      • d-pinitol regulates Th1/Th2 balance via suppressing Th2 immune response in ovalbumin-induced asthma

        d-pinitol has been demonstrated to exert insulin-like and anti-inflammatory activities. However, its anti-allergic effect in the Th1/Th2 immune response is poorly understood. Recently, it was shown that T-bet and GATA-3 are master Th1 and Th2 regulatory transcription factors. In this study, we have attempted to determine whether d-pinitol regulates Th1/Th2 cytokine production, T-bet and GATA-3 gene expression in OVA-induced asthma model mice. We also examined to ascertain whether d-pinitol could influence eosinophil peroxidase (EPO) activity. After being sensitized and challenged with ovalbumin (OVA) showed typical asthmatic reactions. These reactions included an increase in the number of eosinophils in bronchoalveolar lavage (BAL) fluid, an increase in inflammatory cell infiltration into the lung tissue around blood vessels and airways, airway luminal narrowing, and the development of airway hyper-responsiveness (AHR). The administration of d-pinitol before the last airway OVA challenge resulted in a significant inhibition of all asthmatic reactions. Accordingly, this study may provide evidence that d-pinitol plays a critical role in the amelioration of the pathogenetic process of asthma in mice. These findings provide new insight into the immunopharmacological role of d-pinitol in terms of its effects in a murine model of asthma, and also broaden current perspectives in our understanding of the immunopharmacological functions of d-pinitol.

        Ahnak functions as a tumor suppressor via modulation of TGFβ/Smad signaling pathway

        We provide detailed mechanisms of Ahnak-mediated potentiation of transforming growth factor β (TGFβ) signaling, which leads to a negative regulation of cell growth. We show that Smad3 interacts with Ahnak through MH2 domain and that Ahnak stimulates Smad3 localization into nucleus leading to potentiating TGFβ-induced transcriptional activity of R-Smad. Moreover, overexpression of Ahnak resulted in growth retardation and cell cycle arrest through downregulation of c-Myc and cyclin D1/D2. We describe results from analyses of Ahnak<SUP>−/−</SUP> mouse model expressing middle T antigen in a mammary gland-specific manner (MMTV<SUP>Tg/+</SUP>Ahnak<SUP>−/−</SUP>), which showed significantly progressed hyperplasia of mammary glands compared with MMTV<SUP>Tg/+</SUP>Ahnak<SUP>+/+</SUP>. Finally, we screened multiple human breast cancer tissues and showed that the expression of Ahnak in cancer tissues is lower than that in control tissues by 50%. Taken together, these data indicate that Ahnak mediates a negative regulation of cell growth and acts as novel tumor suppressor through potentiation of TGFβ signaling.

        In this study, introduce about processing method of fluid passage channel in fuel processor by powder blasting. A processing meterial of the channel used LCD glass which thermal resistance and corrosion resistance are superior. For the comparison of heat transfer performance of methanol evaporator, numerical analysis using FLUENT code was carried out. Thereafter, processed fluid passage channel for fuel processor of LCD glass by powder blasting. After processing, analyzed about comparison of several channel forming cases channel depth, width and rib width.

        Effects of Dietary Fat Types on Growth Performance, Pork Quality, and Gene Expression in Growing-finishing Pigs

        This study was performed to determine the effects of dietary fat sources, i.e., beef tallow, soybean oil, olive oil and coconut oil (each 3% in feed), on the growth performance, meat quality and gene expression in growing-finishing pigs. A total of 72 crossbred pigs (Landrace${\times}$Large White${\times}$Duroc) were used at $71{\pm}1$ kg body weight (about 130 d of age) in 24 pens ($320{\times}150$ cm) in a confined pig house (three pigs per pen) with six replicate pens per treatment. The growing diet was given for periods of $14{\pm}3$ d and the finishing diet was given for periods of $28{\pm}3$ d. The fat type had no significant effect either on growth performance or on chemical composition or on meat quality in growing-finishing pigs. Dietary fat type affected fatty acid composition, with higher levels of unsaturated fatty acids (UFAs) and monounsaturated fatty acids (MUFAs) in the olive oil group. Microarray analysis in the Longissimus dorsi identified 6 genes, related to insulin signaling pathway, that were differentially expressed among the different feed groups. Real time-PCR was conducted on the six genes in the longissimus dorsi muscle (LM). In particular, the genes encoding the protein kinase, cAMP-dependent, regulatory, type II, alpha (PRKAR2A) and the catalytic subunit of protein phosphatase 1, beta isoform (PPP1CB) showed the highest expression level in the olive oil group (respectively, p<0.05, p<0.001). The results of this study indicate that the type of dietary fat affects fatty acid composition and insulin signaling-related gene expression in the LM of pigs.

