      • Association Between MDM2 SNP309 T>G and Risk of Gastric Cancer: A Meta-analysis

        Background: As a negative regulator of P53, MDM2 plays an important role in carcinogenesis; a polymorphism in its promoter region. SNP309 T>G, is known to increase the expression of MDM2, thus being considered related to higher susceptibility to neoplasia. However, no agreement has been achieved regarding its effects on gastric cancer. Methods: The present systematic meta-analysis was performed based on comprehensive literature search from Pubmed, Web of science and CBM databases. Results: It was suggested from 6 independent studies that the GG genotype is associated with a significantly increased risk of gastric cancer (Recessive: OR = 1.43, 95% CI = 1.08-1.91, P = 0.013), and subgroup analysis also confirmed the relationship (English publications-recessive model: OR = 1.45, 95% CI = 1.10-1.91, P = 0.009; Studies in China-recessive model: OR = 1.58, 95% CI = 1.08-2.30, P = 0.017). No publication bias was detected. Conclusion: The meta-analysis indicated a significant inverse association between GG genotype carriage and elevated risk of gastric cancer. However, more studies and detailed information are needed to fully address the topic.

        NaCl + CaCl<sub>2</sub> + H<sub>2</sub>O 및 KCl + CaCl<sub>2</sub> + H<sub>2</sub>O 삼성분계에 대한 50℃에서의 용해도 측정

        NaCl-$CaCl_2$-$H_2O$와 KCl-$CaCl_2$-$H_2O$계의 용해도와 물리화학적 성질(굴절률)을 $50^{\circ}C$에서 측정하였으며, 그 결과를 상평형 그림과 조성에 따른 물리화학적 성질에 대한 도표로 나타내었다. 삼성분계의 상 평형 그림은 불변점이 하나, 일변수 곡선이 둘, NaCl (또는 KCl) 및 $CaCl_2{\cdot}2H_2O$에 대응되는 결정화 지역이 두 개가 있음을 보여주었다. 이들 계에 대하여 혼합 파라미터인 ${\theta}_{M,Ca}$와 ${\Psi}_{M,Ca,Cl}$ (M = Na or K), 평형상수 $K_{sp}$를 최소제곱법을 이용하여 결정하였으며, 이때, NaCl, KCl 및 $CaCl_2$의 단일-염 Pitzer 파라미터 ${\beta}^{(0)}$, ${\beta}^{(1)}$, ${\beta}^{(2)}$ and $C^{\Phi}$는 참고문헌을 통하여 직접 계산하였다. 이 결과들은 실험결과와 잘 일치하였다. The solubility and the physicochemical property (refractive index) in the NaCl-$CaCl_2$-$H_2O$ and KCl-$CaCl_2$-$H_2O$ systems were determined at $50^{\circ}C$ and the phase diagrams and the diagrams of physicochemical property vs composition were plotted. One invariant point, two univariant curves, and two crystallization zones, corresponding to sodium Chloride (or potassium chloride), dihydrate ($CaCl_2{\cdot}2H_2O$) showed up in the phase diagrams of the ternary systems. The mixing parameters ${\theta}_{M,Ca}$ and ${\Psi}_{M,Ca,Cl}$ (M = Na or K) and equilibrium constant $K_{sp}$ were evaluated in NaCl-$CaCl_2-H_2O$ and KCl-$CaCl_2-H_2O$ systems by least-squares optimization procedure, in which the single-salt Pitzer parameters of NaCl, KCl and $CaCl_2$ ${\beta}^{(0)}$, ${\beta}^{(1)}$, ${\beta}^{(2)}$ and $C^{\Phi}$ were directly calculated from the literature. The results obtained were in good agreement with the experimental data.

      • Epigenetic Regulation of miR-129-2 Leads to Overexpression of PDGFRa and FoxP1 in Glioma Cells

        miR-129-2 is frequently downregulated in multiple cancers. However, how it is silenced in cancers remains unclear. Here we investigated the expression profile and potential biological function of miR-129-2 in glioblastoma (GBM), the most common and lethal form of brain tumors in adults. We showed that miR-129-2 is lost in GBM patient specimens and cultured cell lines. miR-129-2 expression could be restored upon treatment with a histone deadetylase inhibitor (trichostatin A) but not a DNA methylation inhibitor (5-Aza-2'-deoxycytidine), and more profound effect was observed with the treatment of these two drugs in combination. Furthermore, forced expression of miR-129-2 repressed the expression of major oncogenic genes such as PDGFRa and Foxp1 in GBMs. Consistently, expression of miR-129-2 significantly inhibits GBM cell proliferation in vitro. These results reveal that miR-129-2 is epigenetically regulated and functions as a tumor suppressor gene in GBMs, suggesting it may serve as a potential therapeutic target for GBM treatment.

