      • 중합효소연쇄 반응법에 의한 인형거대세포바이러스(human cytomegalovirus)의 신속한 검출

        목 적 : 인형거대세포바이러스(HCMV)는 면역 저하 환자에서 폐렴, 망막염, 간염 등 치명적인 감염증의 원인이다. 전통적인 세포 배양법으로 HCMV를 분리하기까지는 1주에서 4주가 걸리므로 이 바이러스에 의한 질환이 의심되는 경우 신속한 진단법을 이용한 진단이 바람직하다. 저자들은 한국에서 분리되는 야생주 HCMV를 검출하는데 중합효소 연쇄 반응법을 이용할 경우의 진단적 유용성을 알기 위하여, 전통적인 세포배양법으로 HCMV가 분리된 검체를 이용하여 중합효소 연쇄 반응법의 특이도와 민감도를 평가하였다. 방 법 : 국내 야생주 HCMV 15주, 그리고 전통적인 바이러스 배양법으로 HCMV가 분리된 소변 15검체를 대상으로 중합효소 연쇄 반응을 실시하였다. 중합효소 연쇄 반응은 Towne주의 immediate early antigen의 유전자에서 유리된 primer MIE와 AD169주의 immediate early antigen의 유전자에서 유리된 primer IE를 이용하였다. 증폭 산물은 겔 전기영동 후 EtBR으로 염색하여 자외선 하에서 관찰하였다. Primer IE에 의한 증폭산물은 또한, DNA blot hybridization 방법으로 확인하였다. 결 과 : 1) 국내 HCMV 야생주 15주를 대상으로 중합효소 연쇄 반응을 한 결과 primer IE를 이용한 경우에는 100%(15/15)에서 177bp의 증폭 산물이 관찰되었고, 이들 증폭 산물은 모두 probe IE와 보합 결합되었다. primer MIE를 이용한 경우에는 93%(14/15)에서 435bp의 증폭 산물이 관찰되었다. 2) 세포 배양법으로 HCMV가 분리된 소변 15검체를 대상으로 primer IE를 이용하여 중합효소 연쇄 반응을 한 결과 direct gel analysis 법으로는 73%(11/15)에서, 보합 결합법으로는 87%(13/15)에서 관찰되었다. Primer MIE를 이용한 경우에는 direct gel analysis법으로 87%(13/15)에서 435bp의 증폭 산물이 검출되었다. 결 론 : Primer IE와 MIE를 이용한 중합효소 연쇄 반응법은 국내 야생주 HCMV를 신속히 검출하는데 유용한 검사법이다. Background : Human cytomegalovirus(HCMV) can cause pneumonitis, hepatitis, retinitis and other serious diseases in the immunocompromised patients. It takes 1 to 4 weeks to diagnose HCMV infection by conventional virus culture. Therefore, when HCMV diseases are suspected, a rapid diagnostic method such as polymerase chain reaction(PCR), antigen assay or shell vial culture is desirable. We evaluate the sensitivity and specificity of a PCR for the rapid detection of HCMV wild strains in Korea. Methods : We used 2 sets of primers ; primer IE and primer MIE derived from the sequence for immediate early gene of AD169 strain and Towne strain, respectively. Fifteen clinical isolates of HCMV, suspended in MRC-5 cells, were amplified by PCR. Fifteen urine specimens which were positive for HCMV by conventional virus culture were also amplified. Amplification products were analyzed by polyacrylamide gel electrophoresis. The products from PCR with primer IE were also identified by DNA blot hybridization. Results : PCR using primer IE gave the PCR products in all of the 15 HCMV wild strains. All of these were hybridized with probe IE. When primer MIE were used, 93%(14/15) of the wild strains showed amplified bands by direct gel analysis. When the urine specimens were amplified by PCR with primer IE, amplified bands were seen in 73%(11/15) by direct gel analysis ; 87%(13/15) by hybridization method. When primer MIE were used, 87%(13/15) of the urine specimens showed the PCR products by direct gel analysis. Conclusion : Polymerase chain reaction with primer IE and MIE may be a specific and sensitive diagnostic method for rapid detection of HCMV wild strains in Korea.

