Whole-genome resequencing analysis of 20 Micro-pigs

Background Miniature pigs have been increasingly used as mammalian model animals for biomedical research because of their similarity to human beings in terms of their metabolic features and proportional organ sizes. However, despite their importance, there is a severe lack of genome-wide studies on miniature pigs. Objective In this study, we performed whole-genome sequencing analysis of 20 Micro-pigs obtained from Medi Kinetics to elucidate their genomic characteristics. Results Approximately 595 gigabase pairs (Gb) of sequence reads were generated to be mapped to the swine reference genome assembly (Sus scrofa 10.2); on average, the sequence reads covered 99.15% of the reference genome at an average of 9.6-fold coverage. We detected a total of 19,518,548 SNPs, of which 8.7% were found to be novel. With further annotation of all of the SNPs, we retrieved 144,507 nonsynonymous SNPs (nsSNPs); of these, 5968 were found in all 20 individuals used in this study. SIFT prediction for these SNPs identified that 812 nsSNPs in 402 genes were deleterious. Among these 402 genes, we identified some genes that could potentially affect traits of interest in Micro-pigs, such as RHEB and FRAS1. Furthermore, we performed runs of homozygosity analysis to locate potential selection signatures in the genome, detecting several loci that might be involved in phenotypic characteristics in Micro-pigs, such as MSTN, GDF5, and GDF11. Conclusion In this study, we identified numerous nsSNPs that could be used as candidate genetic markers with involvement in traits of interest. Furthermore, we detected putative selection footprints that might be associated with recent selection applied to miniature pigs.
Recent Advances in Anti-Aging Medicine

A rapidly aging population in Korea has led to increased attention in the field of anti-aging medicine. The purpose of anti-aging medicine is to slow, stop, or reverse the aging process and its associated effects, such as disability and frailty. Anti-aging medicine is emerging as a growing industry, but many supplements or protocols are available that do not have scientific evidence to support their claims. In this review, the mechanisms of action and the clinical implications of anti-aging interventions were examined and explained. Calorie restriction mimetics define compounds that imitate the outcome of calorie restriction, including an activator of AMP protein kinase (metformin), inhibitor of growth hormone/insulin-like growth factor-1 axis (pegvisomant), inhibitor of mammalian target of rapamycin (rapamycin), and activator of the sirtuin pathway (resveratrol). Hormonal replacement has also been widely used in the elderly population to improve their quality of life. Manipulating healthy gut microbiota through prebiotic/probiotics or fecal microbiota transplantation has significant potential in anti-aging medicine. Vitamin D is expected to be a primary anti-aging medicine in the near future due to its numerous positive effects in the elderly population.
학교 스포츠클럽 참여 초등학생의 재미특성요인이 학교생활만족에 미치는 영향

The purpose of this study was to analyze enjoyment characteristics of elementary students attending school sports club and the effect on school satisfaction. The questionnaire used in this study was revised and adjusted to purpose of the study. A total of 227 students attending at elementary school in J- city were used as final validity sample. For data analysis, PASW 18.0 version was used for frequency analysis, exploratory factor analysis, reliability analysis, correlation analysis and multiple regression analysis. The results are as follow. First, enjoyment characteristics factor had positive influence on “rule compliance” sub-factor of school life satisfaction. Second, enjoyment characteristics factor had positive influence on “teacher relation satisfaction” sub-factor of school life satisfaction. Third, enjoyment characteristics factor had positive influence on “peer relation satisfaction” sub-factor of school life satisfaction. Fourth, enjoyment characteristics factor had positive influence on “learning life satisfaction” sub-factor of school life satisfaction.
테니스 동호인의 라켓결정요인이 만족 및 재구매의도에 미치는 영향

The purpose of this study was to analyze the determinants of racquet purchase and to investigate the effect on satisfaction and repurchase intention of tennis club members. Subjects were 214 members who activated in J-tennis and data were collected using self-administered questionnaire. The data analysis methods were analyzed by frequency analysis, factor analysis, reliability analysis, correlation analysis, and multiple regression analysis using PASW Statics 18.0 statistical package. The results of this study were as follows. First, determinants of racquet purchase were a significantly positive effect on functional pragmatic factors and brand satisfaction factors, and socio-environmental satisfaction factors. Second, determinant of racquet purchase was a significantly negative effect on repurchase intention factors. Third, satisfaction factors were a significantly negative effect on repurchase intention factor and recommendation intention factors of others. Therefore, further study is needed to investigate the effect of selection according to manufacturers on racquet determination factor of tennis club members.

P-10 : Role of Polypyrimide Tract-Binding Protein in HCV RNA Replication

Background : Translation initiation of the hepatitis C virus (HCV) is regulated by an internal ribosome entry site (IRES) which requires polypyrimidine tractbinding protein (PTB), which binds to the 5``-untranslated region (UTR) and the 3``-UTR and the end of the core coding region of HCV RNA, for its function. Aims of this Study are to determine the role of PTB in HCV replication. Methods : siRNA technology was used to specifically knockdown the PTB expression in the cell. We determined whether down-expression of PTB affects the EMCV IRES-mediated translation, and examined whether interferon (IFN) down-regulates PTB expression. Results : In this study, we discovered that the level of PTB expression was efficiently decreased by PTB-specific siRNA. PTB knockdown expression by siRNA significantly reduced the efficiency of cell colony formation induced by HCV RNA replication. PTB siRNA resulted in a significant reduction of PTB expression and therein HCV RNA replication in a subgenomic HCV replicon-bearing Huh7 cell line, suggesting that PTB is required for efficient HCV replication in vivo. Result: PTB siRNA does not significantly affect the efficiency of the EMCV IRES-mediated translation. IFN-α resulted in an inhibition of PTB expression, suggesting a potential mechanism by which IFN inhibits HCV replication. Discussion : PTB siRNA efficiently knockdowns the level of PTB expression and subsequently resulted in a significant reduction of HCV RNA replication, indicating that PTB is required for efficient HCV RNA replication in vivo. It appears that IFN-α inhibits PTB expression, suggesting that inhibition of HCV replication by IFN might be due to suppression of cellular proteins that are required for HCV RNA replication.
