        Identification and Characterization of Nitric Oxide Synthase in Salmonella typhimurium

        The presence of the nitric oxide synthase (NOS) enzyme from Salmonella typhimurium (S. typhimurium) was identified by measuring radiolabeled L-$[^3H]$citrulline and NO, and Western blot analysis. NOS was partially purified by both Mono Q ion exchange and Superose 12HR size exclusion column chromatography, sequentially. The molecular weight of NOS was estimated to be 93.3 kDa by Western blot analysis. The enzyme showed a significant dependency on the typical NOS cofactors; an apparent Km for L-arginine of 34.7 mM and maximum activity between $37^{\circ}C$ and $43^{\circ}C$. The activity was inhibited by NOS inhibitors such as aminoguanidine and $N^{G}$ $N^{G}$-dimethyl-L-arginine. taken together, partially purified NOS in S. typhimurium is assumed to be a different isoform of mammalian NOSs.OSs.

      • Giant Intraosseous Cellular Schwannoma of the Sacro-pelvic Region in a Young Woman

        Giant intraosseous schwannoma is very rare. Schwannomas arising around the sacrum can reach a considerable size before becoming symptomatic because of their insidious and slow growth, which can allow the displacement of nerves and internal organs, often resulting in extensive bone erosion. The treatment of these giant schwannomas causes significant neurological disability. A 29-year-old woman was suffering from paresthesia of her left posterior thigh. Her symptom was aggravated since the last 5 months. Neurological examination showed paresthesia in the left S2 and S3 dermatome. There was no anal sphincter dysfunction or voiding difficulty. Plain radiography demonstrated foraminal widening of the left S2. Pelvic computed tomography (CT) and magnetic resonance images (MRI) showed a huge mass extending from the sacrum (S2) to the presacral area. The size of the lobulated mass was 4×4×6 cm. The patient underwent laminectomy and gross total removal of the tumor. Through a posterior approach, the tumor was removed without any neural or visceral organ injury, including the left S2 root. After a 71-month follow-up, the neurologic examination was normal, the urodynamic tests continued to show no abnormal signs, and the MRI studies revealed no evidence of residual tumor or recurrence.

        Heterolayered, One-Dimensional Nanobuilding Block Mat Batteries

        <P>The rapidly approaching smart/wearable energy era necessitates advanced rechargeable power sources with reliable electrochemical properties and versatile form factors. Here, as a unique and promising energy storage system to address this issue, we demonstrate a new class of heterolayered, one-dimensional (1D) nanobuilding block mat (<I>h</I>-nanomat) battery based on unitized separator/electrode assembly (SEA) architecture. The unitized SEAs consist of wood cellulose nanofibril (CNF) separator membranes and metallic current collector-/polymeric binder-free electrodes comprising solely single-walled carbon nanotube (SWNT)-netted electrode active materials (LiFePO<SUB>4</SUB> (cathode) and Li<SUB>4</SUB>Ti<SUB>5</SUB>O<SUB>12</SUB> (anode) powders are chosen as model systems to explore the proof of concept for <I>h</I>-nanomat batteries). The nanoporous CNF separator plays a critical role in securing the tightly interlocked electrode–separator interface. The SWNTs in the SEAs exhibit multifunctional roles as electron conductive additives, binders, current collectors and also non-Faradaic active materials. This structural/physicochemical uniqueness of the SEAs allows significant improvements in the mass loading of electrode active materials, electron transport pathways, electrolyte accessibility and misalignment-proof of separator/electrode interface. As a result, the <I>h</I>-nanomat batteries, which are easily fabricated by stacking anode SEA and cathode SEA, provide unprecedented advances in the electrochemical performance, shape flexibility and safety tolerance far beyond those achievable with conventional battery technologies.       • Agrobacterium을 이용한 고추의 형질전환체계개발