        Dual Effect of Hepatic Macrophages on Liver Ischemia and Reperfusion Injury during Liver Transplantation

        Ischemia-reperfusion injury (IRI) is a major complication in liver transplantation (LT) and it is closely related to the recovery of grafts' function. Researches has verified that both innate and adaptive immune system are involved in the development of IRI and Kupffer cell (KC), the resident macrophages in the liver, play a pivotal role both in triggering and sustaining the sterile inflammation. Damage-associated molecular patterns (DAMPs), released by the initial dead cell because of the ischemia insult, firstly activate the KC through pattern recognition receptors (PRRs) such as toll-like receptors. Activated KCs is the dominant players in the IRI as it can secret various pro-inflammatory cytokines to exacerbate the injury and recruit other types of immune cells from the circulation. On the other hand, KCs can also serve in a contrary way to ameliorate IRI by upregulating the anti-inflammatory factors. Moreover, new standpoint has been put forward that KCs and macrophages from the circulation may function in different way to influence the inflammation. Managements towards KCs are expected to be the effective way to improve the IRI.

        Distinct evolution of toll‑like receptor signaling pathway genes in cetaceans

        Background The relatively rapid spread and diversity of marine pathogens posed an initial and ongoing challenge for cetaceans (whales, dolphins, and porpoises), descendants of terrestrial mammals that transitioned from land to sea approximately 56 million years ago. Toll-like receptors (TLRs) play important roles in regulating immunity against pathogen infections by detecting specific molecular patterns and activating a wide range of downstream signaling pathways. The ever-increasing catalogue of mammalian genomes offers unprecedented opportunities to reveal genetic changes associated with evolutionary and ecological processes. Objective This study aimed to explore the molecular evolution of TLR signaling pathway genes in cetaceans. Methods Genes involved in the TLR signaling pathway were retrieved by BLAST searches using human coding sequences as queries. We tested each gene for positive selection along the cetacean branches using PAML and Hyphy. Physicochemical property changes of amino acids at all positively selected residues were assessed by TreeSAAP and visualized with WebLogo. Bovine and dolphin TLR4 was assessed using human embryonic kidney cell line HEK293, which lacks TLR4 and its co-receptor MD-2. Results We demonstrate that eight TLR signaling pathway genes are under positive selection in cetaceans. These include key genes in the response to Gram-negative bacteria: TLR4, CD14, and LY96 (MD-2). Moreover, 41 out of 65 positively selected sites were inferred to harbor substitution that dramatically changes the physicochemical properties of amino acids, with most of them situated in or adjacent to functional regions. We also found strong evidence that positive selection occurred in the lineage of the Yangtze finless porpoise, likely reflecting relatively recent adaptions to a freshwater milieu. Species-specific differences in TLR4 response were observed between cetacean and terrestrial species. Cetacean TLR4 was significantly less responsive to lipopolysaccharides from a terrestrial E. coli strain, possibly a reflection of the arms race of host–pathogen co-evolution faced by cetaceans in an aquatic environment. Conclusion This study provides further impetus for studies on the evolution and function of the cetacean immune system.

        Sodium tanshinone IIA sulfonate attenuates angiotensin II-induced collagen type I expression in cardiac fibroblasts in vitro

        Cardiac fibrosis occurs after pathological stimuli to the cardiovascular system. One of the most important factors that contribute to cardiac fibrosis is angiotensin II (Ang II). Accumulating studies have suggested that reactive oxygen species (ROS) plays an important role in cardiac fibrosis and sodium tanshinone IIA sulfonate (STS) possesses antioxidant action. We therefore examined whether STS depresses Ang II-induced collagen type I expression in cardiac fibroblasts. In this study, Ang II significantly enhanced collagen type I expression and collagen synthesis. Meanwhile, Ang II depressed matrix metalloproteinase- 1 (MMP-1) expression and activity. These responses were attenuated by STS. Furthermore, STS depressed the intracellular generation of ROS, NADPH oxidase activity and subunit p47phox expression. In addition, N-acetylcysteine the ROS scavenger, depressed effects of Ang II in a manner similar to STS. In conclusion, the current studies demonstrate that anti-fibrotic effects of STS are mediated by interfering with the modulation of ROS.         The application of a regularization method to the estimation of geometric errors of a three-axis machine tool using a double ball bar

        Geometric accuracy is crucially important for machine tools. Identification of geometric errors, especially position-dependent geometric errors, is still a challenging issue. This paper presents a systematic and fast approach to identify the geometric error components of a precision machine tool using double ball bar (DBB). The approach can be implemented in three steps: (1) polynomial based error modeling that relates the DBB radius error directly to the geometric error parameters of machine tool; (2) spatial measurement trajectory planning with a single installation of DBB in order to avoid producing extra setup errors; (3) error identification with regularization method that can solve the ill-posed identification problem effectively. Simulations and experiments show the accuracy and effectiveness of the proposed identification approach. The results of the DBB test show that, utilizing the proposed identification method, the roundness errors of the three circular paths in xy-, yz- and xz-plane are reduced from 27.3 μm, 20.7 μm and 24.1 μm to 9.2 μm, 12.3 μm and 7.8 μm, respectively, with error compensation.

