        Early engraftment of G-CSF-primed allogeneic bone marrow transplantation in pediatric patients regardless of donor-recipient weight differences.

        <P>Harvesting sufficient progenitor cells from bone marrow (BM) for pediatric patients is a challenging process, especially from smaller donors. Growth factor administration to donors prior to harvest results in an enrichment of the graft and leads to early engraftment. A total of 41 patients received a human leukocyte antigen-identical sibling transplantation using granulocyte colony-stimulating factor (G-CSF)-primed BM. All donors received G-CSF 10?μg/kg/day for 2?days prior to harvest. The median weight difference between donor and recipient was 3.9?kg (range, -29.8 to 32?kg), and the median number of CD34(+) cells harvested was 4.16??10(6)/kg (range, 1.17-31.9??10(6)/kg). The median time to neutrophil engraftment was 12?days (range, 10-27 days), and the time for platelet engraftment was 20?days (range, 12-64 days). The cumulative incidence of acute grade 2 to 3 graft-versus-host disease (GVHD) and chronic GVHD was 4.9% and 5.1%, respectively. An analysis according to the weight difference between donor and recipient showed there was no significant difference in harvested CD34(+) cell dose and in time required for engraftment between smaller and heavier donor recipients. G-CSF-primed BM allows successful engraftment and provides a valuable alternative to unstimulated BM and peripheral blood stem cells with good engraftment and tolerable GVHD even in patients with smaller donors.</P>

        <i>APEX1</i> Polymorphism and Mercaptopurine-Related Early Onset Neutropenia in Pediatric Acute Lymphoblastic Leukemia

        <P><B>Purpose</B></P><P>Mercaptopurine (MP) is one of the main chemotherapeutics for acute lymphoblastic leukemia (ALL). To improve treatment outcomes, constant MP dose titration is essential to maintain steady drug exposure, while minimizing myelosuppression. We performed two-stage analyses to identify genetic determinants of MP-related neutropenia in Korean pediatric ALL patients.</P><P><B>Materials and Methods</B></P><P>Targeted sequencing of 40 patients who exhibited definite MP intolerance was conducted using a novel panel of 211 pharmacogenetic-related genes, and subsequent analysis was performed with 185 patients.</P><P><B>Results</B></P><P>Using bioinformatics tools and genetic data, four functionally interesting variants were selected (<I>ABCC4</I>, <I>APEX1</I>, <I>CYP1A1</I>, and <I>CYP4F2</I>). Including four variants, 23 variants in 12 genes potentially linked to MP adverse reactions were selected as final candidates for subsequent analysis in 185 patients. Ultimately, a variant allele in <I>APEX1</I> rs2307486was found to be strongly associated with MP-induced neutropenia that occurred within 28 days of initiating MP (odds ratio, 3.44; p=0.02). Moreover, the cumulative incidence of MP-related neutropenia was significantly higher in patients with <I>APEX1</I> rs2307486 variants, as GG genotypes were associated with the highest cumulative incidence (p < 0.01). <I>NUDT15</I> rs116855232 variants were strongly associated with a higher cumulative incidence of neutropenia (p < 0.01), and a lower median dose of tolerated MP throughout maintenance treatment (p < 0.01).</P><P><B>Conclusion</B></P><P>We have identified that <I>APEX1</I> rs2307486 variants conferred an increased risk of MP-related early onset neutropenia. <I>APEX1</I> and <I>NUDT15</I> both contribute to cell protection from DNA damage or misincorporation, so alleles that impair the function of either gene may affect MP sensitivities, thereby inducing MP-related neutropenia.</P>

        <P>We demonstrate that hybrid structures of graphene and single-walled carbon nanotubes (SWNTs) are precisely controlled at the liquid-gas interface. The functionalized SWNT Langmuir monolayers anchor single-layer graphene nanosheets (GNSs) suspended in water <I>via</I> Coulomb interaction at the interface. This GNS/SWNT hybrid multilayer electrode can be a promising anode material for Li-ion batteries, offering high specific capacity, outstanding power capability, and excellent cyclability.</P>

      • Neuroprotective effect of loganin against Aβ25-35-induced injury via the NF-κB-dependent signaling pathway in PC12 cells

