<P>Alterations in cell morphology involve changes in the actin cytoskeleton and play crucial roles in determining chondrocyte phenotypes. Although the effects of simvastatin (SV) have been demonstrated in various cell types, the mechanisms and effects of SV on chondrocyte differentiation and actin cytoskeletal rearrangement are still unclear. Here, we investigated the roles of actin filament rearrangement on SV-induced differentiation of rabbit articular chondrocytes. Treatment with SV caused actin remodeling in comparison with that in untreated chondrocytes, as determined by immunofluorescence staining. Moreover, treatment with cytochalasin D (CD) and jasplakinolide (JAS), which modulate actin filament formation, resulted in reorganization of the actin cytoskeleton compared with that induced by SV in chondrocytes. In addition, CD synergistically enhanced the SV-induced increase in type II collagen expression, whereas JAS dramatically inhibited SV-induced differentiation. We also found that differentiation via SV-dependent actin cytoskeleton changes was regulated by the extracellular signal-regulated kinase (ERK)-1/2 and p38 kinase pathways. These results demonstrated that actin cytoskeletal rearrangement by SV regulated type II collagen expression and suggested that ERK-1/2 and p38 kinase pathways may play important roles in SV-induced type II collagen expression by altering actin cytoskeletal reorganization in rabbit articular chondrocytes.</P>

<P><B>Abstract</B></P> <P>Inhibitor of DNA binding (ID) proteins, ID1, ID2, ID3, and ID4 are transcriptional regulators that have a helix-loop-helix (HLH) domain but not a basic DNA binding domain. ID proteins inhibit the functions of basic HLH transcription factors and regulate cell proliferation and differentiation. However, the expression and function of <I>ID1</I>, <I>ID2</I>, <I>ID3</I>, and <I>ID4</I> at the maternal-conceptus interface are not fully understood in pigs. Therefore, we determined the expressions of <I>ID1</I>, <I>ID2</I>, <I>ID3</I>, and <I>ID4</I> in porcine endometrium, conceptus, and chorioallantoic tissues. <I>ID1</I>, <I>ID2</I>, <I>ID3</I>, and <I>ID4</I> mRNAs were expressed in the endometrium, with lower levels of <I>ID1</I>, <I>ID2</I>, and <I>ID4</I> on Day 12 of pregnancy than during the estrous cycle. <I>ID1</I>, <I>ID2</I>, <I>ID3</I>, and <I>ID4</I> mRNAs were also detected in conceptus and chorioallantoic tissues during pregnancy. ID2 protein was mainly localized to luminal epithelia and weakly to vascular smooth muscle cells in the endometrium and conceptus trophectoderm. Increasing doses of interleukin-1β decreased levels of <I>ID2</I> mRNA, while estradiol-17β increased levels of <I>ID3</I> mRNA in endometrial explants. The expressions of <I>ID2</I> and <I>ID4</I> mRNAs were higher in endometria from gilts with somatic cell nuclear transfer-derived conceptuses compared with endometria from gilts carrying conceptuses derived from natural mating on Day 12. These results indicate that the expressions of ID family genes are dynamically regulated at the maternal-conceptus interface, suggesting that ID proteins may play critical roles in the regulation of endometrial epithelial cell function and conceptus development to support the establishment and maintenance of pregnancy in pigs.</P> <P><B>Highlights</B></P> <P> <UL> <LI> <I>ID1</I>, <I>ID2</I>, <I>ID3</I>, and <I>ID4</I> expression in porcine uterine endometrium is pregnancy status- and stage-specific. </LI> <LI> ID2 protein is detected primarily in endometrial LE cells. </LI> <LI> IL1B decreases the expression of <I>ID2</I> mRNA in endometrial tissues. </LI> <LI> The expressions of <I>ID2</I> and <I>ID4</I> mRNAs increase in endometria of gilts with SCNT-derived conceptuses. </LI> </UL> </P>
Development of Virtual Reality-based Visual Perception and Cognitive Rehabilitation Service