      • Effects of the Hippo Signaling Pathway in Human Gastric Cancer

        Background/Aim: The Hippo signaling pathway is a newly discovered and conserved signaling cascade, which regulates organ size control by governing cell proliferation and apoptosis. This study aimed to investigate its effects in human gastric cancer. Methods: Tumor tissues (n=60), adjacent non-tumor tissues (n=60) and normal tissues (n=60) were obtained from the same patients with primary gastric cancer (GC). In addition, 70 samples of chronic atrophic gastritis (CAG) tissues were obtained from patients with intestinal metaplasia (IM) by endoscopic biopsy. Hippo signaling molecules, including Mst1, Lats1, YAP1, TAZ, TEAD1, Oct4 and CDX2, were determined by quantitative polymerase chain reaction (qPCR). Protein expression of Mst1, Lats1, YAP1, TEAD1 and CDX2 was assessed by immunohistochemistry and Western blotting. Results: Mst1, Lats1 and Oct4 mRNA expression showed an increasing tendency from GC tissues to normal gastric tissues, while the mRNA expression of YAP1, TAZ and TEAD1 was up-regulated (all P<0.01). Mst1 and Lats1 protein expression presented a similar trend with their mRNA expression. In addition, YAP1 and TEAD1 protein expression in GC was significantly higher than in the other groups (all P<0.01). CDX2 mRNA and protein expression in the CAG group were higher than in the other groups (all P<0.01). In GC, mRNA expression of Mst1, Lats1, Oct4, YAP1, TAZ, TEAD1 and CDX2 had a close correlation with lymphatic metastasis and tumor TNM stage (all P<0.01). Furthermore, protein expression of Mst1, Lats1, YAP1, TAZ, TEAD1 and CDX2 had a close correlation between each other (P<0.05). Conclusion: The Hippo signaling pathway is involved in the development, progression and metastasis of human gastric cancer. Therefore, manipulation of Hippo signaling molecules may be a potential therapeutic strategy for gastric cancer.

        Design and testing of CFRP sleeve for a high-speed permanent magnet synchronous motor with surface-mounted rotor

        A high-speed (HS) permanent magnet (PM) synchronous motor (HSPMSM) with a carbon fiber-reinforced plastic (CFRP) protective sleeve in the surface-mounted rotor was explored in this study. In view of retaining the PMs at HS operations, the high-strength CFRP sleeve was designed on the basis of a process that could be summarized as follows. First, a multi-physics analysis of the rotor was conducted. The requirements to the CFRP sleeve were obtained from electromagnetic analysis, loss analysis, heat flow coupling computational fluid dynamics analysis, structural analysis, and rotor dynamics analysis. Second, the CFRP sleeve was designed by theoretical analysis. From the results, the thickness values of the carbon fibers in the circumferential and helical directions were chosen to be 4 and 1 mm, respectively, while the helical angle of carbon fibers was chosen to be 70°. Then, a plateshaped CFRP product and a cylindrical CFRP product were fabricated to verify the strength and stiffness of the CFRP sleeve. The performance test results show that the hoop strength and elastic modulus and the axial strength and elastic modulus of the plate-shaped CFRP products at 20 °C are 1963 MPa @ 156 GPa and 550 MPa @ 36 GPa, respectively. The hoop strain and axial strain of the cylindrical CFRP product under 35 MPa are about 3000 and 1900 με, which meet the design requirements of the HSPMSM.

        The Age-Related Orientational Changes of Human Semicircular Canals

        Objectives. Some changes are found in the labyrinth anatomy during postnatal development. Although the spatial orientation of semicircular canals was thought to be stable after birth, we investigated the age-related orientational changes of human semicircular canals during development. Methods. We retrospectively studied the computed tomography (CT) images of both ears of 76 subjects ranged from 1 to 70 years old. They were divided into 4 groups: group A (1–6 years), group B (7–12 years), group C (13–18 years), and group D (>18 years). The anatomical landmarks of the inner ear structures were determined from CT images. Their coordinates were imported into MATLAB software for calculating the semicircular canals orientation, angles between semicircular canal planes and the jugular bulb (JB) position. Differences between age groups were analyzed using multivariate statistics. Relationships between variables were analyzed using Pearson analysis. Results. The angle between the anterior semicircular canal plane and the coronal plane, and the angle between the horizontal semicircular canal plane and the coronal plane were smaller in group D than those in group A (P<0.05). The JB position, especially the anteroposterior position of right JB, correlated to the semicircular canals orientation (P<0.05). However, no statistically significant differences in the angles between ipsilateral canal planes among different age groups were found. Conclusion. The semicircular canals had tendencies to tilt anteriorly simultaneously as a whole with age. The JB position correlated to the spatial arrangement of semicircular canals, especially the right JB. Our calculation method helps detect developmental and pathological changes in vestibular anatomy.