      • Digital 方式에 의한 直流電動機의 速度制御에 관한 硏究

        1. Object and Importance for Study In various industrial process controls it is often necessary to be able to adjust a DC motor's speed over a wide range with good speed resolution and reproducibility. Conventional analog control methods suffer on several accounts, including nonlinearlity in the analog speed transducer and difficulty in accurately transmitting the analog signal after it has been obtained from the transducer. Also, while manipulating the signal to effect control action on the motor, errors are incurred which are related to temperature, component aging, and extraneous disturbances. A digital speed control system is superior in that there is no nonlinearity in the speed transducer, the digital signal representing speed can be transmitted long distances with no degradation of the original accuracy, and the digitally developed control signal is not subject to temperature variations, component changes, or noise. There were a few papers on the study of digital methods for DC motor speed control in foreign countries, but no papers available in our country until we started to take this thesis. Hence, in this paper a digital method for DC motor speed control is proposed, and is to be tested through experiment for the performance of the proposed system. 2.Contents of Study A digital method for DC motor speed control is proposed. 3 phase SCR rectifier circuit can be phase-controlled by using the principle of digital phase shift generator, and six magnitude comparators provide proper firing pulse trains for SCRS. An arc-cosine conversion is performed by using PROM to improve linearity of the relationship between applied cosine wave and firing angle. In order to give amount of feedback digital signal by comparing and detecting the difference between reference speed and motor speed, there is provided with a speed control part which includes subtractor, adder, and etc. Through experiment we confirmed that the digital control system had no influnce of errors by noise and offset voltage, and that the simplification of SCR firing pulse synchronization for 3 phase supply voltage waveforms was obtained by six magnitude comparators and the linearity for the relationship between reference speed and motor speed was greatly improved. 3. Results of Study As a digital speed control method which provides the advantages of freedom from drift and offset errors, immunity of the control circuit from transients and distortion of supply voltage, a digital control method which uses the principle of digital phase shift generator is proposed. In addition, phase locked loop(PLL) control is provided which permits speed control with high degree of precision. Conclusions are as follows; (1) There phase SCR rectifier circuit can be phase-controlled by using the principle of digital phase shift generator. (2) An arc-cosine conversion by using PROM greatly improves the linearity of the relationship between firing angle and motor speed, and hence, motor speed can be controlled accurately. (3) By adding the speed control part as shown in Fig.9 to the system, motor speed follows accurately reference speed as shown in Fig.14. (4) By adding to the output side of the phase shift generator six magnitude comparators, each output of which has phase difference of sixty degrees. And so, there needs synchronization of firing pulses only for a single phase cosine voltage wave instead of synchronizations for three phase cosine voltage waves, that is, simplification of synchronization is obtained. (5) The principle of this control system can be extended to higher horse-power drives or to other phase-controlled converter circuits.

      • 칼만증후군(Kallmann's Syndrome) 1례

        Kallmann's syndrome is the most common form of isolated gonadotropin deficiency, characterized by hypogonadotropic hypogonadism due to GnRH deficiency, delayed puberty and smelling difficulty. It occurs sporadic or familial pattern, and the mode of inheritence has not been fully documented. The defect in patient of Kallmann's syndrome occurs at suprapituitary level involving mechanism that regulate GnRH synthesis or release, so this syndrome classified as a secondary hypogonadotropic hypogonadism. The gonadotropin or pulsatile GnRH administration enable successful stimulation of spermatogenesis and fertility. We have experienced 1 patient with Kallmann's syndrome and presented with the review of the literature.

