of Hanirijinho-tang on hyperlipidemia induced by alcohol. Hanirjinho-tang group showed significant decrease of total cholesterol and triglyceride in comparison with those of the control group. Haninjinho-tang group showed significant increase of HDL-cholesterol in comparison with those of the control group. Haninjinho-tang group didn't show significant increase of body weight in comparison with those of the control group in 4weeks and 8weeks. 2. At second, we observed effects of Haninjinho-tang on liver damage induced by alcohol. Haninjinho-tang group showed significant decrease of LDH, GOT in comparison with those of the control group. Haninjinho-tang group didn't show significant change of ALP, GPT in comparison with those of the control group. Haninjinho-tang group showed significant increase of liver weight in comparison with those of the control group. Conclusions : Reviewing these experimental results, it appears that water extracts from Haninjinho-tang have pharmaceutical efficacy on hyperlipidemia and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of these.

      • Xanthomonas sp. EPS-1의 Exopolysaccharide생산을 위한 무기염류의 영향

        For the production of high viscosity exopolysaccharide from Xanthomonas sp. EPS-1 which isolated and identified from soils, optimum concentration of inorganic salts were investigated : 0.2% KH_2PO_4, 0.05% MgSO_4·7H_2O, 0.3% NaCl, 0.005% CaCO_3, 0.007% FeSO_4·7H_2O, 0.005% MnSO_4·7H_2O. When EPS-1 strain was cultivated at 30℃ for 72 hours in a jar fermentor (impeller speed 300 rpm, aeration rate 3 vvm, 2 liter), polymer content, product rate and yield(Y_p/s) was 18.4g/ℓ, 0.26g/ℓ/h and 37%, respectively.