        본 실험은 고추의 형질전환체 선발을 위한 최소의 항생제농도를 결정하고 그 결과를 이용하여 형질전환체를 유기하고 효율을 최대화하기 위하여 수행한 바 그 결과는 다음과 같다. 1. 항생제 감응성 검정결과 kanamycin 40㎎/ℓ와 hygromycin 20 ㎎/ℓ에서 신초형성이 완전히 억제되었다. 2. Agrobacterium 제거에 사용되는 carbenicillin과 cefotaxime 처리시 재분화에 아무런 영향을 미치지 않았으며 cefotaxime 200㎎/ℓ 처리에서는 무처리구보다 신초형성이 더 활발하였다. 3. 회복친인 135Q와 276F에서는 형질 전환시 높은 재분화율을 보였으나 형질전환율은 각각 10%와 11.1%의 낮은 빈도를 보였다. 4. 재분화된 신초들을 대상으로 histochemical GUS assay를 실시한 결과 부분적으로 푸른발색이 확인되었다. 5. GUS활성을 보인 신초들에 대한 NPTⅡprimer를 이용한 PCR검정에서 NPTⅡ 유전자의 존재가 확인되었다. A transformation system using Agrobacterium tumefaciens was developed with different genotypes of hot peppers (Capsicum annuum). Four antibiotics commonly used in Agrobacterium-mediated transformation were tested for their effects on shoot regeneration. Shoot induction was completely inhibited by kanamycin at 40㎎/ℓ and hygromycin at 20㎎/ℓ or higher, but not by carbenicillin and cefotaxime. Cotyledon explants of hot peppers were cocultivated with Agrobacterium turnefaciens LBA4404 which carries a binary. vector pBI121 harboring the neomycin phosphotransferase, type Ⅱ (NPT Ⅱ) gene and β-glucuronidase (GUS) gene. For selection of transformed cells, 40㎎/ℓ kanamycin were used because the sensitivity test showed that 40㎎/ℓ kanamycin completely inhibited shoot induction from nontransformed cells. Histochemical GUS assay showed the presence of the GUS gene and PCR analysis confirmed the NP TⅡ gene was integrated. Transformation frequency was about 10.0% and 11.1% of total regenerants from 135Q and 276F, respectively.

        오디 시설재배 조기 확산을 위한 수용요인분석

        본 연구는 오디 시설재배기술이라는 신기술을 수용하지 않은 농가 200명을 대상으로 혁신확산이론과 기술수용이론을 결합한 연구모델을 통해 각각의 요인들이신기술 수용 및 확산에 미치는 영향 정도를 분석하기 위하여 기술수용모형을 바탕으로 유용성, 용이성, 위험성이라는 내생변수를 통해 신기술 수용 및 확산에영향을 미치는 외부요인간의 관계를 구조방정식 모형 분석을 실시하여 검증하였다. 분석결과는 채택된 지표중에서 인지된 유용성에 영향을 미치는 개인의 혁신성과 인지된 위험성 변수의 교육지원 품질, 기술지원 품질과 자금지원정도 지표는 부(-)의 영향을 미치고 나머지 지표들은 정(+)의 영향을 미치는 것으로 나타났다. 분석결과를 바탕으로 오디 시설재배의 조기확산을 위해서는 먼저 기술보급 담당자의 전문성과 기술수준을 높이고, 신기술 수용농가와 더불어 애로사항발생에 따른 해결을 위한 인적 네트워크를 구축하여 운영할 필요가 있다. 다음은 오디재배 농가들은 시설재배기술이 쉽지만 시설투자에 대한 부담 때문에 유용성에는 회의적인 반응을 보이므로 시설투자비에 대한 개인별 맞춤형 자금보조가필요하다. 끝으로 기술수용에 부정적인 농가들을 대상으로 재배기술교육과 신기술수용현장견학과 경영성과에 대한 홍보를 지속적으로 실시하면 빠른 시일 내에 보급이 가능할 것이다. This study conduct for 200 farmhouses who didn’t accept new technologylike mulberry Protected Cultivation. Through research model combinedInnovation diffusion theory(IDT) and Technology Acceptance Model(TAM), itanalysed which factors affect for new technology acceptance and diffusion. Toverify relations between external factors which affect new technologyacceptance and diffusion by endogenous variables such as perceived usefulness,perceived ease of use and perceived riskiness, with external factors, mothedused to analyse it. Most of adopted variables were showed positive resultsexcept for individual innovativeness and reliability. Based on the results ofthese analyses, we can suggest rapid. First, technique diffusion agents shouldhave heighten the level of expertise and technology, and build up the networkto solve difficulties with technique supply with mulberry farmhouses. Secondlyfarmhouses of mulberry outdoor cultivation showed positive attitude protectedcultivation, but negative attitude it bacause of the burden of facilitiesinvestment. Thus, it is needed to capital investment for individual farmhousehold. Finally, through cultivation technology education, field trip aboutmulberry protected cultivation, we can diffuse technology receive for negativefarmers.

