A quantitative, real-time PCR method was developed to enumerate Lactobacillus plantarum IWBT B 188 during the malolactic fermentation (MLF) in Grauburgunder wine. The qRT-PCR was strain-specific, as it was based on primers targeting a plasmid DNA sequence, or it was L. plantarum-specific, as it targeted a chromosomally located plantaricin gene sequence. Two 50 l wine fermentations were prepared. One was inoculated with 15 g/hl Saccharomyces cerevisiae, followed by L. plantarum IWBT B 188 at 3.6 × 106 CFU/ml, whereas the other was not inoculated (control). Viable cell counts were performed for up to 25 days on MRS agar, and the same cells were enumerated by qRT-PCR with both the plasmid or chromosomally encoded gene primers. The L. plantarum strain survived under the harsh conditions in the wine fermentation at levels above 105/ml for approx. 10 days, after which cell numbers decreased to levels of 103 CFU/ml at day 25, and to below the detection limit after day 25. In the control, no lactic acid bacteria could be detected throughout the fermentation, with the exception of two sampling points where ca. 1 × 102 CFU/ml was detected. The minimum detection level for quantitative PCR in this study was 1 × 102 to 1 × 103 CFU/ml. The qRT-PCR results determined generally overestimated the plate count results by about 1 log unit, probably as a result of the presence of DNA from dead cells. Overall, qRT-PCR appeared to be well suited for specifically enumerating Lactobacillus plantarum starter cultures in the MLF in wine.
Integration over the quantum diagonal subgroup and associated Fourier-like algebras

<P>By analogy with the classical construction due to Forrest, Samei and Spronk, we associate to every compact quantum group G, a completely contractive Banach algebra A(Delta) (G), which can be viewed as a deformed Fourier algebra of G. To motivate the construction, we first analyze in detail the quantum version of the integration over the diagonal subgroup, showing that although the quantum diagonal subgroups in fact never exist, as noted earlier by Kasprzak and Soltan, the corresponding integration represented by a certain idempotent state on C(G) makes sense as long as G is of Kac type. Finally, we analyze as an explicit example the algebras A(Delta) (O-N(+)), N >= 2, associated to Wang's free orthogonal groups, and show that they are not operator weakly amenable.</P>
PHYTOREMEDIATION POTENTIAL OF IXORA COCCINEA IN PB CONTAMINATED SOILS

Numerous methods have been operated to relieve the environment of contamination of heavy metals. One of which is the use of chemical and thermal technology. However, this method was proven to be costly and operates far less than the optimum. It was discovered that plants can detoxify soils, therefore resulting in cleaner groundwater free from heavy metal contamination, a method referred to as phytoremediation. In this study, the ability of Ixora Coccinea to aid in phytoremediation was evaluated. Sixty-day old Ixora Coccinea plants were replanted in pots containing 1.3kg of soil mixed thoroughly with a lead (Pb) and distilled water solution with a concentration of 16.9 mg/ml. Every 15 days, for a period of 90 days, three plants were harvested for laboratory analysis testing. The samples were acid digested and analyzed of its Pb content using Inductively Coupled Plasma (ICP). The reduction of Pb gradually increased with respect to time, attaining a maximum of 38.1% reduction on day 60 of culture, but declined afterwards. On average, the Pb uptake rate of Ixora Coccinea was 15.3%. In a patch of land, plants spaced 240mm away from each other would absorb a maximum of 127.1 mg/kg of Pb with a depth of 200mm.

Chemical sensor layers for environmental applications require optimal selectivity, sensitivity, and long term stability, which can be achieved in artificial matrices. For detecting thiols in air, reversible affinity interactions can be optimized by varying the stoichiometry of molybdenum disulphide nanoparticles to achieve sulphur deficiencies. Generating MoS1.9 increases the quartz crystal microbalance (QCM) sensor responses towards butane thiol by a factor of three. Artificial recognition sites are accessible by molecular imprinting: acrylate copolymers can be tuned in polarity to interact selectively with atrazine in water leading to detection limits below one ppb with QCM sensors. Finally, sensor arrays coated with six different molecularly imprinted polymers (MIP) correctly reproduce the ethyl acetate concentration of a composter over a period of two weeks validated by GC-MS measurements.

Complete characterization of quantum-limited timing jitter in passively mode-locked fiber lasers.

<P>We characterize the timing jitter of passively mode-locked, femtosecond, erbium fiber lasers with unprecedented resolution, enabling the observation of quantum-origin timing jitter up to the Nyquist frequency. For a pair of nearly identical 79.4MHz dispersion-managed lasers with an output pulse energy of 450pJ, the high-frequency jitter was found to be 2.6fs [10kHz, 39.7MHz]. The results agree well with theoretical noise models over more than three decades, extending to the Nyquist frequency. It is also found that unexpected noise may occur if care is not taken in optimizing the mode-locked state.</P>
Modification of Starch Composition Using Downregulation of GBSS1 gene in Japonica Rice

There is a great consideration on rice eating quality aside from improving its tolerance to various stresses. High yielding and pest and disease tolerant rice is highly desirable but it is more commercially important if it also has a high eating quality. There are various factors contributing to the good eating quality of rice. This study focuses on modifying the expression of GBSS1 genes which are responsible for amylopectin and amylose synthesis in rice by using RNAi and antisense techniques. We have developed 40 transgenic plants with RNAi-GBSS1 gene and 60 transgenic lines with antisense-GBSS1 gene. The transgenic plants show diverse amylose contents in rice seed. We selected candidate lines according to PCR, RNA expression and amylose contents. A semi-quantitative RT-PCR was carried out to measure the expression level of GBSS1 gene at several time points after the flowering of transgenic plants. The expression level of GBSS1 gene in rice grains decreases over time and the mRNA expression among the transgenic plants were lower compare to its wild type. In the SEM analysis, the starch granule of wild type Gopumbyeo has very large structures accompanied with small ones around the area. However, the starch structures in transgenic plants were smaller and more uniform in size and shape throughout the viewing area