      • Effect of grain-boundary flux pinning in MgB<sub>2</sub> with columnar structure

        We studied the flux pinning properties by grain boundaries in MgB<SUB>2</SUB> films prepared by using a hybrid physical chemical vapor deposition method on the c-axis oriented sapphire substrates. All the films we report here had the columnar grains with the growth direction perpendicular to the substrates and the grain sizes in the range of a few hundred nanometers. At very low magnetic fields, no discernable grain-boundary (GB) pinning effect was observed in all measuring temperatures, but above those fields, the effect of GB flux pinning was observed as enhanced critical current densities (J<SUB>c</SUB>s) and reduced resistances when an external magnetic field (B) was aligned parallel to the c-axis. We interpret the B dependence of J<SUB>c</SUB> in the terms of flux line lattice shear inside the columnar grains activated by dislocations of Frank-Read source while the flux lines pinned by GB act as anchors for dislocations. Magnetic field dependence of flux pinning force density for B parallel to the c-axis was reasonably explained by the above model.

        한우난소 황체세포의 체외배양에 있어서 Progesterone 분비에 대한 LH 및 PGF2α 첨가 효과

        The effects of exogeneous LH and PGF₂α on the progesterone secretion in the Hanwoo were studied using in vitro culture of luteal cells. The corpora lutea(CL) were collected and pooled from the Korean Native Cattle (Hanwoo) ovaries within 2-3 hrs from a local slaughter house. After enzymatic dissociation, combined large and small luteal cells(LLC and SLC)(I × 10^6 live cells/㎖) were incubated in D-MEM(Dulbicco's-Modified Eagle Medium) in the presence of 10% FCS and antibiotics with or without LH and PGF2α, alone or in various combinations, for 7 days with the medium change every 24 hr. Luteal tissues were apparently formed by two luteal cell populations which were the small and large luteal cells(about 10:1) on day 1 of culture. Treatment with LH(10, 100ng/㎖) markedly stimulated in vitro luteal cell development and proliferation until day 7 of culture. Progesterone secretion in vitro from luteal cell culture was the highest on day 1 and then dramatically decreased to day 7 of culture, and was positively stimulated by LH or PGF2α on day I of culture. However, addition of exogeneous PGF₂α (200ng/㎖) in the presence of LH(10, 100ng/㎖) significantly(P$lt;0.05) inhibited progesterone secretion on day 1 of culture. These observations indicate that luteolysis of luteal cell by PGF₂α may be regulated by inhibition of pituitary LH function and other factors.

        키토산 발효사료의 첨가가 비육돈의 도체특성 및 육질에 미치는 영향

        처리구 당 돼지 10마리씩을 선발하여 비육후기에 0.2%, 0.5%의 키토산 발효사료를 첨가하여 6주간(43일) 급여한 사양시험 결과는 증체량에 있어서는 0.2% 처리구가 가장 높았으며 (p<0.05) 사료요구율에 있어서는 0.5% 처리구가 가장 낮게 나타났다. 0.2% 수준에서의 첨가는 사료의 효율을 증가시키고 증체량을 개선하였다. pH값은 등심의 경우에 있어서 대조구가 5.27로서 가장 낮았고 0.2%와 0.5% 처리구는 각각 5.47과 5.54를 나타내었으며 처리구간에 있어서 유의한 차이를 보여 주었다(p<0.05). 등심의 경우 L*값이 0.2%와 0.5%의 두 처리구에 있어서 대조구보다 유의하게(p<0.05) 낮게 나타났으며, 목심의 경우네는 0.5% 처리구가 다른 처리구보다 유의하게(p<0.05) 낮은 것으로 나타났다. 3가지 처리에 따른 등심부위의 지방산 조성을 보면 현저한 차이를 발견할 수는 없으나 키토산 발효사료 처리구에는 C12:0 이나 C16:0과 같은 포화지방산의 비율이 높아진 반면 탄소수가 18개의 불포화 지방산 함량은 다소 낮아 진 것을 알 수 있다(대조구 : 30.15%, 0.2% 처리구 : 26.03%, 0.5% 처리구 : 26.94%). 관능검사항목 중에서 다즙성의 경우에만 처리구간별로 유의한 차이를 나타내었으며, 0.5% 처리구가 가장 높은 것으로 나타났다(p<0.05). 또한 연도에 있어서도 0.5% 처리구가 가장 부드러운 것으로 나타났다. 삽겹살에 대해서는 다즙성, 풍미 그리고 기호성을 조사한 바, 풍미와 기호성 측면에서도 0.5% 처리구가 가장 높은 것으로 나타났다(p<0.05) This experiment was carried out to investigate the effect of fermented chitosan additive(FCA) as a probiotics in finishing pig. The levels of 0.2% and 0.5% of FCA powder was added to the regular feed. A total of 30 pigs were used to evaluate the growing performance. Those FCA supplemented feeds were supplied to the pigs at 17 weeks of age for 6 weeks. After slaughtering, 4 pigs of each treatment were used to compare the physico-chemical characteristics and sensory evaluation. Addition of 0.2% of FCA improved the average daily weight gain and feed efficiency. The pH of the FCA treated loin was slightly higher than control. The L^*, a^*, b^* values of the treatment groups were generally low compared to control group. So the effect of FCA to meat color was expression of purplish red color. Juiciness of the pork shoulder treated with 0.5% FCA was significantly(p<0.05) higher than the other groups. Flavor and overall preference scores for bacon treated with 0.5% of FCA were significantly(p<0.05) higher than the other groups.

