        Dimethylnitrosamine 유발 급성 간 손상 흰쥐에서 ^(99m)Tc-Lactosylated Serum Albumin을 이용한 간 기능의 평가

        목적: ^(99m)Tc-lactosylated serum albumin (^(99m)Tc-LSA)은 간세포에 특이적으로 결합하는 간수용체 영상용 방사성의약품으로 새로이 합성되었다. 간섬유화를 유발하는 dimethylnitrosamine (DMN)을 투여한 간 손상 휜쥐 모델에서 ^(99m)Tc-LSa의 역동학적인 간섭취를 조사하고 간효소치의 변화와 조직학적 소견을 비교하여, LSA의 간섭취가 간기능의 변화를 반영하는지를 연구하였다. 대상 및 방법: SD계 흰쥐에 DMN를 27 mg/kg으로 복강 내 주사하여 급성 간손상을 유도하고 대조군과 비교하였다. DMN을 주사한 흰쥐를 3일(DMN-3), 8일(DMN-8), 21일(DMN-21)에 ^(99m)Tc=LSA (1,665 mg/kg) 29 MBq를 정맥 주사하여, 30분 동안 동적 영상을 획득하고 간과 신장부위에 관심영역을 설정하여 간과 심장부위의 시간방사능 곡선을 얻었다. 간기능 평가를 위해 시간방사능 곡선을 이용하여 간섭취지수와 혈중제거지수를 구하였고 곡선 최적화를 시행하였다. DMN 투여군과 대조군의 간효소치의 변화와 간조직의 광학현미경 소견을 비교하였다. 결과: 대조군에서는 ^(99m)Tc-LSA가 빠르게 간에 섭취되고 혈중에서 제거되었으나 DMN을 처리한 군에서는 간섭취가 낮았다. 간섭취지수의 비교에서 대조군에 비해 DMN 처리군에서 유의하게 간섭취지수가 낮았다(DMN-3: 0.842, DMN-8: 0.898, DMN-21: 0.91, 대조군: 0.96, p<0.05). 혈중제거지수의 비교에서도 대조군에 비해 DMN 처리군에서 혈중제거지수가 유의하게 높았다(DMN-3: 0.731, DMN-8: 0.654, DMN-21: 0.604, 대조군: 0.473, p<0.05). 비선형 회귀분석에서 R_(2) 값은 0.9이상으로 좋은 일치를 보였고, 대조군에ㅓ K값이 DMN처리군에 비해 크고(DMN-3: 0.28, DMN-8: 0.41, DMN-21: 0.46, 대조군: 0.97, p<0.05), T_(1/2)값은 작았다(DMN-3: 2.5, DMN-8: 1.7, DMN-21: 1.5, 대조군: 0.7, p<0.05). 간효소치의 변화는 DMN-3군에서는 대조군에 비해 상승하였으나 DMN-8, DMN-21군에서는 간효소치의 상승이 관찰되지 않았다. 간조직 소견의 경우 DMN-3군에서 중심정맥 주위에 괴사가 관찰되었으나 DMN-8군, DMN-21군에서는 미약한 정도의 염증세포 침윤만이 관찰되었다. 결론: ^(99m)Tc-LSA 간신티그래피의 간섭취 정도는 간손상과 반비례하였으며 간섭취의 변화는 조직학적 손상이 심한 정도와 간손상후 회복되는 과정을 반영하여 주었다. ^(99m)Tc-LSA 간신티그래피가 간손상을 평가하고 간손상후 회복되는 과정을 추적하는 간수용체 영상용 방사성 의약품으로 사용될 수 있을 것으로 생각된다. Objects: ^(99m)Tc-lactosylated human serum albumin(LSA) is a newly synthesized radiopharmaceutical that binds to asialoglycoprotein receptors, which are specifically presented on the hepatocyte membrane. Hepatic uptake and blood clearance of LSA were evaluated in rat with acute hepatic injury induced by dimethylnitrosamine(DMN) and results were compared with corresponding findings of liver enzyme profile and these of histologic changes. Materials and Methods: DMN (27 mg/kg) was injected intraperitoneally in Sprague-Dawley rat to induce acute hepatic injury. At 3(DMN-3), 8(DMN-8), and 21(DMN-21) days after injection of DMN, LSA injected intravenously, and dynamic images of the liver and heart were recorded for 30 minutes. Time-activity curves of the heart and liver were generated from regions of interest drawn over liver and heart area. Degree of hepatic uptake and blood clearance of LSA were evaluated with visual interpretation and semiquantitative analysis using parameters (receptor index : LHL3 and index of blood clearance : HH3), analysis of time-activity curve was also performed with curve fitting using Prism program. Results: Visual assessment of LSA images revealed decreased hepatic uptake in DMN treated rat, compared to control group. In semiquantitative analysis, LHL3 was significantly lower in DMN treated rat group than control rat group (DMN-3:0.842, DMN-8: 0.898, DMN-21: 0.91, Control: 0.96, p<0.05), whereas HH3 was significantly higher than control rat group (DMN-3: 0.731, DMN-8: 0.654, DMN-21: 0.604, Control: 0.473, p<0.05). AST and ALT were significantly higher in DMN-3 group than those of control group. Centrilobular necrosis and infiltration of inflammatory cells were most prominent in DMN-3 group, and were decreased over time. Conclusion: The degree of hepatic uptake of LSA was inversely correlated with liver transaminase and degree of histologic liver injury in rat with acute hepatic injury.