Purpose: Patients with brain damage suffer from limitations in performing the activities of daily living (ADL) because of their motor function and visual perception impairment. The aim of this study was to help improve the motor function and visual perception ability of patients with brain damage by providing them with virtual reality-based contents. The usability results of the patients and specialists group were also evaluated. Methods: The ADL contents consisted of living room, kitchen, veranda, and convenience store, similar to a real home environment, and these were organized by a rehabilitation specialist (e.g., neurologist, physiotherapist, and occupational therapist). The contents consisted of tasks, such as turning on the living room lights, organizing the drawers, organizing the kitchen, watering the plants on the veranda, and buying products at convenience stores. To evaluate the usability of the virtual reality-based visual cognitive rehabilitation service, general elderly subjects (n=11), stroke patients (n=7), stroke patients with visual impairment (n=4), and rehabilitation specialists (n=11) were selected. The questionnaires were distributed to the subjects who were using the service, and the subjective satisfaction of individual users was obtained as data. The data were analyzed using SPSS 21.0 software. The general characteristics of the users and the evaluation scores of the experts were analyzed using descriptive statistics. Results: The usability test result of this study showed that the mean value of the questionnaire related to content understanding and difficulty was high, between 4-5 points. Conclusion: The virtual reality rehabilitation service of this study is an efficient service that can improve the function, interest, and motivation of stroke patients.
Altered pharmacokinetics of zalcitabine by concurrent use of NSAIDs in rats

Aim: To investigate the pharmacokinetic interactions between zalcitabine and nonsteroidal anti-inflammatory drugs (NSAIDs) in rats. Methods: Zalcitabine was administered to rats via an iv injection (20 mg/kg) in the presence or absence of ketoprofen or naproxen (20 mg/kg),_and the pharmacokinetic parameters were determined by using non-compartmental analysis. Results: Compared with the control (zalcitabine alone), pretreatment with ketoprofen or naproxen 30 min prior to intravenous administration of zalcitabine significantly altered the pharmacokinetic profiles of zalcitabine in rats. Renal clearance of zalcitabine was reduced by approximately 3-4-fold in the presence of ketoprofen or naproxen. Consequently, systemic exposure (AUC) to zalcitabine in the rats pretreated with ketoprofen or naproxen was significantly greater than that for the control group given zalcitabine alone. The terminal plasma half-life of zalcitabine was also prolonged by 4-5-fold in the presence of ketoprofen or naproxen. Conclusion: The NSAIDs ketoprofen and naproxen effectively altered the pharmacokinetics of zalcitabine. Therefore, concomitant use of ketoprofen or naproxen in patients being treated with zalcitabine may necessitate close monitoring for potential drug interactions.

Co-60 및 Ir-192 등의 방사성 동위원소가 비파괴 검사(Non-Destructive Test; NDT) 등의 분야에서 널리 쓰 임에 따라 방사선 안전관리가 매우 중요시되고 있다. 본 연구에서는 요오드화수은(Mercury(Ⅱ) Iodide; HgI2) 의 선원추적 시스템 적용 가능성을 평가하였다. HgI2로 제작된 Unit cell 센서의 신뢰도 검증을 위한 전기적 특성평가를 수행한 후, 방사선에 대한 센서의 위치의존성을 분석하고, Planning system의 선량 분포와 비교 하였다. 평가결과, R-sq>0.99 이상의 선형성과 CV<0.015 이하의 재현성을 보이며 신뢰도가 높은 것으로 나타났다. 또한, 위치의존성 평가에서는 센서의 isocenter에서 최댓값이 측정되었으며, 거리에 따라 점진적 감소를 나타냈다. 그러나 Planning system 상의 선량 분포 데이터와는 최대 30%의 차이를 보였는데, 센서는 단일지점으로부터 데이터를 수집하는 Planning system과 달리 면적으로부터 수집하기 때문으로 사료된다. Radiation safety management is being considered very important since radioactive isotopes such as Co-60 and Ir-192 are widely used in fields such as non-destructive test(NDT). In this study, the applicability of Mercury(Ⅱ) Iodide(HgI2) source for tracing system was evaluated. To make sure the unit cell sensor’s reliability, we evaluated the electrical properties of the sensor made with HgI2, and then position dependence of the sensor was analyzed and compared with the dose distribution from the planning system. As a result of the evaluation, high reliability of the sensor was shown through the linearity of R-sq > 0.990 and reproducibility of CV < 0.015. In the position dependence evaluation, the maximum value was measured at the isocenter of the sensor and gradually decreased according to the distance. However, the dose distribution data from the planning system was turned out that has difference with that of the sensor up to 30%. This seems to come from the difference between single-point measuring based planning system and area measuring based sensor.