        <P>Amyloid-beta (Aβ) protein, the main constituent of senile plaques, is believed to play a pivotal role in the pathogenesis of Alzheimer's disease (AD). AD is closely associated with inflammatory reactions which are considered to be responses to Aβ deposition. The present study investigated the effect of loganin on Aβ25-35-induced inflammatory damage and the underlying molecular mechanism of its neuroprotective action. Loganin predominantly prevented Aβ25-35-stimulated cell death through suppressing ROS generation, and attenuating apoptosis by inhibiting caspase-3 activity and regulating cell cycle. Furthermore, loganin suppressed the level of TNF-α and protein expression of iNOS and COX-2 in Aβ25-35-injured PC12 cells. These inhibitions appeared to correlate with the suppression of NF-κB activation by loganin, as pre-treating cells with loganin blocked the translocation of NF-κB into the nuclear compartment and degradation of the inhibitory subunit IκB. Loganin substantially inhibited phosphorylation of MAPKs including ERK1/2, p38 and JNK, which are closely related to regulation of NF-κB activation. Taken together, the results implied that loganin attenuated neuroinflammatory responses through the inactivation of NF-κB by NF-κB dependent inflammatory pathways and phosphorylation of MAPK in Aβ25-35-induced PC12 cells.</P>

        Exposure and Risk Assessment of Operators to Insecticide Acetamiprid during Treatment on Apple Orchard

        Occupational exposure and risk assessment were conducted to evaluate the safety of operators when insecticide acetamiprid was applied to apple orchard using a speed sprayer. Dermal patches, cotton gloves, socks, and masks were used to monitor the dermal exposure, and personal air pump with solid sorbent was used to measure the potential inhalation exposure. In validation to analytical methods, the limit of detection and limit of quantitation were 0.25 ng and 1 ng, respectively. Good reproducibility (coefficient variation < 4%), linearity (coefficient determination > 0.999), and recovery (85.3-118.2%) were obtained. Trapping efficiency of solid sorbent was 96.4% while breakthrough did not occur. Only hand exposure was measured on the gloves during mixing/loading to give $33-1,132{\mu}g$. Exposure amount of operator 3 among 4 workers was noticeably high. The total volumes of spray liquid for operators were $535-1,235mL{\cdot}h^{-1}$, corresponding to 0.03-0.08% of the applied spray solution. Highest contaminated parts of body were thighs, chest, and lower legs. The inhalation exposure ratio to the total application amount was significantly low. However, wind seemed to affect the inhalation exposure of operator. For risk assessment, margin of safety was calculated by the application of cloth and dermal penetration rate to obtain values of much larger than 1 in all cases. Therefore, health risk of operators during treatment of acetamiprid in apple orchard could be of least possibility.

        COVID-19’s Radiologic, Functional, and Serologic Consequences at 6-Month and 18-Month Follow-up: A Prospective Cohort Study

        Background: We evaluated the radiologic, pulmonary functional, and antibody statuses of coronavirus disease 2019 (COVID-19) patients 6 and 18 months after discharge, comparing changes in status and focusing on risk factors for residual computed tomography (CT) abnormalities. Methods: This prospective cohort study was conducted on COVID-19 patients discharged between April 2020 and January 2021. Chest CT, pulmonary function testing (PFT), and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) immunoglobulin G (IgG) measurements were performed 6 and 18 months after discharge. We evaluated factors associated with residual CT abnormalities and the correlation between lesion volume in CT (lesionvolume), PFT, and IgG levels. Results: This study included 68 and 42 participants evaluated 6 and 18 months, respectively, after hospitalizations for COVID-19. CT abnormalities were noted in 22 participants (32.4%) at 6 months and 13 participants (31.0%) at 18 months. Lesionvolume was significantly lower at 18 months than 6 months (P < 0.001). Patients with CT abnormalities at 6 months showed lower forced expiratory volume in 1 second (FEV1) and FEV1/forced vital capacity (FVC), and patients with CT abnormalities at 18 months exhibited lower FVC. FVC significantly improved between 6 and 18 months of follow-up (all P < 0.0001). SARS-CoV-2 IgG levels were significantly higher in patients with CT abnormalities at 6 and 18 months (P < 0.001). At 18-month follow-up assessments, age was associated with CT abnormalities (odds ratio, 1.17; 95% confidence interval, 1.03–1.32; P = 0.01), and lesionvolume showed a positive correlation with IgG level (r = 0.643, P < 0.001). Conclusion: At 18-month follow-up assessments, 31.0% of participants exhibited residual CT abnormalities. Age and higher SARS-CoV-2 IgG levels were significant predictors, and FVC was related to abnormal CT findings at 18 months. Lesionvolume and FVC improved between 6 and 18 months.         A Case of Infantile Nephrotic Syndrome associated with Neuroblastoma

