        Product-Rate Correlations for Solvolyses of 2,4-Dimethoxybenzenesulfonyl Chloride

        The solvolysis rate constants of 2,4-dimethoxybenzenesulfonyl chloride (1) in 30 different solvents are well correlated with the extended Grunwald-Winstein equation, using the N T solvent nucleophilicity scale and Y Cl solvent ionizing scale, with sensitivity values of 0.93 ± 0.14 and 0.65 ± 0.06 for l and m, respectively. These l and m values can be considered to support a S N 2 reaction pathway. The activation enthalpies (H  ) were 12.4 to 14.6 kcal·mol 1 and the activation entropies (S  ) were 15.5 to 32.3 cal·mol 1 ·K 1 , which is consistent with the proposed bimolecular reaction mechanism. The solvent kinetic isotope effects (SKIE) were 1.74 to 1.86, which is also in accord with the S N 2 mechanism and was possibly assisted using a general-base catalysis. The values of product selectivity (S) for solvolyses of 1 in alcohol/water mixtures was 0.57 to 6.5, which is also consistent with the proposed bimolecular reaction mechanism. Third-order rate constants, k ww and k aa , were calculated from the rate constants (k obs ), together with k aw and k wa calculated from the intercept and slope of the plot of 1/S vs. [water]/[alcohol]. The calculated rate constants, k calc (k ww , k aw , k wa and k aa ), are in satisfactory agreement with the experimental values, supporting the stoichiometric solvation effect analysis.

        HII 영역 분광자료를 통한 M31과 M33의 화학원소 결정

        Chemical evolution of galaxies can be understood by studying the spatial distribution of heavy elements. We selected two nearby galaxies, M31 and M33 and investigated spectrum of their HII regions: a) the elec-tron densities have been derived from the [S II] 6717/6731 ratio along with the most recent atomic constants (Hyung & Aller 1996); b) the electron temperatures were determinated from the Pagel's empirical method. Nebula Model (Hyung 1994) has been employed to predict the spectral line intensities which gives the proper chemical abundances. The model would predict the line intensities correctly only when various input parameters such as the effective central star temperatures, gravity log g, model atmosphere as well as the geometry and the nebula physical condition are appropriate. Thus, the determination of chemical abundances of O, S, N of the two nearby galaxies M31 and M33 has been done, which shows a radial dependance of O/H and N/H: decrease with the distance, or increasing electron temperature due to the elemental deficiency. Abundances of M31 appear to be enhanced than those of M33.

        Climate change causes an adverse impact on biodiversity and human life. Correspondingly, global communities have proposed visions and goals to mitigate the adverse impact of climate change without considering local land use planning. Thus, in this study, we propose a framework for local land-use scenarios with an environment-friendly vision and goal. Through literature survey, we summarized our scenario framework in three steps: 1) development of a system, 2) storyline development, 3) review and distribution. First, we prepared a system that can help set a plan in motion. Then, we developed the storyline by selecting 25 main variables, which were classified into seven categories. Climate change adaptation and mitigation, type of ecosystem, effects of urban areas, and land use policies were selected as direct variables, while the other 21 variables were considered as indirect variables. After we applied the framework to the Ansan-si region in South Korea, we conducted a Strengths, Weaknesses, Opportunities, Threats (SWOT) analysis to link the key variables, and then proposed a direction for the application of the scenario-based strategy. After establishing five scenarios, we constructed narrative storylines using the key direct variables identified in each scenario. As a result of the application of our framework to the Ansan-si region, we established a narrative storyline framework for local land use planning considering a quantification scenario. Thus, the proposed land use planning framework considers the impact of climate change and ecosystem service changes at the local level.

        Reliability of the Supraspinatus Muscle Thickness Measurement by Ultrasonography

        Objective To assess the intrarater and interrater reliability of the supraspinatus thickness measured by ultrasonography (US) in normal subjects and to identify the relationship between the supraspinatus thickness measured by US and cross sectional area (CSA) of the supraspinatus muscle by magnetic resonance imaging (MRI) in hemiplegic patients.Method We examined 20 shoulders of normal subjects and 10 shoulders of hemiplegic patients. In normal subjects, one examiner measured the supraspinatus thickness twice by US at the scapular notch and another examiner measured the supraspinatus thickness several days later. The intrarater and interrater reliability of supraspinatus thickness measurements were then evaluated. In hemiplegic patients, the supraspinatus thickness at the scapular notch was measured by US in affected side and compared with CSA of the supraspinatus muscle at the scapular notch and the Y-view of MRI.Results One examiner’s supraspinatus thickness measurement average was 1.72±0.21 cm and 1.74±0.24 cm, and the other examiner’s supraspinatus thickness measurement average was 1.74±0.22 cm in normal subjects. Intraclass correlation coefficients of intrarater and interrater examination were 0.91 and 0.88, respectively. For hemiplegic patients, the supraspinatus thickness measured by US was 1.66±0.13 cm and CSA by MRI was 4.83±0.88 cm2 at the Y-view and 5.61±1.19 cm2 at the scapular notch. The Pearson Correlation Coefficient between the supraspinatus thickness at the scapular notch and the CSA at the Y-view was 0.72 and that between the supraspinatus thickness and CSA at the scapular notch was 0.76.Conclusion The supraspinatus thickness measurement by US is a reliable method and is positively correlated with the CSA of the supraspinatus muscle in MRI in hemiplegic patients. Therefore, supraspinatus thickness measurement by US can be used in the evaluation of muscle atrophy and to determine therapeutic effects in hemiplegic patients.

