        Sphingosine 1-Phosphate and Sphingosine Kinase Activity during Chicken Embryonic Development

        The chicken embryo has been weil used in studies of the developmental process, and during development sphingosine and sphingosine 1-phosphate (So1P) are considered critical mediators of cell death and survival. In this study, we compared the sphingolipid contents of chicken embryos during the early embryonic development period from day 3 to day 6. HPLC analyses of sphingosine and So1P in chicken embryos revealed that sphingosine levels were greatly reduced on day 4 whereas So1P levels were not significantly changed. Sphingosine kinase(Sphk) activities, which require sphingosine as substrate to produce So1P, were also greatly reduced on day 4. Collectively, we found sphingosine levels and Sphk activities, but not So1P levels are changed in early stage of chicken embryos development.

        To detect the numerical changes of the chromosome 13 and 17, containing tumor suppressor genes (retinoblastoma gene and p53) in normal tissue, intraepithelial neoplasm and carcinoma of the prostate, the Fluorescence In Situ Hybridization technique with paraffin embedded tissue was done. Materials observed in this study consisted of 14 cases of lacalized carcinoma(TB1NOMO) and 1 case of locally infiltrating cancer(TC1NOMO)> The chromosomal number changes were counted at normal tissue, intraepithelial neoplasm and neoplastic gland at the same radical prostatectomy specimen. From the 4 patients, studied prospectively, fresh cancer tissue and peripheral whole blood were collected for the primary culture of the cancer cells and lymphocytic culture. There cultured cells were used fro karyotyping. The results were as follows : 1. There were no significant chromosomal number changes between 13 and 17 in each group. 2. The normal non-neoplastice duct, intraepithelial neoplasm and neoplastic gland of experimental group showed no significant difference in chromosomal change. 3. There were significant changes more than 30% in chromosomal numbers between control Y chromosome and experimental chromosome 13 and 17 in each group. 4. The karyotyping on primary cultured prostate cancer cells and lymphocyte showed significant difference in chromosomal number in spite of the same host, representing local chromosomal changes.

        용량별 죽력 투여가 생쥐의 간과 신장의 조직 및 혈액학적 변화에 미치는 영향

        Objectives : Bambusae Caulis in Liquamen extracts (BE) has been used for various curative effect that remove intense heat from the heart, replenish deficiency Yin, nourish deficiency blood, resolve phlegm. This study was performed to investigate the toxicity of BE to mice according to the administered dosage. Methods : The experimental groups divided four. Control group treated saline, J1 group treated BE and distilled water in the ratio of 1:15(0.05ml/kg), F2 group treated BE and distilled water in the ratio of 1:7(0.05ml/kg), J3 group treated BE and distilled water in the ratio of 1:17(0.05ml/kg). We administered BE to the mice for 24 weeks at interval of 48 hours, and investigated the body weight, changes in hematological profile, activities of the transaminase(GOT, GPT) and two hepatic antioxidant enzyme(SOD, catalase), and histological changes of the liver and kidney. Results : Leukocytes of J3 group were increased in the administered for 12 weeks, but decreased in the administered for 24 weeks. Platelets were decreased about 36% in J3 group treated for 12 weeks, but increased about 50% in the same group treated for 24 weeks. Blood total proteins were decreased slightly in the all experimental groups compared with control group. The activities of the transaminase and superoxide dismutase(SOD) were increased, but decreased activities of catalase in the J2 and J3 groups administered for 12 or 24 weeks. In the light microscopical observation, ballooned and injured hepatocytes in portal vein area of J1 group occurred, and dilated hepatocytes and disoriented sinusoid observed in J2 group administered extracts to mice for 24 weeks. Apoptosis occured in the ballooned and injured hepatocytes of the portal vein area in J3 group. A agglutination of RBC in glomerular capsule of the kidney observed in the J2 group, and destructed convoluted tubule observed in the J3 group treated for 24 weeks. Conclusions : These results indicate that mice treated with high dose BE develop a hematological changes, dysfuncton of the liver and kidney.