        Nephrotic syndrome in the first year of life, characterized by renal dysfunction and proteinuria, is associated with a heterogeneous group of disorders. These disorders are often related to genetic mutations, but the syndrome can also be caused by a variety of other diseases. We report an infant with nephrotic syndrome associated with a neuroblastoma. A 6-month-old girl was admitted with a 10% weight loss over 10 days and nephrotic-range proteinuria. She was ill-looking, and her blood pressure was higher than normal for her age. Her cystatin-C glomerular filtration rate was decreased, and levels of plasma renin, aldosterone, and catecholamines were elevated. Renal ultrasonography and abdominal computed tomography showed a retroperitoneal prevertebral mass encasing both renal arteries and the left renal vein. The mass was partially resected laparoscopically, and the pathologic diagnosis was neuroblastoma. Findings on a simultaneous renal biopsy were unremarkable. The patient was treated with chemotherapy and several anti-hypertensive drugs, including an alpha blocker. Two months later, the mass had decreased in size and the proteinuria and hypertension were gradually improving. In an infant with abnormal renin-angiotensin system activation, severe hypertension, and nephrotic-range proteinuria, neuroblastoma can be considered in the differential diagnosis.

        Evaluating the Prevalence of Foodborne Pathogens in Livestock Using Metagenomics Approach

        Food safety is the most important global health issue due to foodborne pathogens after consumption of contaminated food. Foodborne bacteria such as Escherichia coli, Salmonella enterica, Staphylococcus aureus, Campylobacter spp., Bacillus cereus, Vibrio spp., Yersinia enterocolitica and Clostridium perfringens are leading causes of the majority of foodborne illnesses and deaths. These foodborne pathogens often come from the livestock feces, thus, we analyzed fecal microbial communities of three different livestock species to investigate the prevalence of foodborne pathogens in livestock feces using metagenomics analysis. Our data showed that alpha diversities of microbial communities were different according to livestock species. The microbial diversity of cattle feces was higher than that of chicken or pig feces. Moreover, microbial communities were significantly different among these three livestock species (cattle, chicken, and pig). At the genus level, Staphylococcus and Clostridium were found in all livestock feces, with chicken feces having higher relative abundances of Staphylococcus and Clostridium than cattle and pig feces. Genera Bacillus, Campylobacter, and Vibrio were detected in cattle feces. Chicken samples contained Bacillus, Listeria, and Salmonella with low relative abundance. Other genera such as Corynebacterium, Streptococcus, Neisseria, Helicobacter, Enterobacter, Klebsiella, and Pseudomonas known to be opportunistic pathogens were also detected in cattle, chicken, and pig feces. Results of this study might be useful for controlling the spread of foodborne pathogens in farm environments known to provide natural sources of these microorganisms.

        Complete genome sequence of Salmonella enterica strain K_SA184, multidrug resistance bacterium isolated from lamb (Ovis aries)

        Salmonella enterica is a representative foodborne pathogen in the world. The S. enterica strain K_SA184 was isolated from the lamb (Ovis aries), which was collected from a local traditional market in South Korea. In this study, the S. enterica strain K_SA184 was sequenced using PacBio RS II and Illumina NextSeq 500 platforms. The final complete genome of the S. enterica strain K_SA184 consist of one circular chromosome (4,725,087 bp) with 52.3% of guanine + cytosine (G + C) content, 4,363 of coding sequence (CDS), 85 of tRNA, and 22 of rRNA genes. The S. enterica strain K_SA184 genome includes encoding virulence genes, such as Type III secretion systems and multidrug resistance related genes.

        Morphology Control of Surfactant-Assisted Graphene Oxide Films at the Liquid–Gas Interface

        <P>Control of a two-dimensional (2D) structure of assembled graphene oxide (GO) sheets is highly desirable for fundamental research and potential applications of graphene devices. We show that an alkylamine surfactant, i.e., octadecylamine (ODA), Langmuir monolayer can be utilized as a template for adsorbing highly hydrophilic GO sheets in an aqueous subphase at the liquid–gas interface. The densely packed 2-D monolayer of such complex films was obtained on arbitrary substrates by applying Langmuir–Schaefer or Langmuir–Blodgett technique. Morphology control of GO sheets was also achieved upon compression by tuning the amount of spread ODA molecules. We found that ODA surfactant monolayers prevent GO sheets from sliding, resulting in formation of wrinkling rather than overlapping at the liquid–gas interface during the compression. The morphology structures did not change after a graphitization procedure of chemical hydrazine reduction and thermal annealing treatments. Since morphologies of graphene films are closely correlated to the performance of graphene-based materials, the technique employed in this study can provide a route for applications requiring wrinkled graphenes, ranging from nanoelectronic devices to energy storage materials, such as supercapacitors and fuel cell electrodes.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/langd5/2014/langd5.2014.30.issue-8/la403255q/production/images/medium/la-2013-03255q_0011.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/la403255q'>ACS Electronic Supporting Info</A></P>