      • Anti-Proliferative Effect of an Aqueous Extract of <i>Prunella vulgaris</i> in Vascular Smooth Muscle Cells

        <P>The abnormal proliferation of vascular smooth muscle cells (VSMCs) in arterial walls is an important pathogenic factor of vascular disorders such as diabetic atherosclerosis. We have reported the anti-inflammatory effect of an aqueous extract from <I>Prunella vulgaris</I> (APV) in vascular endothelial cell. In the present study, APV exhibited inhibitory effects on high glucose-stimulated VSMC proliferation, migration, and invasion activities, inducing G<SUB>1</SUB> cell cycle arrest with downregulation of cyclins and CDKs and upregulation of the CKIs, p21<SUP>waf1/cip1</SUP> and p27<SUP>kip1</SUP>. Furthermore, APV dose dependently suppressed the high glucose-induced matrix metalloproteinase activity. High glucose-induced phosphorylation of ERK, p38 MAPK, was decreased by the pretreatment of APV. NF-<I><I>κ</I></I>B activation by high glucose was attenuated by APV, as an antioxidant. APV attenuated the high glucose-induced decrease of nuclear factor E2-related factor-2 (Nrf2) translocation and heme oxygenase-1 (HO-1) expression. Intracellular cGMP level was also increased by APV treatment. These results demonstrate that APV may inhibit VSMC proliferation via downregulating ROS/NF-<I><I>κ</I></I>B /ERK/p38 MAPK pathways. In addition, APV has a beneficial effect by the interaction of Nrf2-mediated NO/cGMP with HO-1, suggesting that <I>Prunella vulgaris</I> may be useful in preventing diabetic atherosclerosis.</P>

        In recent years, gonorrhea has been pandemic and remains one of the most common STDs in the world, especially in developing countries. For the detection of a more effective therapeutic regimen and assessing the prevalence of Penicillinase Producing Neisseria gonorrhoeae(PPNG), we have been trying to study the patients who have visited the Venereal Disease Clinic of Choong-Ku Public Health Center in Seoul since 1980 by menas of the chromogenic cephalosporin method. In 1998, 93 strians of N. genorrhoeae were isolated, among which 60(64.5%) were PPNG. The prevalence of PPNG in Seoul, which had been decreased to 39% in 1996 after a peak of 74.3% in 1993, is increased to 64.5% in 1998.

        Acid rain was strongly raised in regional air pollution problems. All of researchs have been focused in plt tendency in a limited area. But, chemical composition data are important because of the control strategy are also necessary to reduce source emission from various air pollution sources. Longtime may be helpful to evaluate acid rain effect to regional area for a long time. We have accumulated acid data from Sep. 1993 to Jan. 2003. In this study, data which was given from Sept., 1993 to 2000 is measured pH and EC, analyzed cations anions by ion chromatography. All of analyzed data were calculated to fit average and standard by SAS package.

