Supplement of Allicin during IVM Enhances the Oocyte Maturation and Developmental Competence

The efficiency of the in vitro production of oocytes is significantly lower than that of in vivo culture. Allicin (AL) is generally known to regulate the cellular redox, proliferation, viability, and cell cycle of different cells against extracellular-derived stress. This study performed to reveal the effects of allicin treatment on porcine oocyte maturation and developmental competence. Cumulus oocyte complexes were cultured in IVM medium, containing 0, 0.01, 0.1, 1, 10, and 100 μM AL for 44 h. The rate of polar body emission was tended to be higher in the 0.1 AL-treated group than in the control group (p<0.1). The rates of cleavage and blastocyst formation significantly increased in the 0.1 AL-treated group (p<0.05). The reactive oxygen species level at metaphase II did not significantly differ among all groups. The expression of maternal marker (BMP15 and CCNB1) and anti-apoptotic genes significantly increased in the 0.1 AL-treated group at MII stage, respectively (p<0.05). The rate of phosphorylated p44/42 MAPK to p44/42 MAPK was ~4-fold higher in the 0.1 AL-treated group than in the control group (p<0.05). Consequently, these results indicate that supplementation of oocyte maturation medium with allicin during IVM improves the maturation of oocytes and the subsequent developmental competence of porcine oocytes. In conclusion, allicin enhances the efficiency of IVM to establish a novel IVC system, and we suggested that it will be helpful reagent as assisted reproductive technology (ART) in the future.
Pioglitazone Improves Oocyte Maturation and Subsequent Developmental Competence of Porcine Embryos

The optimization of culture condition is both basic and important for improving oocyte maturation, subsequent development of preimplantation embryo, and further implantation. This study performed to confirm effect of pioglitazone (PIO) treatment directly on porcine oocyte during in vitro maturation, further development of embryo. Immature oocytes were cultured in IVM medium, containing 0, 0.01, 0.1, 1, and 10 μM pioglitazone (0, 0.01, 0.1, 1, and 10 PIO, respectively) for 44 h. Although both oocyte survival and polar body emission did not significantly differ between the control and PIO-treated groups, percentage of normal spindle formation and chromosome alignment was increased in the 1 PIO-treated group than in the control group (control, 60.9±8.2%; and 1 PIO, 81.5±5.9%, p<0.1). Level of GSH was significantly increased in 1 PIO-treated group than in the control group (control, 100.0±5.0%; and 1 PIO, 119.8±5.2%, p<0.05), while the level of ROS was not differ in all groups. The rate of blastocyst was significantly higher in the 1 PIO-treated group than in the control at day 6 (control, 29.6±2.5%; and 1 PIO, 37.9±2.3%, p<0.05) and 7 (control, 35.1±1.6%; and 1 PIO, 41.8±2.7%, p<0.05), respectively. These results indicated that PIO treatment during IVM showing beneficial effects on oocyte and embryonic development. Pioglitazone will contribute to enhancing efficiency of in vitro production.

The pretreatment for substrate was carried out in change of gun pressure of bar using wet blasting. The size of powder was about . As the results, the surface roughness of cemented carbide substrate was improved with increment of gun pressure of wet blasting. A new surface layer was formed and Co particles were uniformly distributed over the entire surface after pretreatment. The adhesion of the pretreated substrate in same PVD-TiAlN film was improved and in approximately shown the best adhesion value.

쏘일네일 벽체는 국낸에서 과거 10년간 가시설 및 영구구조물에 상당수 시공되어 왔다. 그러나 쏘일네일링 공법은 많은 장점이 있음에도 불구하고 인접한 건물의 하부에 쏘일네일이 삽입되는 것을 허용하지 않아 제한적으로 사용되고 있는 실정이다. 따라서 본 논문에서는 쏘일네일이 인접한 건물의 하부에 삽입되는 문제를 해결하고 철근을 재활용할 수 있는 방안으로 제거식 쏘일네일링 공법을 소개하였다. 제서식 쏘일네일링 공법은 연직굴착시 가시설 흙막이벽체로 주로 사용할 수 있다. 제거식 쏘일네일의 인발특성을 규명하기 위해 총 23회의 현장인발실험을 실시하였으며, 이를 통해 고정자 소켓의 설치간격과 시멘트 그라우트의 W/C 비가 중요한 설계인자라는 것을 알 수 있었다. 또한 본 논문에서는 국내에서 처음으로 시공되는 제거식 쏘일네일 벽체의 설계 및 시공과 시공상태 등을 소개하였으며, 제거식 쏘일네일의 철근을 제거하기 전과 후에 벽체의 변형을 계측자료로부터 분석하였다.

Increasing environmental pressures to reduce NOx emission are being placed on coal-fired boilers. To meet the environmental requirements, Doosan Heavy Industries & Construction Co., Ltd.(Doosan) has developed low NOx pulverized coal burner. Low NOx pulverized coal burner has already delivered, and it’s combustion performance was evaluated to the NOx and Unburned Carbon(UBC) during the commissioning tests. The test results are shown that the strong relationship is existed between NOx and OFA flow rate, and also fuel-N fraction of coal has effected on NOx emission.
