Objectives: The purpose of this study was to assess the correlation relationship between personal exposure and urinary hippuric acid in biological monitoring for airborne toluene in an industrial complex and in general environments. Methods: Personal exposure to toluene and its metabolite, hippuric acid, in urine were simultaneously measured in occupational environments area near an industrial complex and in general environments. The study subjects were divided into three types: 137 workers who use organic solvents in the workplace, 210 residents living near a dyeing industrial complex, and 379 residents living in general environments. The toluene exposures of workers and residents were measured by a passive sampler for four days. The urine of participants was sampled when the passive samplers of personal exposure were collected. Results: The toluene and hippuric acid concentrations of workers were the highest, followed by the concentrations of residents living near a dyeing industrial complex and residents living in general environments. The coefficient of correlation between the concentrations of toluene and hippuric acid among workers was 0.749 (p<0.01) in the workplace. On the contrary, correlations between the concentrations of toluene and hippuric acid among residents living near a dyeing industrial complex and residents living in general environments were all not significant. The relationship between the concentrations of hippuric acid and toluene in three types could be described by the exponential growth model. Conclusions: This study analyzed the relationships between toluene exposure and the concentrations of hippuric acid in urine in high, middle, and low exposure environments, and could be described by the exponential growth model.

Cell-free DNA (cfDNA) sequencing has demonstrated great potential for early cancer detection. However, most large-scale studies have focused only on either targeted methylation sites or whole-genome sequencing, limiting comprehensive analysis that integrates both epigenetic and genetic signatures. In this study, we present a platform that enables simultaneous analysis of whole-genome methylation, copy number, and fragmentomic patterns of cfDNA in a single assay. Using a total of 950 plasma (361 healthy and 589 cancer) and 240 tissue samples, we demonstrate that a multifeature cancer signature ensemble (CSE) classifier integrating all features outperforms single-feature classifiers. At 95.2% specificity, the cancer detection sensitivity with methylation, copy number, and fragmentomic models was 77.2%, 61.4%, and 60.5%, respectively, but sensitivity was significantly increased to 88.9% with the CSE classifier (p value < 0.0001). For tissue of origin, the CSE classifier enhanced the accuracy beyond the methylation classifier, from 74.3% to 76.4%. Overall, this work proves the utility of a signature ensemble integrating epigenetic and genetic information for accurate cancer detection.

Objective: The ARTISENTIAL instruments aim at bringing advanced articulation to laparoscopy to overcome challenges in narrow anatomical spaces. We present the first single-center results of a series of single-port laparoscopic hysterectomies performed with ARTISENTIAL. Methods: Between September 2021 and August 2022, 34 patients who underwent single-port laparoscopic hysterectomy using ARTISENTIAL needle holder for benign or malignant disease by a single surgeon were enrolled retrospectively. The ARTISENTIAL needle holder was used only for vaginal vault closure. The handle of ARTISENTIAL was placed left to the camera head and held with the surgeon's right hand. A conventional grasper was placed right to the camera head and held with the surgeon's left hand. The two devices were crossed in peritoneal space (reverse hand grip technique). All suturing process was videotaped to measure vaginal vault closure time and technique. Perioperative and short-term postoperative data were analyzed. Results: The patients had a median age of 48 years (range 35-75 years). The median time of the vault suture was 411 seconds (range 156-1083 sec), and the median total operative time was 92 min (range 60-160 min). There was one case of vaginal vault dehiscence and two cases of vaginal vault hematoma. The median suture time for the Initial 10 cases and the last 10 cases were 900 sec (range 576 ~ 1083) and 364 sec (range 156 ~ 470 sec), respectively (p <0.001). After 5 cases of the initial experience, the suture times did not exceed 10 minutes in any patient. Conclusion: Intracorporeal vaginal vault closure with ARTISENTIAL needle holder in single-port hysterectomy is feasible and safe. All patients had a successful surgical outcome. The Cumbersomeness of the handle of ARTISENTIAL can be solved using the reverse hand grip technique, which confers more suitable ergonomics to the surgeon. We will now seek to evaluate the benefits of ARTISENTIAL in comparison with standard laparoscopic instruments through a larger study.
Oral Supplementation with Cocoa Extract Reduces UVB-Induced Wrinkles in Hairless Mouse Skin

<P>Cacao beans contain various bioactive phytochemicals that could modify the pathogeneses of certain diseases. Here, we report that oral administration of cacao powder (CP) attenuates UVB-induced skin wrinkling by the regulation of genes involved in dermal matrix production and maintenance. Transcriptome analysis revealed that 788 genes are down-or upregulated in the CP supplemented group, compared with the UVB-irradiated mouse skin controls. Among the differentially expressed genes, cathepsin G and serpin B6c play important roles in UVB-induced skin wrinkle formation. Gene regulatory network analysis also identified several candidate regulators responsible for the protective effects of CP supplementation against UVB-induced skin damage. CP also elicited antiwrinkle effects via inhibition of UVB-induced matrix metalloproteinases-1 expression in both the human skin equivalent model and human dermal fibroblasts. Inhibition of UVB-induced activator protein-1 via CP supplementation is likely to affect the expression of matrix metalloproteinases-1. CP supplementation also downregulates the expression of cathepsin G in human dermal fibroblasts. 5-(3',4'-Dihydroxyphenyl)-gamma-valerolactone, a major in vivo metabolite of CP, showed effects similar to CP supplementation. These results suggest that cacao extract may offer a protective effect against photoaging by inhibiting the breakdown of dermal matrix, which leads to an overall reduction in wrinkle formation.</P>