      • 극 저 에너지 비소 및 붕소 이온 주입시 효과적인 분자 역학 방법을 사용한 불순물 분포와 격자 결함 분포 예측

        본 논문은 분자 역학 방법(molecular dynamics method)을 사용하여 극 저 에너지 비소 및 붕소 이온 주입의 원자단위에 관한 연구로서 국부 격자결함 축적 모델(local damage accumulation model)을 제안하고, 이를 사용한 이온 주입 시뮬레이션 결과를 보여주고 있다. 국부 격자결함 확률 축적 함수는 각 단위 셀에 축적된 에너지, 이온빔 전류, 기판 물질, 주입 이온과 이온 발생 순서 등으로 구성되어 있다. 시뮬레이션 결과는 SIMS 측정치 및 다른 분자 역학 시뮬레이션과 잘 일치한다. 격자결함을 고려하지 않는 MDRANGE는 격자 결함으로 인한 채널링 억제 효과가 나타나지 않기 때문에 불순물 분포의 꼬리 부분에서 많은 차이를 보였다. 또한 국부 격자결함 축적 모델을 사용한 경우와 이를 고려하지 않은 경우의 불순물과 격자결함에 관한 2차원 분포를 계산하여 국부적으로 축적된 격자결함이 이온의 진행에 미치는 영향에 관하여 연구하였다. We have investigated the effects of atomic dynamics for ultra-low energy As and B ion implant using highly efficient molecular dynamics(MD) scheme. We simulate ion implantation by MD simulation using energetic particle approximation method and local damage accumulation model proposed in the paper. Local damages accumulation probability function is composed of deposited energy in a unit cell, implant dose rate, target material, projectile atom, and recoil event number. The simulated results were good agreement with the experimental and other simulated results. The MDRANGE results considering no damages were different from SIMS data in the tail region. Using the local damage accumulation model and energetic particle approximation method, we simulated 2 dimensional dopant and damage profiles.

      • 배기가스 재순환에 의한 실린더 및 피스톤 마모에 미치는 영향에 관한 연구

        The effects of recirculated exhaust gas on the wear of cylinder liner, piston and piston rings have been investigated by the experiment with a two-cylinder, four cycle, indirect injection disel engine operating at 75% load and 1600 rpm spped. For the purpose of comparison between the wear rates of two cylinders with and without EGR, the recirculated exhaust gas is sucked into one of two cylinders after the soot among exhaust emissions is removed by an intentionally designed cylinder-type scrubber equipped with 6 water injectors(A water injector has 144 nozzles of 1.0 mm diameter), while only the fresh air into another cylinder. These experiments are carried out on the fuel injection at a fixed 15.3˚BTDC timing. It is found that firstly the mean wear amount of cylinder liner with EGR is more increased in the measurement positions of the second half than of the first and the mean wear amoount without EGR is almost uniform regardless of measurement positions, secondly the wear rates of the first and second piston ring(compression ring) thickness with EGR are more than twice, but the wear rate of oil ring thickness without EGR is more increased than that with EGR, and finally the wear rate of piston skirt with EGR is a little bit increased, but the piston head diameter is rather increased owing to soot adhesion and corrosion wear, and especially larger with EGR.

      • 견인용 동기전동기의 시동 및 운전특성 해석에 관한 연구

        It needs a large capacity of equipments to test the starting performance of middle and large synchronous motors in a factory. This paper propose a new method which is the combination of single phase AC applied test and usual routine test with a small synchronous test machine and testing equipments. We can obtain the results from these proposed tests as follows ; (1) The starting performance and stabilized operation at variable speed is predicted from this results. (2) The armature winding leakage reactance is resulted from the no-load saturation curve, the short-circuit charactristic curve and the armature-reaction magnetomotive force converted to the field current. (3) This test must be performed twice, one for short-circuiting the field winding, the other for joining a resistance between the field terminals, as a result the unknown constants in equivalent circuits are identified. (4) Inverter-fed three phase voltage with variable frequency is applied to armature terminals, as a result, We show the actual armature leakage reactance and reaction of the induced harmonic voltages from rotor.

