        Serine proteinase (SP) and serine protease inhibitor (serpin) gene expression after hormone treatment in the silkworm Bombyx mori (Lepidoptera: Bombycidae)

        Many members of the Serine protease inhibitor (serpin) family participate in regulation by inhibiting Serine proteinases (SPs) or Serine protease homologs (SPHs) isozymes and maintaining the balance in the internal environment. SPs were expressed in the midguts of mid-fifth-instars and exhibited slightly higher expression in female compared to male late-fifth-instars. However, serpinsweremainly expressed in the silk glands, achieved a higher level of expression in the late fifth-instar and had an expression advantage in the males. The ratio of the cocoon shell was significantly higher in Jingsong compared to the other varieties in both the males and females, which was consistent with the higher mRNA and protein expression of Spi1 in the silk gland of Jingsong compared to the other varieties. These results reflected the positive correlation between TRP protein expression and the ratio of the cocoon shell and suggested that the higher mRNA and protein expression levels of the serpin gene wasmolecular evidence ofmore efficient silkmaterial synthesis. The SP/SPHand serpin genes presented different regulatory trends after treatment of the day 3 fifth-instar silkworm larvae with 20-hydroxyecdysone (20E) and juvenile hormone analogues (JHA). Themajority of the surveyed SP/SPH geneswere down-regulated and serpin geneswere up-regulated under treatmentwith the high concentration 20E. After JHA treatment, all of the investigated genes were up-regulated 48 h after treatment with JHA. The hormone regulatory effectwas primarily influenced by the developmental stage and the hormone dose. The tissue specificity and sex differences in the SP/ SPH and serpin genes implied that they participated in tissue-specific transcriptional regulation. The genes were expressed at different levels during different developmental periods and following hormone treatment. The results demonstrated that the genes played a role in scheduling and provided insights into the mechanismunderlying silk material formation and accumulation.

        The pivotal role of the NLRC4 inflammasome in neuroinflammation after intracerebral hemorrhage in rats

        The NLRC4 inflammasome, a member of the nucleotide-binding and oligomerization domain-like receptor (NLR) family, amplifies inflammation by facilitating the processing of caspase-1, interleukin (IL)–1β, and IL-18. We explored whether NLRC4 knockdown alleviated inflammatory injury following intracerebral hemorrhage (ICH). Furthermore, we investigated whether NLRC4 inflammasome activation can be adjusted by the regulator of G protein signaling 2/leucine-rich repeat kinase-2 pathway. Fifty microliters of arterial blood was drawn and injected into the basal ganglion to simulate the ICH model. NLRC4 small interfering RNAs (siRNAs) were utilized to knockdown NLRC4. An LRRK2 inhibitor (GNE7915) was injected into the abdominal cavity. Short hairpin (sh) RNA lentiviruses and lentiviruses containing RGS2 were designed and applied to knockdown and promote RGS2 expression. Neurological functions, brain edema, Western blot, enzyme-linked immunosorbent, hematoxylin and eosin staining, Nissl staining, immunoprecipitation, immunofluorescence assay and Evans blue dye extravasation and autofluorescence assay were evaluated. It was shown that the NLRC4 inflammasome was activated following ICH injury. NLRC4 knockdown extenuated neuronal death, damage to the blood-brain barrier, brain edema and neurological deficiency 3 days after ICH. NLRC4 knockdown reduced myeloperoxidase (MPO) cells as well as tumor necrosis factor (TNF)-α, interleukin (IL)-6, IL-1β and IL-18 following ICH. GNE7915 reduced pNLRC4 and NLRC4 inflammasome activation. RGS2 suppressed the interaction of LRRK2 and NLRC4 and NLRC4 inflammasome activation by regulating pLRRK2. Our study demonstrated that the NLRC4 inflammasome may aggravate the inflammatory injury induced by ICH and that RGS2/LRRK2 may relieve inflammatory injury by restraining NLRC4 inflammasome activation.