        Genome-Wide Analysis of DNA Methylation before- and after Exercise in the Thoroughbred Horse with MeDIP-Seq

        Athletic performance is an important criteria used for the selection of superior horses. However, little is known about exercise-related epigenetic processes in the horse. DNA methylation is a key mechanism for regulating gene expression in response to environmental changes. We carried out comparative genomic analysis of genome-wide DNA methylation profiles in the blood samples of two different thoroughbred horses before and after exercise by methylated-DNA immunoprecipitation sequencing (MeDIP-Seq). Differentially methylated regions (DMRs) in the pre-and post-exercise blood samples of superior and inferior horses were identified. Exercise altered the methylation patterns. After 30 min of exercise, 596 genes were hypomethy-lated and 715 genes were hypermethylated in the superior horse, whereas in the inferior horse, 868 genes were hypomethylated and 794 genes were hypermethylated. These genes were analyzed based on gene ontology (GO) annotations and the exercise-related pathway patterns in the two horses were compared. After exercise, gene regions related to cell division and adhesion were hypermethylated in the superior horse, whereas regions related to cell signaling and transport were hypermethylated in the inferior horse. Analysis of the distribution of methylated CpG islands confirmed the hypomethylation in the gene-body methylation regions after exercise. The methylation patterns of transposable elements also changed after exercise. Long interspersed nuclear elements (LINEs) showed abundance of DMRs. Collectively, our results serve as a basis to study exercise-based reprogramming of epigenetic traits.

      • Overlapping and unique toxic effects of three alternative antifouling biocides (Diuron, Irgarol 1051<sup>®</sup>, Sea-Nine 211<sup>®</sup>) on non-target marine fish

        <P><B>Abstract</B></P> <P>The use of alternative biocides has increased due to their economic and ecological relevance. Although data regarding the toxicity of commercial alternative biocides in marine organisms are accumulating, little is known about their toxic pathways or mechanisms. To compare the toxic effects of commercial alternative biocides on non-target pelagic fish (flounder) embryos, we investigated the adverse effects of developmental malformation and transcriptional changes. Three biocides including Diuron, Irgarol 1051<SUP>®</SUP> and Sea-Nine 211<SUP>®</SUP> produced a largely overlapping suite of developmental malformations, including tail-fin fold defects and dorsal body axis curvature. In our test, the potencies of these biocides were ranked in the following order with respect to malformation and mortalities: Sea-Nine 211<SUP>®</SUP> > Irgarol 1051<SUP>®</SUP> > Diuron. Consistent with the toxicity rankings, the expression of genes related to heart formation was greater in embryonic flounder exposed to Sea-Nine 211<SUP>®</SUP> than in those exposed to Irgarol 1051<SUP>®</SUP> or Diuron, while expression of genes related to fin malformation was greater in the Irgarol 1051<SUP>®</SUP> exposure group. In analyses of differential gene expression (DEG) profiles (fold change of genes with a cutoff <I>P < 0.05</I>) by high-throughput sequencing (RNA-seq), genes associated with nervous system development, transmembrane transport activity, and muscle cell development were significantly changed commonly. Embryos exposed to Diuron showed changes related to cellular protein localization, whereas genes associated with immune system processes were up-regulated significantly in embryos exposed to Irgarol 1051<SUP>®</SUP>. Genes related to actin filament organization and embryonic morphogenesis were up-regulated in embryos exposed to Sea-Nine 211<SUP>®</SUP>.         급성 췌장염으로 발현된 낭성 변화를 이릉킨 종격동 부갑상선 선종 1예

        Hypercalcemia due to hyperparathyroidism is rarely associated with acute pancreatitis. But, the relationship between hypercalcemia and pancreatitis still remains controvesial. Ectopic parathyroid adenoma with cystic change is one of the rare causes of hyperparathyroidism, and is usually located in neck and mediastinum. We report a case of mediastinal parathyroid adenoma with cystic change associated with acute pancreatitis. A 54-year-old male presented with epigastric pain for 3 days. The serum calcium, phosphate, elastase were 16.8mg/dL, 1.1 mg/dL, 2772.0 ng/mL respectively and his parathyroid hormone level in serum was 651.84 pg/mL. Chest CT showed a mediastinal mass with well defined inhomogenously enhanced density located between SVC and aorta. The patient was diagnosed to hyperparathyroidism with acute pancreatitis due to mediastinal parathyroid adenoma with cystic change, and was surgically removed. The surgical biopsy showed parathyroid adenoma with cystic change. After operation his general condition was improved and serum calcium, phosphate, amylase, lipase level were normalized (J Kor Soc Endocrinol 13:473-479, 1998).

