      • Detection of Individuals Restoring Fertility by DNA Fragment Converted into STS (sequence tagged site) in Red pepper

        Co-segregation of male fertility with DNA markers selected by RAPD analysis as being potentially linked to the restorer gene (Rf) for Cytoplasmic male sterility (CMS) was analyzed using segregating F2 population. One RAPD marker directly linked to the Rf locus was identified. Amplification of OPT-02/570 using the STS primers generated a monomorphic band of each fertile plants randomly selected F2 progenies. From these results, this specific marker would be strongly linked to be restoring gene. The use of STS marker is effective in overcoming the reliability of the RAPD phenotype and improving their utility for MAS, co-dominant STS markers are especially very useful.

      • 병원 건축물의 외벽마감재에 따른 배색 이미지에 관한 연구

        The purpose of this study is to find out the coloring image satisfaction according to exterior finishing material of main building on hospital in Gwang-ju City. For this study, six exterior materials were selected such as brick, ston, matal, painting, tile, bare concrete and ninety-one building samples used these exterior materials. The color of main building on hospital in Gwang-ju City was measured by CR-200, color was divided by coloring type. As a result, we could estimate that outer wall images can be effective in case they were taken by color contrast or tone change because clearness factor works depending on un-vagueness, familiarity and similarity of various kinds of colors with high brightness and low chroma even though we use natural materials or artificial materials.

      • 생쥐 대장 평활근 세포에서 내향 정류성 칼륨 전류의 특성 연구

        Objectives: K^(+) channels play an important role in regulating cellular excitability. The aim of this study was to know whether or not inward rectifier K^(+) channel exists in colonic smooth muscle cells. Methods: Mouse colonic smooth muscle cells were isolated using collagenase, and then we recorded their membrane currents using a whole-cell patch clamp technique. Results: With 90 mM K^(+) in bath, hyperpolarization-induced inward currents from -120 mV to 20 mV with 400 ms duration at a holding potential of -10 mV showed rapid activation, inactivation and inward rectification. The inactivation showed single exponential time course. Reduction of external K^(+) to 60 mM decreased the amplitudes of the currents in whole test voltage range and shifted the reversal potential to more negative potential. The inactivation process and peak currents of hyperpolarization-induced inward currents were not affected by removing external Na^(+). External Ba^(+) blocked hyperpolarization-induced inward currents by dose-dependent manner and pure Ba^(+)-sensitive currents showed strong inward rectification. Cs^(+) also suppressed hyper- polarization-induced inward currents. Ba^(+) and Cs^(+)-induced inhibitiOn of hyperpolarization-induced inward currents was voltage-dependent, and the extent of inhibition increasing with membrane hyperpolarization. Conclusions: These results suggest that inward rectifier KU channels may exist in proximal colonic smooth muscle and may play an important role in regulating membrane potential.

        The junction of tension materials, connected with high-strength bolt, should be designed to bear net section destruction, fastener fracture, and fracture of block shear considering yielding of the shear plane and the impact of shear retardation. Block shear among them means that tension side vertical and shear plane parallel to and shear plane horizontal in the direction of weight is torn in the form of "square piece;" it is a type of fracture well known in the junction such as simple beam or gasset plate connected with the bolts. In other words, block shear is a situation of limitations where member resistance is determined by shear strength generated by the destruct line (DL) and tension strength generated by right-angled sections. Since the design law of limitation status of steel architecture was established in 1997, investigation into detailed block shear calculation formula is not thoroughgoing enough, and it is not put into practice very well. This study showed that fracture shape and the proof stress of the fracture of 'ㄱ' steel (angle) are emphasized based on the previous results in order to find out feasibility of design criteria of nlock shear in terms of design law of limitation status and verify calculation formula of block shear.

      • 생분해성 amidoximated cellulose계 chelate 수지의 합성 및 Cu^(2+)흡착에 관한 연구

        2h of premixing, rapid agitation was found to enduce cyanoethylation of cellulose at room temperature, and 80℃, 3h of reaction condition in cosolvent (DMSO:H_(2)O = 50:50) was suitable for over 90% amidoximation of the corresponding cyanoethylcellulose. Amidoximated cellulose did not adsorb copper ion at all at pH below 3 and adsorbed more copper ion in higher pH with the range of 3 to 7. These copper ion adsorbability was determined by atomic absorption spectroscopy. AMC adsorbed more metal ions in the order of Cu^(2+)>Cr^(3+)>Ni^(2+). In addition, the fourth regenerated AMC by treatment in 1N H_(2)SO_(4) for 1h showed a same copper ion adsorbability as original AMC did.