        Eosinophilic endotype of chronic obstructive pulmonary disease: similarities and differences from asthma

        Approximately 25% to 40% of patients with chronic obstructive pulmonary disease (COPD) have the eosinophilic endotype. It is important to identify this group accurately because they are more symptomatic and are at increased risk for exacerbations and accelerated decline in forced expiratory volume in the 1st second. Importantly, this endotype is a marker of treatment responsiveness to inhaled corticosteroid (ICS), resulting in decreased mortality risk. In this review, we highlight differences in the biology of eosinophils in COPD compared to asthma and the different definitions of the COPD eosinophilic endotype based on sputum and blood eosinophil count (BEC) with the corresponding limitations. Although BEC is useful as a biomarker for eosinophilic COPD endotype, optimal BEC cut- offs can be combined with clinical characteristics to improve its sensitivity and specificity. A targeted approach comprising airway eosinophilia and appropriate clinical and physiological features may improve identification of subgroups of patients who would benefit from biologic therapy or early use of ICS for disease modification.

        Identification of Wiener Model with Discontinuous Nonlinearities using Differential Evolution

        This paper deals with the identification of Wiener models with discontinuous nonlinearities. The identification of the Wiener model is formulated as an optimization problem. Differential evolution algorithm, a powerful and robust evolutionary algorithm, is used to search the optimal parameter of the Wiener model such that the error between the output of true model and that of the identified model is minimized. The proposed method can identify the parameters of linear dynamic subsystems and static nonlinear function of the Wiener model simultaneously, and overcome the difficulty of un-availability of the intermediated signal. Two application examples verify that the proposed method can accurately estimate the parameters of the Wiener model even in a low SNR environment.

      • Nonlinear thermal post-buckling analysis of graphene platelets reinforced metal foams plates with initial geometrical imperfection

        Although some scholars have studied the thermal post-buckling of graphene platelets strengthened metal foams (GPLRMFs) plates, they have not considered the influence of initial geometrical imperfection. Inspired by this fact, the present paper studies the thermal post-buckling characteristics of GPLRMFs plates with initial geometrical imperfection. Three kinds of graphene platelets (GPLs) distribution patterns including three patterns have been considered. The governing equations are derived according to the first-order plate theory and solved with the help of the Galerkin method. According to the comparison with published paper, the accuracy and correctness of the present research are verified. In the end, the effects of material properties and initial geometrical imperfection on the thermal post-buckling response of the GPLRMFs plates are examined. It can be found that the presence of initial geometrical imperfection reduces the thermal post-buckling strength. In addition, the present study indicates that GPL-A pattern is best way to improve thermal post-buckling strength for GPLRMFs plates, and the presence of foams can improve the thermal post-buckling strength of GPLRMFs plates, the Foam- II and Foam- I patterns have the lowest and highest thermal post-buckling strength. Our research can provide guidance for the thermal stability analysis of GPLRMFs plates.

      • An Optimized Motion Estimation Algorithm and Application in the FRUC System

        Based on the optimized three-step search algorithm. Combining threshold judgment as well as local full search, a more efficient motion estimation algorithm is proposed. The algorithm not only inherited the traditional three-step algorithm’s quick speed but also kept the advantages of a relatively small amount of calculation, besides it can avoid the local optimum problem in the three-step search algorithm (TSS). In addition, the algorithm combined with the threshold judgment and local full search algorithm, so it also maintains satisfactory visual quality. Comparing the algorithm with TSS and local full search algorithm (LFS). The algorithm has great performance in search points and peak signal-to-noise ratio. Experimental results show that compared with LFS, search points drop by 34.61% ~ 54.47% .While compared with the TSS, the search points only rise by 6.15% ~ 12.21%. The average PSNR of proposed Algorithm is 0.24dB higher than LFS and 3.30dB higher than TSS.