        중증 고혈압증의 임상적 관찰

        N/A Clinical studies were carried out in the 110 cases of severe hypertension who had been admitted from January 1979 to August 1985 and the following results were obtained: 1) Underlying diseases of severe hypertension were essential hypertension in 67 cases, renal parenchymal hypertension in 36 cases, and renovascular hypertension in 7 cases. 2) Plasma renin activity was the highest in the group of renovascular hypertension which was 22.07±9.20 ng/ ml/hr, and as a whole, the higher diastolic blood pressure, the higher plasma renin activity was. Plasma renin activity in the group of K-W grade IV retinopathy was higher than those of the other groups. 3) The incidence of ventricular hypertrophy fullfilling voltage criteria and with accompanying ST-T changes on electrocardiolgram was 81.5% in the group of essential hypertension and there was no difference in relation to the level of pressure or fundoscopic changes. 4) The serum creatinine level was the highest in the group of renal parenchymal hypertension (7.93±2.98 mg/dl), and in the group of K-W grade IV retinopathy (6.86±3.50mg/dl), but there was no significant difference in relation to the level of diastolic blood pressure, 5) There were no significant difference in cardiothoracic ratio on chest X-ray in relation to underlying diseases, diastolic blood pressure or fundoscopic changes. 6) The major complications were congestive heart failure (49.1%), hypertensive encephalopathy (17.3%), and cerebrovascular accident (8.2%). 7) In the group of essential hypertension blood pressure was controlled by conservative treatment in 81.0% but in 35.7% in the group of renal hypertension. The patients of chronic renal failure who were treated with dialysis blood pressure was controlled in 87.0%. In all 6 cases with renal transplantation, nephrectomy or renal angioplasty blood pressure was well controlled. 8) In cases refractory to triple antihypertensive regimen blood pressure was controlled by minoxidil in 66.7% and by captopril in 75.0%. 9) In 45 cases who were followed up for more than 6 months diastolic blood pressure was controlled under 100mmHg in 77.8% and there were improvement of renal function, fundoscopic findings and electrocardiographic findings in 20.0%, 33.3% and 16.7%, respectively.

        The present report deals with the analysis and evaluation of the results of the Fifth World Junior Hand Ball Games held in the period of Oct. 19through Oct. 30 1985 in Seoul. The purpose of this report is to prepare the basic data for selection of the players, training program and the game plannings, with a view toward enhancing the atheletic performance of our national team in the international meetings including the 1988 Seoul Summer Olympic Games. Since the subjects of the study are the players and teams who would be the main forces representing each nation in the 1988 games, the results presented here would be valuable for the future team from Korea-the hand ball medalist country in the last 1984 Summer Olympic Games, L.A, U.S.A. 1. As for the physical strengths, it is an urgent requirement to secure large sized players exceeding 180cm in height and 75kg in weight, with more than 40 of the body weight ratio. This is worstly urgent for the pivot man. 2. The Korean team should try to further develop the right and the left side shot shill which is possible even for the players of mediocre height, and to be better prepared with the fast break offense strategy. 3. It is desirable to use the maximum number of the team members during the game in order to minimize the physical fatigue of the players. It is also recommended to specialize the function of each player-offense or defense. 4. The physical characteristics of the team members must be fully considered in order to minimize the scores for the opponents due to field shot by the tall back man and line shot by the large sized pivot man of the opponent teams. The aggresive defence method must be developed by exploiting the fast maneuvering so as to perform the zone defence of the various formations and the man to man defence at the same time. 5. The speedy pass work and the set play by the feint motion must be further studied in this direction would over-come the blocking and the save by the large sized opponent players. 6. All players should complement their stamina as to avoid the rapid fatigue especially in the first 15 minutes of the second half and to be fully prepared for the overall game time.