        Inhibitory effects of the atypical antipsychotic, clozapine, on voltage-dependent K<sup>+</sup> channels in rabbit coronary arterial smooth muscle cells

        To investigate the adverse effects of clozapine on cardiovascular ion channels, we examined the inhibitory effect of clozapine on voltage-dependent K<sup>+</sup> (Kv) channels in rabbit coronary arterial smooth muscle cells. Clozapine-induced inhibition of Kv channels occurred in a concentration-dependent manner with an half-inhibitory concentration value of 7.84 ± 4.86 µM and a Hill coefficient of 0.47 ± 0.06. Clozapine did not shift the steady-state activation or inactivation curves, suggesting that it inhibited Kv channels regardless of gating properties. Application of train pulses (1 and 2 Hz) progressively augmented the clozapine-induced inhibition of Kv channels in the presence of the drug. Furthermore, the recovery time constant from inactivation was increased in the presence of clozapine, suggesting that clozapine-induced inhibition of Kv channels is use (state)-dependent. Pretreatment of a Kv1.5 subtype inhibitor decreased the Kv current amplitudes, but additional application of clozapine did not further inhibit the Kv current. Pretreatment with Kv2.1 or Kv7 subtype inhibitors partially blocked the inhibitory effect of clozapine. Based on these results, we conclude that clozapine inhibits arterial Kv channels in a concentration-and use (state)-dependent manner. Kv1.5 is the major subtype involved in clozapine-induced inhibition of Kv channels, and Kv2.1 and Kv7 subtypes are partially involved.

        ABUNDANCES OF PLANETARY NEBULAE IN M 31 AND M 32

        Planetary nebulae provide a direct way to probe elemental abundances, their distributions and their gradients in populations in nearby galaxies. We investigate bulge planetary nebulae in M 31 and M 32 using the strong emission lines, H$\alpha$, He I, [O III], [N II], [S II] and [Ne III]. From the [O III] 4363/5007 line ratio and the [O II] 3727/3729, we determine the electron temperatures and number densities. With a standard modeling procedure (Hyung, 1994), we fit the line intensities and diagnostic temperatures, and as a result, we derive the chemical abundances of individual planetary nebulae in M 31 and M 32. The derived chemical abundances are compared with those of the well-known Galactic planetary nebulae or the Sun. The chemical abundances of M 32 appear to be less enhanced compared to the Galaxy or M 31.

        <P>Phosphatidylinositol 3-kinase (PI3K) has been implicated in synaptic plasticity and other neural functions in the brain. However, the role of individual PI3K isoforms in the brain is unclear. We investigated the role of PI3K gamma in hippocampal-dependent synaptic plasticity and cognitive functions. We found that PI3K gamma has a crucial and specific role in NMDA receptor (NMDAR)-mediated synaptic plasticity at mouse Schaffer collateral-commissural synapses. Both genetic deletion and pharmacological inhibition of PI3K gamma disrupted NMDAR long-term depression (LTD) while leaving other forms of synaptic plasticity intact. Accompanying this physiological deficit, the impairment of NMDAR LTD by PI3K gamma blockade was specifically correlated with deficits in behavioral flexibility. These findings suggest that a specific PI3K isoform, PI3K gamma, is critical for NMDAR LTD and some forms of cognitive function. Thus, individual isoforms of PI3Ks may have distinct roles in different types of synaptic plasticity and may therefore influence various kinds of behavior.</P>

        Inhibition of MicroRNA-221 and 222 Enhances Hematopoietic Differentiation from Human Pluripotent Stem Cells via c-KIT Upregulation

        The stem cell factor (SCF)/c-KIT axis plays an important role in the hematopoietic differentiation of human pluripotent stem cells (hPSCs), but its regulatory mechanisms involving microRNAs (miRs) are not fully elucidated. Here, we demonstrated that supplementation with SCF increases the hematopoietic differentiation of hPSCs via the interaction with its receptor tyrosine kinase c-KIT, which is modulated by miR-221 and miR-222. c-KIT is comparably expressed in undifferentiated human embryonic and induced pluripotent stem cells. The inhibition of SCF signaling via treatment with a c-KIT antagonist (imatinib) during hPSC-derived hematopoiesis resulted in reductions in the yield and multi-lineage potential of hematopoietic progenitors. We found that the transcript levels of miR-221 and miR-222 targeting c-KIT were significantly lower in the pluripotent state than they were in terminally differentiated somatic cells. Furthermore, suppression of miR-221 and miR-222 in undifferentiated hPSC cultures induced more hematopoiesis by increasing c-KIT expression. Collectively, our data implied that the modulation of c-KIT by miRs may provide further potential strategies to expedite the generation of functional blood cells for therapeutic approaches and the study of the cellular machinery related to hematologic malignant diseases such as leukemia.