        공사조직간 지식관리활동의 비교연구 : 중앙부처 공무원 및 대기업 직원의 인식조사를 중심으로

        정부조직의 문제해결능력 및 역량의 제고는 정부의 효율성 및 국민에의 대응성을 목표로 하는 것으로 공공부문에서의 지식관리는 민간조직의 지식관리 못지 않게 중요하다. 그러나 우리 정부조직의 경우 지식관리제도 도입시부터 민간기업과는 달리 종합적이고 체계적인 마스터플랜 없이 시작한 부처가 많았으며 지식관리시스템을 구축한 이후 지식관리에 관심을 가지기 시작한 경우가 대부분이었다. 본 연구는 지식획득, 지식축적, 지식공유 및 지식활용 등 지식관리활동정도에 대하여 정부조직과 민간기업의 지식관리활동 정도를 비교분석하여 양 부문간 지식관리 활동정도의 차이가 있는지를 살펴보았다. 그 결과 민간부문에 비해 공공부문의 지식관리 활동이 덜 활발한 것으로 나타났다. 이러한 분석결과를 기반으로 정부조직 지식관리 정책방향을 제시하고 있다.

        The purpose of this study was to offer the basic materials on the secondary physiscl education. The subjects were 129 physiscl education teachers at K provience. The results were as follows: In teaching learning methods, the more educated experience teacher had the more improvement feeled. In the degree of difficulty of current physiscl curriculum, the more educated experience teacher had, the higher it was to be thought. In the goal achievement, the more educated experience had, the more it affected the goal achievement. The efficiency and ability of methods was not shown a significant difference among educated experience group, but shouled be consider it. The equipment and implement of goal achievement was not show a significant difference among educated experience group, but should be consider it. The evaluation of propriety and objectivity and on psychomotor, affective, and cognitive domain was not shown a significant difference.

      • Retinol Palmitate가 연골발생에 미치는 영향에 관한 형태학적 연구

        Vitamin A, an isoprenoid which has cyclohexynyl ring chemically, is known to promote the growth and differentiation during development and regeneration, to maintain the state of health and to improve the vision, and is known to be essential in mucous secretion, reproduction and maintenance of epithelium. Recently, the anticarcinogenic and antimutagenic effects of vitamin A and the teratogenic effects were reported, however the mechanism of action of vitamin A was not confirmed thoroughly. In this experiment, the morphological changes after administration of retinol palmitate during the development of hindlimb and differentiation of chondrocytes were observed in the rat fetuses. SpragueDawley rats were used as an experimental animal. The rats were divided into 2 groups : control and vitamin A(retinol palmitate) treated group. In retinol palmitate treated group, 500mg/kg of retinol palmitate(Sigma Chemicals Co.) dissolved in corn oil(Sigma Chemicals Co.) was administered orally with gastric tube once at the day 8.5 of gestation. In the control group, corn oil of equal volume as in retinol palmitate treated group was administered. On the gestation day 12.5, 13.5, 14.5, 15.5 and 16.5, the rats were sacrificed under ether anesthesia, laparotomized and hysterectomized. Under stereoscope the pregnant sacs and the fetuses were observed, and the body weight of each fetus was measured. For the observation of external development of hindlimb, the embryos were stained with Nile blue sulfate, and compared the differences between 2 groups. The histological changes were observed light microscopically after routine processing, and H-E & alcian blue staining, and the ultrastructural changes of chondrocytes observed electron microscopically. The results were as follows ; 1. The survival rate of fetuses was lowered in the retinol palmitate treated group significantly except the day 12.5 of gestation. 2. In the retinol palmitate treated group, the development of hindlimb bud and aggregations of precarti lagenous mesenchymal cell were decreased significantly on the day 12.5 and 13.5 of gestation. 3. Cartilage matrix formation was found initially on the day 13.5 of gestation in control group and on the day 14.5 of gestation in retinol palmitate treated group. 4. Electron microscopic changes of chondrocytes after retinol palmitate administration were as the followings : mitochondrial and IR swelling, decreased filopodia, reduced intercellular space and cartilage matrix, abnormal aggregation of glycogen particles and increased intercellular junctions. According to the above results, it is considered that retinol palmitate has an inhibitory effect on the hindlimb developmnet by reducing the aggregations of precartilagenous mesenchymal cell and chondrocytic structural and metabolic changes.

      • 단독주택의 외관형태와 이미지평가에 관한 연구

        This study analyzes the types of external forms of detached housing buildings, and conducts the S.D test with it and is to structurize the image of external form of buildings. Types of 24 detached housing buildings are analyzed in this vein and the results are as follows: (1) Mass type in the primary cognitive factor of building is mostly simple or multiple box type. (2) As a result of analyzing the secondary cognitive factor of building, horizontal type is most and it shows that it is the favorable structure in the stability of form. Roof shape and side pattern are mostly gable and plat roofs and it is known that traditional gable roof and flat roof influenced by modern construction are shown in housing. (3) To examine the tertiary cognitive factor, surface shape is mostly flat type and it shows a little monotonous composition. Also finishing materials are concrete and brick. S.D test of detached houses doesn't show the extreme psychological test disposition and does somewhat affirmative evaluation. As a result of conducting the factor analysis based on S.D test, nine psychological factors including 「originality」, 「pleasantness」, 「harmony」, 「ornamentation」, 「openness」, 「balance」, 「rhythm」, 「stability」 and 「orientation」. These can be called psychological factor axis which can evaluate the modern detached house.