        LncRNA CRNDE Promotes the Progression of B-cell Precursor Acute Lymphoblastic Leukemia by Targeting the miR-345-5p/CREB Axis

        The imbalance between the proliferation and apoptosis of B-cell precursors is an important contributor to the pathogenesis of B-cell precursor acute lymphoblastic leukemia (BCP-ALL), while its specific regulatory mechanism remains perplexing. This study aimed to expound the underlying mechanism of the proliferation and apoptosis of BCP-ALL cells from the perspective of non-coding RNA. In this study, long non-coding RNA colorectal neoplasia differentially expressed (LncRNA CRNDE) was upregulated in the bone marrow of BCP-ALL patients and BCP-ALL cell lines (NALM-6 and RS4;11). Functionally, LncRNA CRNDE knockdown restrained cell proliferation and boosted cell apoptosis in NALM-6 and RS4;11 cells. The subsequent investigation confirmed that LncRNA CRNDE bound to miR-345-5p and negatively regulated miR-345-5p expression. The overexpression of miR-345-5p suppressed cell proliferation and boosted cell apoptosis in NALM-6 and RS4;11 cells. Further experiments revealed that miR-345-5p downregulated cyclic AMP response element-binding protein (CREB) expression by targeting its mRNA directly. CREB overexpression reversed the effect of miR-345-5p mimic on cell proliferation and apoptosis in NALM-6 and RS4;11 cells. Finally, in vivo experiments showed that LncRNA CRNDE knockdown prolonged the survival of mice xenotransplanted with NALM-6 cells. In conclusion, LncRNA CRNDE upregulated CREB expression by suppressing miR-345-5p, thus promoting cell proliferation and reducing cell apoptosis in BCP-ALL.

        Positive effects of porcine IL-2 and IL-4 on virus-specific immune responses induced by the porcine reproductive and respiratory syndrome virus (PRRSV) ORF5 DNA vaccine in swine

        The purpose of this study was to investigate the effects ofporcine interleukin (IL)-2 and IL-4 genes on enhancing theimmunogenicity of a porcine reproductive and respiratorysyndrome virus ORF5 DNA vaccine in piglets. Eukaryoticexpression plasmids pcDNA-ORF5, pcDNA-IL-2, andpcDNA-IL-4 were constructed and then expressed in Marc-145cells. The effects of these genes were detected using an indirectimmunofluorescent assay and reverse transcriptionpolymerase chain reaction (RT-PCR). Characteristicfluorescence was observed at different times after pcDNAORF5was expressed in the Marc-145 cells, and PCR productscorresponding to ORF5, IL-2, and IL-4 genes were detected at48 h. Based on these data, healthy piglets were injectedintramuscularly with different combinations of the purifiedplasmids: pcDNA-ORF5 alone, pcDNA-ORF5 + pcDNA-IL-2,pcDNA-ORF5 + pcDNA-IL-4, and pcDNA-ORF5 + pcDNAIL-4 + pcDNA-IL-2. The ensuing humoral immune responses,percentages of CD4+ and CD8+ T lymphocytes, proliferationindices, and interferon-γ expression were analyzed. Resultsrevealed that the piglets co-immunized with pcDNA-ORF5 +pcDNA-IL-4 + pcDNA-IL-2 plasmids developed significantlyhigher antibody titers and neutralizing antibody levels, hadsignificantly increased levels of specific T lymphocyteproliferation, elevated percentages of CD4+ and CD8+ Tlymphocytes, and significantly higher IFN-γ production thanthe other inoculated pigs (p < 0.05).

      • Nonlinear resonances of nonlocal strain gradient nanoplates made of functionally graded materials considering geometric imperfection

        When studying the resonance problem of nanoplates, the existing papers do not consider the influences of geometric nonlinearity and initial geometric imperfection, so this paper is to fill this gap. In this paper, based on the nonlocal strain gradient theory (NSGT), the nonlinear resonances of functionally graded (FG) nanoplates with initial geometric imperfection under different boundary conditions are established. In order to consider the small size effect of plates, nonlocal parameters and strain gradient parameters are introduced to expand the assumptions of the first-order shear deformation theory. Subsequently, the equations of motion are derived using the Euler-Lagrange principle and solved with the help of perturbation method. In addition, the effects of initial geometrical imperfection, functionally graded index, strain gradient parameter, nonlocal parameter and porosity on the nonlinear forced vibration behavior of nanoplates under different boundary conditions are discussed.