        韓國人 靑少年 身長과 體重의 時代的 變遷에 따른 統計學的 推定値에 관한 硏究

        As compared with body height and body weight by ages and sexes, by means of the data reported under other researchers from 1967 to 1994 for 33 years, this study obtained the estimate value of body height and body weight by ages and sexes for the same period, and figured out prediction value of body height and body weight in the ages of between 6 and 14 from 1995 to 2000. These surveys and measurements took for one year from October 1st 1994 to September 30th. As shown in the <Table 1>, in order to calculate the establishment, estimate value and prediction value of the chronological regression model of body height and body weight, by well-grounded 17 representative research papers, this research statistically tested propriety of liner regression model by the residual analysis in advance of being reconciled to simple liner regression model by the autonomous variable-year and the subordinate variable-body weight and measured prediction value, theoretical value from 1962 to 1994 by means of 2nd or 3rd polynomial regression model, with this result did prediction value from 1995 to 2000. 1. Chronological Change of Body Height and body Weight The analysis result from regression model of the chronological body height and body weight for the aged 6-16 in both sexes ranging from 1962 to 1994, comed from the <Table 2-20>. On the one hand, the measurement value of respective researchers had a bit changes by ages with age growing, but the other hand, theoretical value, prediction value showed the regular increase by the stages and all values indicated a straight line on growth and development with age growing. That is, in case of the aged 6, males had 109.93cm in 1962 and females 108.93cm, but we found the increase that males had 118.0cm, females 113.9cm. In theoretical value, prediction value, males showed the increase from 109.88cm to 117.89cm and females from 109.27cm to 115.64cm respectively. There was the same inclination toward all ages. 2. Comparision to Measurement Value and Prediction Value of Body Height and Body Weight in 1994 As shown in the <Table 21>, in case of body height, measurement value and prediction value of body height and body weight by ages and sexes almost showed the similiar inclination and poor grade, in case of body weight, prediction value in males had a bit low value by all ages, and prediction value in females had a high value in adolescence, to the contrary, a low value in adult. 3. Prediction Value of Body Height and Body Weight from 1995 to 2000 This research showed that body height and body weight remarkably increased in adolescence but slowly in adult. This study represented that Korean physique was on the increase and must be measured continually hereafter.

        Potential application of antibody-mimicking peptides identified by phage display in immuno-magnetic separation of an antigen

        Phage display was performed against human IgG (hIgG) through five rounds of 'biopanning'. Each round consisted of: (1) incubating a library of phage-displayed 12-mer peptides sequences on hIgG-coated magnetic beads, (2) washing the unbound phages, and (3) eluting the bound phages. The eluted phages were either amplified to enrich the pool of positive clones or subjected to the next round without amplification. Through ELISA, four clones (F9, D1, G5, and A10) showing specific binding affinity to hIgG were identified. Among these, F9 had the highest affinity (K<SUB>d</SUB>=6.2nM), only one order of magnitude lower than the native anti-hIgG antibody (0.66nM). Following the DNA sequences of the selected clones, four 12-mer peptides were chemically synthesized. Among them, D1 peptide showed the highest binding affinity to hIgG via SPR biosensor measurements. This peptide was conjugated to biofunctionalized magnetic beads, and its immuno-binding ability was compared with that of the native antibody immobilized to magnetic beads. The mol-to-mol binding efficacy of the peptide-coated magnetic beads was approximately 1000-fold lower than that of the antibody-coated magnetic beads. Our results suggest a feasibility of using antibody-mimicking peptides identified by phage display technique for immuno-magnetic separation of an antigen.