      • 대관령의 일조시간을 이용한 월별 일사량의 추정

        The monthly mean daily global solar radiation and sunshine duration, have been employed in this study to develop correlation model for Daegwallyeong during 2011. The corresponding monthly value of extraterrestrial solar radiation and possible duration of sunshine are calculated for Daegwallyeong meteorological data. Forecasting performance parameters such as coefficient of determination (R2), mean bias error (MBE) and root mean square error (RMSE) are presented as analyzed for the model. The model with multi-linear regression has the highest R2 value of 0.95 and average MBE and RMSE values of −0.11 MJ/m2day and 1.63 MJ/m2day, respectively. The model exhibited negative MBE which shows that the model resulted in underestimation of the global solar radiation relative to the meteorological measured value. The equation could be employed in the estimation of solar radiation at Daegwallyeong, other locations with similar meteorological characteristics.

      • 에어로빅스 하이킥시 운동화 중저의 경도에 따른 지면반력의 변화 및 발의 안정성에 관한 연구

        본 연구는 에어로빅스 운동 중 하이 킥 동작시 운동화 중저의 경도 변화에 따른 바른 안정성과 지면반력의 변화를 연구함으로써 적당한 운동화의 경도를 연구하는 데 있다. 피험자는 에어로빅스 운동을 하는 20대 여성 7명을 대상으로 하였으며, 운동화는 경도를 40(연질), 50(중질), 60(경질)으로 분류하였다. 지면반력과 3차원 영상분석을 통해 운동학적, 운동역학적 변인들의 측정치를 비교 분석한 결과는 다음과 같다. 첫재, 중저의 경도는 제 1 이지시 아킬레스 건각과 후족각에는 영향을 미치나 그 밖의 각도 변인에는 영향을 미치지 않았다. 그리고 경도가 작을수록 회내 운동이 크게 나타나서 후족의 안정성에는 부정적인 영향을 미쳤다. 둘재, 중저의 경도는 최대 충격력, 최대 수직 지면반력, 최대 수직 지면반력 도달 시간, 충격량, 착지 속도에는 크게 영향을 미치지 않는 것으로 나타났는데, 이러한 변인들은 단지 동작의 변화에 따라서 크게 영향을 받는 것으로 사료된다. 즉, 본 연구 결과 중저의 경도가 증가함에 따라서 발이 안정되는 경향을 나타내지만 지면반력에는 큰 영향을 미치지 않는 것으로 나타났다. This study was performed to investigate the influence of midsole hardness of aerobic shoes on motion and attenuation during high kick in order to find out the optimal midsole hardness of aerobic shoes suitable for young females. The subjects on this study were 7 young females who had experienced in aerobics. Three types of aerobic shoes with the midsole hardness of 40, 50, 60 durometers were selected. The lower leg motion during the high kick were measured using a force platform and two 16mm high speed camera. The findings of the study were as follows : 1. The midsole hardness influenced on the initial achilles tendon and rear foot pronation angles, but did not influence the the others angles. As the midsole hardness decreased, the pronation was increased. It brought the nagative effect on the rearfoot stability. 2. The midsole hardness did not influnce on maximal vertical ground reaction force and time to maximal vertical ground reaction force, impluse, take off velocity.

      • 다양한 절삭이송속도로 가공한 A1 7075-T6 소재의 쇼트피닝 전후 피로수명에 관한 연구

        Aluminum alloy 7075-T6 is ideally used for highly stressed parts such as aircraft components because of its excellent strength to weight ratio. This paper presents a study on the effect of feedrate changes on the fatigue life of A1 7075-T6 specimens before and after shotpeening. For this purpose, five groups of specimens, each of which consists of eight specimens, were first machined at five different feedrates (0.05, 0.10, 0.15, 0.20 0.25mm/rev). Half of eight specimens in each group were then peened and the others remained un-peened. Finally, the fatigue life of each specimen was measured by using 4-point rotatry bending machine. In this paper, comparison of fatigue life among ten sets of specimens is presented. One observation is that the fatigue life of unpeened specimens are inversely proportional to the feedrates. On the other hand, the fatigue life of peened specimens are similarly improved regardless of feedrates except 0.05. Based on the results, it is concluded that shotpeening may improve the fatigue life of Aluminium alloy regardless of feedrates. Therefore, machining productivity may also be increased by choosing higher feedrates for structural conponents that need to be strain-hardened by shotpeening after machining