      • 골격근 근형질세망의 ATP유도 ^45Ca-uptake에 대한 Thapsigargin및 Vanillylnonanamide의 영향

        To investigate the effect of thapsigargin(THP) and vanillylnonanamide(VN), derivative of capsaicin, on the ATP-induced ^45Ca-uptake in the sarcoplasmic reticulum(SR) of the skeletal muscle, the SR vesicles were prepared from the back muscle of the rabbit, and ^45Ca-uptake was carried out. The results as follows: ATP-induced ^45Ca-uptake of skeletal muscular SR was significantly increased by 1 mM ATP. It was significantly blocked by 5 μM THP, but slightly decreased by 20 μM VN. The combined effect of THP and VN on the 45Ca-uptake of the SR vesicles was much potentiated than the sum of each effect of them. The above results suggest that the action of VN, being slightly influential to ATP-induced Ca-uptake but potentiating the effect of THP on the Ca-uptake, might be related with direct perturbation of the SR membrane or exposure of the THP-binding site.

        제2형 근관의 확대에 따른 최종근관장파일 크기의 비교

        제2형의 근관형태를 갖는 치아에서 각각의 근관을 모두 근관장까지 확대한 경우,여러 확대기구에 따른 최종근관장파일의 크기를 상호 비교하여 얼마나 많은 변화가 있었는지를 분석하고자 치근 우식이 없고 치근단공 형성이 완료되었으며 2개의 근관 입구에서 시작하여 하나의 치근단공을 갖는 제 2형의 근관형태를 보이는 상악 소구치,상악 대구치의 근심협측 치근 및 하악 대구치의 근심 치근을 선택하여 확대기구에 따라 KF 군, PT 군,HS 군, K3 군으로 분류하고 치경부 조기 확대를 시행하였다. 초기근관장파일이 15번 크기인 지아를 각 군당 20개씩 무작위로 선택하였고,모든 근관을 각 군에 해당되는 30번 크기의 파일까지 제조회사의 지시에 따라 확대를 시행한 후,촉감과 universal testing machine을 이용하여 최종근관장파일의 크기를 결정하였다. 각 군의 최종근관장파일의 크기와 제거 시 요구되는 힘의 크기에 대한 유의성 검증은 one-way ANOVA를 이용하여 비교 분석하였으며,사후검정은 Tukey HSD test를 이용하여 0.05의 유의수준에서 분석하였다. 본 연구의 결과 제2형의 근관형태를 갖는 치아의 근관확대 시,모든 근관을 각각의 근관장으로 확대하는 방법은 확대에 마지막으로 이용된 기구의 크기보다 한,두 단계 정도 증가된 최종근관장파일을 나타냈다. 그러므로 제2형 근관형태를 나타내는 치아의 근관치료 시 임상가들은 근관충전에 앞서 치근단 받침을 다시 확인하고 형성하는 과정이 필요하리라 사료된다. Type II root canal was defined that two canals leave the chamber and merge to form a single canal at short of the apex. The aim of this study was to analyse the master apical file (MAF) size according to various instrumentation techniques in the type II root canal when each canal was enlarged to working length. Eighty mesial roots of molar with ISO #15 initial apical file (IAF) size in type II root canals were randomly divided into four experimental groups with 20 teeth each. According to enlarging instruments, four groups are: K-FLEXOFILE^(?) (KF), engine-driven Ni-Ti PRoT_(APER)^(?)(PT), HERO Shaper^(?)(HS), K³™ (K3). All canals were enlarged to each working length with ISO #30 size: #30 in KF, F3 in PT, .04/30 in HS, and .06/30 in K3. The master apical file (MAF) size was confirmed by tactile sensation and universal testing machine (EZ test, Shimadzu Co., Kyoto, Japan). The mean MAF size was statistically compared using one-way ANOVA and Tukey HSD test at the 0.05 probability level. These results show that the MAF size was appeared one or two sizes larger than the final enlarging instrument when all canal in type II configuration were enlarged to each working length. Therefore, the clinician have to confirm the apical stop once more after instrumentation of type II root canal.
