Oral Delivery of Probiotics in Poultry Using pH-Sensitive Tablets

As alternatives to antibiotics in livestocks, probiotics have been used, although most of them in the form of liquid or semisolid formulations, which show low cell viability after oral administration. Therefore, suitable dry dosage forms should be developed for livestocks to protect probiotics against the low pH in the stomach such that the products have higher probiotics survivability. Here, in order to develop a dry dosage forms of probiotics for poultry, we used hydroxypropyl methylcellulose phthalate 55 (HPMCP 55) as a tablet-forming matrix to develop probiotics in a tablet form for poultry. Here, we made three different kinds of probiotics-loaded tablet under different compression forces and investigated their characteristics based on their survivability, morphology, disintegration time, and kinetics in simulated gastrointestinal fluid. The results indicated that the probiotics formulated in the tablets displayed higher survival rates in acidic gastric conditions than probiotics in solution. Rapid release of the probiotics from the tablets occurred in simulated intestinal fluid because of fast swelling of the tablets in neutral pH. As a matrix of tablet, HPMCP 55 provided good viability of probiotics after 6 months under refrigeration. Moreover, after oral administration of probiotics-loaded tablets to chicken, more viable probiotics were observed, than with solution type, through several digestive areas of chicken by the tablets.

Succinic acid production by genetically engineered C. glutamicum from lignocellulosic biomass requires the hydrolysis of carbohydrate polymers into fermentable syrup. A variety of toxic compounds are produced such as aldehydes and organic acids, while the hydrolysis of hemicellulose with dilute acid. In this study, we have investigated the toxicity of representative aldehydes (furfural, 5-hydroxymethylfurfural, syringaldehyde, and vanillin) and organic acids (ferulic, 4-hydroxybenzic, vanillic, protocatechuic acid) on growth and succinic acid accumulation of C. glutamicum NC-1. In the presence of various inhibitors of growth experiment, furfural, 5- hydroxymethylfurfural appeared less toxic to growth of C. glutamicum NC-1, syringaldehyde almost completely inhibitor growth of C. glutamicum NC-1, vanillin has inhibited the growth of 67%, of organic acids, only ferulic appeared toxic to growth of C. glutamicum NC-1. Of succinic acid accumulation experiment under oxygen deprivation, all the organic acids compounds showed little inhibition on the glocuse consumption and succinic acid accumulation of C. glutamicum NC-1, but furfural, 5- hydroxymethylfurfural and vanillic have decreased the production of succinic acid. In addition, the actual inhibitor mixtures from the acid hydrolysate of corn cobs have reduced the accumulation of succinic acid. Across further research showed, a reason of succinic acid yield decrease was the malic enzyme activity was inhibited.

In our cell based screening of antitumor ingredients from plants, the EtOH extract of Garcinia bracteata displayed antiproliferative effect against human lung adenocarcinoma A549 cells, human breast cancer MCF-7 cells, and human prostate cancer PC3 cells. Phytochemical investigation of this active extract produced nine ingredients, and their structures were established by analysis of MS and NMR spectra. Antiproliferative evaluation of isolated ingredients on A549, MCF-7 and PC3 cells indicated that a xanthone named isobractatin (1) exhibited potent antiproliferative activity against the above three human cancer cell lines with $IC_{50}$ values ranging from 2.90 to $4.15{\mu}M$. Treatment of PC3 cells with 1 led to an enhancement of the cell apoptosis, and arrested cell cycle in the G0/G1 phase. The G0/G1 phase cycle-related proteins analysis showed that the expressions of cyclins D1 and E were reduced by 1, whereas the protein level of cyclin dependent kinase (CDK) inhibitor P21 was induced. Additionally, 1 enhanced PC3 cell apoptosis by activations of Bax, caspases 3 and 9, and by inhibition of Bcl-2. Our combined data illustrated that isobractatin (1) was the antiproliferative ingredient of G. bracteata against three human cancer cell lines, which exerted its antiproliferatrive effect via cell cycle arrest and induction of apoptosis.

Two new ursane-type triterpenoid saponins,bodiniosides M (1) and N (2), along with three knownsaponins, oblonganosides I (3), pseudobuxussaponin B (4)and bodinioside A (5), were isolated from the aerial partsof Elsholtzia bodinieri. The structures of compounds 1 and2 were characterized by spectroscopic data as well as acidhydrolysis and GC analysis as 3-O-b-D-xylopyranosyl-19ahydroxy-23-acetoxy-urs-12(13)-en-28-oic acid 28-O-a-Lrhamnopyranosyl-(1-2)-b-D-glucopyranoside and 3-O-b-Dglucopyranosyl-2a,19a-dihydroxy-urs-12(13)-en-28,20blactone. Compounds 1 and 5 exhibited potent anti-HCVactivities in vitro with a selective index of 6.53 and 4.41,respectively.
Health management of laser rangefinder considering maximum continuous working time

High-power lasers have severe thermal effects. The heat buildup during longterm operation will increase the threshold current of the laser and the internal defects, which will seriously affect the lifetime of the device. A temperature sensor is difficult to install on the laser rangefinder due to the small size of the solid-state laser device. Considering this situation, this work analyses the thermal mechanism of the laser rangefinder and proposes a temperature calculation method for the crystal working substance inside the laser rangefinder based on the finite element simulation to obtain the crystal center temperature and thermal model with the working time. Then, the prediction model of the maximum continuous working time of the laser can be obtained. The proposed model can provide an early scientific warning for the remaining continuous working time of the solid-state laser and improve the single-shot utilization of the solid-state laser to a certain extent. A comprehensive health management framework of laser rangefinders including FMEA is also proposed.
Anti-proliferation Effects of Interferon-gamma on Gastric Cancer Cells

IFN-${\gamma}$ plays an indirect anti-cancer role through the immune system but may have direct negative effects on cancer cells. It regulates the viability of gastric cancer cells, so we examined whether it affects their proliferation and how that might be brought about. We exposed AGS, HGC-27 and GES-1 gastric cancer cell lines to IFN-${\gamma}$ and found significantly reduced colony formation ability. Flow cytometry revealed no effect of IFN-${\gamma}$ on apoptosis of cell lines and no effect on cell aging as assessed by ${\beta}$-gal staining. Microarray assay revealed that IFN-${\gamma}$ changed the mRNA expression of genes related to the cell cycle and cell proliferation and migration, as well as chemokines and chemokine receptors, and immunity-related genes. Finally, flow cytometry revealed that IFN-${\gamma}$ arrested the cells in the G1/S phase. IFN-${\gamma}$ may slow proliferation of some gastric cancer cells by affecting the cell cycle to play a negative role in the development of gastric cancer.
Pepsin and Laryngeal and Hypopharyngeal Carcinomas

Laryngeal and hypopharyngeal carcinomas are common malignant tumors of the head and neck, and the incidence of both is increasing. Laryngopharyngeal reflux refers to the retrograde flow of gastric contents into the larynx, oropharynx, and/or nasopharynx. It remains controversial whether laryngopharyngeal reflux is a risk factor for laryngeal and hypopharyngeal cancers. The refluxing substances mainly include hydrochloric acid, pepsin, and occasionally bile acids and bile salts, as well as bacteria that colonize the gastrointestinal tract. Loss of epithelium in the mucous membrane of the larynx and hypopharynx is thought to be caused by pepsin. Here, we review the relationships between laryngopharyngeal reflux and both laryngeal and hypopharyngeal carcinomas, as well as the significance of pepsin, methods of clinical detection, and the mechanism of carcinogenesis.