        KSR-Ⅲ 과학로켓의 자세제어기 설계와 비행시험

        KSR-Ⅲ 로켓은 액체추진 엔진을 사용한 과학로켓이며, 추력 비행중 피치 및 요 자세제어를 위해 추력벡터제어 방식을 사용하고, 롤 자세를 제어하기 위해 냉가스 추력기를 사용하였다. 본 논문은 KSR-Ⅲ 로켓의 3축 자세제어를 위해 설계된 자세제어기의 구조와 이득 스케쥴링, 자세 안정성 분석결과에 대해 소개한다. 설계된 자세제어기는 국산화 개발된 관성 항법시스템의 비행소프트웨어로 구현되었는데 비행시험에서 완벽히 작동하였다. 비행에서 측정된 데이터는 시뮬레이션 결과와 거의 일치되었다. The KSR-Ⅲ rocket is a liquid propellant sounding rocket and thrust vector control actuators and cold gas thrusters are used to control pitch and yaw, roll attitude respectively during thrusting phase. In this paper, the structure of designed attitude controller and gain scheduling, results of stability analysis for KSR-Ⅲ rocket are presented. The attitude controller is implemented with flight software in the domestically developed INS and successfully performed its function in the flight test. The flight data are coincident with simulation results.

      • Granulocyte colony-stimulating factor-induced immature myeloid cells inhibit acute graft-versus-host disease lethality through an indoleamine dioxygenase-independent mechanism

        <P>Summary</P><P>Granulocyte colony-stimulating factor (G-CSF)-mobilized donor graft tissue used for peripheral blood stem cell transplantation contains a large number of immature myeloid cells that suppress alloreactive donor T cells, resulting in an inhibition of acute graft-versus-host disease (GVHD). However, the molecular mechanism underlying the suppressive function of immature myeloid cells is not fully understood. Here, we investigated whether indoleamine 2,3-dioxygenase (IDO) is related to the suppressive mechanism of G-CSF-induced immature myeloid cells (gMCs). We found that Gr-1<SUP>+</SUP> CD11b<SUP>+</SUP> cells were highly induced in G-CSF-injected donor graft tissue, which is a phenotype of immature myeloid cells, resulting in an inhibition of acute GVHD lethality by suppressing alloreactive donor T-cell expansion. IDO was not detected in primary isolated gMCs; however, this enzyme was markedly induced after treatment with interferon-&ggr; (IFN-&ggr;). This level was significantly higher in IFN-&ggr;-treated gMCs than in bone marrow myeloid cells, which promote alloreactive T-cell responses. We next investigated the functional role of IDO in gMC-mediated inhibition of acute GVHD lethality. We found no changes in gMC-mediated survival or alloreactive donor T-cell suppression when IDO activity was blocked using 1-methyl tryptophan. In addition, there was no difference in gMC-mediated survival rates between recipients transferred with either wild-type gMCs or IDO<SUP>−/−</SUP> gMCs. Taken together, our data suggest that gMC-mediated inhibition of lethal acute GVHD is through an IDO-independent mechanism.</P>

        렘수면행동장애 환자에서 수면무호흡증이 인지기능에 미치는 영향

        Objectives : We investigated the effects of sleep apnea syndrome (SAS) on cognitive functions in patients with REM sleep behavior disorder (RBD). We also studied correlations between sleep variables related with SAS and parameters of cognitive function tests. Methods : Twenty two RBD patients with SAS and 20 RBD patients without SAS participated in the study. The diagnoses of RBD and SAS were confirmed on polysomnography. All the participants performed neuropsychological tests by the Korean version of the Consortium to Establish a Registry for Alzheimer’s disease (CERAD-K) Assessment Packet. Results : There were no differences between two groups in age and sex, education, body mass index, and results of the Beck depression inventory (BDI) and the Epworth sleepiness scale (ESS). No difference was found in any of parameters of the CERAD-K between two groups. The cognitive functions were not correlated with BDI, ESS, or variables related with SAS. Conclusion : SAS had no influence on cognitive functions of RBD patients. The result could be explained by negligible effects of SAS on cognitive functions among aged population and no difference in daytime sleepiness between the RBD patients with and without SAS.

        Analysis of the Genetic Variation of Horse Gap Junction Protein Alpha 4 in Thoroughbreds

        The purpose of this study was to analyze the novel single nucleotide polymorphisms (SNPs) of the gap junction protein alpha 4 gene (GJA4) identified in horse muscle RNA-seq and to predict structural changes of proteins by SNPs. In our previous study, we observed differentially expressed genes (DEGs) in Thoroughbreds before and after exercise through RNA-seq analysis. In addition, we conducted an evolutionary analysis using Thoroughbred and Jeju horse re-sequencing data. As a result, we discovered a novel SNP present in GJA4 (LOC22385534 C>G) in the evolutionarily selected gene in the Thoroughbred horse. Transcription factor (TF) binding sites in the 5′-regulatory region of this gene were identified via PROMO. Additionally, bioinformatics tools were used to predict the effect of non-synonymous SNPs (nsSNP) on function and stability. We identified the change of protein structure owing to the amino acid sequence change, which was proline to arginine according to nsSNP data. Our analysis will be useful as a basis for studying genes and SNPs that affect horses.
